Antifungal Activity of Securinine against Some Plant Pathogenic Fungi

  • The alkaloid securinine was assessed against spore germination of some plant pathogenic and saprophytic fungi (Alternaria alternata, Alternaria brassicae, Alternaria brassicicola, Curvularia lunata, Curvularia maculans, Curvularia pallenscens, Colletotrichum musae, Colletotrichum sp., Erysiphe pisi, Helminthosporium echinoclova, Helminthosporium spiciferum, Heterosporium sp.). Spore germinations of all the tested fungi were inhibited. Alternaria brassicicola, C. lunata, C. pallenscens and H. spiciferum were highly sensitive as complete inhibition of spore germination was observed at very low concentrations (200 ppm).

  • Withametelin, a steroidal compound isolated from leaves of Datura metel L.(Solanaceae), showed antifungal activity against some plant pathogenic as well as saprophytic fungi tested in vitro. Except Curvularia maculans and Colletotrichum sp., spore germination of all the other 23 fungi was inhibited significantly at 125 to 1000 ppm. Out of the 25 fungi tested, Curvularia sp., Cercospora abelmoschi, Heterosporium sp., Erysiphe cichoracearum and Ustilago cynodontis were most sensitive as there was complete inhibition of germination at 1000 ppm. Similarly, spores of Alternaria brassicae, Curvularia lunata and Helminthosporium pennisetti showed less than 3% germination at the same concentration. Rest of the fungi showed more than 15% spore germination at the same concentration.

Effect of Different Carbon and Nitrogen Compounds on the Growth and Sporulation of Curvularia clavata (야자나무 고조병균(枯調病菌)의 생장(生長)과 포자형성(胞子形成)에 대한 탄소 및 질소의 효과)

  • The effect of different carbon and nitrogen compounds on the growth and sporula­tion of Curvularia clavata Alcorn (Herberium No. IMI264075) has been studied. All the carbon sources tried were well utilized by the pathogen though glucose, and sucrose supported the best growth while glucose, maltose and sucrose the sporulation of the fungus. Of the nine nitrogen compounds, L-glutamic acid supported the best growth while aspartic acid and L-glutamic acid the sporulation of the fungus. Growth and sporulation were generally better with organic than inorganic nitrogen sources. Ammonium sulphate was the best inorganic source. A sudden drop of pH value of the culture media after 4 days of incubation did not favour good growth of the fungus.

  • Phytoterpenoid isolated from Artabotrytis odoratissimus inhibited spore germination of some plant pathogenic as well as saprophytic fungi e.g. Alternaria alternata, A. solani, Cercospora sp., Curvularia maculans, C. pennisetti, Fusarium udum, Helminthosporium echinochlova, H. frumentacie, H. penniseti and Ustilago cynodontis. In Curvularia maculans and H. frumentacie, spore germination was completely inhibited at 2000 ppm. However, Curvularia maculans and C. pennisetti showed considerable sensitivity to this chemical even at 500 ppm.

  • Natural sclerotium formation in two species of Curvularia was observed. The sclerotia were spherical to elongated. The scanning electron microscopical observations of sclerotia revealed that the sclerotia were of two distinct layers of cells, outer loosely woven hyphae and inner contact layer of cells. Different lights, viz. red, blue, green, fluorescent or addition of culture filtrate of Sclerotium rolfsii in the medium did not affect sclerotium formation.

  • The alkaloid fuyuziphine was isolated from the whole plant of Fumaria indica. It had inhibitive effect against spore germination of some plant pathogenic fungi (Collectotrichum sp., C. gloeosporioides, C. falcatum, Curvularia maculans, C. lunata, Erysiphe cichoracearum, Helminthosporium pennisetti, Oidium erysiphoides, Ustilago cynodontis, Alternaria chieranthi, A. mel-ongenae, A. brassicicola and A. solam). Curvularia lunata, Oidium erysiphoides, Alternaria brassicicola and A. solani did not germinate at 750 and 1000 ppm and Colletotrichum gloeosporioides, C. falcatum, Curvularia maculans were inhibited at 1000 ppm for 24 hr incubation. Germination of most fungi was significantly inhibited at $100{\sim}750ppm$.

