피부의 노화와 주름 억제, 미백효과를 가지는 전통 한약재의 탐색 (Research of Traditional Herbal Medicines for Anti-aging, Inhibition Effect of Wrinkle and Whitening Effect in the Skin)

  • The aims of this study are to search herbal medicines that have anti-aging, anti-wrinkle, anti-oxidant and whitening effects. This is preliminary study as a finding for excellent and specific natural agents which inhibit the skin aging and whitening formation. In this study, the articles and the documents, which have been published in domestically or internationally, have been searched and analyzed. Articles were gathered by taking advantage of Society of Cosmetic Scientists of Korea's documents, Pub med database, Korean Medical Database and Korean Studies Information Service System. Among the many species of herbal medicines, we selected the herbal medicines showing superoxide scavenging activities, and 1.1-diphenyl-2-picrylhydrazyl(DPPH) scavenging effects. The herbal medicines were showed anti-wrinkle effects on Metrixmetallo proteinase (MMP-1) inhibition activity and elastase inhibition activity. Also we found herbal medicines that have highly effect of tyrosinase inhibition, L- DOPA inhibition and melanin synthesis inhibition.

식물성 한약의 Tyrosinase 활성 저해 효과 검색 (1) (Screening of Tyrosinase Inhibitory Activity of Plant Oriental Medicines (1))

  • Tyrosinase is a key enzyme to control the biosynthesis of melanin pigments and has two enzyme activities, namely of 1-tyrosine hydroxylase and of 1-dopa oxidase. Thus, tyrosinase is regarded as a target in skin-whitening and therapeutic intervention of local hyperpigmentation diseases. We have tested tyrosinase inhibitory activity on the water extracts of 50 species oriental medicinal plant. Among them, five medicinal plants, Linderae Radix, Clematidis Radix, Cinnamomi Cortex Spissus, Fritillariae Thunbergii Bulbus and Bulbus Fritillariae Cirrhosae were investigated strong inhibition effect. Five medicinal plants were fractionated using organic solvents (methylene chloride, ethyl acetate, n-butanol, water). Cinnamomi Cortex Spissus (ethyl acetate fraction) was investigated strong inhibition effect. Tyrosinase inhibitory activity below $IC_{50}\;40{\mu}g/ml$ is confirmed in five herbal plants that are Linderae Radix, Clematidis Radix, Cinnamomi Cortex Spissus, Fritillariae Thunbergii Bulbus and Bulbus Fritillariae Cirrhosae. Tyrosinase inhibitory levels ($IC_{50}\;{\mu}g/ml$) of each plants were 15.56, 35.02, 25.14, 15.20 and 39.77. We also investigate the effect of effective plant's fraction. in dose of $100{\mu}g/ml$, Cinnamomi Cortex Spissus (P-36) EtOAc fraction significant inhibitory effect over 50%. Clematidis Radix (P-35) and Cinnamomi Cortex Spissus (P-36) MC fraction inhibit tyrosinase each 36.60% and 43.21%. inhibitory rates of Fritillariae Thunbergii Bulbus (P-40) EtOAc and $H_2O$ fraction are 31.40% and 31.51%. Bulbus Fritillariae Cirrhosae (P-45) BuOH fraction regulate tyrosinase activity to 37.71%. We examined tyrosinase inhibitory activity of natural products and these results suggest that several herbs have potential as a new whitening material.

白급이 B16 흑색종세포의 멜라닌 형성 억제에 미치는 영향 (Inhibitory Effect of Rhizoma Bletillae on Melanogenesis of B16 Melanoma Cell)

