Kinetic Properties of Manganese Peroxidase from the Mushroom Stereum ostrea and its Ability to Decolorize Dyes

  • Manganese peroxidase (MnP) was isolated from the culture filtrate of the wood log mushroom Stereum ostrea (S. ostrea), grown on Koroljova medium, and then purified by ammonium sulfate [70% (w/v)] fractionation, DEAE-cellulose anion exchange chromatography, and Sephadex G-100 column chromatography, with an attainment of 88.6-fold purification and the recovery of 22.8% of initial activity. According to SDS-PAGE the molecular mass of the MnP was 40 kDa. The optimal pH and temperature were found to be 4.5 and $35^{\circ}C$, respectively. The enzyme was stable even after exposure to a pH range of 4.5 to 6.0, and at temperatures of up to $35^{\circ}C$ at a pH of 4.5 for 1h. The $K_m$ and $V_{max}$ values for the substrate phenol red were found to be $8{\mu}m$ and 111.14 U/mg of protein, respectively. The MnP also oxidized other substrates such as guaiacol, DMP, and veratryl alcohol. Sodium azide, EDTA, SDS, $Cu^{2+}$, and $Fe^{2+}$, at 1-5 mM, strongly inhibited enzyme activity, whereas $Ca^{2+}$ and $Zn^{2+}$ increased enzyme activity. The participation of the purified enzyme in the decolorization of dyes suggests that S. ostrea manganese peroxidase could be effectively employed in textile industries.

폐정격(肺正格) 자침이 촌관척(寸關尺) 맥파 변화에 미치는 영향 (The Effect of Sa-Am Lung Tonifying Acupuncture on Radial Pulse in Healthy Human Subjects)

  • Objectives : The purpose of this study is to find the effects of Sa-Am lung tonifying acupuncture on radial pulse through the parameters difference. Methods : Forty healthy subjects participated in this study, acupuncture group and control group were divided. Compared various parameters of radial pulse between two groups, and the significantly changed 16 parameters ware measured using 3 dimensional pulse imaging system(DMP-3000) before, right after, 30 minutes after, and 60 minutes after acupuncture at Chon, Gwan, Cheok in acupucture group. Results : After lung tonifying acupuncture, radial pulse 16 parameters were showed changes significantly according to the time at each measuring location. 1. Heart rate, T2/T, T5, T5/T and W/T significantly decreased, W significantly increased. 2. Modulus of elasticity significantly decreased in left Cheok. 3. Amplitude of H4 significantly increased in left Gwan and right Cheok. 4. AIx, AIxHR significantly decreased in left Gwan and significantly increased in right Chon. 5. Area of pulse, area of W significantly increased in right Cheok. 6. Pulse energy, energy/min, EIx significantly decreased in left Cheok. 7. Maximum variation time of each parameter is showed different according to the parameter. Conclusions : Lung tonifying acupuncture exerts an influence on radial purse parameters. Further radial pulse change study on various acupuncture treatment is required.

  • 연구 데이터 관리(Research Data Management: RDM)는 연구데이터를 생산, 수집, 이용, 보전하는데 있어 방향을 제시하고 지원하는 인력, 정책, 자원 및 기술을 포괄하는 시스템이다. RDM은 연구비 신청시 작성하는 DMP(Data Management Plan)의 작성지원, 데이터 컬렉션과 리파지토리 구축, 연구 데이터의 디지털 보전과 유통 등을 포함하는 광범위한 활동들로 구성된다. 선진국의 경우 각 기관들이 RDM을 위한 시스템과 관련 조직을 구성하여 운영하고 있으나 우리나라의 경우에는 연구 데이터에 관한 인식수준이 낮아 미흡한 실정이다. 본 논문에서는 각 조직의 현실에 적합한 연구데이터 관리체계 구축방안을 제안한다. 특히, 최근들어 각 분야마다, 조직마다 빅데이터의 생성과 관리를 위한 빅데이터 플랫폼 구축이 급증하고 있어 이를 조직내 RDM 구축에 반영할 필요가 있다. 또한 블록체인 기술을 활용하여 연구자의 데이터 주권 확보를 지원하고, 데이터 프로비넌스 보장과 P2P 방식의 분산 RDM 구축 방안도 제안한다.

