아토피피부염은 만성 염증성 피부질환으로 습진성병변, 피부건조 및 가려움증의 특징적 증상을 보이는 질환이다. 현재 사용되고 있는 아토피피부염 치료약제들은 오래 사용시 여러 부작용이 있어서 이를 대신할 새로운 대체 치료법의 개발이 요구되고 있다. Curcumin은 항염, 항알러지와 같은 효과를 나타내는 천연 폴리페놀(polyphenol)이며, 발광다이오드(light emitting diode, LED) 광치료는 아토피피부염과 상처치유에 효과가 있는 것으로 알려져 있다. 본 연구는 DNCB로 유도한BALB/c 마우스의 아토피피부염에서 curcumin 투여와 630 nm LED 조사를 병용하여 그 치료 효과를 높일 수 있는가를 조사하기 위해 시도하였다. 아토피피부염의 치료 효과를 조사하기 위해 modified SCORAD 지수, 조직병리학적 분석, 면역조직화학염색 및 TUNEL 검사를 사용하였다. 그 결과 curcumin 투여와 630 nm LED 조사가 각각 SCORAD 지수, 비만세포 및 탈과립세포의 수, 호산구 수, 표피세포의 증식능 및 자멸세포의 수를 의미 있게 감소시키고, 비만세포의 탈과립을 억제하여 섬유모세포에 의한 콜라겐 생성을 의미 있게 감소시켰으며, curcumin 투여와 LED 조사를 병용하였을 경우 단일 처리군에 비해 이들 수치가 의미 있게 감소하는 것을 확인하였다. 이러한 결과는 아토피피부염 치료에 curcumin 투여와 LED 조사가 각각 효과가 있음을 의미하며, 병용 치료 시 그 효과가 좀 더 향상된다는 것을 의미하는 것으로, 아토피피부염의 대체치료 전략으로 curcumin 투여와 630 nm 조사의 병용 치료를 제안한다.
Atopic dermatitis (AD) is an allergic, inflammatory skin disease characterized by chronic eczema and mechanical injury to the skin, caused by scratching. Korean red ginseng (RG) has diverse biological activities, but the molecular effects of RG on allergic diseases, like AD, are unclear. The present study was designed to investigate whether RG inhibits 1-chloro-2,4-dinitrobenzene (DNCB)-induced AD in a mouse model. DNCB was applied topically on the dorsal surface of Balb/c mice to induce AD-like skin lesions. We observed the scratching behavior and examined the serum IgE level and interleukin (IL)-4 and IL-10 in splenocytes compared with dexamethasone. We also evaluated the DNCB-induced mitogen-activated protein kinases (MAPKs), NF-${\kappa}B$, and Ikaros activities after RG treatment using reverse transcriptase-polymerase chain reaction, Western blotting, and ELISA. Our data showed that the topical application of RG significantly improved the AD-like skin lesions and scratching behavior. RG decreased not only the mRNA expression of IL-4 and IL-10, but also the secretion of IL-4 protein and serum IgE in mice. Additionally, RG treatment decreased the DNCB-induced MAPKs activity and subsequent Ikaros translocation irrespective of NF-${\kappa}B$. We suggest that RG may be useful as a therapeutic nutrition for the treatment of AD.
본 연구에서는 1-chloro-2,4-dinitrobenzene (DNCB)에 의해 유도된 아토피성 피부염 마우스 모델에서 고삼, 감초 및 백선피 추출물과 그들 복합 추출물(GGB)의 항아토피 효능을 평가하였다. 아토피성 피부염의 병리학적 형태인 홍반, 귀부종, 부종, 각질과 상처는 복합 추출물 중 고삼, 감초 및 백선피 추출물이 3:1:1로 혼합된 GGB-1 처리군에서 다른 시료 처리군들에 비하여 유의적인 억제 효능을 나타내었다. 또한 면역 기관인 비장의 무게도 GGB-1 처리군에서 감소하였고, 간의 무게는 정상군과 시료 처리군의 값이 유사한 결과를 나타내었다. 아울러 GGB-1 처리군에서 비만 세포의 침윤이 감소되었으며, 혈청 중 IgE의 농도 역시 GGB-1 처리군에서 가장 낮은 값을 보였다. 비록 단일 한약재와 그들의 혼합물 처리군에서는 면역 사이토카인(IFN-${\gamma}$, IL- 1, 4, 5, 6, 13, $1{\beta}$, and TNF-${\alpha}$) 의 값이 유의적으로 감소되었으나, 여러 가지 결과들을 종합하여 볼 때 고삼, 감초 및 백선피 추출물이 3:1:1로 혼합된 복합 추출물인 GGB-1이 아토피성 피부염의 개선에 효과적인 약물로 판단된다.
