Effects of Plant Growth Regulators on the Regrowth of Perennial Ryegrass (Lolium perenne L.) (식물 생장 조절물질이 페레니얼 라이그라스의 재생에 미치는 영향)

  • Plant growth regulators were treated on the cut perennial ryegrass (Lolium perenne L. cv. Reveille) to investigate the effect on the regrowth after cutting. The growth showed better result when 0.1 or 0.5 mg/L cytokinin were treated. Among cytokinins, kinetin or 2iP gave the better effect on the growth than BAP. In 2.4-D as an auxin. cut plants grew best at the concentration of 0.1 mg/L. The initial regrowth was very vigorous when GA$_3$ was treated as a growth regulator, but the growth was retarded after 2 weeks later of cutting. Co-treatment of kinetin as a cytokinin and 2,4-D as an auxin showed synergistic effect on the regrowth of cut perennial ryegrass. Both plant growth regulators gave the same result at the same concentrations in the suspension culture of perennial ryegrass cells.

  • Calcium promoted ethylene production from mungbean hypocotyl segments incubated in the presence of either auxin or cytokinin (kinetin). Time course studies indicated that the calcium effect on ethylene production had a longer latent period (about 6 h) in combination with kinetin than with auxin. Studies on the effects of agents that are known to interfere with either action or transport (uptake) of calcium on ethylene biosynthesis indicated different patterns between auxin- and kinetin-treated tissues. Auxin-induced ethylene production was inhibited by the calmodulin inhibitor, trifluoperazine (TFP), and this inhibition was overcome by high concentrations of calcium applied, but TFP had no significant effect on kinetin-induced ethylene production regardless of calcium in the medium. The calcium channel blocker, verapamil, inhibited auxin-induced, but had little effect on kinetin-induced, ethylene producton. In vivo activity of "ethylene forming enzyme (EFE)" was found to be substantially promoted by calcium treatment. The enzyme activity was further increased by kinetin when segments were simultaneously treated with calcium, but auxin did not have such an effect.an effect.

  • The most effective cytokinin for shoot multiplication in vitro of Prunus persica cv. Baekmijosaeng, Okubo, and Yumyeong was 2.0 mg/L BA. As the result of combinational treatment of BA and auxin sources (IAA, IBA and NAA), 2.0 mg/L BA with 1.0 mg/L IAA was the most effective for shoot multiplication of cv. Baekmijosaeng. The most effective auxin source for rooting was IAA and the concentration was 5.0 mg/L and 3.0 mg/L for cv. Baekmijosaeng and Okubo, respectively.

  • Callus-inducing ability of Alnus hirsuta was examined by culturing various tissues (leaf, hypocotyl, cotyledon and seed) on NT (Nagata & Takebe) medium, supplemented with 2.5$\mu$M 2,4-D. Leaf-originated callus was cultured on media varying in auxin (IBA and NAA) and cytokinin (BAP) concentrations to examine the effects of auxin and cytokinin on callus growth. Maximum growth was obtained at 10 $\mu$M IBA+10$\mu$M BAP and 10$\mu$M NAA without cytokinin. Cell suspensions established from cotyledon-originated callus yielded viable protoplasts after incubation for 16-18 hours in an enzyme mixture (1% (w/v) Onozuka R-10 0.5% (w/v) Macerozyme, CPW salts and 13% (w/v) mannitol, pH 5.8). Protoplasts were cultured on NT medium, supplemented with glucose, hormones and coconut milk. After 6 weeks of culture, protoplasts sustained cell divisions to form microcallus, which showed various colors from red to white.

Physiological and morpho-anatomical analyses of hyperhydric Arabidopsis thaliana influenced by media components

  • Hyperhydricity is a physiological anomaly that significantly affects the growth and proliferation rate of crops cultivated by tissue culture techniques. To better understand the mechanisms that govern hyperhydricity incidence, we examined the effects of several media components, particularly cytokinin and gelling agents. These elements were found to be influential in both in vitro propagation and the development of hyperhydricity. Our study revealed that Arabidopsis thaliana seedlings had a greater manifestation of hyperhydricity symptoms when exposed to high cytokinin concentrations compared with the control. The presence of gelrite led to the manifestation of hyperhydric symptoms by elevated water build-up in the apoplast. The phenomenon of stomata closure was observed in the hyperhydric leaves, resulting in an increased ability to retain water and a decrease in the transpiration rates when compared to their respective control leaves. Additionally, histological examinations of the cross sections of hyperhydric leaves revealed an irregular cellular arrangement and large intercellular spaces. Furthermore, hyperhydric seedlings displayed impaired cuticular development in comparison to their normal seedlings.

