Anti-Inflammatory Principles from the Fruits of Evodia rutaecarpa and Their Cellular Action Mechanisms

  • The fruits of Evodia rutaecarpa Benth (Rutaceae) has long been used for inflammatory disorders and some anti-inflammatory actions of its constituents such as dehydroevodiamine, evodiamine and rutaecarpine were previously reported. Since the pharmacological data is not sufficient to clearly establish the scientific rationale of anti-inflammatory medicinal use of this plant material and the search for its active principles is limited so far, three major constituents (evodiamine, rutaecarpine, goshuyuamide II) were evaluated for their anti-inflammatory cellular action mechanisms in the present study. From the results, evodiamine and rutaecarpine were found to strongly inhibit prostaglandin $E_2$ synthesis from lipopolysaccharide-treated RAW 264.7 cells at $1-10{\mu}M$. Evodiamine inhibited cyclooxygenase-2 induction and NF-kB activation, while rutaecarpine did not. On the other hand, goshuyuamide II inhibited 5-lipoxygenase from RBL-1 cells $(IC_{50}=6.6{\mu}M)$, resulting in the reduced synthesis of leukotrienes. However, these three compounds were not inhibitory against inducible nitric oxide synthase-mediated nitric oxide production from RAW cells up to $50{\mu}M$. These pharmacological properties may provide the additional scientific rationale for anti-inflammatory use of the fruits of E. rutaecarpa.

Effect of trans-10, cis-12 Conjugated Linoleic Acid on Production of Prostaglandin E2, Cyclooxygenase-2 and 5-lipoxygenase in Lipopolysaccharide-Stimulated Porcine Peripheral Blood Mononuclear Cells

  • The objective of this study was to examine the effect of trans-10, cis-12 conjugated linoleic acid (t10c12-CLA) on the expression of cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LOX) pathway in lipopolysaccharide (LPS)-stimulated porcine peripheral blood mononuclear cells (PBMCs). t10c12-CLA was treated with different concentrations in culture medium of LPS$na{\ddot{i}}ve$ and LPS-stimulated PBMCs. The mRNA expressions of prostaglandin $E_2$ ($PGE_2$)-synthase, COX-2 and 5-LOX were measured using quantitative real-time PCR. In addition, the production levels of $PGE_2$ and 5-LOX in culture supernatant from PBMCs with or without LPS were assessed by ELISA. In LPS$na{\ddot{i}}ve$ PBMCs, treatment of t10c12-CLA significantly (p < 0.05) increased the mRNA expressions of PGE2 synthase and 5-LOX compared to vehicle control. Expression of COX-2 mRNA did not show significant difference compared to vehicle control by t10c12-CLA treatment in LPS$na{\ddot{i}}ve$ PBMCs. However, the addition of LPS in PBMCs markedly (p < 0.05) increased the mRNA expression of COX-2, $PGE_2$ synthase and 5-LOX, and also significantly (p < 0.05) enhanced the production of $PGE_2$ and 5-LOX relative to LPS$na{\ddot{i}}ve$ PBMCs, respectively. However, the addition of t10c12-CLA significantly (p < 0.01) suppressed the LPS-induced excessive expression of COX-2, $PGE_2$ synthase, and 5-LOX compared to those of PBMCs treated with LPS alone. The production levels of $PGE_2$ and 5-LOX in culture supernatant from LPS-stimulated PBMCs were also significantly (p < 0.05) inhibited by the treatment of t10c12-CLA compared to LPS alone. These results suggested that t10c12-CLA has an anti-inflammatory effect via dual inhibition of COX-2 and 5-LOX with gene expression and production level in LPS-stimulated porcine PBMCs. Therefore, it was thought that t10c12-CLA can attenuate the inflammatory response by down-regulation of eicosanoids production.

