흑마늘과 남해특용작물을 혼합 첨가하여 제조한 소시지의 품질특성 (Quality Characteristics of Sausage Prepared with Black Garlic Extract and Dried Powder of Specialized Crops Cultivated in Namhae)

  • $15^{\circ}Bx$ 흑마늘 추출액 1%에 울금(ST1), 백년초(ST2), 아스파라거스(ST3) 및 알로에(ST4) 분말을 각각 0.3%씩 첨가한 소시지를 제조하여 $8^{\circ}C$에서 4주간 저장하면서 품질 변화를 살펴보았다. 색도는 흑마늘 추출물이 첨가된 대조구에 비해 백년초 분말을 첨가한 ST2에서 명도, 적색도가 가장 높게 측정되었으며, 황색도는 아스파라거스 분말을 첨가한 ST3에서 높게 측정되었다. 전단가는 저장 0일에 대조구가 $854.19{\pm}38.21cm/kg^2$으로 가장 높게 측정이 되었다. 산도는 pH와 유사한 경향으로 ST4가 가장 높았으며 대부분의 처리구가 저장 2주까지 증가하다가 감소하는 경향이었다. 총당은 저장 0일에 ST4가 $17.46{\pm}0.92mg/100g$으로 가장 높은 함량이었으며 전체적으로 저장기간의 경과에 따라 증가하였다. TBARS 함량은 처리구가 대조구에 비해 낮아지는 경향을 보였으나 전체적으로 저장기간이 경과함에 따라 모든 시료에서 유의적으로 증가하였다. ABTS 라디칼 소거능은 $1000{\mu}g/mL$ 농도에서 저장 0일에 $89.11{\pm}0.66{\sim}99.22{\pm}0.09%$의 높은 소거능을 보였다. 결과적으로 화학적 및 물리적 특성 결과와 비교했을 시 흑마늘에 특화작물의 첨가가 상품화 가능성이 있는 것으로 판단되었다.

천연 추출물을 이용한 화학감작제 후보물질 탐색 (Screening of Chemosensitizer Candidates Using Natural Extracts)

  • 본 연구에서는 다양한 천연 추출물의 P-gp 저해능의 고속 탐색을 통하여 새로운 화학감작제 후보물질을 발굴하고자 하였다. P-gp 활성에 대한 천연 추출물의 저해능은 P-gp이 과발현된 L-MDR1 세포주를 이용하여 대표적인 P-gp 기질 약물인 calcein AM의 세포 내 축적 정도를 측정함으로써 평가하였다. 강황 및 울금은 가장 강력한 P-gp 기능 저해를 나타내었고, 이외에도 Mentrasto, 취호초, 봉출, Rakta chandan, 강진향, 소목, 노회 등의 순으로 P-gp 기능 저해능을 보였다. 이들 추출물에서 P-gp 저해능을 보이는 성분을 확인하기 위하여 LC/MS/MS 분석을 수행한 결과, 기존에 P-gp 활성을 저해한다고 잘 알려진 curcumin 이외에, 다양한 플라보노이드 화합물이 질량 스펙트럼 DB 검색을 통하여 확인되었다. In vitro 연구 결과를 통하여 상기의 천연 추출물이 P-gp 활성을 저해하는 성분을 함유하고 있음을 확인하였다. 향후 이들 천연 추출물의 화학감작제 후보물질로의 사용 가능성에 대한 in vivo 연구가 필요할 것이다.

  • An in vitro and in vivo studies were conducted to evaluate the antibacterial efficacy of certain botanicals viz., rhizomes of turmeric (Curcuma longa) and leaves of amla (Phyllanthus emblica), asparagus (Asparagus racemosus), bael (Aegle marmelos), boerhavia (Boerhavia diffusa), garlic (Allium sativum) and basil (Oscimum basicilum) against bacterial pathogens viz., Staphylococcus sp., Bacillus sp. and Klebsiella cloacae, of silkworm, Bombyx mori. Asparagus and basil, amla and boerhavia, basil and bael at concentration of 20, 000 ppm showed higher antibacterial activity against Staphylococcus sp., Bacillus sp., K. cloacae respectively, both in vitro and in vivo studies.

