Vascular Plants and Specific Plants of Bulgapsan (불갑산 지역의 관속식물상과 특정식물)

  • The vascular plants in Bulgapsan, Younggwang-gun, Jeollanam-do, were consisted of 437 taxi; 104 families, 289 genus, 382 species, 51 varieties, 4 forma. From the floristic point of view, Bulgapsan is belongs to the South Province in Korea represented as Platycarya strobilacea, Cudrania tricuspidata, Mallotus japonicus, Lindera glauca, Euscaphis japonica, Meliosma myriantha, Euryu japonica, Halorrhagis micrantha, Hydeocotyle maritima, H. ramifloru, Chionanthus retusa, Trachelospermum asiaticum var. intermedium and Cymbidium goeringii, The Specific plants which is categorized to the Degree by the Ministry of Environment appeared to 25 taxa; Degree V was 2 species, Degree IV was 15 species, Degree III was 7 species and Degree II was 1 species. The mark evaluated by Degrees of five evaluation unit was 186.

The Vegetation of Seoimal-Lighthouse area in koje Island (거제 서이말등대 주변의 식생)

  • The vegetation of Seoimal-lighthouse area was investigated from August, 2001 to July, 2002. In order to analyze the vegetation of this area, synthesis table, actual vegetation map were prepared. The main community structures of this area represent evergreen broad-leaf community(about 48%), which are very good condition(nearly last sere) to preserve and 11communities(Camellia japonica community, Castanopsis cuspidnta var. thunbergii community, Neolitsea sericea community, Quercus serrata community, Quercus variabilis community, Carpinus corenna community, Srtrax japonica community, Pinus thunbergii community, Quercus acutissima community, Zelkova serrata community, Cinnamimum japonicum community) were classified by vegetation type. And Daphne kiusiana, Asirum maculatum, Cymbidium goeringii, Chloranthus glaber, Ligularia taguetii should be worthwhile species to conserve in this area.

Use of Gelatin Particle Agglutination Test for the Detection of Cymbidium mosaic virus in Cattleya Plants

  • Gelatin particle agglutination test (GPAT) was used to detect Cymbidum mosaic virus (CymMV) in Cattleya plants. Gelatin particles were coated with purified anti-CymMV immunoglobulin of 25-100 $\mu\textrm{g}$/ml and were subjected to several different concentrations of purified CyMfV as well as varying dilutions of orchid leaf extracts. The GPAT detected purified CymMV up to a minimum concentration of 10 $\mu\textrm{g}$/ml. CymMV was detected from crude sap extract of infected Cattleya leaves and roots up to 1:51,200 and 1:25,600 dilutions, respectively. However, the optimum range of leaf and root sap dilutions was between 50-100. Non-specific reactions were not encountered from any of the healthy orchid plants tested. The entire GPAT process was completed within 2-3 hours. This test was found to be very useful for the detection of CymMV in orchids because it is sensitive, economical, and easy to perform.

Identification of Streptomyces sp. Producing Antibiotics Against Phytopathogenic Fungi, and Its Structure

  • In order to develop a biocontrol agent that can effectively control Fusarium wilt on Cymbidium genus, the effectiveness of antagonistic microbes against the cause pathogen was screened. The selected microbe showed a broad spectrum of antifungal activity, and the culture broth of this microbe had better preventive effect on Fusarium wilt than the commercial chemical agent in the pot assay. This isolated strain, GBA-12, was identified as Streptomyces kasugaensis, and the antifungal substance was purified from a broth culture of GBA-12. This purified substance was identified as a polyene macrolide (YS-822A) that was newly discovered from Streptomyces kasugaensis, and it exhibited antifungal activity against several phytopathogenic fungi.