본 연구에서는 DNCB에 의해 인위적으로 접촉피부염을 유발시킨 후 천궁추출물을 표피에 처리하여 피부표면, 피부두께, 혈장내 성분, 혈장내 IgE 농도 및 항산화효소을 조사하여 표피의 회복능에 미치는 효과를 조사하였다. 천궁추출물은 DNCB에 의해 유발된 알레르기성 접촉피부염에 도포 시에 환부의 피부염이 급격히 호전되었으며, 피부표면은 정상군에 비교될 정도로 각질이 없어진 깨끗하고 매끄러운 상태로 되었으며, 표피층의 두께도 점차 정상군과 같은 상태로 회복되고 있는 것으로 관찰되었다. 또한 알레르기성 접촉피부염이 유발되어 체내 지질함량이 급격히 증가된 경우 지질함량을 낮춰 정상수준으로 회복시키는 지질함량의 개선에 효과적으로 작용하고 있으며, 직접적으로 혈액 내 IgE 수준의 감소에 관여하여 피부의 과민반응 해소에 관여할 것으로 생각된다. SOD와 catalase의 활성은 천궁추출물의 처리에 의해 정상군 수준으로 낮아지는 경향을 보이고 있어 항산화 효소의 활성을 낮춰 정상수준으로 회복시켜주는 효과를 갖는 것으로 생각된다. 결론적으로 천궁 추출물은 알레르기성 접촉피부염의 회복에 매우 효과적으로 작용하는 천연 유용자원으로 이용할 수 있을 것으로 생각된다.
Background: The skin acts as a barrier to protect organisms against harmful exogenous agents. Compound K (CK) is an active metabolite of ginsenoside Rb1, Rb2 and Rc, and researchers have focused on its skin protective efficacy. In this study, we hypothesized that increased expression of the serine protease inhibitor Kazal type-5 (SPINK5) may improve skin barrier function. Methods: We screened several ginsenosides to increase SPINK5 gene promoter activity using a transactivation assay and found that CK can increase SPINK5 expression. To investigate the protective effect of CK on the skin barrier, RT-PCR and Western blotting were performed to investigate the expression levels of SPINK5, kallikrein 5 (KLK5), KLK7 and PAR2 in UVB-irradiated HaCaT cells. Measurement of transepidermal water loss (TEWL) and histological changes associated with the skin barrier were performed in a UVB-irradiated mouse model and a 1-chloro-2,4-dinitrobenzene (DNCB)-induced atopic dermatitis-like model. Results: CK treatment increased the expression of SPINK5 and decreased the expression of its downstream genes, such as KLKs and PAR2. In the UVB-irradiated mouse model and the DNCB-induced atopic dermatitis model, CK restored increased TEWL and decreased hydration and epidermal hyperplasia. In addition, CK normalized the reduced SPINK5 expression caused by UVB or DNCB, thereby restoring the expression of the proteins involved in desquamation to a level similar to normal. Conclusions: Our data showed that CK contributes to improving skin-barrier function in UVB-irradiated and DNCB-induced atopic dermatitis-like models through SPINK5. These results suggest that therapeutic attempts with CK might be useful in treating barrier-disrupted diseases.
