회화 및 울금에 의한 단독 염색 견직물 그리고 쪽과 회화 및 쪽과 울금에 의한 복합 염색 견직물의 염색성 및 항균성 (Dyeability and Antimicrobial Activity of Silks Dyed Singly with Sophora japonica L., Curcuma longa L., Combination-Dyed with Polygonum tinctorium L. and Sophora japonica L., and with Polygonum tinctorium L. and Curcuma longa L.)

  • 이 연구는 회화 및 울금으로 단독 염색한 견직물 그리고 쪽과 회화 및 쪽과 울금으로 복합 염색한 견직물의 염색성 및 항균성에 대하여 검토한 것이다. 회화 및 울금으로 단독 염색한 견직물의 Munsell 표색계의 색상은 각각 5.0Y 및 0.6Y로 노란색 계열을, 쪽과 회화 그리고 쪽과 울금 추출액으로 복합 염색한 견직물의 색상은 각각 2.6BG 및 1.5BG로 청록색 계열을 나타내었다. 쪽과 회화로 복합 염색한 견직물의 일광견뢰도는 3등급으로 비교적 양호하였고, 드라이클리닝견뢰도 및 마찰견뢰도의 경우, 네 종류의 염색 견직물 모두 4~5등급으로 우수하게 나타났다. 회화 추출액으로 염색한 견직물의 Staphylococcus aureus에 대한 항균활성은 99.5% 그리고 나머지 세 종류의 염색 견직물은 99.9%의 뛰어난 항균성을 나타내었다. 또한 Klebsiella pneumonia에 대한 항균활성에 있어서도 회화 추출액으로 염색한 견직물은 99.6%, 그 나머지 세 종류의 염색 견직물은 99.9%의 우수한 항균성을 나타내었다.

남부도서지역에서 차광처리가 울금 (Curcuma aromatica Salisbury)의 주요 형질에 미치는 영향 (Effect of Shade-method on Agronomic Characteristics of Curcuma aromatica Salisbury in Southern Islands of Korea)

  • 우리나라 남부 도서지역인 진도 지방에서 울금 다수확의 기 초 자료를 얻고자 차광 재배를 실시 한 결과는 다음과 같다. 진도 지역에서 울금을 4월 20일 정식할 경우 4주 지난 후에 출현되고, 출현율은 83­88%이었다. 초장은 129cm­147cm이였고, 차광의 정도가 높을수록 초장이 커지는 경향이었다. 엽장은 47.5cm­49.4cm로 차광 30­60%에서 약간 양호하였다. 또한 엽폭은 16.9­17.9cm로 차광이 높을수록 짧아지는 경향이었으나 유의성은 인정되지 않았다. 울금의 1주 근경중은 941­993g으로 비대가 잘되었고, 10a당 3,980­4,200kg을 생산 할 수 있었으며, 15%와 30%차광이 수량이 약간 증수되었으나 45%이상의 차광에서는 오히려 감소되는 경향으로 차광의 효과는 인정되지 않았다. 이러한 결과로 보아 울금을 재배할 경우 15%차광에서 생육이 가장 양호한 것으로 나타나고, 근경 수량이 약간 증수되나 경제성을 고려하여 재배하여야 할 것으로 생각된다.

강황(薑黃)이 MIA 유도 골관절염 모델에 미치는 영향 (Effects of Curcuma longa Rhizoma on MIA-induced Osteoarthritis in Rat Model)