Isolations of the Orchid mycorrhizal Fungi from the Roots of the Korean native Orchids and Inoculations of the Isolates to Four different Orchids (한국 자생란에 난 균근균의 분리와 유묘난에 접종)

  • Ten isolates of the orchid mycorrhizal fungi were isolated from the roots of Korean native orchid plants (Cymbidium goeringii) which inhabitate mainly in southern and western areas of Korea. The growth rates and color of the isolates in potato dextrose agar (PDA) were various. Microscopic observations of the hyphae isolated were identified as Rhizoctonia repens and R. endophytica var endophytica or their related species. R. repens was isolated from the roots of the Korean native orchids, but R. endophytica var endophyica was only isolated from the roots of the commercial orchids introduced from foreign countries. Also, the polymorephic patterns of genomic DNA extracted from selected isolates were compared with those of DNA extracted from the orchid mycorrhizal fungi isolated previously and similar band patterns were observed among those isolates. Five isolates of R. repens were selected and cultured at the oatmeal agar for investigating their symbiosis with orchid plants. The symbiotic specificity between orchid plants and isolated orchid mycorrhizal fungi was observe by growing orchids about six months in the greenhouse. The symbiotic responses of the commercial orchid plants with selected isolates were quite different form different isolates due to the genetic variations.

Complementary DNA Cloning of Genomic RNA in Orchid Strain of Tobacco Mosaic Virus

  • Viral RNA was extracted from a purified orchid strain of tobacco mosaic virus (TMV-O) from Cymbidium "Grace Kelly". Polyadenylated viral RNAs were primed with Not I-oligo (dT) primer-adapter. First-strand cDNAs were reversely transcribed by Moloney murine leukaemia virus reverse transcriptase (RNAse H-), and then second-strand cDNAs were synthesized by RNase H and DNA polymerase I. The resulting double-stranded cDNAs were ligated into pSPORT1 vector and transformed into competent E. coli strain JM109 cells. The size of cDNAs within the recombinant plasmids was ranging from 0.9 to 3.9 kb. Among the selected clones, pTMO-0205 and -0210 covered the 3' half and the 5' half of the viral genomic RNA, respectively, which were covering more than 99% of the viral genemo size based on sequencing analysis. Two cDNA fragments which were 3.1 kb BamHI and NotI fragement released from pTMO-0.205 and 3.3 kb SalI and BamHI fragment released from pTMO-0210 were ligated with T4 DNA ligase. The clone was almost entire length, lacking only 31 nucleotides from the 5' terminus based on the sequencing result. This method was shown to be efficiently applicable to other plant viral gnomic RNA for the construction of cDNA.n of cDNA.

Vascular Plants of Manbokdae, Goribong and Suieongbong in the Baekdudaegan (백두대간 만복대.고리봉.수정봉 일대의 관식물상)

  • The vascular plants in Manbokdae-Yeowonjae section of the Baekdudaegan were consisted of 502 taxa; 104 families,305 genus,439 species, 60 varieties, 3 forma. From the floristic point of view, the Baekdudaegan of Manbokdae-Yeowonjae section belongs to the southern province in Korea. The rare and endangered plants designated by Korea Forest Service were Abies koreana, Pedicularis verticillata, Patrinia saniculaefolia, Hololeion maximowiczii, Lilium callolsum, Paeonia obovate, Tricyrtis dizutata and VioEa diamantica, and Cymbidium goeringii. The Korean endemic species were Abies koreana, Patrinia saniculaefolia, Vicia unijuga, Stewartia koreana, Cornus walteri, Weigela subsessilis, Saussurea seoulensis, Tyicyrtis dilatata, Hosta capitata, Clematis trichotoma et at.

Black Leaf Spot of Dendrobium phalaenopsis Caused by Fusarium moniliforme (Fusarium moniliforme에 의한 덴파레 검은점무늬병)

  • Black leaf spot disease occurred on Dendrobium phalaenopsis grown in farmer's fields located in Bonggangmyon, Gwangyang-eup, Jeonnam, Korea. Black small spots occurred on leaves at initial stage of infection and the infected leaves turned yellow from the tip. The yellow leaves were falled, resulting in stem blighting or eventual death of the entire plant. White mycelial colony of the causal fungus grown on potato dextrose agarturned dark violet later and optimum temperature for the mycelial growth was $25^{\circ}C$. The causal fungus isolated from the black leaf spot on D. phalaenopsis was identified as Fusarium moniliforme based on the mycological characteristics and pathogenicity. The fungus also caused same symptoms on leaves of Phalaenopsis sp. and Cymbidium sp. as well sa D. phalaenopsis by wound inoculation. This is the first report on black leaf spot of D. phalaenopsis caused by F.moniliforme in Korea.

