Antioxidant Activities and Antimicrobial Effects of Solvent Extracts from Lentinus edodes (표고버섯(Lentinus edodes) 용매 추출물의 항산화 활성 및 항균 효과)

  • The aim of this study was to investigate the antioxidant and antimicrobial activities of various solvent (acetone, ethyl acetate, and ethanol) extracts from Lentinus edodes. The antioxidant activities were evaluated by measuring total polyphenol and flavonoid contents, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity. Total polyphenol content and ABTS radical scavenging activity were highest in ethanol extract. ABTS radical scavenging activity of ethanol extract showed the highest value (98.5%), which was similar to that of ascorbic acid (95.7%). The ethyl acetate extract from Lentinus edodes showed relatively high total flavonoid content and DPPH radical scavenging activity. Negative correlations were found between total polyphenol contents and DPPH radical scavenging activities in Lentinus edodes extracts. Antimicrobial activities of the extracts were determined against Bacillus subtilis, Staphylococcus aureus, Micrococcus luteus, Escherichia coli, Pseudomonas aeruginosa, and Enterobacter cloacae by the disc diffusion method. The acetone and ethanol extracts showed moderate antimicrobial activities against almost all tested microorganisms except E. coli and S. aureus, respectively. The ethyl acetate extract showed a significant growth inhibition effect against E. coli, Ent. cloacae, and B. subtilis.

Antioxidant Activity and Quality Characteristics of Pine Needle Cookies (솔잎 쿠키의 항산화활성 및 품질특성)

  • Pine needle powder has medical and functional uses. In this study, cookies were prepared using different 0.5%, 1%, 3%, 5% of pine needle powder. The total phenolic content was measured by the Folin-Ciocalteu method, and antioxidant activity was evaluated by DPPH assay. The antioxidant activity was highly correlated with the total phenolic composition of pine needle cookies (r=0.989). In addition, the quality characteristics of the pine needle cookies were estimated based on the bulk density and pH of the dough, spread factor, loss rate, leavening rate, color, texture profile analysis, and sensory evaluations. The bulk density of the dough and the hardness, total polyphenol contents and DPPH free radical scavenging activity of cookies significantly increased with increasing pine needle powder, while the pH of the dough, spread factor and L values of the cookies decreased with increasing pine needle powder content (p<0.05). Additionally, the consumer acceptability scores for the 3% pine needle cookie groups ranked significantly (p<0.05) higher than those of the other groups in appearance, taste, flavor, texture and overall preference. Taken together, the results of this study suggest that pine needle powder is a good ingredient for increasing the consumer acceptability and the functionality of cookies.

Effects of polyphenol and anthocyanin contents of Punica granatum fruit parts on their antioxidant and anticancer activities (석류 과실 부위별 폴리페놀 및 안토시아닌 함량이 항산화 및 항암 활성에 미치는 영향)

  • The purpose of this study was to determine the content of phenolics and various anthocyanin compounds, and antioxidant and anticancer activities of the extracts from pomergranates, which were produced in two different regions including Goheung, Korea and California, USA. These pomergrantes were divided into juice, pericarp, and seed parts and each part was extracted with 95% methanol. Content of total phenolics [mg chlorogenic acid equivalents/kg DW] was highest in pericarp, followed by juice and seeds from pomergrantes in both regions. The anthocyanins identified in pomegranate fruit were cyanidin-3,5-diglucoside chloride (Cy3,5G), cyanidin-3-O-glucoside chloride (Cy3G), delphini din-3,5-di-O-glucoside chloride (Dp3,5G), delphinidin-3-O-glucoside chloride (Dp3G), pelargonidin-3,5-di-glucoside chloride (Pg3,5G), and pelargonidin-3-glucoside chloride (Pg3G). Among these, cyanidin-3,5-diglucoside chloride (Cy3,5G) was the major anthocyanin in California pomegranate fruit juice and Goheung pomegranate fruit pericarp. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was dose-dependently increased, and was higher in pericarp part than juice or seed parts from pomergrantes in both regions. By MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay, pericarp extracts from pomergrantes in both regions showed the highest anticancer activity, which was higher on Calu-6 for human pulmonary carcinoma than SNU-601 for human gastric carcinoma. Correlation between polyphenols and anticancer activity on Calu-6 was determined to be in the range of $r^2=0.8904$ to 0.9706.