Transactivators for the Odontoblast-specific Gene Targeting

  • Dentin, a major component of teeth, is formed by odontoblasts which produce the dentin matrix beneath the dental epithelium and induce the mineralization of dentin. To date, the biochemical properties of dentin matrix proteins have been well characterized, but upstream regulators of these proteins are not yet well known. Recently in this regard, several transcription factors have been identified as potential regulators of matrix proteins. Most transcription factors are generally involved in diverse biological processes and it is essential to identify those that are odontoblast-specific transactivators to further understand the process of dentin formation. We thus analyzed the expression pattern of dentin matrix proteins and the activities of established transactivators containing a Cre-locus. Expression analyses using in situ hybridization showed that dentin matrix proteins are sequentially expressed in differentiating odontoblasts, including type-I collagen, Dmp-1 and Dspp. The activities of the transactivators were evaluated using ${\beta}$-galactosidase following the generation of double transgenic mice with each transactivator and the ROSA26R reporter line. The ${\beta}$-galactosidase activity of each transactivator paralled the expression of the matrix proteins. These results thus showed that these transactivators could be utilized for odontoblastspecific conditional gene targeting. In addition, time- and tissue-specific conditional gene targeting might also be achieved using a combination of these transactivators. Odontoblast-specific conditional gene targeting with these transactivators will likely also provide new insights into the molecular mechanisms underlying dentin formation.

Production of a Recombinant Laccase from Pichia pastoris and Biodegradation of Chlorpyrifos in a Laccase/Vanillin System

  • The recombinant strain P. pastoris GS115-lccC was used to produce laccase with high activity. Factors influencing laccase expression, such as pH, methanol concentration, copper concentration, peptone concentration, shaker rotate speed, and medium volume were investigated. Under the optimal conditions, laccase activity reached 12,344 U/L on day 15. The recombinant enzyme was purified by precipitating and dialyzing to electrophoretic homogeneity, and was estimated to have a molecular mass of about 58 kDa. When guaiacol was the substrate, the laccase showed the highest activity at pH 5.0 and was stable when the pH was 4.5~6.0. The optimal temperature for the laccase to oxidize guaiacol was $60^{\circ}C$, but it was not stable at high temperature. The enzyme could remain stable at $30^{\circ}C$ for 5 days. The recombinant laccase was used to degrade chlorpyrifos in several laccase/mediator systems. Among three synthetic mediators (ABTS, HBT, VA) and three natural mediators (vanillin, 2,6-DMP, and guaiacol), vanillin showed the most enhancement on degradation of chlorpyrifos. Both laccase and vanillin were responsible for the degradation of chlorpyrifos. A higher dosage of vanillin may promote a higher level of degradation of chlorpyrifos, and the 2-step addition of vanillin led to 98% chlorpyrifos degradation. The degradation of chlorpyrifos was faster in the L/V system ($k_{obs}$ = 0.151) than that in the buffer solution ($k_{obs}$ = 0.028).

동맥경화 평가를 위한 연령별 맥파 주요인자 분석 (Analysis of Pulse Wave Parameters According to Aging for Arteriosclerosis Evaluation)

  • Objectives : The aim of this study is to propose the W area of pulse (AW) as a new index which can confirm the arteriosclerosis by analyzing parameters of 5-level pressure pulse waveform measurement system for normotensive group according to aging. Methods : We measured radial pulse waveforms of normotensive group (20 to 60 years old) using 3-dimensional pulse imaging analyser (DMP-3000, DAEYOMEDI Co., Korea). And then we analyzed various parameters for sclerosis of the arteries such as Height (h1, h2, h3, h4, h5), Time (t1, t2, t3, t4, t5), AW, AW rate, Total area of pulse (At) and Augmentation Index (AIx). Results : As a result of analyzing parameters according to the aging, h2, h3, AS (systolic area rate to AT), AIx and AW were increased but t2/t, t3/t, t5/t and AD (diastolic area rate to AT) were decreased. Conclusions : We checked blood vessel conditions for normotensive group according to aging and confirmed various parameters. Also, we found that AW was analogous to AIx which has been used for diagnosing arteriosclerosis. Furthermore, we confirmed the usefulness of AW as a new parameter for checking vessel condition and characteristic compared with the AIx.

  • Objectives : The purpose of this study is to find the effects of Sa-am liver tonifying and sedating acupuncture on radial pulse. Methods : Sixty healthy people were divided into a liver tonifying acupuncture(LTA) group, liver sedating acupuncture(LSA) group, and control group. The LTA group and LSA group received acupuncture for 20 minutes with a supine position. The Control group took a rest without receiving acupuncture. Radial pulse was measured by three dimensional pulse imaging system(DMP-3000) at four different time points (before, right after, 30 minutes after, and 60 minutes after acupuncture). We compared the three groups and figure out determined the parameters which significantly changed after acupuncture treatment. Results : 1. Pulse period, T1/T, T4/T, (T-T4)/T, T4/(T-T4), T5/T significantly changed after acupuncture. 2. H4, pulse area, systolic pulse area, diastolic pulse area, radial augmentation index(RAI), and pulse power volume / min significantly changed after acupuncture. 3. Frequency of Fourier components, ratio of frequencies of Fourier components, magnitude of Fourier components, and ratio of magnitudes of Fourier components significantly changed after acupuncture. Conclusions : LTA and LSA have an effect on the radial pulse parameters. Further studies on radial pulse change using Sa-Am acupuncture are needed.

