Synthesis and Characterization of Cationic and Anionic Cyclodextrin Oligomers and Their Use in Layer-by-Layer Film Formation

  • Ionically modified ${\beta}$-cyclodextrins, which have excellent water-solubility, have been interested in purification technology as well as drug carrier system. The present study summarizes the synthesis and characterization of cationic and anionic ${\beta}$-cyclodextrin (${\beta}$-CyD) products using by polycondensation. The oligo (${\beta}$-CyD)s are synthesized from ${\beta}$-CyD, epichlorohydrin (EP) and choline chloride (CC; for cationic polymer) or chloroacetic acid (CAA; for anionic polymer) through one step polycondenstaion process. Unlike the previous studies, we successfully purified the ionic ${\beta}$-CyD condensation products from the ${\beta}$-CyD reaction mixtures and accomplished a great level of structural analysis. The detailed structural analysis of these ionic ${\beta}$-CyD compounds is done by $^1H$ NMR, MALDI-TOF as well as GPC analysis and confirms the formation of oligomers with a few units of ${\beta}$-CyD. We found that the sequence of reactant addition also could effect on the molecular weight of the resulting product as well as the molar ratio of the reactants. Finally, we used the cationic and anionic ${\beta}$-CyD oligomers for fabricating multilayer films by layer-layer process.

Increased Production of Digoxin by Digitoxin Biotransformation Using Cyclodextrin Polymer in Digitalis lanata Cell Cultures

  • Addition of ${\beta}$-cyclodextrin (${\beta}$-CD) polymer during the biotransformation of digitoxin into digoxin using cell suspension cultures of Digitalis lanata enhanced the conversion yield. Digitoxin showed better adsorption to CD polymer compared to digoxin, so that the optimization of addition time was found to be necessary. In the case of adding CD polymer 24 hours after the feeding of substrate digitoxin, the highest digoxin production could be achieved. At this period, digitoxin was almost consumed by cells and productivity was proportionally enhanced according as the amount of substrate was increased. Immobilization of CD polymer did not promote the biotransformation. When 3.33 g/L of CD selective inclusion complex formation could be expected. Adsorption rate was found to be rapid and saturation was obtained within 10 hours of contact.

  • Although ${\beta}$-cyclodextrin (${\beta}$-CD) has been used as medicine, agrichemical, food, cosmetic, antioxidant, anti-volatile agent, anti-hygroscopic agent, fading-protecting agent, and emulsifier due to its ability to form inclusion complex by enclosing another molecule (guest molecule), it has been restricted in practical application because of its low solubility in water and organic solvent. In this study, ${\beta}$-CD derivative as a new polyol with inclusion characteristics against Bisphenol A and cyclopropane, foaming agent for polyurethane (PU), and with improved solubility was synthesized, characterized and used to formulate rigid PU foam with better thermal conductivity and compressive strength compared to that from commercial polyols.

Raman Spectra of Nitrophenol Molecules Included in Cyclodextrin Polymers Cross-linked with Epichlohydrine

  • Inclusion complexes of the p-nitrophenol with ${\beta}$-cyclodextrin (CD), sulfated ${\beta}$-CD, and ${\beta}$-CD polymer cross-linked with epichlorohydrine (EP) were prepared and characterized by Raman spectroscopy. The intensity of vibration peaks of the C-O and C-N at 1284 and $856cm^{-1}$ of the p-nitrophenol in the presence of EP-linked CD polymer was remarkably increased, respectively. The vibration modes at 1284 and $856cm^{-1}$ are assigned to the out-of phase C-C-O stretching mode and the C-N stretching mode, respectively. The vibration peaks at 1284 and $856cm^{-1}$ increased with increasing the content of EP-linked CD polymers, while decreased with increasing the p-nitrophenol content. Furthermore, the vibration mode of the $NO_2$ symmetric stretch at $1344cm^{-1}$ enhanced with increasing the content of p-nitrophenol.

