Tne present studies were camied out to ~nvestigate the allele frequency variations of alcohol dehydrogenase (ADH) in natural populat~ons of Drosophiio melonogoster and the correlations of iwo ADH alleles between fitness and ethanol. ADH alleles were found to be polymorphic in natural populations of D. rnelanogaster. The frequencies of FF, FS and SS genotypes were 47.66, 42 18, and 10.16%. respectively, therefore the F gene frequency (68.75) was shown to be hlgher than the S gene (31.25 %). The FF genotype was slightly superior to the SS genotype in both fecundiiy and eclaslon. The frequency of AdhF allele in the small alt>fic~apl opulaliow originated from natural populations was increased for 20 generations on normal media at 25$^{\circ}$C In resistance to ethanol, the FF genotype was supenor to the SS genotype, too. It meant that ethanol as environmental factor might be the selective factor on ADH locus in natural populat~ons of D meionogoster.
Taste sensitivity to sugars plays an essential role in the initiation of feeding behavior. In Drosophila melanogaster, recent studies have identified several gustatory receptor (Gr) genes required for sensing sweet compounds. However, it is as yet undetermined how these GRs function as taste receptors tuned to a wide range of sugars. Among sugars, fructose has been suggested to be detected by a distinct receptor from other sugars. While GR43A has been reported to sense fructose in the brain, it is not expressed in labellar gustatory receptor neurons that show taste response to fructose. In contrast, the Gr64a-Gr64f gene cluster was recently shown to be associated with fructose sensitivity. Here we sought to decipher the genes required for fructose response among Gr64a-Gr64f genes. Unexpectedly, the qPCR analyses for these genes show that labellar expression levels of Gr64d and Gr64e are higher in fructose low-sensitivity flies than in high-sensitivity flies. Moreover, gustatory nerve responses to fructose in labellar sensilla are higher in Gr64d and Gr64f mutant lines than in mutant flies of the other Gr64a-Gr64f genes. These data suggest the possibility that deletion of GR64D or GR64F may indirectly induce enhanced fructose sensitivity in the labellum. Finally, we conclude that response to fructose cannot be explained by a single one of the Gr64a-Gr64f genes.
KP elements derived from Korean populations (Seoul, Cheonan and Taegu) of Drosophila melunoguster were examined for their molecular structure. The entire 1.15 kb sequence of the three KP elements KC-137 (Cheonan), KS-95 (Seoul) and KT-99 (Taegu) have been obtained by PCR amplification using inverted repeat primers and DNA sequencing. The 1.15 kb fragments of KP elements were cloned into pCRmll vector plasmids, and subsequently sequenced. The sequence of the three KP elements in these populations suggested that there might have been derived from the complete P element by a 1753 bp internal deletion between positions 808 and 2560. Therefore these KP elements were confirmed to be identical to that isolated from M'10 strains widely distributed in most Eurasian populations of D. melanoguster. Sequence comparison with the 2.9 kb complete P element in pn25.1 revealed that KC-137 has only shown to be two base substitutions of A to G and G to A at positions 62 and 242, respectively. The retained sequence of the two KP elements KS-95 and KT-99 shows complete homology to the P factor in pn25.1. Based on this result, the two base substitutions in KC-137 might be due to Taq DNA pollunerase errors. Finally, it is suggested that the high copy numbers of KP elements provieds an explanation for the suppression of P-mediated hybrid dvssenesis in Korean population of D. melunogoster.

