Urinary Estrone Sulfate for Monitoring Pregnancy of Dairy Cows

  • The purpose of this study firstly was conducted to establish a radioimmunoassay (RIA) of estrone sulfate ($E_1S$), secondly to monitor the reproductive status of dairy cows using their urine samples. Urine and blood samples were collected in series within a day from four pregnant Holstein-friesian cows to evaluate the relationship between $E_1S$ levels in blood and urine with or without urinary creatinine basis. The urine was then collected biweekly from three cows in estrous and those artificially inseminated; collection from pregnant cows was made on a monthly basis. Results indicated that sensitivity for the $E_1S$ RIA was 5 pg/tube and the recovery rate was 100%. The daily urinary creatinine concentrations fluctuated within a day, but changes were slighter in midday, whereas the changes of concentrations of $E_1S$ in urine were relatively smaller. The concentrations of serum $E_1S$ during the estrous cycle were undetectable due to the limitation of assay, but the urinary $E_1S$ level could be measured with no obvious changes during the cycle. The urinary $E_1S$ levels increased remarkably around 7.7 to 8.3 ng/ml, 80 to 100 days after pregnancy but the serum $E_1S$ levels did not elevate until 120 to 150 days. The level of $E_1S$ increased gradually during pregnancy and eventually reached its peak before parturition at around 40 ng/ml and finally decreased to its basal level 2 days postparturition. During pregnancy, $E_1S$ concentrations of urine increased earlier than those in blood. The correlation coefficients between urinary and serum $E_1S$ concentration during pregnancy and postparturm were higher than those adjusted with creatinine (creatinine ratio). The concentrations of $E_1S$ in urine could be maintained unchanged for 8 days storing the samples in room temperature, which was extended to 8 days when the samples were pretreated by boiling for 30 minutes or treated with autoclave. In conclusion urinary $E_1S$ concentrations can be used directly for monitoring the pregnant status and fetal viability of dairy cows and can assist accurate confirmation of pregnancy in cows at least 80 to 100 days after insemination much earlier than by serum $E_1S$.

Study on the Plant Growth Variation According to Change of Luminous Flux LED Light in Plant Factory (식물공장의 LED 광강도 변화에 따른 식물성장 변화에 관한 연구)

  • The purpose of this study is to change plant growth according to the light quantity of LED light source and to standardize plant factories technology according to the optimal LED light range. Growth was relatively stable at the 1,400 lx and 1,600 lx intensities, and growth continued with increasing light intensity, but growth slowed down with growth without leaves (Ed- I don't understand). The growth characteristics at 400 lx to 800 lx were sustained to some extent during the experimental period, but the growth of the larvae was maintained until the 7th day and the growth of leaves was not developed. From 1,000 lx to 1,400 lx, brightness, leaf growth and daily growth increased, and the growth cycle exhibited a repeating cycle of growth and slowdown. In addition, 1,600 lx and 1,800 lx showed similar growth to that at 1,400 lx, but leaf growth was high. However, at 1,800 lx, the growth was slightly higher than at 1,400lx and it slowed down over time. In addition, the stronger the light, the higher the temperature around the plant and the greater the dryness of the leaves due to the heat emitted from the light source.

Comparison of Pregnancy Rates by Intrauterine Insemination after Ovulation Trigger with Endogenous LH Surge, GnRH Agonist or hCG in Stimulated Cycles (인공수정시술주기에서 내인성 LH, GnRH Agonist 및 hCG를 이용한 배란유발후 임신율의 비교연구)

  • Objective: This study was designed to evaluate the effects of endogenous LH surge, GnRH agonist (GnRH-a) or human chorionic gonadotropin (hCG) as ovulation trigger on pregnancy rate by intrauterine insemination (IUI). Method: Patients received daily 100 mg of clomiphene citrate (CC) for 5 days starting on the third day of the menstrual cycle followed by human menopausal gonadotropin (hMG) for ovulation induction. Follicles larger than >16 mm in diameter were present in the ovary, frequent LH tests in urine were introduced to detect an endogenous LH surge. Final follicular maturation and ovulation were induced by GnRH-a 0.1 mg (s.c.) or hCG $5,000{\sim}10,000$ IU (i.m.) administration except natural ovulation. Pregnancy was classified as clinical if a gestational sac or fetal cardiac activity was seen on ultrasound. Results: There were no differences in age, duration of infertility and follicle size, but more ampules of hMG were used in GnRH-a group compared to hCG 10,000 IU treated group (p<0.05). Lower level of estradiol ($E_2$) on the day of hCG or GnRH-a injection was observed in hCG 10,000 IU group than other treatment groups (p<0.01). The overall clinical pregnancy rate was 19.8% per cycle (32/162) and 22.2% per patient (32/144). Pregnancy rate was higher in natural-endogenous LH surge group (37.5%, 9/24) than GnRH-a (18.8%) or hCG treated group (20.9% & 13.9%), but this difference was not statistically significant. No patient developed ovarian hyperstimulation. Abortion rate was 22.2% (2/9) in hCG 5,000 IU group. Delivery or ongoing pregnancy rate was 37.5% (9/24), 18.8% (3/16), 16.3% (7/43) and 13.9% (11/79) in endogenous LH surge, GnRH-a, hCG 5,000 IU and hCG 10,000 IU treatment groups, respectively. Conclusion: These results support the concept that use of natural-endogenous LH surge in stimulated cycles may be more effective to obtain pregnancies by IUI than GnRH-a or hCG administration.

