  • 한국산 Drosophila melanogaster species group 8종의 생화학적 유연관계를 조사하기 위하여 3령기 유충, 말기 번데기, 성체의 세 발생단계에서 각각 효소 분석을 실시하였다. Rogers의 공식에 의해 각 발생단계 별로 8종간의 유전적 유사도를 계산하고 그 값을 근거로 UPGMA법에 의한 dendrogram을 작성하였다. 발생단계에 따라 관찰된 효소allele의 빈도는 큰 차이를 나타냈는데 이는 효소의 활성도나 유전자 발현의 발생단계별 차이로 생각되었다 이러한 발생단계별 효소분석 결과의 차이에도 불구하고 세 발생단계에서 모두 같은 pattern의 dendrogram이 얻어졌다. 따라서 대소 분석은 성체뿐만 아니라 유충이나 번데기와 같은 초기발생단계를 재료로 취할 때에도 종간 유연관계의 분석에 유용한 분석법이 될 것으로 생각할 수 있었다 세 발생단계에서 효소분석을 통해 얻어진 결과로 볼 때 D. melanogaster species group 8종은 크게 두개의 계통으로 나뉘었는데 D. melonogaster와 여. simulons, D. lutescens가 한 계통에 속했고 D. suzukii와 D. aurauia, D. biourariu, D. trioururia, D. rufa가 다른 한 계통에 속하고 있는 것으로 나타났다.

Comparative Toxic Effects of Gramoxone in the D. melanogaster and its Sibling Species (노랑초파리(Drosophila melanogaster)와 그 동포종들에 있어서 Gramoxone의 독성에 관한 비교 연구)

  • Physiological toxic and mutagenic effects of gramoxone in Drosophila melanogaster were invetigated. Gramoxone was highly toxic on the development, resulting in of lowering the viability and in prolongation of the developmental times. Adults treated with gramoxone during the developmental stages caused a lowering of the productivity and a little chinge in protein quantity. But the effect on the sex-linked lethal mutagenesis was found to be negative. The order of mortality causing ado리t stoa형e feeding to gramoxone in the D. melanogaster complex was like this ; D. mauritiana, D. sechellia, D. simulans and D. melanogasteu Two species of the D. yakuba complex were alike. Those results were more or less correlation with speciation of the D. melanogaster subgroup.

Comparative Study of Morphology on Male Sex Comb Variation of the Drosophila Species (Drosophila 수종의 성즐변이에 관한 형태적 비교 연구)

  • Sex combs which lack in females are characteristic of some members of the subgenus Sophophora When present, they can be van/ from a single blistle to very extensive structures, but the meianogaster subgroup of the subgenus Sophophora have only on the f~rst tarsal segments of the fore legs. D mauritiona among the melanogaster compler of the melar~ogaster subgroup have significantly more sex comb teeth (mean 12.75) than other specie& and the yakubo complex (range of the mean 6.84-758) have less than the melanogaster complex (range of the mean 835-12.75). In sex comb tooth number of interspecd~ch ybrids among the melonogoster complex, there appeared difference u;ith parental species Thus, analysis of male sex comb tooth in FI hybrid could not exhibit the direction of hereditan/ nature. On the other hand, D auraria (the montium subgroup), D suzuki (the suzuki subgroup) and D, lutecens (the takohashii subgroup) of the melanogaster species have two sex combs on the first and second tarsal segments, and the mean number were different according to the species.

  • Drosophila melanogaster Meigen and Drosophila suzukii Matsumura are taxonomically close Drosophila species belonging to the family Drosophilidae and melanogaster group. D. melanogaster is thought to be evolutionarily adapted to overripe, decaying, and fermented fruits, in which large amounts of chemicals such as ethanol, acetic acid, and 2-phenylethanol are produced, whereas, D. suzukii is attracted to fresh ripening fruit. Considering the distinct habitats of the two flies, D. suzukii is hypothesized to exhibit higher susceptibility to these chemicals than D. melanogaster. Therefore, in this study, we investigated the survival rate of the flies at various concentrations of three chemicals (2-phenlyethanol, acetic acid, and ethanol) and calculated the lethal concentration (LC) values to compare the tolerance and susceptibility of D. melanogaster and D. suzukii to the chemicals. Our results revealed that D. melanogaster exhibited higher tolerance than D. suzukii to all chemicals, supporting the hypothesis of different evolutionary adaptations to distinct habitats of the two flies.