Optimization of Processing Conditions for Making a Black Ginger and Design Mixture for Black Ginger Drinks (흑생강 제조 공정 최적화 및 기능성 흑생강 음료 제조)

  • Black ginger, obtained from steaming and drying process, provides the various functional properties. This study was performed to investigate the optimum processing conditions for black ginger with high content of biologically active substance such as anti-oxidations. Optimum processing conditions such as temperature and time for black ginger was determined by response surface methodology (RSM) with manufacturing process and functionality. The optimum steaming condition was determined 6 hours at 93.2$^{\circ}C$, and 82.7 mg/L DPPH scavenging activities was obtained at this condition. The black ginger drink was made with black ginger extracts, Japanese apricot, and honey. Interaction effects of these ingredients were investigated by modified distance based on design and analyzed by linear, nonlinear regression model, and RSM. The optimization of mixture ratio was made by statistical modeling using DPPH scavenging activities and sensory properties which are the important target constraints in drink. Total flavonoids showed a linear canonical form, while preference and antiradical activity showed a nonlinear canonical form indicating the higher interaction among mixtures. The response trace plot revealed that antiradical activity, sensory properties and total flavonoids were quite sensitive to the drink blending. The optimum formulation of the drink was set at 14.2% of black ginger extracts, 5% of Japanese apricot, and 10.8% honey.

Change of Total Polyphenol Content of Fermented Gastrodia elata Blume and Radical Scavenging (발효천마의 총 폴리페놀 함량 변화 및 라디칼 소거능)

  • This study was conducted to compare and analyze change of content of polyphenol and p-hydroxybenzyl alcohol (HBA), which is an index-component, and antioxidant activity between fermented Gastrodia elata Blume and non-fermented G. elata Blume. The polyphenol contents before and after the fermentation were 108.65 and 389.99 mg/mL respectively, and the content of index-component HBA increased from 2.46 mg/g before fermentation to 7.94 mg/g after fermentation. In comparison between the non-fermented G. elata Blume extract (NFGE) and fermented G. elata Blume extract (FGE) in DPPH, ABTS, and superoxide radical scavenging assay, we found that FGE showed more activity than NFGE as the extract was more concentrated. Especially, the superoxide radical scavenging activity was increased more than twenty times in FGE. In conclusion, we confirmed increase in the electron donating ability and radical scavenging when the dried G. elata Blume is fermented, and its further feasibility as an antioxidant.

The Antioxidative Effect of Ethanol Extracts from Lithospermum erythrorhizon Siebold & Zucc., Xanthium strumarium Linn, and Lonicera japonica (한약재에탄올 추출물의 항산화 효과)

  • In this study, we examined the antioxidative activities of ethanol extracts obtained from three plants; Lithospermum erythrorhizon Siebold & Zucc., Xanthium strumarium Linn, and Lonicera japonica, which have traditionally been used as drugs in Eastern medicine in Korea. Their extraction yields were 7.01%, 2.92%, and 7.95% in Lithospermum erythrorhizon Siebold & Zucc., Xanthium strumarium L, and Lonicera japonica, respectively. The contents of the phenolic compounds were $4.3{\pm}0.1{\mu}g/ml$, $5.4{\pm}0.1{\mu}g/ml$, and $4.6{\pm}0.1{\mu}g/ml$ in Lithospermum erythrorhizon Siebold & Zucc., and Xanthium strumarium L, respectively. Furthermore, the radical scavenging activity measured through the DPPH assay appeared highest in the Lonicera japonica's extract, and its $EC_{50}$ was 0.24 mg/ml. Compared to the control, the xanthine oxidase inhibiting activities of all extracts were effective at 0.01 mg/ml concentration. Superoxide radical scavenging activity in Lithospermum erythrorhizon Siebold & Zucc. and Lonicera japonica was more than 80%, with a concentration of 50 mg/ml. OH radical scavenging activity was 40% in the three plants, with a concentration of 50 mg/ml scavenging activity. From our results, we demonstrated that the ethanol extracts of three medicinal plants have antioxidant activities and could be potential candidates for natural antioxidants.