Effects of Turmeric (Curcuma longa L.) on the Physicochemical Characteristics of Kochujang during Fermentation

  • Physicochemical properties of kochujang prepared with different concentration of turmeric were compared. The number of yeast and bacteria in the kochujang were low in the group with more than 1% of turmeric while the amylase and protease activity increased in the group with less than 1% of turmeric. The L- and b-values increased as the concentration of turmeric increased, however a-value decreased. The titratable acidity was low in the high percentage of turmeric added group. Oxidation-reduction potential increased in the later stage of fermentation with decrease in Aw. Reducing sugar and alcohol were high in than higher turmeric added group. Amino- and ammonia-type nitrogen were the highest in the 0.5% turmeric added group. The overall acceptability of kochujang was significantly more favorable (p<0.05) in the 0.25% turmeric added group than over 1% turmeric added one.

Production of Curcuminoids in Engineered Escherichia coli

  • Curcumin, a hydrophobic polyphenol derived from the rhizome of the herb Curcuma longa, possesses diverse pharmacological properties, including anti-inflammatory, antioxidant, antiproliferative, and antiangiogenic activities. Two curcuminoids (dicinnamoylmethane and bisdemethoxycurcumin) were synthesized from glucose in Escherichia coli. PAL (phenylalanine ammonia lyase) or TAL (tyrosine ammonia lyase), along with Os4CL (p-coumaroyl-CoA ligase) and CUS (curcumin synthase) genes, were introduced into E. coli, and each strain produced dicinnamoylmethane or bisdemethoxycurcumin, respectively. In order to increase the production of curcuminoids in E. coli, the shikimic acid biosynthesis pathway, which increases the substrates for curcuminoid biosynthesis, was engineered. Using the engineered strains, the production of bisdemethoxycurcumin increased from 0.32 to 4.63 mg/l, and that of dicinnamoylmethane from 1.24 to 6.95 mg/l.

Curcumin Inhibits Phorbol Ester-induced Expression of Cyclooxygenase-2 In Vivo through Suppression of Extracelluar Signal-regulated Kinase (ERK) 1/2 and NF-${\kappa}$/B in Mouse Skin

  • Curcumin derived from turmeric (Curcuma longa L.. Zingiberaceae) has been shown to possess marked chemopreventive activities, but the underlying molecular mechanisms remain unclear. In the present work. curcumin was found to inhibit 12-Ο-tetradecanoylphorbol-13-acetate(TPA)-induced expression of cyclooxygenase-2 (COX-2) in female ICr mouse skin as determined by Western and Northern blot analysis as well as immunohistochemical staining. Curcumin treatment atlenuated TPA-stimulated epidermal NF-${\kappa}$B activation. which was associated with its blockade of degradation and phosphorylation of the inhibitory protein l${\kappa}$ Bu and also of subequent translocation of the p65 subunut to nucleus. RAW 264.7 세포에서 발효 울금 추출물의 면역조절 효과 (Immunomodulatory Effects of Fermented Curcuma longa L. Extracts on RAW 264.7 Cells)