Studies on the Fungi Associated with Ear Blight of Rice (벼 이삭마름병해에 관여하는 균류에 관한 연구)

  • This experiment was conducted in order to determine the fungi associated with ear blight of rice and their chemical control. 1, Pyricularia oryzes, Helminthosporium oryzne, Epicoccum purpurascens, Curvularia lunata, Cladosporium cladosporoides and Alternaria tenuis et al. were mainly detected in large quantities from the discolored seeds of recommended varieties but Trichoconis padwickii, Fusarium dimerum, and Nigrospora oryzae were mainly detected from Tongil. 2. Pyricularia oryzae, Nigrospora oryzae, and Curvularia lunate et at. were from the necks; Helminthosporium oryzae, Trichoconis padwickii, Stemphylium sp. , Cladosporium cladosporoides, and Alternaria tenuis et at. were from the branches of ear; and Phoma spp. was mainly detected from the kernels. 3. In the laboratory Difolatan WP $0.13\%$ gave good control of Helminthosporium oryzae, Cladosporium cladosporoides, and Alternaria tenuis, et al.; and Dithane M-45 WP $0.2\% gave especially good control of Pyricularia oryzae, Trichoconis padwickii, and Curvularia lunate. 4. In field experiments, the appropriate date of chemical spray was milk ripening stage or panicle emergence stage. Under these conditions Hinosan Ec and Kasugamin Ec were more effective than Difolatan WP and Dithane M-45 WP. However, the assessment of ear blight was complicated by a severe outbreak of neck blast.

Antifungal Activity of Methanolic of Centella asiatica and Andrographis panicuiata

  • The antifungal activity of methanolic extracts of Centella asiatica and Andrographis paniculata leaves was observed against fourteen fungi, viz., Alternaria alternata, A. brassicae, A. brassicicola, A. solani, A. tenuissima, Cercospora blumae, Curvularia lunata, C. penniseti, and Drechslera monoceras, D. oryzae, D. turitica, Fusarium albizziae and F. udum. Different concentrations of the methanolic extract (1000, 2000, 3000, 5000, 7000, 10000ppm) were used. The effect of mixed leaf extract (1500 ppm of C. asiatica + 1500 ppm of A. paniculata) and its 1:2 ad 1:4 dilutions were also studied. The individual extracts of both the plants showed significant inhibitory effect on spore germination of all the fungi tested. F. udum, F. albizzae, D. oryzae, D. turtica, and D. monoceras were particularly sensitive to these extracts. In general, the extract of C. asiatica showed a higher inhibitory effect in all concentrations against all the fungi as compared to A. paniculata, except for A. brassicae A. solani, D. oryzae, D. penniseti and Curvularia sp. The inhibitory effect of extracts increased when they were used in combination with or without dilutions against A. brassicicola, A. solani A. brassicae, A. alternata, A. tenussima, C. blumae, C. lunata, C. penniseti and Curvularia species. Higher efficacy of active ingredient of these extracts under field condition is envisaged against plant pathogens.

Two Unrecorded Endophytic Fungi Isolated from Funaria hygrometrica in Korea (표주박이끼(Funaria hygrometrica)에서 분리된 2종의 국내 미기록 내생균)

  • In the present study, we isolated endophytic fungal strains from the rhizoids of the moss Funaria hygrometrica. The isolated strains were identified based on morphological characteristics and analysis of the internal transcribed spacer (ITS) and large subunit (LSU) rDNA sequence regions. Consequently, we confirmed the presence of two endophytic fungal species, Curvularia protuberata and Didymella anserina, which have not been reported in Korea previously. Here, we describe the morphological characteristics and molecular analysis results of these fungal species.