  • Objectives: The aim of this study was to investigate the anti-inflammatory effects of Curcuma longa rhizoma extract in an experimental rat model of osteoarthritis. Methods: Osteoarthritis was induced in rats by injecting monosodium iodoacetate (MIA) into the knee joint cavity of rats. The rats were divided into 5 groups (Normal, Control, positive comparison, low (CL) and high (CH) concentration groups). Rats in the low concentration (CL) group had MIA-induced osteoarthritis; they were treated with Curcuma longa rhizoma extract at a dose of 50mg/kg body weight. Rats in the high concentration (CH) group had MIA-induced osteoarthritis; they were treated with Curcuma longa rhizoma extract at a dose of 100mg/kg body weight. Hind paw weight distribution and ROS levels were measured. At the end of all treatments, changes in alanine aminotransferase (ALT), aspartate aminotransferase (AST), blood urea nitrogen (BUN), and creatinine levels were analyzed. In addition, inflammatory protein levels were evaluated by western blot analysis. Results: In this study, hind paw weight distribution significantly improved in the CL and CH groups, while. Reactive oxygen species (ROS) production significantly decreased in both. The levels of ALT, AST, BUN, and creatinine did not significantly change in either group. The production of nicotinamide adenine dinucleotide phosphate oxidase 4 (NOX4), $p47^{phox}$, and Ras-related C3 botulinum toxin substrate 1 (RAC1) decreased in both. Catalase, heme oxygenase-1 (HO-1) and superoxide dismutase (SOD) significantly increased in the CL and CH groups, respectively. Nuclear factor erythroid 2 (Nrf2) increased, but there were no significant differences between the experimental and control groups. Inflammatory cytokines, including nuclear factor-kappa Bp65 (NF-${\kappa}Bp65$), interleukin-1beta (IL-$1{\beta}$), and tumor necrosis factor-alpha (TNF-${\alpha}$), decreased significantly in both the CL and CH groups. Conclusions: Our results showed that Curcuma longa rhizoma extract has anti-inflammatory effects. Anti-inflammatory activity is regulated by the inhibition of inflammatory cytokines and mediators, such as NF-${\kappa}B$, therefore, it suppresses cartilage damage as well.

Modulation Effects on Acute Orofacial Inflammatory Pain in Rats by Curcuma longa L., Curcuma aromatica Salisb., Zingiber officinale Rosc. Extracts

  • Curcuma longa L. (C.L), Curcuma aromatica Salisb. (C.A) and Zingiber officinale Rosc. (Z.O) of Zingiberaceae plants which are well known as effects of natural anti-oxidant, anti-cancer and anti-inflammatory. We examined that the Zingiberaceae plants are involved in development and modulation of orofacial pain in rats. Male, 7- to 8-week-old, Sprague-Dawley rats weighing 240~280 g were used in this study. Experiments were performed using acute pain model that was caused by the injection of 5% formalin into the right vibrissa pad. The number of scratching or rubbing to the injection site was recorded for 9 consecutive 5-minute intervals following injection of formalin. The experimental groups were acute orofacial inflammatory pain; control group (formalin, 5%), vehicle group (5% formalin after sodium carboxymethyl cellulose), single administration group, single mixed administration group, repeated administration group. The experiments were performed various concentrations of Zingiberaceae plants extract. Therefore, oral administration of C.L, C.A, and Z.O (p.o., concentrations of 12.5, 25 mg/mL) in orofacial inflammatory pain model substantially decrease the nociceptive behavior in a concentration dependent manner. And it tended to decrease at low concentration (12.5 mg/mL) of single mixed and repeated administration more than single administration. This result means that Zingiberaceae plants extract affects the modulation of acute orofacial inflammatory pain. Thus, Zingiberaceae plants extract may be a potential therapeutic treatment for orofacial inflammatory pain.