Citronellal, a well-known plant-derived mosquito repellent, was previously reported to repel Drosophila melanogaster via olfactory pathways involving but not directly activating Transient Receptor Potential Ankyrin 1 (TRPA1). Here, we show that citronellal is a direct agonist for Drosophila and human TRPA1s (dTRPA1 and hTRPA1) as well as Anopheles gambiae TRPA1 (agTRPA1). Citronellal-induced activity is isoform-dependent for Drosophila and Anopheles gambiae TRPA1s. The recently identified dTRPA1(A) and ag-TRPA1(A) isoforms showed citronellal-provoked currents with EC50s of $1.0{\pm}0.2$ and $0.1{\pm}0.03mM$, respectively, in Xenopus oocytes, while the sensitivities of TRPA1(B)s were much inferior to those of TRPA1(A)s. Citronellal dramatically enhanced the feeding-inhibitory effect of the TRPA1 agonist N-methylmaleimide (NMM) in Drosophila at an NMM concentration that barely repels flies. Thus, citronellal can promote feeding deterrence of fruit flies through direct action on gustatory dTRPA1, revealing the first isoform-specific function for TRPA1(A).

Autophagy, the lysosomal degradation pathway, is an intracellular recycling system that is necessary for the metabolic benefits of exercise and for producing lasting beneficial effects of exercise in various diseases. However, the most recent studies have only examined the effect of a single bout of exercise or resistance exercise on autophagic responses. To determine the differential effects of acute and chronic exercise on the expression of autophagy-related genes in D. melanogaster, white-eyed mutant D. melanogaster were assigned randomly to four groups: control, acute exercise, 2 hr chronic exercise, and 3 hr chronic exercise. The flies were exercised using a mechanized platform known as the Power Tower. Our results revealed that a single bout of exercise resulted in increased mRNA levels of the Atg8a gene (~20%, p<0.05). However, Atg1 and Atg6 mRNA expression were not induced by acute exercise. Transcript levels of Atg6 (~29%, p<0.05) related to the nucleation of autophagosomes were significantly induced by 2 hr of chronic exercise. However, this chronic exercise was not enough to increase Atg1 and Atg8a mRNA expression. On the other hand, 3 hr of exercise for 7 days significantly increased Atg1, Atg6, and Atg8a gene expression-about 57%, 37%, and 71%, respectively (p<0.05). These results suggest that a single bout of exercise is not enough to induce full activation of selected autophagy-related genes in D. melanogaster. Our results demonstrated that chronic regular exercise induced autophagy-related gene expression, suggesting that chronic regular exercise training might be required to activate autophagic responses important for producing beneficial effects of exercise in various diseases.

In order to cloning of PPO gene as a melanin formation related genes involved in hardening and pigmentation of insect integument or wing, we cloned cDNA and analyzed the sequence of PPO gene of H. axyridis. PPO2 primer were designed based on the sequences of PPO genes of Tribolium castaneum and Drosophila melanogaster, and then plasmid DNA were cloned from PCR products obtained from different two color patterns. When the plasmid DNA band pattern were digested by restriction enzymes, BamH1, Xba1, and EcoR1, we found same size band pattern. However, this sequence was not homologous to sequence of T. castaneum PPO gene. Using the primer designed based on the sequence of D. melanogaster, 209 bp PCR product was observed.

Animals use their odorant receptors to receive chemical information from the environment. Insect odorant receptors differ from the G protein-coupled odorant receptors in vertebrates and nematodes, and very little is known about their protein-protein interactions. Here, we introduce a mass spectrometric platform designed for the large-scale analysis of insect odorant receptor protein-protein interactions. Using this platform, we obtained the first Orco interactome from Drosophila melanogaster. From a total of 1,186 identified proteins, we narrowed the interaction candidates to 226, of which only two-thirds have been named. These candidates include the known olfactory proteins Or92a and Obp51a. Around 90% of the proteins having published names likely function inside the cell, and nearly half of these intracellular proteins are associated with the endomembrane system. In a basic loss-of-function electrophysiological screen, we found that the disruption of eight (i.e., Rab5, CG32795, Mpcp, Tom70, Vir-1, CG30427, Eaat1, and CG2781) of 28 randomly selected candidates affects olfactory responses in vivo. Thus, because this Orco interactome includes physiologically meaningful candidates, we anticipate that our platform will help guide further research on the molecular mechanisms of the insect odorant receptor family.