Antioxidant Activity and Tyrosinase Inhibition Effect of Makgeolli Using Mistletoe Cultivated on Prunus mume (매실나무(Prunus mume) 겨우살이 막걸리의 발효기간 및 겨우살이 함량 차이에 따른 항산화활성 및 Tyrosinase 저해활성)

  • 매실나무겨우살이 막걸리의 생리적 효능을 검정하여 의약 화장품의 소재화 가능성을 검토할 목적으로 발효기간과 겨우살이 함량에 따른 총 폴리페놀, 플라보노이드 함량, DPPH free radical 소거능을 측정하였고, tyrosinase 저해활성은 5분 간격으로 65분간 측정하였다. 막걸리는 백미와 기장을 7:3의 비율로, 겨우살이의 함량을 0%, 5%, 10%로 달리하여 10일 간격으로 최대 30일까지 Shaking incubator($30{\sim}35^{\circ}C$, 100 rpm)에서 발효하였고, 발효가 끝난 막걸리는 80 mesh 망으로 여과하였다. 총 폴리페놀 함량은 10일에 최대치를 나타내었고, 0%는 $633.53{\mu}g/ml$, 5%는 $759.67{\mu}g/ml$, 10%는 $752.2{\mu}g/ml$을 나타내었다. 기간이 경과함에 따라 감소하는 경향이었으며 30일에 0%는 $507.57{\mu}g/ml$, 5%는 $570.13{\mu}g/ml$, 10%는 $551{\mu}g/ml$를 함유하고 있는 것으로 나타났다. 총 플라보노이드 함량 또한 10일에 최대치를 나타내었고, 0%는 $16.18{\mu}g/ml$, 5%는 $63.87{\mu}g/ml$, 10%는 $62.99{\mu}g/ml$였고, 기간이 경과함에 따라 감소하는 경향성을 나타내어 30일에 0%는 $0.59{\mu}g/ml$, 5%는 $10.98{\mu}g/ml$, 10%는 $1.18{\mu}g/ml$를 함유하였다. DPPH free radical 소거능은 0%의 경우 10일에 72.34%로 최대치가 나타났고, 30일에 58.61%로 가장 낮은 소거능을 보여주어 기간이 경과함에 따라 감소하는 경향성을 보여주었다. 하지만, 겨우살이 첨가군인 5%(10일:91.13%, 20일:91.09%)와 10%(10일:91%, 20일:90.8%)는 비교적 차이가 없었고, 발효기간이 30일이 경과한 후 5%는 93.46%, 10%는 92.3%로 최대값을 보여주었다. Tyrosinase 저해활성은 DPPH free radical 소거능이 가장 좋은 30일 발효 막걸리를 측정하였다. 모든 겨우살이 첨가군에서 5분이 경과하였을 때, 0%는 82.68%, 5%는 69.29%, 10%는 79.53%의 저해활성을 나타내었으며, 시간에 유의적으로 저해활성이 감소하였다. 본 연구 결과에 따르면, 겨우살이 막걸리의 총 폴리페놀과 플라보노이드 함량은 기간이 경과함에 따라 감소함에도 불구하고, DPPH free radical 소거능은 증가함을 알 수 있었다. Tyrosinase 저해활성은 5% 첨가 막걸리에서 가장 낮았으며, 겨우살이를 첨가하지 않은 0%에서 가장 저해활성이 높고 오랜시간 유지되는 것을 확인할 수 있었다.