The Ecological Characteristics of Native Habitat of Korean Native Wando Holly (llex X wandoensis) (자생완도 호랑가시나무 자생지의 생태학적 특성)

  • This study was conducted to analysis the ecological characteristics of "Wando HolIy"$(llex{\times}wandoensis)$ which is expecting as a possible ornamental tree with its beautiful leaves, flowers and fruits, and to find special breeding material which have particular characteristics and also provide correct environmental information which are required for commercial cultivation. The hill of Galmoon-Ri, Gunoei-Myon, Jeonnam Province, Korea, were reported as a original habitat of native $llex{\times}wandoensis$. The investigation shows that soil pH of native habitat was 6.18, hardness was 2.39, humidity was 38.6%, direction of slope is SSW, and average inclination was $15^{\circ}. The vegetation around $I.{\times}wandoensis$ is occupied by trees like an Quercus variabilis, Q. serrata, Q. acuta and so on. These are mixed forest of deciduous trees. Tree height reached up to about 12m and its coverage is about 30%. Q. salicina, Camellia japonica, I. integra, Cryptomeria japonica account for sub-tree layer. Tree height is around 8m and coverage is about 20%. Shrub layer was taken by Viburnum furcatum, Callicarpa japonica, Styrax japonica, Eurya japonica, Lindera obtusiloba, Ligustrum japonicum, Smilax china, Cornus walteri etc. Tree height reaches around l.2m and coverage is around 20%. Herb layer includes Trachelospermum asiaticum var. intermedium, Cymbidium goeringii, Dryopteris bissetiana, Disporum viridescens, Disporum smilacinum, Sasa borealis var. gracilis etc. Trachelospermum asiaticum var. intermedium, which are observed in southern evergreen zone, are dominant and its coverage is around 20%. The native habitat was in spoiled condition because of artificial thin out.

Construction of Complementary DNA Library and cDNA Cloning for Cy Strain of Odontoglossum Ringspot Virus Genomic RNA (오돈토글로썸 윤문 바이러스 Cy계통 게놈 RNA의 cDNA 구축 및 유전자 크로닝)

  • Genomic RNA was extracted from Cy strain of odontoglossum ringspot tobamovirus (ORSV-Cy) isolated from infected leaves of tobacco cv. Samsun. Size of the genomic RNA was about 6.6 kb in length. The genomic RNA was fractionated using Sephadex G-50 column chromatography into 2 fractions. They were polyadenylated at their 3'-end using E. coli poly(A) polymerase. Polyadenylated viral RNA was recovered by oligo (dT) primer adapter containing NotI restriction site and Moloney murine leukemia virus SuperScript reverse transcriptase (RNase H-). Second-strand cDNA was synthesized by using E. coli DNA ligase, E. coli DNA polymerase I and E. coli RNase H. Recombinant plasmids containing cDNAs for ORSV-Cy RNA ranged from about 800 bp to 3,000 bp. Among the selected 238 recombinants, pORCY-124 clone was the largest one covering 3'-terminal half of the viral RNA. This clone contained two restriction sites for EcoRI and XbaI and one site for AccI, AvaI, BglII, BstXI, HindIII, PstI, and TthIII 1. respectively. The clone contained partial viral replicase, a full-length movement protein and a complete coat protein genes followed by a 3' untranslated region of 414 nucleotides based on restriction mapping and nucleotide sequencing analyses. Clones pORCY-028, -068, -072, -187 and -224 were overlapped with the pORCY-124. Clones pORCY-014 and -095 covered 5' half upstream from the middle region of the viral RNA, which was estimated based on restriction mapping and partial sequence analysis. Constructed cDNA library covered more than 90% of the viral genome.