In Vitro Propagation of Anthurium andreanum ′Atlanta′ Developed for Pot Culture (분화용 Anthurium andreanum ′Atlanta′의 기내번식)

  • In order to establish micropropagation system Anthurium andreanum 'Atlanta', dwarf type, shoots of A. andreanum were cultured on medium supplemented with cytokinin. Callus was formed from the base of shoots. high frequency callus induction was obtained on medium with 10.0mg/L BA or 10.0mg/L TDZ(thidiazuron) at more than 71.8%. The shoots were cultured on media with various combinations and concentrations of TDZ, BA and 2.4-D to enhance callus induction. Callus was induced at more than 72.6% and grew vigorously on media containing 10.0mg/L BA and 0.0∼0.5mg/L 2.4-D, or 1.0mg/L TDZ. Stimulation effects of cytokinin by 2.4-D did not occur in combined treatments of cytokinin and 2.4-D. Callus was cut into sections(7${\times}$10mm), and then cultured on media with BA alone or BA and 2.4-D to regenerate shoots and to stimulate the callus growth. Shoot regeneration and callus growth were effective on media with 10.0mg/L BA alone, or 10.0mg/L BA and 0.1mg/L 2.4-D. In combined treatments of BA and 2.4-D, stmulation effects of cytocinin by 2.4-D also did not occur. Callus growth was decreased, accordiong to increasing the concentration of 2.4-D. In cimbined treatments of TDZ and 2.4-D in shoot regeneration and callus proliferation, stimulated effects of cytokinin by 2.4-D did not occur entirely. Media with 0.5∼1.0mg/L TDZ ingibited the regeneration and rooting of shoots, and callus growth from callus sections. Addition of 2.4-D on medium with TDZ ingibited the regeneration and rooting of shoots, and callus growth. Rooted plantdts were acclimatized in greenhouse. The plantlets were survived more than 98% in soil of vermiculite alone or mixed perlite 1 and vermiculite 1.

Effect of Cytokinin and Putrescine on Plant Regeneration from Leaf Explant of Rhodiola sachalinesis A. Bor (홍경천 (Rhodiola sachalinensis A. Bor) 잎절편으로부터 식물체 재분화에 미치는 Cytokinin과 Putresine의 영향)

  • The effects of cytokinin and putrescine on adventitious shoot induction from leaf explant of Rhodiola sachalinensis A. Bor were investigated. Among cytokinin used in this experiment, BA was more effective on adventitious shoot induction and shoot elongation than kinetin. Especially, 1 mg/L BA was the best to increase adventitious shoot induction (71%) and shoot elongation (3.0 mm). In addition, 100 mM putrescine in MS medium with 1 mg/L BA was higher in adventitious shoot induction (93%) and shoot elongation (3.8 mm) than single treatment of 1 mg/L BA. Adventitious shoots induced in this experiment rooted on 1/2 MS medium and acclimated over 95% on composed soil (peatmoss:sand=1:1).

Effect of Benzyladenine on the IAA-Induced Ethylene Production in the Primary Roots of Maize (옥수수 일차뿌리에서 benzyladenine이 IAA에 의해 유도된 에틸렌 생성에 미치는 영향)

  • This study was conducted to examine the effect of cytokinin ($N^6$-benzyladenine; BA) and/or an IAA on ethylene production of maize (Zea mays) primary roots. When the two hormones were applied exogenously, both hormones synergistically increased ethylene production, which was greater than the sum of the level of each hormone's effect. For example, the ethylene production was stimulated between about 87% and 170% of the control by $10^{-4}\;M$ BA with $10^{-4}\;M$ IAA for 8 hours respectively, whereas the ethylene production was increased by about 480% of the control when the two hormones were treated simultaneously. Such a synergistic effect was also found in changes in the activity and gene expression level of ACC synthase. However, in the case of ACC oxidase did not show any observable effects. Based on our results, it is possible to conclude that BA and IAA stimulated the ethylene production synergistically by affecting the ACC synthase in maize roots.

Expression of Kip-related protein 4 gene (KRP4) in response to auxin and cytokinin during growth of Arabidopsis thalia

  • The cell cycle is regulated by cyclin-dependent kinase (CDK)-cyclin complexes as well as other regulators. We isolated Kip-related protein 4 (KRP4) cDNA that encodes 289 amino acids including six conserved domains. To investigate the expression pattern of KRP4 as well as of other cell cycle-related genes associated with plant hormones, Arabidopsis seedlings were cultured on MS medium containing auxin or cytokinin. All seedlings treated with phytohormones displayed an increased proportion of cells in S phase. A higher proportion of cells in G2 phase was observed in seedlings treated with NAA. RT-PCR confirmed that the expression of KRP4 was decreased after treatment with phytohormones, and that CDKA and D-type cyclin transcription was increased. Additionally, mitotic cyclins were up-regulated by NAA treatment. These results suggest that KRP4 as well as other cell cycle-related genes might contribute to the control of plant growth in response to exogenous hormones.
