Survival Rates of Frozen-thawed Surf Clam, Spisula sachalinensis Larvae in Five Developmental Stages (북방대합, Spisula sachalinesisr 유생의 발생단계별 냉동-해동후 생존율)

  • This study was peformed to find out the optimum larval stage among trochophore, early Dshaped larva, late D-shaped larva, early umbo and late umbo stages for cryopreservation of surf clam, Spisula sachalinensis larvae. Dimethyl sulfoxide (DMSO)and ethylene glycol were used as cryoprotectant, The larvae were immersed to cryoprotectants for 10 minutes and thereafter, cryopreserved in liquid nitrogen. Survival rates of trochophores frozen-thawed in 2.0 M dimethyl sulfoxide and 2.0 M ethylene glycol were the highest as 97.4% and 85.0%, respectively and post-thaw survival rates were decreased with the larval growth.

  • Develoment precess and characteristics of eggs of the geoduck clam, Panope japonica are reporting in this study. Eggs and sperm were excised from gonad, artificially fertilized in an aquarium, reared under various temperature regimes, and record and record the larval period and the time need to reach a certain larval stage from ferilization. Unfertilized eggs of P. japonica appeared to be oval with a mean diameter of $70\mu$m and they became spherical after fertilization. The eggs of P. japonica can be classified as demersal. At a constant water temperature of $ 11^{citc}C$, it took 4 hours form fertilization to become four-cell stage, two days to become trochophore larvae, three days to become D-shape larvae, twenty-three days to become umbo stage, and thirty-six days to become fully grown veliger ready form settlement. A negative correlation was observed between the water temperature and the larval period of P. japonica. From fertilization to D-shape larvae, it took five days at 8$^{\circ}C$, while it was only two days to become D-shape larvae at $ 17^{citc}C$. Time required to D-shape larvae from fertilization was proportional to temperature, and the relationships were expressed as follows : To 8-cell stage, 1/t=0.0209 w-0.1167 (r=0.9967) To blastula stage, 1/t=0, 0055 w-0.0192 (r=0.9825) To trochophore stage, 1/t=0.0034 w-0.0155 (r=0.9907) To D-shape larvae stage, 1/t=0.0014 w-0.0023 (r=0.9843) (t, time in hours ; w, water temperature) Bioligical minimum temperature for egg development was calculated as 3.82$^{\circ}C$ in average.

Development and growth in fertilized eggs and larvae of Korea swift's scallop Chlamys swiftii reared in the laboratory (실험실에서 사육된 고랑가리비 Chlamys swiftii 수정란 발생과 유생 성장)

  • The development of swift's scallop Chlamys swiftii, reared in the laboratory, has been examined through the investigation of morphological characteristics in fertilized egg, larvae and juvenile. Eggs were fertilized with a dilute sperm solution to improve the survival of fertilized eggs. Developing larvae were maintained at a temperature of $16{\pm}0.5^{\circ}C$ and salinity concentration of 33 ppt. We have investigated the fertilization rates and egg number spawned at several stimulating conditions such as sunlight exposure, air dry, seawater temperature rise ($5^{\circ}C$) and seawater temperature rise ($5^{\circ}C$) after exposure of air dry for spawning induction of swift's scallop Chamys swiftii. Stimulation treated with sunlight exposure and seawater temperature rise ($5^{\circ}C$) have shown the spawning number of 700,000-900,000 and 700,000-800,000 per individual, respectively while stimulation treated with seawater temperature rise ($5^{\circ}C$) after exposure of air dry have shown the high spawning number of 1,000,000-1,500,000 per individual. Survival rate of D-shaped larvae of swift's scallop put into the different seawater temperatures of $8^{\circ}C$, $12^{\circ}C$, $16^{\circ}C$, $20^{\circ}C$ and $24^{\circ}C$ has been 4.1%, 11.6%, 32.7%, 18.6% and 3.2%, respectively. Fertilized eggs with the diameter of $72{\mu}m$ developed into trochophore larvae of $103{\pm}3.8{\mu}m$ shortly after 35 hours and to D larvae of $129{\pm}10.4{\mu}m$ shell length within 72 hours. It took 336 hours to become initial Umbo-stage larva of $145{\pm}16.8{\mu}m$ shell length. Post larvae, which have been $197{\pm}13.6{\mu}m$ shell length, spontaneously have settled in the attachment substances. It have required 528 hours from fertilized eggs to early attached juvenile to become initial juvenile size of $245{\pm}15.8{\mu}m$ shell length.

