Cloning and Characterization of Novel Soluble Acid Invertase Which is Responsible to JA, ABA and GA During Tip Growth of Pea Seedlings (Pisum sativum)

  • The enzyme invertase contributes to sugar unloading, pathogen defense, differentiation and development in plants. We cloned the complete cDNA of a soluble acid invertase from pea seedlings (Pisum sativum) via RT-PCR and the rapid amplification of the cDNA end (RACE) technique. The full-length cDNA of the soluble pea invertase comprised 2237 bp and contained a complete open reading frame encoding 647 amino acids. The deduced amino acid sequence showed high homology to soluble acid invertases from various plants. Northern blot analysis demonstrated the soluble acid invertase gene of P. sativum was strongly expressed in sink organs such as shoot tips and root tips, and induced by abscisic acid, gibberellic acid and jasmonic acid in shoots. Especially, gibberellic acid enhanced the gene expression of the soluble acid invertase in a time-dependent manner. This study presents that the gene expression patterns of a soluble acid invertase from pea are strongly consistent with the suggestion that individual invertase gene product has different functions in the growing plant.

토양에서 분리한 Bacilus flexus로부터 Invertase의 생산 (Production of Invertase from Newly Isolated Strain Bacilus flexus)

  • In the present study, we isolated a new bacterial strain producing invertase (EC 3.2.1.26) and determined optimized culture condition in flask culture. The strain was identified as Bacilus flexus determined by the 16S rDNA sequencing method. The invertase was produced only in the sucrose medium as the sole carbon source. Potassium nitrate was an adequate nitrogen source for enzyme production, whereas meat peptone showed the highest bacterial growth. Enzyme production was increased about 2-fold when $MgSO_4\cdot7H_2O$ was supplemented to the growth media. The optimum temperature was found to be $30^{\circ}C$ for both enzyme production and bacterial growth. Invertase exhibited pH optima in the range 5.0-6.0 and have a temperature optimum at $40^{\circ}C$, similarly to other invertases found from different microbial sources. Several mineral ions (K and Fe) stimulated the invertase activity, whereas some bioelements (Ag, Mg, and Mn) inhibited enzyme activity. Under the optimized culture condition, the maximum enzyme production (over 250 units/mL) was achieved at 20 h. To the best of our knowledge, this is the first time to report on invertase production by Bacilus flexus.

외래 알파아밀라제의 Saccharomyces cerevisiae에서의 생산과 분비효율의 증진 (Improvement of Production and Secretion of Heterologous \alpha-Amylase from Saccharomyces cerevisiae.)

  • Saccharomyces cerevisiae로부터 외래 $\alpha$-amylase의 발현 및 분비를 증진시키기 위하여 여러 실험이 수행되었다. ADC1 promoter와 mouse salivary $\alpha$-amylase cDNA gene의 native signal sequence를 효모의 PRB1 promoter와 invertase leader sequence로 대치한 plasmid vector pCNN(AMY)를 제작하였다. 효모세포에서 생성된 $\alpha$-amylase의 세포외로의 분비율은 mouse o-amylase의 native signal sequence인 경우는 약 89.4%이었으며 invertase leader sequence로 치환된 경우는 96.3%로 분비효율이 증진되었다. 야생주인 K8l/pCNN(AMY)와 호흡결여변이주인 K81/pCNN(AMY)p-의 혐기적 조건하에서의 배양 결과 $\alpha$-amylase 생산량이 K8l/pCNN(AMY)보다 K81/pCNN(AMY)p-가 약 5~8배 정도 증가하였다. $\alpha$-Amylase의 생산에 있어서 배지조성에 따른 K81/pCNN(AMY)의 생산증진의 비교는 배지성분인 yeast extract와 peptone의 구성비율을 비교하였을 때 yeast extract 1%와 peptone 2%, NaCl의 경우 100 mM, 2-mercaptoethanol인 경우에는 0.015%(w/v)을 첨가하였을 때 최대 효소 활성을 나타내었고, 특히 2-mercaptoethanol인 경우에는 대조구에 비해 효소 생산량이 약 3배 정도 증진되었다.

Levan-Producing Bacillus subtilis BS 62 and Its Phylogeny Based on Its 16S rDNA Sequence

  • A viscous substance producer strain BS62, which was isolated from conventional Chungkookjang, was examined for its productivity of levansucrase and levan during soybean fermentation at $37{\circ}C$. After one day of cultivation, the enzyme activity reached the highest level, 8 units $ml^{-1}$. Extracts of fermented soybeans were precipitated by ethanol and hydrolyzed by either 0.1 N HCl or invertase, and the hydrolyzates were analyzed using thin layer and ion chromatographies. Fructose was the only sugar detected. This suggest that fructose was derived from the levan produced by the strain BS62 during soybean fermentation. The aerobic, endospore-forming bacterium BS62 was identified as a Bacillus subtilis sp., based on the composition of its cellular fatty acids and phylogeny, which was determined by its 16S rDNA sequence.