Assessment of Climate Change Impact on Imha-Dam Watershed Hydrologic Cycle under RCP Scenarios (RCP 기후변화 시나리오에 따른 임하댐 유역의 미래 수문순환 전망)

  • This study was to evaluate the RCP climate change impact on hydrological components in the Imha-Dam watershed using SWAT(Soil and Water Assessment Tool) Model. The model was calibrated for six year(2002~2007) and validated for six year(2008~2013) using daily observed streamflow data at three watershed stations. The overall simulation results for the total released volume at this point appear reasonable by showing that coefficient of determination($R^2$) were 0.70~0.85 and Nash-Sutcliffe model efficiency(NSE) were 0.67-0.82 for streamflow, respectively. For future hydrologic evaluation, the HadGEM3-RA climate data by scenarios of Representative Concentration Pathway(RCP) 4.5 and 8.5 of the Korea Meteorological Administration were adopted. The biased future data were corrected using 34 years(1980~2013, baseline period) of weather data. Precipitation and temperature showed increase of 10.8% and 4.9%, respectively based on the baseline data. The impacts of future climate change on the evapotranspiration, soil moisture, surface runoff, lateral flow, return flow and streamflow showed changes of +11.2%, +1.9%, +10.0%, +12.1%, +18.2%, and +11.2%, respectively.

  • 유역 물순환 개선 기술은 기후변화 및 토지이용변화가 진행 중에 있거나 예상되는 지역에 대하여 강우-유출수를 지연, 저류, 침투시켜 지속가능한 물순환체계를 유지?회복하도록 하는 기술이라 할 수 있다. 기후변화 및 토지이용변화에 따른 영향을 평가하고, 지속가능한 유역 물순환체계를 구축하여 적응 전략을 체계적으로 수립하기 위해서는 물순환 개선 기술의 정립 및 개발이 필수적이다. 그러나 현재 유역 물순환 개선기술은 일부 시가화 지역에 국한되어 있어 유역 규모의 개선 전략을 수립하는데 한계가 있다. 또한, 물순환계의 변화량을 평가하기 위한 모형화 기법 역시 해외에서 개발된 기술을 여과 없이 도입하여 국내의 환경을 충분히 반영하기는 어렵다. 개발된 유역 물순환 개선 및 평가시스템은 기존 국가연구개발사업을 통해 개발되고 사업화에 성공한 바 있는 유역 물순환 평가 모형인 CAT(Catchment hydrologic cycle Assessment Tool)을 수정 및 개선하여 기후변화에 따른 영향을 평가하고 적응 대책을 수립하기 위한 실무적인 소프트웨어이다. 침투트렌치, 식생침투트렌치, 습지, 저류지, 빗물탱크 등의 물순환개선시설에 대한 효과를 평가할 수 있도록 개별시설의 제원에 따른 물순환개선 효과를 정량적으로 평가하여 제시한다. 기후변화에 따른 장기간의 유역 물순환을 평가하기 위해서는 물리적 매개변수 기반의 수문해석 보다는 단순환된 개념적 매개변수 기반의 집중형 장기유출 해석이 필요할 수 있다. 따라서 국내외에서 많이 사용되고 있는 장기 일유출 모형(GR4J, GSM, HBV, SYMHYD, TANK, TPHM 등)을 유역 물순환 개선 평가 플랫폼에 탑재함으로써 소유역의 특성을 반영한 기후변화 적응 일유출 해석이 가능하도록 하였으며, 각 모형들의 매개변수는 수동보정 외에도 SCE-UA를 이용한 자동보정이 가능하도록 시스템으로 구축하였다. 개발된 유역물순환 개선 및 평가시스템은 실무적 차원에서 기후변화에 따른 유역 물순환 개선 기술을 적용하고 평가하는데 있어서 매개변수 입력자료 구축에 따른 자원 소요 시간 및 시스템 개발 비용을 획기적으로 단축시켰으며 국외 의존도가 높은 수문 해석모형을 국내 기술로 개발함으로써 기술자립도를 높이고 국내 및 해외의 유역의 성공적인 적용을 통하여 성능을 입증하였다. 기후변화에 따른 수자원의 재평가, 개선시설의 정량적 평가 및 하천유역의 수자원관리 실무에 적용성이 높을 것으로 기대된다.