  • 본 연구에서는 울금의 선호도 및 이용율을 높이기 위해 Aspergillus oryzae를 이용하여 발효시킨 후 용매별 추출을 통하여 얻은 발효 울금 추출물들을 사용하여 면역조절 효과를 살펴보았다. MTT assay를 이용하여 실온 추출물 $200{\mu}g/mL$, 열수 추출물 $400{\mu}g/mL$, 20% 주정 추출물 $200{\mu}g/mL$, 80% 주정 추출물 $100{\mu}g/mL$의 농도가 적정하다고 판단되어 추후 실험에 사용하도록 하였다. 면역조절 효과를 보고자 대식세포의 탐식작용 능력, nitric oxide(NO), TNF-${\alpha}$ 생성, NK 세포 활성, SC-1 세포에서 발현되는 LP-BM5 virus의 LP-BM5 eco 유전자 발현을 측정하였다. NK 세포의 활성을 측정한 결과 NK 세포는 YAC-1 세포에 대한 cytotoxity를 관찰했을 때 20% 주정 추출물이 가장 활성을 나타내는 것으로 확인하였다. 발효 울금 추출물 중 열수 추출물과 20% 주정 추출물은 대식세포를 활성화시킴으로써 탐식능력을 증가시켰다. NO 생성을 관찰했을 때 발효 울금처리에 의해 활성화된 대식세포는 효과를 나타내지 않았으나 80% 주정 추출물을 제외한 추출물에서 TNF-${\alpha}$ 생성을 정상군에 비해 유의적으로 증가시켰다. SC-1 세포에 있는 LP-BM5 eco 유전자 발현을 비교하여 바이러스 복제 억제능을 측정한 결과 모든 추출물이 유의적으로 억제되었으며, 특히 20% 주정 추출물이 가장 많이 억제하고 있는 것을 확인할 수 있다. 이러한 연구 결과로부터 발효 울금이 면역조절 효과를 가진 천연 기능성 소재로 개발에 있어 기초자료로 활용할 수 있을 것으로 기대할 수 있다.

재배(栽培) 울금(鬱金)의 쿨쿠민 함량(含量) (Curcumin Content of Cultivated Tumeric in Korea)

  • Tumeric, Curcuma longa L.(Zingiberaceae) is native to the southern Asia, cultivated mainly in India and has been used for the treatment of hemorrhages in oriental medicine. The ground powder has also been used for flavouring and dyeing agents. In this report, the results of trial cultivation of tumeric in the middle portion of Korea (Kyungi, Shihung) are summarized. The rhizomes used for this trial cultivation were purchased from the Takii Seed Co. (Japan) and planted in the late of April and harvested in the middle of November. The curcumin contents of harvested tumeric were about 0.1% and 0.24% from finger type and bulb type respectively and these are comparable with those of commercial tumerics.

  • Diarylheptanoids [curcumin (1), demethoxycurcumin (2), bisdemethoxycurcumin (3), bisdimethoxymethylcurcumin (4), and 1,2-dihydrobis(de-O-methyl)curcumin (5)] were isolated from the methanolic extract of Curcuma longa L. and A new cyclic diarylheptanoiid (6) and a known compound 7 were isolated from fruits of Alnus japonica S. Diarylheptanoids (1-3) inhibited farnesyl protein transferase (FTPase) with an IC50 of 29-50 $\mu\textrm{m}$. (omitted)

천연염료를 이용한 한지염색에 관한 연구(I) -황색계열의 색상을 중심으로- (Studies on the Dyeing of Hanji by Natural Dye-stuffs(I) -With a Focus on the Color Tone of Yellow Color Series-)

  • Yellow dye-stuffs in natural plant were extracted from a gardenia, saffron, safflower, amur tree and pagoda. And then they were used to color Korean handmade paper(Hanji) on using a mordant. The results of the degree of discoloration are as follows. 1. As for a gardenia(Gardenia jasminodes Ellis for. grandiflora Makino), the effects of coloring were outstanding in the acid area. But for the preservation, it might be desirable that used a lye in a dye-stuff obtained at $40{\pm}5^{\circ}C$. 2. As for saffron(Curcuma longa L.), when used alum as a mordant, it was colored to a medium yellow color with green color. But easily discolorated and was not desirable. And, it didn't fit in a dye-stuff of Hanji. 3. For safflower(Carthamus tinctorius L.), when pH was in the low acid it was colored to the cleaner yellow color. It was the distinction of discoloration that the degree of brightness's increase was low. 4. For amur cork-tree(Phellodendron amurense Rupr.), the effects of yellow coloring were great in the areas of acidity and alkali. But, when used alum, the degree of the discoloration was high and was not effective. 5. For pagoda tree(Styphnolobium Japonica L.), using a calcium hydroxide as a mordant, enabled the more than average yellow to be gained. The degree of discoloration was good.
