Objectives : The purpose of this study is to examine the effects of external application of HC001 on atopic dermatitis. HC001, newly composed herbal medicine combinations contains 9 medicinal herbs which are known to have anti-inflammatory and anti-allergic effects. We investigated the anti-inflammatory effects of HC001 on atopic dermatitis-like skin lesions in mice. Methods : Seven-week-old BALB/c mice were sensitized with DNCB to develop atopic dermatitis. Animals were divide into three group: Normal, DNCB (Negative control group), HC001 (Experimental group, treated with DNCB and HC001). Skin sections were stained with H&E to measure the thicknesses of the epidermis and dermis, respectively. The expression of NF-${\kappa}B$ protein was measured by western blotting analysis in skin lesion. Results : Topical HC001 treatment significantly restored the skin thickening and hyperplasia of the epidermis and dermis compared with DNCB-sensitized group in histopathological analyse. In addition, HC001 inhibited the expression of NF-${\kappa}B$ protein increased in DNCB-induced atopic dermatitis-like skin lesions in mice. Conclusions : These results suggest that HC001 may be useful as an external application agent for atopic dermatitis based on reductions of various inflammatory responses.
Objective: This study was carried out to evaluate the inhibitory effect on immme hypersensitive reaction by Yangdokbackhotang extract. methods: To study, We divided 4 weeks old rats into three groups of 20, which are the constrasive group with nothing treated, the skin transplated only, and the Yangdokbackhotang extract fed before skin transplantation. And did the experiment. Results: 1. The ear swelling in mice treated with YB noticeably decreased than only DNCB treated mice. 2. As shown in Table3, also results in a significant decrease of YB group than ST group in the number of positive cells of $NF-{\kappa}B$, COX-2, IL-12B, and ICAM-1. From the above results, it could be concluded that Yangdokbackhotang showed benificial immunosupressive effect on the hypersensitive reaction.

Atopic dermatitis (AD) is the most common inflammatory pruritic skin disease worldwide, characterized by the infiltration of multiple pathogenic T lymphocytes and histological symptoms such as epidermal and dermal thickening. This study aims to investigate the effect of vinpocetine (Vinp; a phosphodiesterase 1 inhibitor) on a 1-chloro-2,4-dinitrobenzene (DNCB)-induced AD-like model. DNCB (1%) was administered on day 1 in the AD model. Subsequently, from day 14 onward, mice in each group (Vinp-treated groups: 1 mg/kg and 2 mg/kg and dexamethasone-treated group: 2 mg/kg) were administered 100 µl of a specific drug daily, whereas 0.2% DNCB was administered every other day for 30 min over 14 days. The Vinp-treated groups showed improved Eczema Area and Severity Index scores and trans-epidermal water loss, indicating the efficacy of Vinp in improving AD and enhancing skin barrier function. Histological analysis further confirmed the reduction in hyperplasia of the epidermis and the infiltration of inflammatory cells, including macrophages, eosinophils, and mast cells, with Vinp treatment. Moreover, Vinp reduced serum concentrations of IgE, interleukin (IL)-6, IL-13, and monocyte chemotactic protein-1. The mRNA levels of IL-1β, IL-6, Thymic stromal lymphopoietin, and transforming growth factor-beta (TGF-β) were reduced by Vinp treatment. Reduction of TGF-β protein by Vinp in skin tissue was also observed. Collectively, our results underscore the effectiveness of Vinp in mitigating DNCB-induced AD by modulating the expression of various biomarkers. Consequently, Vinp is a promising therapeutic candidate for treating AD.
The aim of this study was to examine inhibitory effects of pine needle ethanol extracts (PNEE) on atopic dermatitis (AD). To determine inflammatory activity PNEE was added to LPS-induced murine peritoneal macrophages for an in-vitro test. In addition, anti-AD test was carried out by spreading PNEE on the dorsal skin of 2,4-dinitrochlorobenzene (DNCB)- induced BALB/c mice. It was confirmed that the nitric oxide (NO) secretion was suppressed when $1{\sim}50{\mu}g/mL$ of PNEE were added to LPS-induced murine peritoneal macrophages. Moreover, levels of TNF-${\alpha}$, IL-6, and IL-$1{\beta}$, were decreased. For the anti-AD test, PNEE alleviated symptoms of the erythema in DNCB-induced mice. Furthermore, the IFN-${\gamma}$ secretion of the group treated with PNEE was increased in splenocytes from DNCB-induced mice compared to the positive control, while IL-4 secretion diminished. Through these results, we can conclude that PNEE can inhibit AD by modulating the IFN-${\gamma}$, IL-4 cytokines production and inhibiting inflammation.