Antioxidant Activity of Korean Gomchwi (Ligularia fischen) Extracts (국내산 곰취(Ligularia fischen) 추출물의 항산화 활성)

  • In this study, Gomchwi (Ligularia fischen) derived from Taebaek-si, Gangwon-do was extracted with 70% ethanol (LFE) and antioxidant activity was measured. The following experimental techniques were used to evaluate the antioxidant efficacy of LFE. Total phenolic contents, ABTS/DPPH radical scavenging analysis, cell viability assay, NO assay, and quantitative real-time PCR technique. The content of polyphenol and flavonoid was each 113.97±0.37 mg GAE/g or 29.22±2.06 mg QE/g in LFE. DPPH radical scavenging activity was measured to be 25 ㎍/㎖ 11.26±0.95%, 50 ㎍/㎖ 17.12±0.63%, 100 ㎍/㎖ 29.54±0.36%, 250 ㎍/㎖ 68.31±0.28%, 500 ㎍/㎖ 75.12±0.05%, and 1000 ㎍/㎖ 75.75±1.57%. In addition, ABTS radical scavenging activity was identified as LFE 25 ㎍/㎖ 13.75±0.21%, 50 ㎍/㎖ 26.71±0.20%, 100 ㎍/㎖ 56.92±0.22%, 250 ㎍/㎖ 91.30±0.12%, 500 ㎍/㎖ 93.40±0.02, and 1000 ㎍/㎖ 93.19±0.04%. There was no significant cytotoxicity of LFE. NO production was significantly decreased to LFE 50 ㎍/㎖ 79.40±2.64%, 100 ㎍/㎖ 55.01±5.36%, and 200 ㎍/㎖ 30.93±3.11%. Also, the NOS2 gene expression was significantly reduced to LFE 50 ㎍/㎖ 0.94±0.11, 100 ㎍/㎖ 0.59±0.05, and 200 ㎍/㎖ 0.32±0.04. This result objectively confirmed the antioxidant effect of Gomchwi. We will continue to conduct in-depth research. Therefore, it is believed that the possibility of using Gomchwi as a cosmetic and functional food material can be established.

Effects of Methanolic Extract from Salvia miltiorrhiza Bunge on in vitro Antithrombotic and Antioxidative Activities (단삼 메탄올 추출물의 항혈전 및 항산화 효과)

  • Salvia miltiorrhiza Bunge is known to potentially prevent arteriosclerosis and hypertension, but its effects on platelet function are not clear. In this study, we evaluated the in vitro antithrombotic activities of the edible plant extract. Methanol extract of S. miltiorrhiza Bunge exhibited about 70% fibrinolytic activity compared to the plasmin control, and inhibited ADP- and collagen-induced platelet aggregation in a concentration-dependent manner with $IC_{50}$ values of 0.42 and 0.07 mg/mL, respectively. S. miltiorrhiza Bunge extract significantly prolonged the activated partial thromboplastin time (APTT) and prothrombin time (PT) compared with control. Moreover, 0.05 mg/mL S. miltiorrhiza Bunge extract contained 87.3% l,l-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity. In conclusion, S. miltiorrhiza Bunge seemed to enhance antithrombotic activity due to its radical scavenging activity. Based on these data, further examination is required to determine the mechanism of platelet-dependent antithrombosis and the effect of polyphenols on platelet function.

Evaluation of 1,1-Diphenyl-2-Picrylhydrazyl Radical Scavenging Effect, Cytotoxicity and Tyrosinase Inhibition Activities in 4 Species of Herb Plants (허브 식물 4종의 1,1-Diphenyl-2-Picrylhydrazyl 라디칼 소거능, 세포 독성 및 tyrosinase 저해활성 검정)

  • The present study was conducted to evaluate of the 1,1-Diphenyl-2-Picrylhydrazyl (DPPH) radical scavenging effect, cytotoxicity and tyrosinase inhibition activities using methanol extracts from different parts of four herb plants. The results showed that whole and root extracts of yarrow the highest total polyphenol and flavonoid contents as well as whole of yarrow revealed the highest DPPH radical scavenging effect. In cytotoxicity test against three cancer cell lines, HeLa (uterus), SK-Hep-1 (liver), and YD-15 (oral), the whole extract of feverfew showed the highest toxicity with $IC_{50}$ values of $102.58-138.68{\mu}g/mL$. Also, mallow root extract ($71.24{\mu}g/mL$) exhibited potent tyrosinase inhibitory activity comparable to arbutin ($69.56{\mu}g/mL$), which was used as the control.