  • Arachidonic acid (AA), which is stored in membrane glycerophospholipids, is liberated by phospholipase $A_2$ (PLA$_2$) enzymes and is sequentially converted to cyclooxygenases (COXs) and lipoxygenases (LOXs) then to various bioactive PGs, and LTs. In order to find the specific inhibitors of AA metabolism especially PLA$_2$, COX-2, 5-LO and lyso PAF acetyltransferase, 120 Korean residential plants extracts were evaluated for their inhibitory activity on PGD$_2$, LTC$_4$ production from cytokine-induced mouse bone marrow-derived mast cells (BMMC) and arachidonic acid released from phospholipid and PAF production from lyso PAF. From this screening procedure, methanol extract of ten indigenous plant such as Salix gracilistyla, Sedum kamtschaticum, Cirsium chanroenicum, Hypericum ascyron, Astilbe chinensis, Agrimonia pilosa, Aristolochia manshuriensis, Vodia daniellii, Pyrola japonica, Styrax obassia were found to inhibit production of inflammatory mediators in vitro assay system.

The Role of Cyclooxygenase Metabolites in the Pathogenetic Mechanism of Endotoxin-Induced Acute Lung Injury in Domestic Pigs (내독소에 의한 돼지의 급성 폐손상에서 Cyclooxygenase 대사물의 역할에 관한 연구)

  • Background:It has been suggested that the cyclooxygenase metabolites play an important role in changes of early hemodynamic parameters in the endotoxin-induced acute lung injury. But there have been many debates about their role in the late increase of alveolar-capillary permeability, and it is not known whether they act directly or indirectly through oxygen free radicals which have been known to be produced during the metabolic process of cyclooxygenase pathway. So we performed this study to identify the pathogenetic role of cyclooxygenase metabolites in the endotoxin-induced acute lung injury in domestic pigs. Method: We infused endotoxin into 8 domestic pigs; endotoxin only (n=3), and pretreatment with indomethacin (n=5). We observed the sequential changes in hemodynamic parameters, the concentration of plasma oxidized glutathione (GSSG) in pulmonary arterial and venous blood, and albumin content in bronchoalveolar lavage fluid (BALF). Results: 1) While cardiac output decreased, mean pulmonary arterial pressure, pulmonary vascular resistance, and alveolar-arterial oxygen difference increased over phase 1 (0-2hr) and phase 2 (2-4.5hr) by endotoxin, indomethacin attenuated the decrease in cardiac output during phase 1 and increase in mean pulmonary arterial pressure, pulmonary vascular resistance, and alveolar-arterial oxygen difference during both phases. 2) The increase in plasma GSSG content during phase 2 was not attenuated by indomethacin. 3) The content of BALF albumin was significantly lower in indomethacin groups than that of endotoxin group. Conclusion: These results suggest that it is likely that cyclooxygenase metabolites have an effect on endotoxin-induced acute lung injury during both phases probably through direct action.

Antioxidant and Anti-inflammatory Activities of Ethanol Extracts from Medicinal Herb Mixtures (단일 생약재 및 생약복합제 에탄올추출물의 항산화 및 항염증 활성)

  • In this study, antioxidant and anti-inflammatory effects of ethanol extracts from medicinal herb mixtures were investigated in vitro. Contents of total polyphenols in ethanol extracts from Houttuynia cordata Thunb. (HC), Carthamus tinctorius L. (CT), and MIX-2 (Houttuynia cordata Thunb., Acanthopanax gracilistylus Smith, Carthamus tinctorius L., Kalopanax pictus) were 58.98, 60.79 and 57.74 mg/g respectively. Radical scavenging activities of the ethanol extracts were examined using ${\alpha},{\alpha}$-diphenyl-${\beta}$-picrylhydrazyl radicals and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) assay. Anti-inflammatory effects of ethanol extracts from medicinal herbs composites were examined using 5-lipoxygenase (5-LO), cyclooxygenase-2 (COX-2) and nitric oxide (NO) inhibition assays. The ethanol extracts from medicinal herb mixtures showed higher 5-LO and COX-2 inhibition activities than those from single medicinal herbs. The ethanol extracts from medicinal herb mixtures also showed strong anti-inflammatory activity through inhibition of NO production.