비단가리비 인공종묘 생산

  • 비단가리비의 인공 종묘 생산을 목적으로 전라남도 신안군 대흑산도 주변에 서식하는 비단가리비를 대상으로 인공 종묘 생산을 위한 산란 유발, 수정란의 발생 과정, 유생 사육, 채묘 및 중간 육성 등 양식 생물학적 연구를 실시하였다. 어미의 각종 산란 유발 자극에 대한 반응은 Serotonin 주사, 온도 자극, 혼합 자극에서 반응율이 가장 높았으며, 자외선 조사 해수 자극은 수컷만 반응을 하였고 간출 자극은 반응이 없어 실효성이 없는 것으로 나타났다. 수정란의 크기는 $69.5{\mu}m$이었고, 수정 후 약 2시간에 2 세포기, 8시간 후에 8 세포기, 20시간 후에는 담륜자 유생으로 부화하였으며 40시간 후에는 D상 유생으로 되었다. 수온별 비단가리비 유생의 성장은 수온 $20^{\circ}C$에서 각장 $178.9{\mu}m$로 가장 좋았으며, 이 때의 생존율은 15.5%이었다. 그러나 수온 $15^{\circ}C$에서는 낮은 성장을 보여 각장 $135.9{\mu}m$로 성장하였으며, 생존율도 9.8%로 저조하였다. 수용 밀도별 사육 시험에서 $1m{\ell}$당 1개체와 5개체에서 성장 및 생존율이 양호하여 성장 및 생존율을 볼 때 적정 사육 밀도는 $1m{\ell}$당 5개체 이하로 나타났다. 먹이 생물 종류에 따른 유생의 성장을 알기 위하여 I, gal-baba, C, calcitrans, N, oculata를 단독 또는 혼합으로 공급하였을 때 실험 종료후 각장의 성장은 I, galbana+C. calcitrans+N. oculata구가 $194.2{\mu}$로 가장 좋았고 N. oculata구는 $162.2{\mu}m$로 가장 낮은 성장을 보였다. 생존율에서는 I. galbana+C. calcitransoculata구는 9.4%로 가장 낮은 생존율을 보였다. 채묘기별 유생의 부착률은 염화비닐판을 수평으로 놓은 것과 패각이 각각 3.43%와 3.17%로 가장 양호하였으나, 양파망과 염화비닐판을 수직으로 놓은 것은 각각 1.52%와 1.61%로 비교적 저조하였다. 수정 후 40일째부터 90일째까지 측정한 부착치패의 경과 일수에 따른 각장의 성장은 $SL=184.44e^{0.0335X}(r^2=0.9861)$의 회귀직선식으로서 나타났다. 중간육성 시험에서 수심별 성장을 분석한 결과, 비단가리비 치패는 저층보다 표층이 각장 5.92mm, 전중량 6.07g 정도 더 빨리 성장하였다.

Spawning and Larval Development of the Jicon Scallop, Chlamys farreri (비단가리비, Chlamys farreri의 산란과 유생사육)

  • This study focused on spawning season, induce spawning, spawning and larval development of the Jicon scallop in Daehuksan Island of southwestern waters in Korea. The condition index and gonadosomatic index were used to investigate the reproductive pattern of the Jicon scallop. The major spawning season was from July to August, showing an unimodal gametogenic cycle per year. Several different tests were carried out to induce spawning of the mature male and female C. farreri. For females, the injection of serotonin, temperature induction technique and the combination of the both treatments produced significantly faster gamete release. Unlike females, males spawned only in response to the UV rays irradiation stimulation. Mean size of fertilized eggs was 69.5 $\mu$m in diameter. After fertilization, the zygote could be divided into 2 cells as early as 2 hours. It took about 8 hours to develop the 8-cell stage, about 20 hours to hatch trochophore larvae, and about 40 hours to be D-shaped larvae.

Distribution of Bivalve Larvae in Deukryang Bay (득량만에서 조개류 유생의 분포)

  • Larval distribution of bivalve from 29 stations in Deukryang Bay was examined from May to November in 1995. The abundance of total larvae increased from May ($3,736\;inds/m^3$) and reached maximum in August ($174,616\;inds/m^3$). But it decreased continuously until November ($2,171\;inds/m^3$). D-shape, early umbone and advanced umbone stage larvae composed of 76%, 18% and 6%, respectively, of total bivalve larvae. Nine larval species of bivalve were identified in this study. The mean abundance of Scapharca subcrenata larvae was $14,030\;inds/m^3$, which was 33.5% of total identified larvae. S. broughtonii (16.2%), Crassostrea gigas (12.5%), Mytilus edulis (10.2%) were also major species in order of their abundance. Considering the monthly larval occurrence in Deukryang Bay, main spawning month of bivalves except M. edulis and M. senhausia was estimated as July. Even though the spatial distribution of bivalve larvae in Deukryang Bay was different by the species, the larvae were generally abundant in the inner and west area of the bay, where was shallower and higher water temperature than the entrance and east area of the bay.

  • To develop techniques for the production of artificial seedling of Corbicula japonica, protocols for induction of spawning and larval development were developed. During the assumed spawning period of August to mid-September, attempts were first made to induce spawning by gonadal incision and UV-irradiation but all in vain. At the end of August, elevated thermal induction evoked 90 % positive response in animals maintained at $3\textperthousand$ salinity. Immersion in (1/1000~3/1000 N) ammonium hydroxide ($NH_4$OH) also induced spawning in 15~45%) of the treated animals at $3\textperthousand$ salinity. Fertilized eggs measured 86$\mum. At 23.0~24.5$^{\circ}C$, the fertilized egos developed into 4-cell stage embryos within 2 hours, trochophores 15 hours, D-shaped larvae 2 days, umbo 9 days and fully grown veligers, ready to infiltrate into the sediment, within 16 days.

  • The development of Japanese geoduck, Panopea japonica, grown under culture conditions, has been examined through the morphological characteristics in fertilized egg, larvae and juvenile. Gametes were stripped from ripe broodstock and placed into two separate containers. Eggs were washed through a $40{\mu}m$ sieve and fertilized with dilute sperm solution. Developing larvae were maintained at $19{\pm}1^{\circ}C$. Fertilized eggs with $81.6{\mu}m$ diameter developed to trochophores within 14 h and to D-stage larvae ($116{\mu}m$ shell length) within 27 h. Larvae were spontaneously settled at shell length of $311{\mu}m$ after 20 days. The hatching from fertilized eggs and larval rearing were normally available in $18.5-21.5^{\circ}C$, and the growth was good in a cashmilon substrate, as well as sand. After rearing of day 108 from metamorphosis, the shell length of juvenile P. japonica reached 13 mm, and growth rate of shell length of the juvenile was $117.5{\mu}m/d$.