Metabolic Rate and Thermolabile Properties of Ognev's Great Tube-nosed Bat Murina leucogaster in Response to Variable Ambient Temperature

  • The winter-resident Korean bats, Murina leucogaster ognevi, show a circadian cycle of thermoregulation and locomotion in summer, as do other bat species in temperate regions. They are most active between dusk and dawn with body temperature (Tb) of 35-4OC, and are usually torpid in their roost sites for the rest of day with their Tb close to ambient temperature (Ta) of around 15C. The present study was conducted to determine thermogenic and thermolabile properties of the heterothermic bats that would influence their daily feeding activities and ultimately, their energy conservation strategy. Testing on active male Murina, resting metabolic rate (RMR, gauged by oxygen consumption rate) at the lower limit of thermoneutral zone (31C) was 2.0 L kq-1 h-1. The regression slope of RMR below the thermoneutral zone (an index of metabolic thermal sensitivity) was -0.38 L $kg^{-l} h^{-1} C^{-1}$. The metabolic rate at the roost Ta (15C) was 4.5 times the lowest RMR in the active state but becomes nearly zero in the torpid state. This implies that by being torpid during daytime (between dawn and dusk), the individual bats would save about 4.7 kcal each day in mid-summer. Interspecific comparisons of thermal metabolic response over a mass scale suggest that the smaller bats show a relatively higher metabolic rate in thermoneutral zone and a greater thermal sensitivity of metabolism, which follows the general principle seen in homeothermic metabolism. Thermolabile features in metabolic responses seem to be fairly common for these bats in conditions other than a fully active state. Types of thermolabile responses and their energetic significance are discussed.

Effects of ANTORIN R-10 on Ovarian Morphology, Follicular Development and Serum Estradiol Level in Immature Wistar Rats (미성숙 Wistar 랫트에서 ANTORIN R-10이 난소형태, 난포발육 및 혈중 호르몬 농도에 미치는 영향)

  • This study evaluated the effect of ANTORIN R-10(pFSH), a commercially available follicle stimulating hormone on ovarian morphology, on follicular development and serum estradiol levels in rats. Immature female Wistar S/T rats(27 day old; 80-100 g B.wt) maintained under controlled environmental conditions($22{\pm}2^{\circ}C$; 50% humidity; 12 h light/12 h dark cycle) with free access to standard laboratory feed and tap water were utilized. Animals were allowed to acclimatize to the new environment for at least 2 weeks before being included in the experiment. Rats were randomly allotted to 5 groups(Control, SL 0.1AU, SH 0.2AU, TL 0.1AU and TH 0.2AU). ANTORIN R-10 was subcutaneously injected twice daily for 3 days. Twenty hours after hormone treatment, blood was collected to estimate the serum estradiol $17-\beta$ concentration. Immediately, all rats were sacrificed and the ovarian morphology, ovary weight and number of follicles were recorded. Ovaries were fixed for histomorphological examination. Higher standard and treatment groups were significantly increased on ovary weight and the number of follicles more than 1mm compared with lower standard and treatment. However, no difference revealed between standard and treatment groups. ANTORIN R-10 was similar effects of follicles development and maturation compared with House standard FSH.