Antioxidative and Antiallergic Effects of Aronia (Aronia melanocarpa) Extract (아로니아(Aronia melanocarpa)로부터 유래한 추출물의 항산화 및 항알레르기 효능)

  • The purpose of this study was to investigate the antioxidative and antiallergic effects of aronia extract. The aronia extract was tested for various antioxidative potentials (scavenging activity of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and superoxide anion radical) and inhibitory effect of 5-lipoxygenase (5-LO) and cyclooxygenase (COX). In aronia extract, polyphenol content was $81.6{\pm}3.3\;mg/g$ and flavonoid content was $5.15{\pm}0.16\;mg/g$. DPPH radical-scavenging activity ($SC_{50}$) of aronia extract was $6.15{\pm}0.343\;ppm$. $SC_{50}$ value of aronia extract against superoxide anion radical was $6.99{\pm}1.28\;ppm$. In addition, $IC\_{50}$ values of aronia extract against 5-LO, COX-1, and COX-2 were $42.07{\pm}0.15\;ppm$, $24{\pm}4.29\;ppm$, and $24.3{\pm}3.5\;ppm$, respectively. These results suggest that aronia extract may be useful for the prevention or treatment of allergic disease.

Anti-Inflammatory Activity of the Total Flavonoid Fraction from Broussonetia papyrifera in Combination with Lonicera japonica

  • To establish the anti-inflammatory activity of the total flavonoid fraction of the root barks of Broussonetia papyrifera (EBP) and a new formula, the ethanol extract of the root barks of B. papyrifera was fractionated with ethylacetate, yielding the hydrophobic prenylated flavonoid-enriched fraction. EBP and the ethanol extract of the whole Lonicera japonica (ELJ) plant were then mixed at a ratio of 1:1 (w/w) to give a new preparation (BL) in the hope of obtaining an optimal formula with a higher anti-inflammatory activity. Evaluation of the effects of these preparations on A23187-treated rat basophilic leukemia (RBL-1) cells revealed that EBP potently inhibited 5-lipoxygenase (5-LOX), while ELJ showed weak inhibition. Additionally, the mixture (BL) clearly showed stronger inhibitory effects against 5-LOX than either preparation alone. These preparations also inhibited cyclooxygenase-2-catalyzed $PGE_2$ and inducible nitric oxide (NO) synthase-catalyzed NO production by lipopolysaccharide-treated RAW 264.7 cells. When tested against arachidonic acid-induced mouse ear edema, EBP showed strong inhibitory activity at doses of 5-200 mg/kg when administered orally, but BL had obviously stronger inhibitory effects. When tested against ${\lambda}$-carrageenan-induced paw edema in mice, BL showed a potent and synergistic anti-inflammatory effect. In addition, in the acetic acid-induced writhing test, BL was found to have strong analgesic activity at 50-400 mg/kg. Taken together, these results indicate that each of these preparations exert anti-inflammatory activity in vitro and in vivo. In particular, BL showed stronger anti-inflammatory activity than EBP, and these anti-inflammatory effects were partially related to the inhibition of eicosanoid and NO production. BL may be useful for the treatment of human inflammatory disorders.

Modulation of arachidonic acid metabolism and inflammatory process in macrophages by different solvent fractions of Glasswort (Salicornia herbacea L.) extract (큰포식세포에서 퉁퉁마디 추출물의 아라키돈산 대사효소조절 및 항염증 활성)