Dissolution and Stability Enhancements of Trimethoprim by ${\beta}-Cyclodextrin$ Polymer Inclusion Complexation (트리메토프림과 ${\beta}$-시클로덱스트린 고분자 간의 포접복합체 형성에 의한 용출 및 안정성 향상)

  • To improve the solubility and dissolution rate of trimethoprim (TMP), which is slightly soluble drug, its inclusion complexes were prepared and studied in this experiment. Inclusion complexes of TMP with ${\beta}-cyclodextrin$ and ${\beta}-cyclodextrin$ polymer (CDPS) were prepared according to Fenyvesi method. These were compared with TMP and its physical mixture with CDPS. Water, diluted hydrochloric acid and phosphate buffer solution were used as dissolution media. And accelerated stability test was studied at $50,\;70\;and \;80^{\circ}C$. It was found that solubility and dissolution rate of inclusion complexes were increased in water. Especially, the solubility and dissolution rate of TMP was found to be markedly increased by inclusion complexation with CDPS. In stability test, ${\beta}-cyclodextrin$ inclusion complexes were more or less stable than TMP alone. This tendency was not led in CDPS. Consequently, CDPS was useful in increasing dissolution rate and stability of TMP.

  • Polypyrrole is a typical electrical conducting polymer, which has an excellent charge transport property. Cyclodextrins are a group of toxic-free and cyclic oligosaccharide molecules, capable of capturing low molecular weight chemicals. Considering these advantages, hybrid materials of polypyrrole and cyclodextrin can be used to detect hazardous compounds. Cyclodextrin molecules can accommodate toxic chemicals by the formation of host-guest complexes and generate electric signals, which are effectively delivered by polypyrrole backbone. In this study, the polypyrrole/cyclodextrin hybrid material was prepared using a facile wet method and included into a hydrogel. Subsequently, it was applied to a simple sensor system with a gold-patterned electrode for the detection of potentially hazardous material, methyl paraben. Compared with pristine polypyrrole, it was found that the polypyrrole/cyclodextrin hybrid showed an improved performance. This study can be an example of using environmentally benign conducting polymer/cyclodextrin hybrids as sensing media.

Synthesis and Characterization of Hyaluronic Acid-α-Cyclodextrin Conjugate as the Potential Carrier of PEGylated Drugs

  • The hyaluronic acid (HA) conjugate bearing $\alpha$-cyclodextrin ($\alpha$-CD) was synthesized as the potential carrier of poly(ethylene glycol) (PEG)-drug conjugates. The HA conjugate was prepared by the reaction between the carboxylic acid of HA and the primary amine of $\alpha$-CD in the presence of 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide and 1-hydroxybenzotriazole. The chemical structure of the conjugate was confirmed using $^1H$ NMR and FT-IR spectroscopy. The conjugate could form nano-sized particles in the presence of PEG by forming the inclusion complexes between $\alpha$-CD at the backbone of HA, which was demonstrated using electrophoretic light scattering and field emission transmission electron microscopy. It is anticipated that this novel kind of nanoparticles can serve as a useful delivery system for PEGylated drugs.

Effect of Guest Molecules on Structure and Properties of Polymer/beta-Cyclodextrin Inclusion Compound Hybrid Films (고분자/베타-사이클로덱스트린 포접 화합물로 이루어진 고분자 혼성체 필름의 물성 및 구조에 미치는 게스트 분자의 영향)

  • In this study, the effect of molecular features of guest molecules on the structure, property, and formation of poly(vinyl alcohol) (PVA)/beta-cyclodextrin (bCD) inclusion compound hybrid films was investigated using three types of guest molecules such as hydroquinone (HQ), arbutin (AB), and tranexamic acid (TA). First, the successful formation of inclusion compounds between bCD and the guest molecules, and polymer/inclusion compound hybrid were proved using Raman spectroscopy. The effect of bCD-based inclusion compounds on the structure and property of PVA matrix composites containing inclusion compounds was also studied using X-ray diffraction (XRD) and thermal analyses such as differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA). It was notable that the effect of TA to the crystalline structure of the PVA was significantly different from that of using other guest molecules including HQ and AB. It was also supported by a simple molecular simulation result. This article will be a good example for demonstrating the effect of molecular characteristics on the inclusion compound formation in polymer films, which can provide important information for relevant future research.