The Role of Rice Vacuolar Invertase2 in Seed Size Control

  • Sink strength optimizes sucrose import, which is fundamental to support developing seed grains and increase crop yields, including those of rice (Oryza sativa). In this regard, little is known about the function of vacuolar invertase (VIN) in controlling sink strength and thereby seed size. Here, in rice we analyzed mutants of two VINs, OsVIN1 and OsVIN2, to examine their role during seed development. In a phenotypic analysis of the T-DNA insertion mutants, only the OsVIN2 mutant osvin2-1 exhibited reduced seed size and grain weight. Scanning electron microscopy analysis revealed that the small seed grains of osvin2-1 can be attributed to a reduction in spikelet size. A significant decrease in VIN activity and hexose level in the osvin2-1 spikelets interfered with spikelet growth. In addition, significant reduction in starch and increase in sucrose, which are characteristic features of reduced turnover and flux of sucrose due to impaired sink strength, were evident in the pre-storage stage of osvin2-1 developing grains. In situ hybridization analysis found that expression of OsVIN2 was predominant in the endocarp of developing grains. A genetically complemented line with a native genomic clone of OsVIN2 rescued reduced VIN activity and seed size. Two additional mutants, osvin2-2 and osvin2-3 generated by the CRISPR/Cas9 method, exhibited phenotypes similar to those of osvin2-1 in spikelet and seed size, VIN activity, and sugar metabolites. These results clearly demonstrate an important role of OsVIN2 as sink strength modulator that is critical for the maintenance of sucrose flux into developing seed grains.

Characterization of a new staphylococcal site-specific recombinase sin and genetic organization of its flanking region

  • A new site-specific recombinase sin, as a component of a putatie transposon has been cloned and its base sequence has been determined. The proposed sin shows a hish degree of homology with pI9789-sin and pSK1-sin. There is a large (16 bp) inverted repeat downstream of proposed sin and the postulate dhelix-turn-helix motif is located at the extreme C-terminus of the poposed Sin. The transposase gene (tnpA) and .betha.-lactamase gene (blaZ) are located upstream of sin and arsenate reductase gene (arsC) and arsenic efflux pump protein gene (ars B) are downstream. This genetic arrangement seems to be a part of a new putative transposon because there is no known transposon with a gene arrangement of tnpA-blaZ-sin-arsC.

Cloning of the Genomic DNA Which Complements the Drug-Hypersensitivity of Saccharomyces cerevlsiae

  • The yeast Saccharomyces cerevisiae, mutant CH117, shows a drug-hypersensitivity (dhs) to cycloheximide, bleomycin, actinomycin D, 5-fluorouracil. nystatin, nigericin and several other antibiotics. CH 117 was also temperature-sensitive (ts). being unable to grow at $37^{\circ}C$ and secreted more invertase and acid phosphatase into the medium than the parent yeast. CH117 grows very slowly and the cell shape is somewhat larger and more sensitive to zymolyase than the wild type cells. Light microscopic and electron microscopic observation also revealed abnormality of the mutant cell wall. These characteristics indicate that CH117 has a defect in an essential component of the cell surface and that the cell wall which performs barrier functions has become leaky in the mutant. We screened a genomic library of wild type yeast for clones that can complement the mutation of CH117. A plasmid, pCHX1, with an insert of 3.6 kilobases (kbs) could complement the dhs and ts of CH117. Deletion and subcloning of the 3.6 kb insert showed that a gene for the complementation of mutant phenotypes was located in 1.9 kbs Puvll-Hindlll fragment.

Secretion of Bovine $\beta$-Casein by Saccharomyces cerevisiae

  • Yeast expression plasmids containing an appropriate leader sequence and bovine $\beta$-casein cDNA were constructed to produce $\beta$-casein for the study of its functional characteristics. Two kinds of expression systems for $\beta$-casein were constructed using pCGYl444 as a precursor plasmid. This plasmid is a yeast-E. coli shuttle vector which contains the chelatin promoter. The plasmid pISB202 contains the invertase leader sequence and $\beta$-casein gene. The plasmid pDEB303 contains the original bovine $\beta$-casein leader sequence gene. These two plasmids were introduced into S. cerevisiae AB116 which is a strain deficient in the major yeast proteinases. Each clone was grown in minimal media for 24 h before induction by $CuSO_4$. The cells were thus grown under expression conditions. Both strains harbouring pISB202 and pDEB303 expressed bovine $\beta$-casein. The $\beta$-casein was detected using immunochemical staining after western blot. Secretion of $\beta$-casein was detected in the culture broth. The estimated amount of secreted $\beta$-casein was approximately 50 ${\MU}g$/l.

Characterization of biphenyl biodegradation, and regulation of iphenyl catabolism in alcaligenes xylosoxydans

  • Alcaligenes xylosoxydans strain SMN3 capable of utilizing biphenyl grew not only on phenol, and benzoate, but also on salicylate. Catabolisms of biphenyl and salicylate appear to be interrelated since benzoate is a common metabolic intermediate of these compounds. Enzyme levels in the excatechol 2. 3-dioxygenas which is meta-cleavage enzyme of catechol, but did not induce catechol 1, 2-dioxygenase. All the oxidative enzymes of biphenyl and 2, 3,-dihydroxybiphenyl (23DHBP) were induced when the cells were grown on biphenyl and salicylate, respectively. Biphenyl and salicylate could be a good inducer in the oxidation of biphenyl and 2, 3-dihydroxybiphenyl. The two enzymes for the degradation of biphenyl and salicylate were induced after growth on either biphenyl or salicylate, suggesting the presence of a common regulatory element. However, benzoate could not induce the enzymes responsible for the oxidation of these compounds. Biphenyl and salicylate were good inducers for indigo formation due to the activity of biphenyl dioxygenase. These results suggested that indole oxidation is a property of bacterial dioxygenase that form cis-dihydrodiols from aromatic hydrocarbon including biphenyl.
