쑥부쟁이 추출물의 라디칼 소거활성 평가 (Determination of Radical Scavenging Activity of Aster yomena (Kitam.) Honda)

  • 본 연구에서는 1.1-Diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical ($^{\cdot}OH$), 그리고 superoxide radical ($O_2{^-}$) 소거능 실험을 통해 쑥부쟁이의 항산화 효과를 평가하였다. 쑥부쟁이는 에탄올로 추출한 뒤, n-hexane, methylene chloride ($CH_2Cl_2$), ethylacetate (EtOAc), 그리고 n-butanol (n-BuOH)로 분획하였다. DPPH 라디칼 소거능 실험에서 모든 추출 및 분획물이 $10-100{\mu}g/mL$의 농도에서 농도 의존적으로 라디칼 소거능이 증가하였으며, 특히 EtOAc 분획물은 가장 강한 DPPH 라디칼 소거능을 나타내었다. 또한 쑥부쟁이는 $50{\mu}g/mL$ 농도에서 80% 이상의 $^{\cdot}OH$ 라디칼 소거율을 보였다. 특히 EtOAc 분획물은 $0.03{\mu}g/mL$에서 IC50 값을 나타내어 추출물과 분획물 중 가장 강한 $^{\cdot}OH$ 라디칼 소거능을 보여주었다. 또한, EtOAc 분획물은 $O_2{^-}$ 라디칼 소거능에서도 가장 강한 소거활성을 보였다. 이러한 결과는 쑥부쟁이가 산화스트레스로 인한 질병을 예방하는 천연 항산화제로서의 활용 가능성이 있음을 보여준다.

경옥고가미방의 베타글루칸, 진세노사이드 함량, 2,2-diphenyl-1-picrylhydrazyl (DPPH) Free Radical 소거 활성 및 단회 투여 독성 연구 (A Study on the β-glucan, Ginsenoside Content, 2,2-diphenyl-1-picrylhydrazyl (DPPH) Free Radical Scavenging Activity and Single Dose Toxicity Assessment of Modified Kyungohkgo)

  • Objectives This study was conducted to investigate the beta-glucan, ginsenoside content, antioxidant activity and safety of modified Kyungohkgo added to Sparassis crispa and Hericium erinaceum. Methods The marker compounds contents, antioxidant activity and safety of modified Kyungohkgo were tested. The contents of beta-glucan and ginsenoside Rb1, Rg1, and Rg3 marker compounds were measured, the antioxidant activity was measured using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, and a safety test was conducted via single dose toxicity assessment. Results Analyzing the contents of marker compounds showed 351.75 mg/g of beta-glucan, 0.0327 mg/g of ginsenoside Rb1 and 0.0802 mg/g of ginsenosai Rg3. In the DPPH free radical scavenging activity, the inhibition concentration 50% of modified Kyungohkgo was 0.2880%. The scavenging activity of modified Kyungohkgo was 5.49% activity at 0.05% concentration, 89.66% activity at 0.5% concentration, 94.68% activity at 1% concentration, and 96.06% activity at 5% concentration. In the single dose toxicity test of modified Kyungohkgo, a dose of 2,000 mg/kg B.W. was set at its highest capacity and observed after oral administration to female and male rats. No toxicological findings were recognized. It was observed that the resulting lethal dose can be set to 2,000 mg/kg B.W. or higher for both females and males. Conclusions The results of the experiment on modified Kyungohkgo showed that the marker compounds contents were beta-glucan and ginsenoside Rb1 and Rg3, that antioxidant activity was observed through the DPPH free radical scavenging activity, and safety was confirmed through the single dose toxicity assessment.