  • Objectives : The purpose of this study is to observe the effects of Sa-am acupuncture with tonifications of kidney and urinary bladder on radial pulse in healthy subjects. Methods : Sixty healthy subjects participated in this study, and were divided into a kidney tonifying acupuncture group(KI group), urinary bladder tonifying acupuncture group(BL group) and control group. Radial pulse was measured by 3 dimensional pulse imaging system(DMP-3000) before, immediately after, 30 minutes after, and 60 minutes after acupuncture at Cun, Guan, and Chi in each time. Results : 1. BL group exhibited significantly reduced T1 / T, T4 / T, T4 / (T-T4), T5 / T and increased(T-T4) / T compared to KI group. 2. BL group showed increase of radial augmentation index / heart rate(RAI / HR) in left Cun, H5, Ad / Ap in left Chi, H1, As / Ap in right Chi compared to KI group. BL group showed decrease of As / Ap, Aw / Ap in left Chi, H1, pulse depth, Ad / Ap in right Chi compared to KI group. 3. KI group showed increase of Aw / Ap, H1, H2, H4, pulse area in right Guan, pulse depth in right Chi compared to BL group. KI group showed decrease of RAI in left Cun, applied pressure, pulse depth in left Guan, Aw / Ap in right Chi compared to BL group. Conclusions : The effects of Sa-am acupuncture with tonifications of kidney and urinary bladder in healthy humans have been observed on various parameters. The parameters analysed in this study can be used to differentiate the effects of Sa-am acupuncture with tonifications of kidney and urinary bladder on radial pulse.

  • Objectives : The purpose of this study is to find parameters to compare the effects of Sa-Am acupuncture with large intestine tonifying and sedating on radial pulse in healthy subjects. Methods : Sixty healthy subjects participated in this study, divided into large intestine tonifying acupuncture group, large intestine sedating acupuncture group and control group. Radial pulse was measured by 3 dimensional pulse imaging system(DMP-3000) before, right after, 30 minutes after and 60 minutes after acupuncture at Cun, Guan, and Chi in the acupuncture group and in the control group at the same time points. Results : 1. Angle of main peak and magnitude of fourier component significantly changed by integrated analysis of Cun, Guan and Chi. 2. Amplitude of H1, Pulse Power volume / min, elasticity, AIx / HR, Frequency and magnitude of fourier component significantly changed by analysis of Cun, Guan, and Chi. Conclusions : The effect of Sa-Am acupuncture with large intestine tonifying and sedating in healthy human may be observed on time, amplitude, pulse area, augmentation index and fourier components parameters. The parameters analysed in this study may be used to differentiate the effects between Sa-Am acupuncture with large intestine tonifying and sedating on radial pulse. Further studies on the effects of Sa-Am acupuncture using radial pulse are needed.

  • Objectives : The purpose of this study is to investigate the effects of Sa-Am spleen-tonifying acupuncture on radial pulse in healthy human subjects. Methods : Forty healthy human subjects participated in this study, divided into acupuncture group and control group. Radial pulse was measured by 3 dimensional pulse imaging system(DMP-3000) before, right after, 30 minutes after and 60 minutes after acupuncture in the acupuncture group. The subjects in the control group didn't received acupuncture but took a rest and then the radial pulse was measured at the same time points as the acupuncture group. The parameters were analyzed by gender, measuring location, and measuring time point. However the time related parameters exceptionally were analyzed without distinction of measuring location. Results : 1. T, variance of period, T2/T, T4/T, (T-T4)/T, T4/(T-T4), and W significantly changed after acupuncture. 2. Pressure, amplitude of H1, amplitude of H2, Amplitude of H4 and pulse energy significantly changed after acupuncture. 3. Pulse area, systolic pulse area and diastolic pulse area significantly changed after acupuncture. 4. Elasticity, AIx and AIx/HR significantly changed after acupuncture. Conclusions : The effect of Sa-Am spleen-tonifying acupuncture in healthy human may be observed on time, amplitude, pulse area and augmentation index. Further studies on the effects of Sa-Am acupuncture using radial pulse are needed.