BACKGROUND/OBJECTIVES: Grifola frondosa, commonly referred to as the maitake mushroom, has been studied extensively to explore its potential health benefits. However, its anti-inflammatory effects in skin disorders have not been sufficiently elucidated. This study aimed to elucidate the anti-inflammatory role of the ethanol extract of G. frondosa in atopic dermatitis (AD) using in vivo and in vitro models. MATERIALS/METHODS: We investigated its impact on skin and spleen inflammatory responses in Dermatophagoides farinae extract (DFE)/1-chloro-2,4 dinitrochlorobenzene (DNCB)-induced AD-like skin lesions in a mouse model. Additionally, we determined the immunosuppressive response and mechanism of G. frondosa by inducing atopic-like immune reactions in keratinocytes through tumor necrosis factor (TNF)-α/interferon (IFN)-γ stimulation. RESULTS: Our study revealed that G. frondosa ameliorates clinical symptoms in an AD-like mouse model. These effects contributed to the suppression of Th1, Th2, Th17, and Th22 immune responses in the skin and spleen, leading to protection against cutaneous inflammation. Furthermore, G. frondosa inhibited the production of antibodies immunoglobulin (Ig)E and IgG2a in the serum of AD mice. Importantly, the inhibitory effect of G. frondosa on inflammatory cytokines in TNF-α/IFN-γ-stimulated AD-like keratinocytes was associated with the suppression of MAPK (Mitogen Activated Protein Kinase) pathway activation. CONCLUSIONS: Collectively, these findings highlight the potential of G. frondosa as a novel therapeutic agent for AD treatment and prevention.
This study was conducted to determine if YJ-1, a novel herbal complex from a mixture of six oriental herbs (Hydnocarpi Semen, Sesami Semen, Dictamni Radicis Cortex, Momordicae Semen, Xanthii Fructus, and Sophorae Radix), has therapeutic properties for the treatment of atopic dermatitis (AD). Using AD like symptom-induced NC/Nga mice by 1-chloro 2,4-dinitrobenzene (DNCB), the effectiveness of YJ-1 on AD was evaluated. Elidel cream$^{(R)}$ (1% pimecrolimus) was used as a control. Dermal application of YJ-1 reduced major clinical signs of AD such as erythema, pruritus, lichenification, edema/escoriations, and dryness. Interestingly, YJ-1 more improved AD-related symptoms including decrease of spleen weight, IL-4, and IgE level in the serum as well as reduction of scratching counts and clinical skin severity in the NC/Nga AD mouse model. Especially, treatment of YJ-1 at 20% in NC/Nga mice more effected than Elidel cream. These results suggest that the ointment of YJ-1 may enhance the process of AD healing by alleviating allergic reaction and has potential for therapeutic reagent for the treatment of AD.

Atopic dermatitis (AD) is a chronic inflammatory skin disease with a complex etiology that encompasses immunologic responses. AD is frequently associated with elevated immunoglobulin (Ig) E levels, and common environmental factors contribute to its pathogenesis. Several recent studies have documented the role of specific lactic acid bacteria in the treatment and prevention of AD in humans and mice. In this study, the efficacy of Duolac ATP, a probiotic preparation, was determined in a mouse model with AD-like skin lesions. Alterations in the cytokine levels and histological staining suggested the alleviation of AD. The in vivo test showed that T helper (Th)2 cytokines, IgE, interleukin (IL)-4, and IL-5, were significantly downregulated, whereas Th1 cytokines, IL-12p40 and interferon (IFN)-${\gamma}$, were upregulated in all groups of mice treated with Duolac ATP compared to that observed in the group of mice treated with 1-chloro-2,4-dinitrobenzene (DNCB) alone. Moreover, the scratch score decreased in all mice treated with Duolac ATP. Staining of the dorsal area of the mice in each group with hematoxylin and eosin and toluidine blue further confirmed the alleviation of AD in mice orally treated with Duolac ATP. These results suggest that Duolac ATP inhibits the development of AD-like skin lesions in NC/Nga mice by suppressing the Th2 cell response and increasing the Th1 cell response. Thus, Duolac ATP is beneficial and effective for the treatment of AD-like skin lesions.
