The Galactose-${\alpha}1$,3-galactose (${\alpha}1$,3Gal) epitope is responsible for hyperacute rejection in pig-to-human xenotransplantation. Human decay-accelerating factor (hDAF) is a cell surface regulatory protein that serves as a complement inhibitor to protect self cells from complement attack. The generation of ${\alpha}1$,3-galactosyltransferase (GGTA1) knock-out pigs expressing DAF is a necessary step for their use as organ donors for humans. In this study, we established GGTA1 knock-out cell lines expressing DAF from pig ear fibroblasts for somatic cell nuclear transfer. hDAF expression was detected in hDAF knock-in heterozygous cells, but not in normal pig cells. Expression of the GGTA1 gene was lower in the knock-in heterozygous cell line compared to the normal pig cell. Knock-in heterozygous cells afforded more effective protection against cytotoxicity with human serum than with GGTA1 knock-out heterozygous and control cells. These cell lines may be used in the production of GGTA1 knock-out and DAF expression pigs for xenotransplantation.
In this study, we divided the process operation scenarios into three categories based on raw water temperature and turbidity. We will select and operate the process operation scenario according to the characteristics of the raw water. The number of algae in the DAF treated water has been analyzed to be less than 100 cells/mL. These results indicated that the DAF process is effective in removing the algae. In addition, the scenario of the integrated management decision algorithm of the DAF process was developed. DAF pilot plants ($500m^3/day$) process has shown a constantly sound performance for the treatment of raw water, yielding a significantly low level of turbidity (DAF treated water, 0.21~1.56 NTU).
Dissolved air flotation (DAF) is an effective solid/liquid separation process for low density floc particles such as algal, color-alum and clay-alum flocs produced from low turbidity water. The removal of taste and odor-causing organics (2-mthylisoboneol and geosmin) originating from algae in drinking water is a local and worldwide concern. Although DAF has been effectively applied for the removal of suspended solid, its application for the treatment of dissolved organic carbon is very limited. In this study, a new hybrid system consisting of adsorption and DAF processes was introduced for the simultaneous removal of algae and taste and odor-causing organics. Powdered activated carbon (PAC) was used as an adsorbent. In this proposed system, the major concern of eliminating the spent PAC from the system was also addressed. It was found that zeta potential of algae and PAC was increased with coagulant dosage, and the removal efficiency in DAF was also enhanced up to 90~95% under the given experimental conditions. Based on this study, the hybrid process was found to be a promising technology for the simultaneous removal of algae and dissolved organic pollutants.

In this study, a lab-scale test was performed to apply the ozone process in drinking water treatment plant using dissolved ai, flotation(DAF). The kinetic study on the ozone decay and hydroxyl(OH) radical formation was investigated by several parameters, such as I.D(Instantaneous ozone demand), $k_c$(ozone decomposition rate), ozone-Ct and OH radical-Ct. Ozonation of several target waters, such as raw water, DAF treated water and filtrate, was conducted to select the optimum position and dosage of ozone process. The highest value of Ozone-Ct and OH radical-Ct was observed at DAF treated water at initial run time($0{\sim}30\;min$). From the results of ozonation, the intermediate ozonation was proposed as the optimum position and the effective dose of ozone was determined to be $1{\sim}2\;mg/L$.

The demand for smaller, faster, and multi-functional mobile devices in increasing at a rapidly increasing rate. In response to these trends, Stacked Chip Scale Package (SCSP) is used widely in the assembly industry. A film type adhesive called die attach film (DAF) is used widely for bonding chips in SCSP. The DAF requires high flowability at high die attachment temperatures for bonding chips on organic substrates, where the DAF needs to feel the gap depth, or for bonding the same sized dies, where the DAF needs to penetrate bonding wires. In this study, the mixture design of experiment (DOE) was performed for three raw materials to obtain the optimized DAF recipe for low elastic modulus at high temperature. Three components are acrylic polymer (SG-P3) and two solid epoxy resins (YD011 and YDCN500-1P) with different softening points. According to the DOE results, the elastic modulus at high temperature was influenced greatly by SG-P3. The elastic modulus at $100^{\circ}C$ decreased from 1.0 MPa to 0.2 MPa as the amount of SG-P3 was decreased by 20%. In contrast, the elastic modulus at room temperature was dominated by YD011, an epoxy with a higher softening point. The optimized DAF recipe showed approximately 98.4% pickup performance when a UV dicing tape was used. A DAF crack that occurred in curing was effectively suppressed through optimization of the cure accelerator amount and two-step cure schedule. The imizadole type accelerator showed better performance than the amine type accelerator.