  • Recently many efforts were focused to understand the mechanical insights of melanogenesis to develop the agents for hyper-pigmentation and hypo-pigmentation. In the melanin biosynthetic pathway, tyrosinase is the rate limiting enzyme, and ${\alpha}$-melanocyte stimulating hormone(MSH) or cAMP-elevating agents stimulate melanogenesis and enhance the melanin synthesis and the tyrosinase activity. The author has analyzed the effects of Rhizoma Bletillae on the basal melanogenic activities of B16/F10 mouse melanoma cells, and on the ${\alpha}$-MSH or forskolin-induced melanogenesis. Rhizoma Bletillae alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. Pretreatment of the cells with Rhizoma Bletillae also suppressed the increase of ${\alpha}$-MSH (100 nM) or forskolin (20 ${\mu}M$)-induced melanin content and tyrosinase activity. The decrease in the tyrosinase activity was paralled by a decrease in the abundance of tyrosinase protein and tyrosinase promoter activity. Pretreatment of the cells with Rhizoma Bletillae also inhibited the increase of forskolin(20${\mu}M$) induced the amount of tyrosinase protein and tyrosinase promoter activity. The results of DOPA staining revealed that pretreatment of the cells with Rhizoma Bletillae showed less intensity than B16 melanoma cells stimulated with ${\alpha}$-MSH or forskolin. These results suggest that Rhizoma Bletillae inhibits melanogenesis and abrogates ${\alpha}$-MSH and cAMP-induced melanogenesis in B16 melanoma cells.

Isolation of a Natural Antioxidant, Dehydrozingerone from Zingiber officinale and Synthesis of lts Analogues for Recognition of Effective Antioxidant and Antityrosinase Agents

  • In the present study, the antioxidative and inhibitory activity of Zingiber officinale Rosc. Rhizomes-derived materials (on mushroom tyrosinase) were evaluated. The bioactive co mponents of Z. officinale rhizomes were characterized by spectroscopic analysis as zingerone and dehydrozingerone, which exhibited potent antioxidant and tyrosinase inhibition activities. A series of substituted dehydrozingerones [(E)-4-phenyl-3-buten-2-ones] were prepared in admirable yields by the reaction of appropriate benzaldehydes with acetone and the products were evaluated in terms of variation in the dehydrozingerone structure. The synthetic analogues were examined for their antioxidant and antityrosinase activities to probe the most potent analogue. Compound 26 inhibited Fe$^{2+}$-induced lipid peroxidation in rat brain homogenate with an IC$_{50}$ = 6.3${\pm}$0.4 ${\mu}$M. In the 1,1-diphenyl- 2-picrylhydrazyl (DPPH) radical quencher assay, compounds 2, 7, 17, 26, 28, and 29 showed radical scavenging activity equal to or higher than those of the standard antioxidants, like ${\alpha}$-tocopherol and ascorbic acid. Compound 27 displayed superior inhibition of tyrosinase activity relative to other examined analogues. Compounds 2, 17, and 26 exhibited non-competitive inhibition against oxidation of 3,4- dihydroxyphenylalanine (L-DOPA). From the present study, it was observed that both number and position of hydroxyl groups on aromatic ring and a double bond between C-3 and C-4 played a critical role in exerting the antioxidant and antityrosinase activity.

Laccase- and Peroxidase-Free Tyrosinase Production by Isolated Microbial Strain

  • Laccase- and peroxidase-free tyrosinase has commercial importance in the production of L-3, 4-dihydroxyphenylalanine (L-DOPA), which is mainly used in the treatment of Parkinson's disease. In the present study, isolation of an actinomycetes microbial strain capable of producing only tyrosinase is reported. Among all soil isolates, three individual colonies revealed black color around the colony in the presence of tyrosine. Further screening for laccase and peroxidase activities using syringaldazine denoted that one of the isolates, designated as RSP-T1, is laccase and peroxidase negative and produces only tyrosinase. The microbe was authenticated as Streptomyces antibioticus based on 16S ribotyping. Effective growth of this isolate was noticed with the use of medium (pH 5.5) containing casein acid hydrolysate (10.0 g/l), $K_2HPO_4$ (5.0 g/l), $MgSO_4$ (0.25 g/l), L-tyrosine (1.0 g/l), and agar (15 g/l). The scanning electron micrograph depicted that the microbe is highly branched and filamentous in nature. The enzyme production was positively regulated in the presence of copper sulfate. The impact of different fermentation parameters on tyrosinase production depicted that the maximized enzyme titer values were observed when this isolate was grown at 6.5 pH and at $30^{\circ}C$ temperature under agitated conditions (220 rpm). Among all the studied physiological parameters, agitation played a significant role on tyrosinase production. Upon optimization of the parameters, the yield of tyrosinase was improved more than 100% compared with the initial yield.

