Growth Characters and Sugar Content During Grain Filling in New Hybrid, Chalok 1/Cooktail 51 Corn

  • 'Chalok 1/Cooktail 51' corns, supersweet corn gene controlled by either brittle-l (bt l) or shrunken-2(sh2) gene introduced into waxy corn, showed agronomic characteristics between supersweet corn and waxy corn. The ears were harvested at different development stages from 15 to 35 days after silking (DAS). Ear diameter of Cocktail 51 and 'Cocktail 51'/Chalok 1 increased from 15 DAS to 30 DAS and little increased thereafter, but that of Chalok 1/Cocktail 51 and 'Chalok l' increased until 35 DAS. Diameter of ear extension increased more in Cocktail 51 and Chalok 1/Cocktail 51 corn than Chalok 1. Ear fresh weight of Cocktail 51 decreased later 30 DAS but those of the other hybrids were vice versa. Rate of super-sweet kernels per ear of Chalok 1/Cocktail 51 corns was about 38 %. Development, and elongation of kernel were much more prominent in supersweet kernel than in waxy kernel, but fresh weight increased higher in waxy kernel than supersweet kernel. Moisture content in kernel decreased from 15 DAS to 35 DAS. Total sugar content of the kernel increased until 25 DAS, and that of Cocktail 51 kernel showed the highest among of them. After cooked by steam, flavor and mastication feeling rate cooked by steam of Cocktail 51 and Chalok 1/Cocktail 51 were increased from 15 DAS to 25 DAS and markedly decreased thereafter. But those of Chalok l/Cocktail 51 and, Chalok 1 were decreased after 30 DAS. These results suggested that the optimum harvest date for fresh supersweet corn (Cocktail 51), Cocktail 51/Chalok 1 seems to be about 20 DAS and Chalok 1/Cocktail 51 and waxy com (Chalok 1) was about 25 DAS.

Numerical Simulation of Fluorescent Whitening Agents (DAS1) in Tokyo Bay (동경만 형광표백제 성분물질(DAS1)의 거동 특성 시뮬레이션)

  • A three-dimensional ecological model(EMT-3D) was applied to DAS1 in Tokyo Bay. The simulated results of DAS1 were in good agreement with the observed values. The result of sensitivity analysis showed that photolysis coefficient and extinction coefficient were important factor for dissolved DAS1, and photolysis coefficient, extinction and POC partition coefficient for PAHs in particulate organic matter. Mass balance of DAS1 in Tokyo Bay was calculated by using the simulated results of EMT-3D.

A Study on the Proposal of DAS Template for Sharing Research Data in Domestic Academic Societies: Focusing on Data Sharing Policy of Foreign Publishers (국내 학회의 연구데이터 공유를 위한 DAS 템플릿 제안에 관한 연구 - 해외 출판사 데이터 공유 정책을 중심으로 -)

  • As sharing of research data has become essential, overseas publishers are providing authors with DAS templates to facilitate data sharing. However, the domestic academic market is still lacking in policies related to such research. This study proposes a data sharing policy and DAS template that can be applied to domestic academic societies in order to activate data sharing. To this end, data sharing policies of 5 foreign publishers including Nature and 12 DAS templates including AMS were investigated and analyzed. As a result of the study, an overview of the data sharing policy applicable to domestic academic conferences and a DAS template consisting of 12 items were derived. It is judged that the data sharing policy and DAS template to be presented as a result of this study can be a practical guide and tool for data sharing.

Common dental anomalies in Korean orthodontic patients: An update

  • Objective: The aim of this study was 1) to investigate the prevalence and pattern of dental anomalies (DAs), 2) to compare DAs according to the type of malocclusion, and 3) to investigate the correlation between tooth impaction and other DAs in the Korean orthodontic population. Methods: A total of 3,240 orthodontic patients were classified as Class I, Class II, or Class III malocclusion groups. The presence and location of common DAs, including impaction, microdontia, agenesis, supernumerary tooth, transposition, and fusion, were identified by examining diagnostic records. Furthermore, samples were classified as Group 1 without impaction or Group 2 with impaction. The prevalence of other DAs concurrent with impaction was investigated and compared to Group 1. Results: Impaction was the most prevalent DA, followed by microdontia, agenesis, and supernumerary. Class I and Class III groups showed the same order of prevalence, but agenesis was more frequent than microdontia in the Class II group. The prevalence of the four DAs was lowest in the Class III group. Overall, 8.6% of patients were classified into Group 2. The incidence of DAs other than impaction and the prevalence of multiple concurrent DAs were significantly higher in Group 2. Impaction showed a significant relationship with supernumerary tooth, transposition, and fusion. Conclusions: The prevalence and pattern of DAs varied depending on the type of malocclusion. As there was a higher risk of other DAs in patients with impacted teeth, early detection of the impacted tooth and a detailed diagnosis of other possible DAs may be essential.