  • Glasswort has attracted an attention because of its interesting physiological actions. In this study, the effects of glasswort on inflammatory events including nitric oxide (NO) synthesis and arachidonic acid metabolism in cultured RAW264.7 macrophages were investigated. A series of solvent fractions, including fractions of hexane (Fr.H), ethyl ether (Fr.E), ethyl acetate, butanol, and water, were prepared from a 70% methanol extract of glasswort. Among the fractions, Fr.E showed the strongest inhibition of NO synthesis and inducible NO synthase (iNOS) expression in lipopolysaccharide (LPS)-stimulated macrophages. At a concentration of $80{\mu}g/mL$, Fr.E decreased the NO and iNOS levels by 73 and 77%, respectively, after 24 h. Fr.E showed the most potent inhibitory effects on the expressions of cytosolic phospholipase $A_2$ and cyclooxygenase-2 with $IC_{50}$ values of 33.4 and $27.9{\mu}g/mL$, respectively. Fr.H and Fr.E also significantly inhibited 5-lipoxygenase expression in LPS-stimulated macrophages. These results suggest that the hydrophobic fractions of glasswort possess anti-inflammatory activities through modulating the arachidonic acid metabolism and NO synthesis.

Beneficial Effects of Phyto-Extract Complex (CME) on Degenerative Arthritis (식물추출복합물(CME)의 퇴행성관절염 개선효과)

  • Objectives : Degenerative arthritis arises from several physiological factors. The purpose of this study is to investigate the beneficial effects of Phyto-extract Complex (CME) on degenerative arthritis. Methods : CME is composed of extracts of mulberry (Morus alba L.) fruit, mulberry leaves and black beans (Glycine max (L.) Merr.). To measure the toxicity of CME, we performed the single-dose toxicity study. For the evaluation of its effects on degenerative arthritis, we examined the inhibition of cyclooxygenase-2 (COX-2) activity, using in vitro enzyme activity assay, the reduction of protein expression of COX-2, 5-lipoxygenase (5-LO), and inducible nitric oxide synthase (iNOS) in RAW264.7 cells which were stimulated by lipopolysaccharide (LPS). We also examined the serum level of prostaglandins (PGs) and injury of the knee joint cartilage, using animal model of degenerative arthritis induced by mono-sodium iodoacetate (MIA). Results : CME did not have any toxicity in single-dose toxicity study. The CME inhibited the activity of COX-2 and could reduce the protein expression of COX-2, 5-LO and iNOS in RAW264.7 cells. The CME also reduced the serum level of PGs and prevented from the cartilage injury of knee joint in animal model of degenerative arthritis induced by MIA. Conclusions : Taken altogether, the CME could be useful for the improvement of degenerative arthritis through its various anti-inflammatory activities and prevention from the cartilage injury of knee joint.

  • Prostaglandin은 인체의 여러 조직에서 생성되어 체내에 널리 분포되어 있지만 그 함유량은 극히 적다. 또한 대사산물의 반감기가 매우 이에 대한 연구결과를 알기 위하여는 조직 또는 체액을 추출하여 세심한 주의가 필요하고 측정방법에 있어서 정밀성이 요구된다. 또 여러가지 대사산물과 전구물질을 분리할 수 있는 방법이 요구된다. Prostaglandin가 생성되고 대사되는 과정이 매우 복잡하기 때문에 보다 정확하게 여러가지 대사산물을 분리할 수 있는 방법이 요구된다. 한편, prostaglandin은 전신의 거의 모든 조직에 분포하며 그 이용도 다양하다. 뿐만 아니라, prostaglandin이 임상적으로 생체기능의 조절(고혈압, 신기능조절, 난소와 자궁의 생리 등) 및 prostaglandin analogue의 길항약물 및 생합성 억제약물로서도 중요한 의미를 가지고 있다. 본 연구에서는 이러한 관점에서 prostaglandin을 측정하는데 있어서: 1. 조직의 추출과 조작에 대한 일반적 방법 2. 생체조직에서 eicosanoid의 추출 방법 3. Prostaglandin, thromboxane 및 leukotriene에 대하여 평활근을 이용한 생물학적 검정 4. Eicosanoids의 방사면역학적 검정 5. 효소면역 검정법 6. Cyclooxygenase의 측정, 정체 및 특성 7. Lipoxygenase의 특성과 측정 8. 지질과산화 반응의 측정 등을 다루었다.