랫트의 간 손상에 대한 울금 추출물의 간 기능 개선 효과 (Enhancement of Liver Function by Curcuma Extract on Acute Hepatotoxicity in Rat)

  • 본 연구는 국내산 울금 추출물, 둥굴레 및 말토덱스트린 혼합물을 이용하여 건강 기능성 식품을 제조한 결과 건강 기능성뿐만 아니라 기호적 품질 향상에도 기여하였다. 울금추출물을 $4{\sim}40mg/kg$ 으로 7일 동안 후 투여(post-treatment)한 SD계 랫트는 정상군과 비교하여 ALT(GPT)의 유의적 차이가 없었다(p>0.05). 울금 추출물을 40 mg/kg으로 7일 동안 후 투여(post-treatment)한 SD계 랫트는 정상군과 비교하여 AST(GOT)의 유의적 감소(p<0.05)를 나타내었다. 또한 울금 추출물을 $4{\sim}40$ mg/kg으로 7일 동안 후 투여(post-treatment)한 SD계 랫트는 대조군과 비교하여 ALT(GPT)와 AST(GOT)의 유의적 감소(p<0.05)를 나타내었다. 울금 추출물을4 mg/kg과 40 mg/kg으로 7일 동안 후 투여(post-treatment)한 SD계 랫트는 대조군과 비교하여 AST(GOT)와 ALT(GPT)의 효소 활성을 각각 82.6%와 78.7% 그리고 78.0%와 79.3%까지 감소함을 보였다. 본 연구 결과 울금 추출물의 경구 투여는 AST(GOT), ALT(GPT) 및 ALKP를 감소하였으며 사염화탄소($CCl_4$)에 의해 손상된 간 기능을 개선할 수 있는 유용한 건강 기능성 식품으로 사용할 수 있다는 것을 입증하였다. 둥굴레 및 말토덱스트린 혼합물은 간 기능 개선 효과는 없었으나 울금 특유의 강한 불쾌 취(off-flavor)감소에 의하여 관능적 품질 향상으로 소비자의 기호성 향상에 기여할 수 있다는 것을 입증하였다.

울금 추출물의 5-Alpha Reductase II 활성 저해 효과 (Inhibitory Effect of Curcuma longa L. Extracts on 5-Alpha Reductase II Activity)

  • 본 연구에서는 울금(Curcuma longa L.)의 다양한 추출물을 대상으로 5-alpha reductase의 활성 저해 정도를 측정하였다. 5-Alpha reductase는 T에서 활성형 남성호르몬인 DHT로 전환을 담당하며, 전립선비대에서는 혈액에서 DHT 농도가 높다. Rat의 전립선조직 균질액과 5-alpha reductase를 과발현시킨 LNCaP 세포 균질액을 대상으로 측정한 결과, $100{\mu}g/mL$의 농도에서 울금의 열수추출물(CL-HW)과 20% 에탄올추출물(CL-E20)이 5-alpha reductase 활성을 효과적으로 저해하였다. 특히 CL-HW는 $100{\mu}g/mL$의 농도에서 80% 이상의 유의적 억제효과를 보였으며, CL-E20은 $50{\mu}g/mL$의 농도부터 80% 이상의 감소효과를 보였다. 이 같은 결과에서 CL-HW 및 CL-E20은 5-alpha reductase의 억제활성이 있으며, 이는 전립선비대 치료소재로 개발될 수 있는 가능성이 있음을 제안한다.

추출 방법에 따른 강황 잎 추출물의 향장 활성 비교 (Comparison of Cosmetical Activities of Curcuma longa L. Leaf Extracts Using Different Extraction Methods)

  • Cosmetic activities of the leaves of Curcuma longa L. were compared according to different drying methods, to expand the use of the by-products of Curcuma longa L. The highest extraction yield of 29.4% was obtained from vacuum drying process (VD), whose value was very close to 29.2% from freeze drying process (FD). Relatively lower extraction yield were observed as 24.5% and 25.3% from the raw leaf (RL) as control and hot air drying process (HD). The cytotoxicity was observed lower FD and VD than RL and HD. It shows that cytotoxicity was closely related to cosmetic activities such as tyrosinase and melanin inhibition activities by showing much better cosmetic activities of the extracts from both FD and VD than those from the RL and HD. It was interesting that was differences of the cosmetic activities and cytotoxicity between FD and VD, which implies that VD method should be a more efficient process than FD method in terms of drying time and operation costs: 6 hours vs 24 hours and 3-5 times higher extraction costs in drying. It was observed that VD is more excellent dry method than others. This result could be utilized to effectively dry other soft plant biomass.