The purpose of the present study is to investigate the electrophoretic patterns of ADH isozymes and activity at various developmental stages of Oregon-R and Kwangju strains of D. melanogaster. The ADH isozymes were seperated by agarose gel plate eletrophoresis and they were stained for ADH activity study. The optical density was measured by using densitometer and the total activity was determined by measuring the enzyme solution's absorbance of 340 nm by spectrophotometer. The results are as follows: 1. At egg stages of Oregon-R and Kwangju strains ADH 2, ADH 3, ADH 4 were observed in te zymograms, but all of ADH 1, ADH 2, ADH 3, ADH 4 and ADH 5 were detected in larval, pupal and imaginal stages. 2. The amount of ADH 1+ADH 2 (anion) was 20-40% and that of ADH 3+ADH 4+ADH 5 (cation) was 60-805. In the developmental stages other, than egg stages, ADH 3 of both strains showed the largest amount among all ADH isozymes, and especially ADH 4 of Kwangju strain showed less amount than that of Oregon-R strain. 3. The total activity of the ADH at the egg stage was found to be the lowest, at larval stage became to be higher and to be lower at pupal stage and then became to be the highest at the imaginal stage. It is noticeable that the activity of the ADH in Kwangju strain was higher than that in Oregon-R strain.
In order to see if NaCl affected the the emergence rate and the SD action when treated it at the pupal stage of Drosophila melanogaster, an experiment was performed in which the resistibility to NaCl at the pupal stage was examined by the emergence rate and the effects of NaCl on the SD action was studied by k values. The four SD strains and two other strains (cn bw and Oregon-R) were used and NaCl media were prepared by adding NaCl at a concentration of 0.0M, 0.1M, 0.3M, 0.5M, 0.7M, and 1.0M to the standard media for the present experiment. The following conclusions were established: 1. The emergence rate (resistibility to NaCl) when treated NaCl at the pupal stage is significantly different not only among the strains but also among concentrations of NaCl. 2. The emergence rate decreases as concentration of NaCl increases but the rate of decrease shows rather constant so that the curves of decrease are not steep. 3. The emergence rate of the cn bw and Oregon-R strains shows higher value than of the four SD strains, implying that the resistibility to the NaCl is lower in the SD strains than in the two strains; not a few flies are emerged from the media of 1.0M of NaCl. 4. The difference in k values is not significant among concentrations of NaCl but among strains, the k values of the original SD strains are rather higher and constant but those of the recombinant SD strains are rather low and unstable. 5. The SD action is not affected by NaCl as far as once emerged from the culture media whether containing NaCl or not in any developmental stage. Thus such an experiment on NaCl effect is of little value to analyze the mechanism of the SD action in D. melanogaster.
The late pupal stage of Drosophila melanogaster occurs immediately before the completion of retinal development, during which the rhabdomere rapidly forms. In this period, the photoreceptor cells were fixed and dehydrated using a high-pressure freezer (HPF) and freeze substitution (FS) technique, which is the most effective in preserving the cell structures, and observed using high-voltage electron microscopy (HVEM) at 1000 KV. The rhabdomere was classified structurally into three types of formation patterns using stereo-tiling image of thick sections. Initially, hexagonal arrays of rhabdomere existed in different angles. In addition, small pieces of rhabdomere could be observed in the cytoplasm of the photoreceptor rolls, which were visible during the profess of rhabdomere formation. In addition, multiple layers of rhabdomere strings were observed. We observed there are at least three types of vesicles related to rhabdomere formation in photoreceptor cells. In addition, it was found that these vesicles initiate the formation of the rhabdomeres during the pupal stage. Collectively, these data suggest that rhabdomeres were mainly formed through vesicles, and that parts of the rhabdomere formed first and then gathered and formed rhabdomeres in the late pupal stage.