  • Soluble protein of the eight species of the Drosophila melanogaster species group was analyzed for three developmental stages of lanra, pupa and adult by 2DE. Genetic distances were calculated by Aquadro and Aviso's equation for three developmental stages, respectively. The dendrosrams showed the same patterns in three stages. The dendrograms showed that the melonogaster species group consisted of two clustered groups. Total soluble protein contents on three developmental stages of the eight species were compared. Closely related species showed resemblant protein changing pattern during development, and their developmental changing patterns were different according to the subgroup.

  • 한국산 노랑초파리 군에 속하는 8종의 유전적 유연관계를 밝히고자 생식적 격리 그리고 수용성 단백질을 전기영동법으로 분석하였다. 생리적 격리 실험에서 교배전격리 실험결과를 Watanane와 Kawanishi model에 근거하여 보면 D. aurario complex 3종 중 D. triauror물가 원시종이며 남 auraria는 팍생종으로 나타났다. 교배후 격리 실험에서 나. melonogaster와 D. simuions의 교배시, D. melonogoster를 수컷으로 하였을 때는 불임의 수컷만이, 또 D. melonogaster를 암컷으로 곯였을 때는 불임의 암컷만이 출현하였다. 그리고 남 ouraria complex 3종간의 교배에서는 수정 능력이 있는 수컷과 암컷이 출현하였는데, 이는 아직 남 auraria Complex가 semispecies단계에 있음을 나타내는 것이라 할 수 있다. SDS-PAGE로 분석한 노랑초파리 군 8종의 band pattern을 densitometer로 scanning한 결과 남 susu상가 가장 특이하였으며, TDE에 의한 유전적 거리(Aquadro and Avise's)는 남 auror지와 봉 triauror지사이가 0.155로 가장 낮았고, D. melonogaster와 고. mfa 사이가 0.422로 가장 높았다. 본 연구의 결과를 UPGMA법 뜨로 분석하면, 한국산 노랑초파리 군 8종은 4개의 아군으로 나뉘어지며 이들은 2개의 다른 큰무리로 구분되었는데, D. suzu가기 아군, D. lutescens의 아군, D. melanogoster와 봉 simulons의 아군이 속한 큰무리와, D. mfa,0. ouroria, D. biauroria 그리고 남 triourario가 속한 다른 큰무리로 나눌 수 있다.

Genetic Analysis of the Polymorphism of Color Pattern in Drosophila auraria (Drosophila auraria 의 반문다형현상의 유전학적분석)

  • D.auraria is a species belonging to the D.melanogaster group, and this species was divided into 3 races(A, B and C race ) by morphoogical difference of the genitals . Korean populations of D.auraria A race are polymorphic with respect to the pigmentation of the abdominal tergites. The female shows two forms of color pattern, dark and light, on the sixth to the ninth tergites, and the male has no distinction shown by the female. Crossing experiments in the laboratory have shown that the difference between these color forms is due to a single pair of allelic genes located on an autosome. In natural populations, the light forms of females are always commoner than the dark one. The number of the light form increases relatively in southern localities and decreases relatively in northern localities. Furthermore, the number of the light form increases relatively in summer and decreases relatively in fall and spring. By genetic analyses of these wild flies, the relation, d/d>d/D>D/D has been disclosed as regards the relative frequencies of the genotypes in natural populations. Three experimental populations have been set up in the laboratory. Homozygous dark forms and homozygous light forms were mixed together and cultured in population cage at the temperature of 25 $^{\circ}C$. Approximately one year later, the frequencies of the light and the dark forms reached an equilibrium , the light one being usually more frequent than the dark one. This indicates that the heterozygous dark form possesses the highest adaptive value, the homozygous light being intermediate and the homozygous dark lowest. In number of adults hached, the homozygous light form was superior to the heterozygous dark form and the homozygous dark form, but the differences are scarcely significant.