Objective : The objective of this study was to demonstrate the effect of Persicae Semen (PS) in DNCB-induced atopic dermatitis mouse and HaCaT cell. Methods : The BALB/c mice were divided into four groups. To develop atopic dermatitis, 200 ㎕ of 1 and 0.5% DNCB solution was put on the back of mice in the Control group, the PS-Low group and the PS-High group once a day. After application of DNCB, 200 ㎕ of the PS extract was also treated. The Normal group was given PBS. The mice dorsal skin was stained with Masson's trichrome, H&E, and toluidine blue to evaluate the thickness of the epidermis and dermis, infiltration of eosinophils and mast cells respectively. ELISA was applied to measure the serum level of IgE and IL-6. Toxicity of PS was measured by MTS assay in HaCaT cell. To investigate the effects of PS on HaCaT cells, cells were pre-treated with PS for 1h, and then stimulated with TNF-α and IFN-γ. After 24 hours, the expression of TARC was analyzed using RT-PCR. Results : PS not only significantly diminished the thickness of the epidermis and dermis, but also reduced the infiltration of eosinophil and mast cell in skin lesion. PS also reduced the serum IgE and IL-6 level which plated important roles in the atopic dermatitis. The expression of TARC was decreased significantly in TNF-α/IFN-γ stimulated HaCaT cell. Conclusion : These results suggest that PS may be effective in alleviating the atopic dermatitis induced by DNCB and inflammation by TNF-α/IFN-γ.

Objectives : The purpose of this study is to investigate effects of Syzygium aromaticum (SAE) spread on the allergic contact dermatitis caused by 2,4-dinitro-chlorobezene (DNCB). Methods : Forty-two mice were divided into six groups ; normal, negative control (DNCB-treated), positive control (DNCB + 1% pimecrolimus), experimental group I, II and III. control and experimental groups were induced allergic contact dermatitis by DNCB. Experimental group I(DNCB + 0.2% SAE), II(DNCB + 1% SAE) and III(DNCB + 5% SAE) were spread SAE and positive control was spread the 1% pimecrolimus. In this study, effect of SAE on clinical aspects on the skin, histopathological change, the blood level of IgE, cytokines, histamine were investigated. In addition, effect of SAE on spleen $CD4^+/CD8^+$ T cell subset was investigated. Results : 1. In experimental group I, II and III, erythemas and edema were more reduced than negative control. 2. In experimental group I, II and III inflammatory edema and the numbers of infiltrated inflammatory cells were more reduced than negative control. 3. In experimental group I, II and III, clinical skin score was more reduced than negative control. 4. In experimental group II and III, the thickness of skin was statistically significant reduced than negative control. 5. In experimental group II and III, histamine release was statistically significant reduced than negative control in dose-dependantly. 6. In experimental group II and III, cytokines (IL-1${\beta}$, TNF-${\alpha}$, IL-4, IL-6, IL-10) were statistically significant reduced than negative control in dose-dependantly. 7. In experimental group I, II and III, the level of total IgE was statistically significant reduced than negative control in dose-dependantly. 8. In experimental group III, $CD4^+$ and $CD8^+$ T cells were statistically significant decreased similar to the positive control. Conclusions : According to above experiments, Syzygium aromaticum(SAE) was effective on allergic contact dermatitis.

Atopic dermatitis (AD) is a chronic inflammatory skin disease with a complex etiology that encompasses immunologic responses. AD is frequently associated with elevated immunoglobulin (Ig) E levels, and common environmental factors contribute to its pathogenesis. Several recent studies have documented the role of specific lactic acid bacteria in the treatment and prevention of AD in humans and mice. In this study, the efficacy of Duolac ATP, a probiotic preparation, was determined in a mouse model with AD-like skin lesions. Alterations in the cytokine levels and histological staining suggested the alleviation of AD. The in vivo test showed that T helper (Th)2 cytokines, IgE, interleukin (IL)-4, and IL-5, were significantly downregulated, whereas Th1 cytokines, IL-12p40 and interferon (IFN)-${\gamma}$, were upregulated in all groups of mice treated with Duolac ATP compared to that observed in the group of mice treated with 1-chloro-2,4-dinitrobenzene (DNCB) alone. Moreover, the scratch score decreased in all mice treated with Duolac ATP. Staining of the dorsal area of the mice in each group with hematoxylin and eosin and toluidine blue further confirmed the alleviation of AD in mice orally treated with Duolac ATP. These results suggest that Duolac ATP inhibits the development of AD-like skin lesions in NC/Nga mice by suppressing the Th2 cell response and increasing the Th1 cell response. Thus, Duolac ATP is beneficial and effective for the treatment of AD-like skin lesions.