찻잎의 수확시기, 돈차의 숙성기간 및 추출온도가 돈차의 생리활성에 미치는 영향 (Effects of Harvesting Time, Aging Period and Extracting Temperature of Wild Green Tea (Camell sinesis) Leaves on Physiological Activity of Don Tea)

  • This study was conducted to determine the potent physiological activity of traditional wild tea ("Don tea"; coin-shaped tea) as affected by different harvesting times, aging periods and extracting temperatures, No difference in anti-oxidative activities in the harvesting time and extracting temperature of tea leaves was observed. However, short aging periods of Don tea showed high ABTS {2-2-azonobis (3-ethylbenzothiazolin-6-sulphonic acid)} activity, ranging from 71.52 to 79.96%. DPPH (${\alpha},\;{\alpha}-diphenyl-{\beta}-picryl-hydrazyl$) radical scavenging activity of Don tea was 71.10 to 91.40%. Especially, longer aging period and an extracting temperature of $100^{\circ}C$ showed higher DPPH radical scavenging activity. With longer aging periods and an extraction at $90^{\circ}C$, nitrite radical scavenging activity of Don tea ranged from 74.04 to 94.92%. On the other hand, angiotensin 1-converting enzyme (ACE) inhibition activity of Don tea was 59.77-81.97%. It showed higher activity when harvested in June and August, aged for longer periods, and extracted at $100^{\circ}C$. These results suggested Korean traditional Don tea exhibited the highest physiological activity when aged over 8 months.

추출용매별 canola meal의 항산화 활성 (Antioxidant Activities of Various Solvent Extracts from Canola Meal)

  • 본 연구에서는 용매별(methanol, ethanol, acetone 및 water)로 추출한 canola meal의 항산화 활성을 조사하였다. 모든 canola meal 용매 추출물들은 첨가 농도가 증가할수록 항산화 활성이 점진적으로 증가하였다. 추출물 중에서 methanol 추출물이 가장 낮은 $EC_{50}$ (DPPH radical scavenging ability; 0.9 mg/mL, ABTS radical scavenging ability; 3.5 mg/mL, reducing power; 1.3 mg/mL and chelating ability; 8.5 mg/mL)과 가장 많은 항산화 물질 (total phenolics; $63.3\;{\mu}g$/mg, total flavonoids; $21.3\;{\mu}g$/mg)을 함유하는 것으로 나타났다. 보다 정확한 canola meal의 항산화 활성을 조사하기 위하여 항산화 활성이 높은 methanol 추출물을 sephadex LH-20 column을 이용하여 순차적으로 MF 1, MF 2 및 MF 3의 3개의 분획물을 얻었으며, 동일한 농도(1.5 mg/mL)에서 측정한 항산화 활성에서 DPPH radical scavenging ability는 MF 2(75.56%)에서, ABTS radical scavenging ability와 reducing power는 MF 3(31.17% and 1.60)에서, chelating ability는 MF 1(77.72%)에서 가장 높게 나타났다.

음료 농축액 및 퓨레의 항산화 활성 (Antioxidant Activities of Beverage Concentrates and Purees)

  • 본 연구에서는 음료 농축액 및 퓨레로 사용되는 석류 농축액, 블루베리 농축액, 양파추출 농축액, 도라지추출 농축액, 흑마늘 농축액, 홍삼 농축액, 아사이베리 퓨레, 새싹유채 퓨레의 이화학적 특성, 총 페놀 함량, 항산화 효능(DPPH radical scavenging, ORAC) 및 아질산염 소거능을 분석하였다. pH는 석류 농축액이 2.81로 가장 낮았으며 양파추출 농축액이 5.12로 가장 높은 수치를 나타냈다. Brix는 홍삼 농축액이 70.30으로 가장 높았고 새싹유채 퓨레가 1.70으로 가장 낮았다. 총 페놀 함량은 아사이베리 퓨레에서 182.71 mg GAE/mL로 다른 시료에 비하여 높은 수치를 나타냈고 양파추출 농축액이 8.80 mg GAE/mL로 가장 낮았다. DPPH 라디칼 소거능 측정은 총 페놀 함량과 마찬가지로 아사이베리 퓨레에서 69.88%로 가장 높은 활성을 나타내었다. ORAC 지수는 블루베리 농축액에서 $27,514{\mu}M$ TE/mL로 가장 높게 나타났으며 아질산염 소거능도 아사이베리 퓨레에서 28.19%로 유의적으로 높은 활성을 보였다. 이상의 연구 결과로부터 천연물 유래 농축액 및 퓨레는 다양한 항산화 평가모델에서 항산화 활성을 나타내었으며 이들 농축액 및 퓨레를 이용한 항산화 활성을 갖는 건강음료의 원료로서 활용이 가능할 것으로 사료된다.