Biosynthesis and Metabolism of Vitamin C in Suspension Cultures of Scutellaria baicalensis

  • The concentrations of L-ascorbic acid (AsA, ascorbate, vitamin C) and its biosynthetic and metabolically-related enzymes such as L-galactono-1,4-lactone dehydrogenase (GLDase), ascorbate peroxidase (APX), and ascorbate oxidase (ASO) were investigated in suspension cultures of Scutellaria baicalensis. Cells growing from 4 days after subculture (DAS) to 9 DAS and from 16 DAS to 19 DAS showed a diauxic growth, and then growth rapidly decreased with further culturing. The AsA content slowly increased to 19 DAS, reached a maximum at 21 DAS (ca $120\;{\mu}g/g$ dry cell wt), and then rapidly decreased with further culturing. GLDase and ASO activity were well correlated with the cell growth curve, showing a maximum at 19 DAS, whereas APX activity showed a good correlation with the changes in AsA content, showing a maximum at 21 DAS. The total ascorbate contents (reduced form, AsA, and oxidized form, dehydroascorbate, DHA) were markedly enhanced at 10 DAS when L-galactose and L-galactono-1,4-lactone (25 mM) were added to SH medium supplemented with 20 g/l sucrose at 9 DAS, by 5.5 and 6.8 times, respectively. DHA composed more than 90% of the total ascorbate contents in suspension cultures of S. baicalensis, even though the ratio of reduced to oxidized form slightly varied with cell growth stage. The results indicate that L-galactose and L-galactono-1,4-lactone are effective precursors of AsA in cell cultures of S. baicalensis, and that in vitro cultured cells provide suitable biomaterials for the study of biosynthesis and metabolism of AsA.

Maturity Effects on Moisture, Total Sugar Contents and Flavor of Fresh Waxy Corn (성숙정도에 따른 풋찰옥수수의 수분, 전당함량 및 맛의 변화)

  • The changes of kernel size, contents of moisture and total sugar, and rates of flavor and stickiness of a waxy corn (cv. Hongcheon native) were observed from 15 days after silking (DAS) to 40 DAS at Suwon and Cheongju in 1984, nespectively. Fresh kernel length and width greatly increased from 15 DAS to 30 DAS and slightly increased thereafter. Fresh 100-kernel weight was markedly increased during 15 DAS to 30 DAS, and slightly and continuously increased thereafter while dry 100-kernel weight almost linearly increased with maturity. Total sugar content of fresh kernel was the highest at 20 DAS and then continuously decreased with maturity. Flavor and stickiness rates of boiled waxy corn were significantly increased during 15 DAS to 30 DAS and flavor rate decreased thereafter. However, stickiness rate maintained the level until 35 DAS and then decreased slightly. The optimum harvest date for fresh waxy corn seems to be about 30 DAS considering kernel size, and flavor and stickiness rates of boiled corn.

A time-course study of flavonoids in buckwheats (Fagopyrum species) (메밀(Fagopyrum species)의 생장에 따른 플라보노이드 함량의 품종별 차이)

  • Flavonoid contents of common buckwheat (cv. Kitawase) and tartary buckwheat (cv. Hokkai T 8, Hokkai T 9 and Hokkai T 10) were determined by high-performance liquid chromatography (HPLC). Moreover, they were measured at different plant developments such as 10, 18, 20, 22 and 30 days after sowing (DAS) and with plant parts including leaf, stem and flower harvested at 30 DAS. Total flavonoids including chlorogenic acid, four kinds of C-glycosylflavones (orientin, isoorientin, vitexin, isovitexin) and rutin of tartary buckwheats (range of 44.2-54.7, mean 44.2) were found 35% higher than those of common buckwheat (28.9 mg/g dry wt.). Among them, rutin was measured above 80% of total flavonoid contents. The other flavonoids (chlorogenic acid and four kinds of C-glycosylflavones) presented the highest level at 10 DAS and decreased according to plant developments. On the other hand, rutin content of Kitawase presented the highest level (33.6 mg/g dry wt.) at 22 DAS and decreased up to 30 DAS. Rutin content in tartary buckwheat temporarily decreased from 10 to 18 DAS and then reversely increased up to 30 DAS presented the highest level as 'U' curve. In Hokkai T 10, rutin content was found the highest level (53.8 mg/g dry wt.) at 30 DAS. In different plant parts harvested at 30 DAS, rutin content of leaf (range of 42.8-68.0, mean 57.0) was 5.3-fold higher than that of stem (range of 8.0-15.9, 10.8 mg/g dry wt.), regardless of cultivar. Significantly, rutin content (78.7) in the flower of Kitawase was 1.8 times higher than in the leaf and 9.8 times in the stem. Especially, chlorogenic acid content (14.6 mg/g dry wt.) in the flower of Kitawase was 63-fold higher than in the leaf, and 20-fold in the stem.

Multiplex PCR Detection for 3 Events of Genetically Modified Maize, DAS-59122-7, TC6275, and MIR604

  • A multiplex polymerase chain reaction (PCR) method was developed to simultaneously detect 3 events of genetically modified (GM) maize. The event-specific primers were used to discriminate the following 3 events of GM maize (DAS-59122-7, TC6275, and MIR604) using multiplex PCR method. The zein gene was used as an endogenous maize reference gene in the multiplex PCR detection. The primer pair Zein-FIR producing a 99 bp amplicon was used to amplify the zein gene. The primer JI-Das-F1/R1 for DAS-59122-7, JI-TC6275-F3/R3 for TC6275, and JI-MIR F1/R1 for MIR604 yielded an amplicon of 130, 162, and 197 bp, respectively. The detection limit of multiplex PCR was 1% for DAS-59122-7, TC6275, and MIR604 for one reaction.

  • In this paper we prepared dextrin dialdehyde (DAS) of a different oxidation ratio (aldehyde content) and examined their properties and discussed their tanning mechanism. DAS was produced by reacting dextrin in aqueous medium with periodate ion used in a molar ratio between periodate and glucose of 0.3 to 1.0 at a temperature of 20 to $45^{\circ}C$. Since DAS reacts with amino group, DAS may have tanning property of aldehyde tanning agent (formaldehyde, glutaraldehyde).