  • This study was purposed to investigate the antioxidative effects, the enzyme activity of the alcohol metabolizing and melanin production of Maesil(Prune mume). The antioxidant activity of Maesil(Prunu mume) was analyzed by measuring thiobarbituric acid reactive substances(TBARS value) and electron donating ability. And we investigated the changes of alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH) activity by measuring the maximum absorbency at 340nm in vitro and human study. The inhibitory effects of Maesil were investigated in vitro and in B-16mouse melanoma cells on melanin biosynthesis that is closely related to hyperpigmentation. The antioxidant activities for TBA values were 29.65% in ascorbicacid, 45.35% in BHT, 15.99% in extract of dehydrated maesil flesh(EDMF) and 25.00% in extract of dehydrated maesil juice(EDMJ). The electron donating abilities by DPPH were 96.69% in ascorbic acid, 77.82% in BHT, 34.25% in EDMF, and 42.99%in EDMJ. Electron donating abilities by DPPH in the presence of 0.02% EDMF and EDMJ were 53.21% and 59.19% respectively. Facilitating rates of ADH activity were 137.92, 131.58, 152.96, 218.70, 111.76, and 144.27% in maesil juice, 5, 10, and 15% GMT, and 0.5 and 1.0% aspartic acid, respectively. ALDH activity increased in the order of Maesil juice > ALDH > GMT > aspartic acid, and facilitating rate of ALDH activity in Maesil juice was the highest at 976.44%. Maesil extracts inhibited tyrosinase activity that converts dopa to dopachrome in the biosynthesis process. B-16 cells treated by Maesil extracts showed that the viability was over 80%. Maesil and maesil products in vitro and B-16 cells inhibited melanin production significantly.

Purification and Some Properties of the polyphenol Oxidase form Ascidian, Halocynthia roretzi

  • Polyphenol oxidase(PPO) isolated from the crude extract of ascidian, Halocynthia roretzi, showed higher affinity for catechol than tyrosine or DL-DOPA. Successful enzyme assay could be performed at $25^{\circ}C$, 10min. by mixing 0.2ml of crude enzyme extract with 2.8ml of 0.13M catechol in 0.1M sodium phosphate buffer(pH 6.4). The specific activity of PPO which had been purified with a combination of ammonium sulfate treatment, ion exchange chromatography on DEAE-cellulose, and gel filtration on Sepharose 6B was 13-fold disc gel electrophoresis. The activity of PPO was stable from pH 5.0 to 8.0 and showed the peak activity at pH 6.4 .The optimum reaction temperature for PPO oxidation on catechol was 35$^{\circ}C$ and those enzyme were heat stable up to 4$0^{\circ}C$. Molecular weigth of the enzyme was estimated about 170kDa. One molecule was found to be composed of gour subunits. Two of them had molecular weigh of 55kDa and the others 30kDa. The {TEX}$K_{m}${/TEX} values, {TEX}$V_{max}${/TEX} and catalytic efficiency({TEX}$V_{max}${/TEX}/{TEX}$K_{m}${/TEX}) for catechol were 0.12mM, 2.5mM/liter/min. and {TEX}$0.18min^{-1}${/TEX} respectively. The substrate affinity and electrophorectic pattern suggested that the enzyme of ascidian was considered to be not tyosine but catechol oxidase.

Effects of Polygonati Rhizoma Extracts on the Collagenase Activity and Procollagen Synthesis in Hs68 Human Fibroblasts and Tyrosinase Activity

