A Comparison Study on SVM MDR and D-MDR for Detecting Gene-Gene Interaction in Continuous Data (연속형자료의 유전자 상호작용 규명을 위한 SVM MDR과 D-MDR의 방법 비교)

  • We have used a multifactor dimensionality reduction(MDR) method to study the major gene interaction effect in general; however, without application of the MDR method in continuous data. In light of this, many methods have been suggested such as Expanded MDR, Dummy MDR and SVM MDR. In this paper, we compare the two methods of SVM MDR and D-MDR. In addition, we identify the gene-gene interaction effect of single nucleotide polymorphisms(SNPs) associated with economic traits in Hanwoo(Korean cattle). Lastly, we discuss a new method in consideration of the advantages that the other methods present.

  • We have used multifactor dimensionality reduction(MDR) method to study interaction effect of statistical model in general. But MDR method cannot be applied in all cases. It can be applied to the only case-control data. So, two methods are suggested E-MDR and D-MDR method using regression tree algorithm and dummy variables. We applied the methods on the identify interaction effects of single nucleotide polymorphisms(SNPs) responsible for longissimus mulcle dorsi area(LMA), carcass cold weight(CWT) and average daily gain(ADG) in a Hanwoo beef cattle population. Finally, we compare the results using permutation test.

  • Diseases of human or economical traits of cattles are occured by interaction of genes. We introduce expanded multifactor dimensionality reduction(E-MDR), dummy multifactor dimensionality reduction(D-MDR) and SNPHarvester which are developed to find interaction of genes. We will select interaction of outstanding gene combinations and select final best genotype groups.

  • To share and standardize information, especially in the database environments, MDR (Metadata Registry) can be used to integrate various heterogeneous databases within a particular domain. But due to the discrepancies of data element representation between organizations, global information integration is not so easy. And users who are searching integrated information on the Web have limitation to obtain schema information for the underlying source databases. To solve those problems, in this paper, we present a 3-layered Information Integration System (LI2S) based on MDRs and Ontology. The purpose of proposed architecture is to define information integration model, which combine both of the nature of MDRs standard specification and functionality of ontology for the concept and relation. Adopting agent technology to the proposed model plays a key role to support the hierarchical and independent information integration architecture. Ontology is used as for a role of semantic network from which it extracts concept from the user query and the establishment of relationship between MDRs for the data element. (MDR and Knowledge Base are used as for the solution of discrepancies of data element representation between MDRs. Based on this architectural concept, LI2S was designed and implemented.

Polyamines in Multi-drug Resistant Cancer Cells (다제 내성 암세포에서의 Polyamine 특성)

  • Since the advent of chemotherapy, certain types of cancer have been particularly resistant to chemotherapeutic treatment. One of the most well-studied types of resistance is resistance to multiple struc-turally dissimialr hydrophobic chemotherapeutic agents, or multidrug resistance (MDR). We found that MDR cells (KBV20C, KB7D) being highly resistant to colchicine, etoposide, and vincristine were found to have very low level of putrescine and low level of spermidine than the drug sensitive parental cells (KB) but they had almost same level of spermine as the drug sensitive cells. Although both MDR and drug sensitive cells had almost same rate of polyamine uptake, MDR cells were much more sensitive to an inhibitor of polyamine synthesis, methylglyoxal-bis guanylhydrazone (MGBG), suggesting that MDR cells might be defective in polyamine synthesis. These results also suggest that HGBG can be used for treatment of MDR in vivo.

  • The purposes of this study were to evaluate the relationship between the genetic polymorphisms in CYP2D6*10 allele, MDR1 (exon 21 and 26) gene and risperidone pharmacokinetics in healthy male Korean subjects. A single dose of 2 mg risperidone tablet was given orally to 23 healthy male Korean volunteers. Blood samples were taken during the 12 hours after the dose. Serum concentrations of risperidone and 9-hydroxyrisperidone were measured using HPLC with UV detector. 23 subjects were genotyped for CYP2D6*10 allele, MDR1 G2677 T and C3435T by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). Timing and predictors of death during treatment in patients with multidrug/rifampin-resistant tuberculosis in South Korea

  • Background/Aims: This study aimed to investigate the timing and predictors of death during treatment among patients with multidrug/rifampin-resistant tuberculosis (MDR/RR-TB) in South Korea. Methods: This was a retrospective cohort study that included MDR/RR-TB cases notified between 2011 and 2017 in South Korea. Results: Among 7,226 MDR/RR-TB cases, 699 (9.7%) died at a median of 167 days (IQR 51-358 d) from the initiation of MDR-TB treatment. The cumulative proportion of all-cause death was 35.5% at 90 days and 52.8% at 180 days from treatment initiation. TB-related deaths occurred at a median of 133 days (IQR 32-366 d), which was significantly earlier than the median of 184 days (IQR 68-356 d) for non-TB-related deaths (p = 0.002). In a multivariate analysis, older age was the factor most strongly associated with death, with those aged ≥ 75 years being 68 times more likely to die (aHR 68.11, 95% CI 21.75-213.26), compared those aged ≤ 24 years. In addition, male sex, comorbidities (cancer, human immunodeficiency virus, and end stage renal disease), the lowest household income class, and TB-specific factors (previous history of TB treatment, smear positivity, and fluoroquinolone resistance) were identified as independent predictors of all-cause death. Conclusions: This nationwide study highlights increased deaths during the intensive phase and identifies high-risk groups including older people and those with comorbidities or socioeconomic vulnerabilities. An integrated and comprehensive strategy is required to reduce mortality in patients with MDR/RR-TB, particularly focusing on the early stages of treatment and target populations.

  • In this paper, the CAD program for designing the microwave waveguide narrow-band bandpass filters has been developed by the passband correction method with filter synthesis for the MDR(Microwave Digital Relay) system. Here, the modified double E-plane structures are employed in the filter structure which is analyzed by the variational method. Using the developed CAD program, 0.01dB equi-ripple chebyshev type 6-section bandpass filters used in the MDR system operating nationally is designed at the center frequency of 11.0GHz, fabricated with tunable type and then measured by tuning process. The experimantal results show good agreements with the theoretical results.

  • It is important to detect the gene-gene interaction in GWAS (genome-wide association study). There have been many studies on detecting gene-gene interaction. The one is D-MDR (dummy multifoactor dimensionality reduction) method. The goal of this study is to evaluate the power of D-MDR for identifying gene-gene interaction by simulation. Also we applied the method on the identify interaction effects of single nucleotide polymorphisms (SNPs) responsible for economic traits in a Korean cattle population (real data).

  • Recently, enterprise systems require the necessity of integration for data sharing and cooperation. As a methodology for integration, Service-Oriented Architecture for service integration and Master Data for integration of data, which is used for service, were appeared. This paper suggests a method that operates BP(Business Process) efficiently. We make XMDR(eXtended Meta Data Registry) as knowledge-repository to support the BP and construct data hubs to operate it. XMDR manages MDM(Master Data Management) to integrate the data, resolves heterogeneity between the data and provides relationship to the business efficiently. This is composed of MDR(Meta Data Registry), ontology and BR(Business Relations). MDR describes relationship between meta data to solve structured heterogeneity. Ontology describes semantic heterogeneity and relationship between data. BR describes relationship between tasks. XMDR data hub supports the management of master data and interaction of different process effectively.