The main advantage of dissolved air flotation (DAF) in water treatment process is the small dimension compared with conventional gravity sedimentation and it can be basically reduced by the separation zone performed with the short solid-liquid separation time. Fine bubbles make such a short time possible to carry out solid from liquid separation as a collector on the course of water treatment. Therefore, the dimension of separation zone in DAF process is practically determined by the rise velocity of the bubble-floc agglomerates, which is a floc attached with several bubbles. To improve flotation velocity and particle removal efficiency in DAF process, many researchers have tried to attach bubbles as much as possible to flocs. Therefore, the maximum number of attached bubble on a floc and the rise velocity of bubble-floc agglomerates considered as the most important factor to design the separation zone of flotation tank in DAF process was simulated based on the population balance theory. According to the simulation results of this study, the size and volume concentration of bubble influenced on the possible number of attached bubble on a floc. The agglomerates attached with smaller bubble was more sensitive to hydraulic loading rate in the separation zone of DAF process. For the design of a high rate DAF process applied over surface loading 40 m/hr. it is required a precise further study on the variation of bubble property and behavior including in terms of bubble size distribution.
The DOF (Dissolved Ozone Flotation) system was used to treat the effluent of the secondary wastewater treatment plant. The DOF system uses ozone instead of air, while DAF (Dissolved Air Flotation) uses air. Moreover, since the solubility of ozone is higher than air, the DOF system produces larger volume of micro-bubbles than the DAF system does. Thus, the DOF system performs better than the DAF system in floating ability. The DOF system could remove 70% of turbidity to an average of 0.59NTU in effluent from 2.31NTU in influent. The removal efficiency of absorbance measured with UV-254 in the effluent of the DOF system was 63%, while only 19% was removed by the DAF system. the DOF system removed 84% of the color from 25~26CU to 4CU, while DAF system removed 42% of the color to 15 CU. The CODMn removal efficiency of the DOF system was 34%, 6.8mg/l of effluent $COD_{Mn}$ concentratin, while it was 20%, 8.3mg/L of effluent $COD_{Mn}$ concentratin, to use the DAF system. Microbial bacteria such as coliform bacteria, and heterotrophic bacteria were removed over 99% by the DOF system, and 42~45% by the DAF system. That is, Microbial bacteria were almost completely destroyed by the DOF system. To sum up with, the DOF system was found to be very effective to treat effluent of the wastewater treatment plant.
Contamination of soil and groundwater by the compounds of hydrocarbon petroleum has been widely accepted as the main cause that harms the environments and health. To remove those pollutants, absorbing clothes, activated carbons, or oil-water separation devices with the gravity method are employed for treatment. However, those materials and devices cannot remove the emulsion pollutants despite of their efficiency for removing free products. Therefore, we investigated the problems which occur during the groundwater treatment for the highly concentrated suspended solid particles, which can be resulted from excavation, and to propose methods to remove TPH(Total Petroleum Hydrocarbon). After coagulation experiment with high molecular polymers, the concentration of SS(Suspended Solids) and COD(Chemical Oxygen Demand) turned to satisfy the groundwater quality criteria within 5 minutes while the concentration of TPH failed to meet the water quality standard of effluent. Consequently, the water quality criteria for effluent could not be met by single DAF(Dissolved Air Flotation) process. However all water quality criteria could be satisfied after 20 minutes when coagulation reactions are carried out simultaneously in the DAF reactor.
The complement (C) regulatory proteins decay accelerating factor (DAF, CD55) and CD59 could protect host cells using different mechanisms from C-mediated damage at two distinct levels within the C pathway. Co-expression of DAF and CD59 would be an effective strategy to help overcome host C-induced xenograft hyperacute rejection. In this study, we made a construct of recombinant expression vector containing DAF and CD59 cDNA and the stable cell lines were obtained by G418 selection. Extraneous genes integration and co-expression were identified by PCR, RT-PCR and Western blot analysis. Human c-mediated cytolysis assays showed that NIH/3T3 cells transfected stably with pcDNA3-CD59, pcDNA3-DAF, and pcDNA3-CD59DAF-DP were protected from C-mediated damage and that synchronously expressed human CD59 and DAF provided the most excellent protection for host cells as compared with either human CD59 or DAF expressed alone. Therefore, the construct represents an effective and efficacy strategy to overcome C-mediated damage in cells and, ultimately, in animals.
In recent years there have been large increases in the hydraulic loading rates used to design dissolved air flotation (DAF) facilities for drinking water applications. High rate DAF processes are now available at loading rates of 20 to $40m^3/m^2{\cdot}h$. This research evaluated dissolved air flotation as a separation method for algae and organic compounds from water treatment plants. During the service period of 2016. 5. to 2017. 6., DAF pilot plants ($500m^3/day$) process has shown a constantly sound performance for the treatment of raw water, yielding a significantly low level of turbidity (DAF treated water, 0.21~1.56 NTU). As a result of analyzing the algae cell counts in the influent source, it was expressed at 100-120 cells/mL. In DAF treated water, the removal efficient of alge cell counts was found to be upto 90%. The stable turbidity and algae removal were confirmed by operating the high rate DAF process under the condition of the surface loading rate of $30m^3/m^2{\cdot}hr$.