MicroRNAs in Human Diseases: From Cancer to Cardiovascular Disease

  • The great discovery of microRNAs (miRNAs) has revolutionized current cell biology and medical science. miRNAs are small conserved non-coding RNA molecules that post-transcriptionally regulate gene expression by targeting the 3' untranslated region of specific messenger RNAs for degradation or translational repression. New members of the miRNA family are being discovered on a daily basis and emerging evidence has demonstrated that miRNAs play a major role in a wide range of developmental process including cell proliferation, cell cycle, cell differentiation, metabolism, apoptosis, developmental timing, neuronal cell fate, neuronal gene expression, brain morphogenesis, muscle differentiation and stem cell division. Moreover, a large number of studies have reported links between alterations of miRNA homeostasis and pathological conditions such as cancer, psychiatric and neurological diseases, cardiovascular disease, and autoimmune disease. Interestingly, in addition, miRNA deficiencies or excesses have been correlated with a number of clinically important diseases ranging from cancer to myocardial infarction. miRNAs can repress the gene translation of hundreds of their targets and are therefore well-positioned to target a multitude of cellular mechanisms. As a consequence of extensive participation in normal functions, it is quite logical to ask the question if abnormalities in miRNAs should have importance in human diseases. Great discoveries and rapid progress in the past few years on miRNAs provide the hope that miRNAs will in the near future have a great potential in the diagnosis and treatment of many diseases. Currently, an explosive literature has focussed on the role of miRNA in human cancer and cardiovascular disease. In this review, I briefly summarize the explosive current studies about involvement of miRNA in various human cancers and cardiovascular disease.

Study of Recognition and Necessity of Oriental Medicinal Menu, and Development of Oriental Therapeutic Menu for Stroke Patients (뇌졸중 환자에서 한방치료식의 인지도 및 요구도 조사와 문헌고찰을 통한 한방 치료식단(藥膳) 개발)

  • The purpose of this study was to develop the oriental therapeutic menu(Yak-sun) through the recognition and necessity of oriental medicine diet in stroke patients admitted to K Oriental Medical Hospital. From May to July 2007, the 73 stroke patients were surveyed for the recognition and necessity of oriental therapeutic menu. The oriental therapeutic menu was developed on the basis of literature of oriental medicine books and research papers. The developed two week cycle of menus were then reviewed by the experts and specialists. The sensory evaluation and nutritional analysis of the menu were followed. The stroke patients who participated in the survey did not know about the oriental medical diet, however, 61.6% of the patients had interest. Most of the patients expected the efficacy of oriental therapeutic diets. Total of forty three medical herbs and ten herb-combined prescriptions were used in the menu. The daily average calorie of developed menu was 2,031 $\pm$ 57.6 kcal and the most of nutrients met Dietary Reference Intakes for Korean(KDRIs). The caloric ratios of carbohydrates, protein, and lipid were 61.3%, 18.7%, 20.0%, respectively. The average values of cholesterol(194 $\pm$ 57.5 mg/day), sodium(3,078 $\pm$ 678.5 mg/day), calcium(1,099 $\pm$ 140.1 mg/day), and dietary fiber(33 $\pm$ 7.0 g/day) were met Korean dietary guide line for stroke patients. Oriental therapeutic diet might be unfamiliar to the hospital patients even though they had interest in and favorable thought about it. Implements of oriental therapeutic diet to the hospital patients might be useful to improve the quality of service in the hospitals.

Studies on the Superovulation and Collection of microinjectable Embryos in Korean Native Goats (Capra hircus aegagrus) (한국 재래산양에서의 과배란유기와 외래유전자 주입에 적합한 수정란의 회수에 관한 연구)

  • This study was carried out to determine the hormone treatment scheme for an efficient superovulation and optimal recovery time for obtaining pronuclear embryos suitable for DNA injection in Korean native goats. For a superovulation, FSH(5.6mg) was given over four days in twice daily injections with (FSH/hCG group) or without(FSH group) hCG(100 IU) co-injection at the time of 7th FSH injection. Estrus cycle was synchronized by norgestomet ear-implantation for 11 days and its removal at the time of 6th FSH injection. Among the treated goats, the percentage of ovulated goats, which were examined at 70 to 76 h following implant removal, was greater in FSH/hCG group than in FSH group (100% vs 36.4%) but there was no significant difference in the mean numbers of ovulation points and fertilization rates between the two groups. To optimize hCG treatment scheme and recovery time, we injected hCG at the time of 7th (FSH/hCGa) or 8th(FSH/hCGb) FSH injection and then examined the developmental stage of the embryos recovered at different times after implant removal. In FSH/hCGa group, significant portions(31 to 44%) were beyond 1-cell stage, which was non-injectable, irrespective of their recovery time. However, in FSH/hCGb group recovered at 70 to 76 h after implant removal, great portions(69%) were fertilized and most of them(96.6%) were injectable 1-cell stage. Considering together the fertilization rate and developmental stage of recovered embryos, it is recommendable to administrate hCG at the time of final 8th FSH injection and collect the embryos at 70 to 76 h after implant removal to obtain injectable embryos as many as possible in Korean native goats.