Antioxidant Effect of Citri Reticulatae Pericarpium Extract on Oxidative Stress-Mediated Cytotoxicity In Cultures

  • In order to examine oxidative stress of reactive oxygen species and the antioxidant effect of Citri Reticulatae Pericarpium (CRP) extract, human skin melanoma cells were treated with various concentrations of hydrogen peroxide ($H_2O_2$). Antioxidant effect of CRP extract on $H_2O_2$-induced cytotoxicity, cell viability, DPPH-radical scavenging activity and superoxide dismutase (SOD)-like activity. In this study, $H_2O_2$ decreased cell viability of cultured human skin melanoma cells in dose- and time-dependent manners, and then, midcytotoxicity value (MCV) was determined at $60\;{\mu}M$ after human skin melanoma cells were cultured for 5 hours in the media containing $20{\sim}60\;{\mu}M$ of $H_2O_2$, respectively. The $H_2O_2$ was on cultured human skin melanoma cells because MCV of $H_2O_2$ was lower than $100\;{\mu}M$. In the antioxidant effect of CRP extract, CRP extract increased cell viability DPPH-radical scavenging activity and SOD-like activity. From these results, it is suggested that $H_2O_2$ was very toxic on cultured human skin melanoma cells. And also, CRP extract has the antioxidant effect on $H_2O_2$-induced cytotoxicity.

Antioxidant Activities of Aroma Extracts in Commercially Available Red Wines in Korea

  • The antioxidant activities of aroma extracts from commercially available red wines in Korea were evaluated. The aroma extracts of the red wines were extracted by simultaneous steam distillation. Antioxidant activity was measured by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity and ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid) radical cation scavenging activity. DPPH radical scavenging activity of the aroma extracts in the red wines increased with increases in the amount of wine used for aroma extraction. Antioxidant activities of domestic wine 1, imported wine 7, and imported wine 12 were 97.16, 96.72 and 94.52%/20 mL wine by DPPH assay and 7.09, 8.07 and 7.28 mg ascorbic acid equivalents per mL wine by ABTS assay, respectively. This study demonstrates potent antioxidant activities of the aroma extracts of commercially available red wines in Korea.

애기거머리말의 항산화 활성 (Antioxidant Activity of the Seagrass Zostera japonica)

  • In this study, crude extract of the seagrass Zostera japonica, and its solvent-partitioned fractions were evaluated for their antioxidant activity. The crude extract was successively fractionated into n-hexane, 85% aqueous methanol (85% aq.MeOH), n-butanol (n-BuOH), and water fractions by liquid-liquid partition. These include DPPH radical scavenging, hydroxyl radical scavenging in HT-1080 cells, peroxynitrite scavenging, and protective effect on DNA damage caused by hydroxyl radicals generated. In all assays, except for DPPH radical, 85% aq.MeOH and n-BuOH fraction showed the strong antioxidant activity. These results suggest that Z. japonica may be used as a potential source of natural antioxidants for the development of cosmetic product or functional food in the future.

찰흑미를 첨가한 식빵의 항산화성에 관한 연구 (A Study on Antioxidant Activity of Bread with Waxy Black Rice Flour Added)

  • The antioxidant activities of the bread made from waxy black rice flour(WBRF) and wheat flour were investigated. The waxy black rice bread was manufactured with 10, 20, 30, or 40% waxy black rice flour(w/w). As the additive amount of WBRF were increased, DPPH(1,1-diphenyl-2-picryl-hydrazyl) radical scavenging activity and TEAC(trolox equivalent antioxidant capacity) were also increased. WBRF group showed the higher activity than the control group in DPPH radical scavenging activity and TEAC. The group with 10% WBRF added was not significantly different from the control group in metal chelating activity, but over 20% of addition, it had the higher metal chelating activity than that of the control group.