Atopic dermatitis is an allergic inflammatory skin disease caused by aberrant and overreactive immune responses including overactivation of $T_H2$ immune responses, high levels of IgE as well as proinflammatory cytokines and chemokines. We examined whether BHOSB, a traditional herbal medicine, has modulatory effects on various immunological factors related to pathogenesis of atopic dermatitis in ONCB treated NC/Nga mice. Oral administration of BHOSB at the concentration of 10.8 mg/mouse/day significantly inhibited the production of IgE compared with control, and the levels of IgG2a and IgG2b, but not IgG1, were also significantly reduced. Production of IL-6 and TNF-a was greatly decreased. The results from flowcytometry of peripheral blood mononuclear cells indicated that the percentages of C03+C069+ cells and C04+ were significantly decreased by BHOSB. Theses results suggested that BHOSB has suppressive effects on aberrant and overreactive immunological activities in ONCB-induced dermatitis mice of NC/Nga.
Objectives: Realgar has been frequently used for skin disorders in history of herbal medicine. However, the efficacy of realgar has not been examined in atopic dermatitis(AD). In this study, the effects of realgar on AD were investigated, especially on pruritus and inflammation. Methods: AD lesions were induced in the shaved backs of BALB/c mice through repeated application of DNCB. The mice were treated for 11 days with 1% realgar ($100{\mu}L/day$). Histological changes in skin thickness were observed. The anti-pruritic effects of realgar were evaluated by the change in numbers of scratching behavior of mice and expression of substance P. The expressions of cytokines IL-4 and IL-6 were measured. Also, anti-inflammatory effects of realgar were examined on expressions of NF-${\kappa}B$, phospho-$I{\kappa}B{\alpha}$ and mitogen-activated protein kinases (MAPKs). Results: Realgar decreased skin thickness (both dermal and epidermal) 38% and 17% respectively, compared to positive control, DNCB group. The scratching behavior of mice was reduced by 42% and expression of substance P was significantly less. Cytokines IL-4 and IL-6 were significantly reduced by 52.6% and 77.6%, respectively. The expressions of NF-${\kappa}B$, phospho-$I{\kappa}B{\alpha}$ and MAPKs (phospho-ERK1/2, -p38 and -JNK) were significantly suppressed with marked effects on phospho-ERK1/2. Conclusions: The collective results suggest that realgar shows anti-pruritic and anti-inflammatory effects on AD. And realgar might be a potential therapeutic candidate for treatment of atopic dermatitis.

Atopic dermatitis (AD) is a common skin disease characterized by chronic and relapsing inflammatory dermatitis with immunological disturbances. Spleen deficiency (脾虛) is one of the major causes of AD, so development of animal model is required for AD research that reflects the pattern identification. The groups that we have used in this study included Senna folium extracts (SFE), 2,4-dinitrochlorobenzene (DNCB), and normal mice. Therefore, the present study was developed to atopic dermatitis mouse model with spleen deficiency in 2,4-dinitrochlorobenzene (DNCB) and senna leaves extracts induced AD in NC/Nga mice. The results demonstrated that senna leaves extract treatment significantly increased the dermatitis clinical score and epidermal thickness in AD-like skin lesions. We also proved beyond doubt that there was occurrence of erythema and skin moisture indices in the senna leaves extract groups. Further, we also found that the level of serum immunoglobulin E (IgE) in the senna leaves extract-treated group was increased. The amount of IL-4, IL-13, $TNF-{\alpha}$ and $TGF-{\beta}$ mRNA determined by real-time PCR was increased remarkably when senna leaves extract groups were treated on dorsal skin. Senna leaves extract groups significantly promoted the number of CD11B+/Gr-1 cell in skin, as well as the number of CD4+/CD8+ cell in dorsal skin compared with control. The review summarizes recent process in our understanding of the immunopathophysiology of spleen deficiency AD and the implications for spleen deficiency mouse models of AD on drug discovery from medical plants.