  • Objectives : This study was designed to investigate the collagen metabolism and tyrosinase activity of Polygonati Rhizoma extracts (PR). It's effects are to tonify spleen qi and augment the spleen yin. It enrichs the yin and moisten the lung. Methods : The effect of PR on type I procollagen production and collagenase (matrix metalloproteinase-1, henceforth referred as MMP-1) activity in human normal fibroblasts Hs68 after ultraviolet B (UVB, 312 nm) irradiation was measured by ELISA method. The tyrosinase activity after treatment of PR was measured. Results : There were no cytotoxicity at concentrations of 10, 30, $100{\mu}g/ml$. The reduced type I procollagen production was recovered by PR in UVB damaged Hs68 cells at a concentration of $100{\mu}g/ml$ ($16.2{\pm}0.0$ ng/ml) from control group ($13.9{\pm}0.5$ ng/ml). However there was no statistical significance. PR reduced The increased MMP-1 activity after UVB damage at concentrations of $10{\mu}g/ml$, $30{\mu}g/ml$, and $100{\mu}g/ml$ in a dose dependent manner ($42.2{\pm}20.5%$, $44.8{\pm}8.5%$, and $22.0{\pm}5.8%$). PR $100{\mu}g/ml$ treatment showed the statistical significace (p < 0.05). PR significantly reduced the tyrosinase activity at a concentration of 10 mg/ml ($32.0{\pm}12.8%$, p < 0.05). However, the L-DOPA oxidation was not changed. Conclusion : PR showed the anti-wrinkle effects and whitening effects in vitro. Although more researches are needed to validate the efficacy, these results suggest that PR may have potential as an anti-aging ingredient in cosmetic herb markets.

꿀풀과 식물 정유의 주성분인 Thymol의 미백활성에 관한 연구 (Anti-Melanogenic Effect of Thymol, a Major Odorant in Essential Oils of Family Lamiaceae)

  • Objectives : Thymol (2-isopropyl-5-methylphenol), a natural monoterpenoid phenol, is one of the major odorant constituents found in natural essential oils of various herbal plants, such as Thymus quinquecostatus and Thymus vulgaris. Multiple biological activities of thymol, including antioxidative, antimicrobial, and anti-inflammatory effects, have been reported in numerous in vitro studies, and recently it was suggested that thymol may could inhibit oxidization of L-dihydroxyphenylalanine (L-DOPA) to dopaquinone required in melanogenesis pathway, as an antioxidant. Methods : MTT assay was performed to test the cytotoxic effect of thymol in B16F10 cells. Inhibitory effect of thymol to tyrosinase activities were examined using both mushroom tyrosinase and intracellular tyrosinase. Expression level of tyrosinase in B16F10 cells were investigated by western blot analysis. Results : The cell viability was decreased by thymol treatment in dose-dependant manner, leading significant cytotoxicity in 500 and $1000{\mu}M$ thymol-treated groups. In the alpha-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis, administration of thymol significantly decreased extracellular (secreted) melanin content in dose-dependent manner. Cellular tyrosinase activity assay and western blot analysis of intracellular tyrosinase showed that thymol has a strong anti-melanogenic effect by inhibition of tyrosinase activity and by decreasing expression of tyrosinase that contribute to melanin synthesis in the B1610 cells. Conclusions : As the first functional study that prove anti-melanogenic effect of thymol and its underlying mechanism in the living cells, our study suggests the applicability of fragrance as the functional materials of cosmetics or health supplement, not as just an additive.

The prominin-like Gene Expressed in a Subset of Dopaminergic Neurons Regulates Locomotion in Drosophila

  • CD133, also known as prominin-1, was first identified as a biomarker of mammalian cancer and neural stem cells. Previous studies have shown that the prominin-like (promL) gene, an orthologue of mammalian CD133 in Drosophila, plays a role in glucose and lipid metabolism, body growth, and longevity. Because locomotion is required for food sourcing and ultimately the regulation of metabolism, we examined the function of promL in Drosophila locomotion. Both promL mutants and pan-neuronal promL inhibition flies displayed reduced spontaneous locomotor activity. As dopamine is known to modulate locomotion, we also examined the effects of promL inhibition on the dopamine concentration and mRNA expression levels of tyrosine hydroxylase (TH) and DOPA decarboxylase (Ddc), the enzymes responsible for dopamine biosynthesis, in the heads of flies. Compared with those in control flies, the levels of dopamine and the mRNAs encoding TH and Ddc were lower in promL mutant and pan-neuronal promL inhibition flies. In addition, an immunostaining analysis revealed that, compared with control flies, promL mutant and pan-neuronal promL inhibition flies had lower levels of the TH protein in protocerebral anterior medial (PAM) neurons, a subset of dopaminergic neurons. Inhibition of promL in these PAM neurons reduced the locomotor activity of the flies. Overall, these findings indicate that promL expressed in PAM dopaminergic neurons regulates locomotion by controlling dopamine synthesis in Drosophila.