울금 분말을 첨가한 쿠키의 품질특성과 항산화 효과 (Quality Characteristics and Antioxidative Effects of Cookie Prepareds with Curcuma longa L. Powder)

  • The purpose of this study was to investigate the quality characteristics and antioxidative effect of cookies prepared with Curcuma longa L. powder (0, 1, 2, 3, 4%) substituted for flour. The pH of cookie dough decreased significantly in response to the addition of all levels of Curcuma longa L. powder (CLP). However, there were no significant differences in dough values among the test groups. Furthermore, when the spread factor values were compared among the groups, they were found to be inversely proportional to the CLP concentration. In addition, Hunter's color L value significantly decreased with increasing CLP content, whereas the a and b values increased. According to textural measurements, substitution of 1~4% for CLP resulted in increased hardness, cohesiveness, springiness, and brittleness when the samples were compared to the control. The results of sensory evaluation showed that the cookies containing 2% CLP had the highest scores. The acid value, peroxide value, and thiobarbituric acid value were all lower in cookies with 2% and 3% CLP than in cookies made with 4% CLP and control cookies.

Fungi-rice bran based Fermentation of Coptis Chinensis and Curcuma Longa Root and its Influence of Silk Dyeing

  • This study examined the dye-properties of natural fabrics dyed with Coptis chinensis and Curcuma longa root fermented with fungi. The optimum culture conditions for the fermentation of microorganisms, the relationship between natural dye color and fermentation conditions were investigated. Two different medical herbs (ground to 80-100 mesh in size) were used as a natural dyeing source. Phellinus linteus (P. linteus), which can grow in different media, such as Agarmedium (only agar containing medium), maltose extract agar (MA) and potato dextrose extract agar (PDA) culture media, were isolated from the medium. P. linteus was confirmed to be the optimum microorganism for the fermentation of Coptis chinensis and Curcuma longa, and the MA medium was confirmed to be the best for culturing. When using the microorganism as the fermenting agent, $32^{\circ}C$ was found to be the optimum fermenting temperature for both natural colorants. Regarding the dyeing property of the fermented natural dye, silk was dyed quite darkly in an appearance by naked eye estimation and the K/S value in the color strength of silk reached a high level of 16 after the fermenting process. The washing fastness of dyed silk after treatment washing was reduced from 4 to under4 and indicates that dyed silk with fermented plant was not unsubstantial. The light fastness was 1 to 2, showing intended to maintain due to the fermentation process.

Inhibitory Effects of Herbal Extracts on Dopa Oxidase Activity of Tyrosinase

  • Tyrosinase catalyzes the rate-limiting steps in melanin biosynthesis which is involved in skin-coloring and local hyperpigmentation of human beings, and unfavorable darkening of food products, Inhibitory effects on 3,4-dihydroxy-phenylalanine (dopa) oxidase activity of tyrosinase by 594 kinds of herbal extracts prepared from herbal medicines and wild plants in Korea were estimated. Two herbal extracts prepared from radicis cortex of Morus alba and rhizoma of Curcuma longa were selected as those exhibiting potent inhibitory effects on the enzyme activity. These herbal extracts were subjected to sequential fractionations with methylene chloride, ethyl acetate, n-butanol, and polar residue. The inhibitory effects on dopa oxidase activity of tyrosinase were shown in ethyl acetate fraction of Morus alba, and in methylene chloride fraction of Curcuma langa. The ethyl acetate fraction of Marus alba exhibited 50% of inhibition on dopa oxidase activity of tyrosinase at the concentration of 12 ${\mu}g/ml$, and methylene chloride fraction of Curcuma langa at 51 ${\mu}g/ml$.
