Formation of protoplasts from pyricularia oryzae (벼 도열병균, pyricularia oryzae의 원형질체 형성)

  • The optimum conditions of protoplasts formation from Pyricularia oryzae were investigated with lytic enzymes and osmotic stabilizers. The mycelia were begun to refease the protoplasts in response to the complex enzyme solution after 30-60 minutes and reached to maximum after 2-3hrs. Among the lytic enzymes tested, the mixture solution containing ${\beta}-Glucuronidase$(0.01 ml/ml), Cellulase ONOZUKA-RS(20mg/ml), Driselase (10mg/ml), and Macerozyme R-10 (10mg/ml) resulted in the highest rate of protoplasts releasing of Pyricularia oryzae. The best stabilizer was 0.6M KCl at pH 7.0. Shen the mycelia were digested with enzyme mixture, the stationary culture was better than shaking culture for higher protoplast formation.

Storage of Waste-Brown Seaweed and Degradation of Alginate Using Microorganism (미생물을 이용한 미역폐기물의 저장 및 알긴산염 저분자화)

  • We studied a storage of waste-brown seaweed at room temperature and degradation of alginate in seaweed by microorganism DS-02. The seaweeds, mixed with 5.0 wt% DS-02 and sealed in vinyl package without any other treatment, could be stored longer than 1 year without spoilage at room temperature. During the storage process, the alginate of seaweed was decomposed by enzyme of DS-02 and the molecular weight of alginate decreased to about 1/10 of initial quantity. DS-02 growed as fast as it had maximum weight after 24 hour culture and it's enzyme had a maximum activity of alginate degradation at $40^{\circ}C.$ The seaweed sample became particles in DS-02 culture solution and the M. W of alginate decreased to about 1/10 of initial value after 24 hour decomposition. The effect of alginate degradation with DS-02 was similar to that of degradation with 3.0 M HCI solution for 24 hour.

  • Water-soluble chitosan and water-insoluble chitosan with molecular weight of 2,000,000, 500,000, 80,000, and 40,000 and more than 90% degree of deacetylation were controlled to evaluate the antibacterial activity of chitosan against a pathogenic bacteria, methicillin resistant Staphylococcus aureus(MRSA), which is being issued in the world. The Shake Flask Method and Modified Shake Flask Method were used to find out the antibacterial activities of 5types of chitosan/acetic acid solution, and the other antibacterial activities test with the cotton filter treated with chitosan /acetic acid solution. Those test methods showed the great differences ,but the results of the antibacterial activites showed the same difference.

The Introduction of Polycrylamide Gel into the Solid Culture of Streptomyces spp

  • It is proposed the polyacrylamide gel, instead of agar, could be used for the solid cultures of microorganisms including Streptomyces strains. Polymerization and gellation of 5% acrylamide solution were done by autoclaving for 5 min at 121.deg.C and no hindered by the addition of nutrient-rich media. In particular, pH buffer solution suitable for corresponding microorganisms must be used in the preparation of culture media. Comparing with agar, it was discussed that polycrylamide gel had many advantages such as gellation within the wide range of strong acid Carbon and Nitrogen sources, requirement tests of growth factors and minerals, sterization at high temperature, diffusion assays of products depending on the pore size of gel, and stability and standarization of microbial cultures.

Microbiological Study of the Extracorporeal Circulation (체외순환법에 대한 미생물학적 고찰)

  • Open heart surgery patients have additional risks arising from the special nature of the operative procedure. And postoperative infections in patients with extracorporeal circulation are associated with high incidence of serious sequelae. To investigate the incidence and organisms of contamination, and the relation between the duration of extracorporeal circulation and contamination, following study was done. Eighty-four of the open heart surgery patients were examined with cultures from the blood and priming solution before and after bypass. 1. Cultures before bypass were sterile, but 2 cases of cultures from the blood and priming solution after bypass were positive, and culture positive group was 5% of all patients. 2. The organisms were Gram-negative, aerobic coliform bacilli; 3. The culture positive group had significantly longer bypass time.

  • For the production of proteolytic enzyme wilth Aspergillus, the examination is made on the culture-time and koji extracting conditions, during producing koji. 1. The highest activity showed up when the culture-time took 50 hours for Aspergillus sojae and 60 hours for Aspergillus flavus. 2. When the cultured koji was extracted by a buffer solution and water, the former gave the product of higher activity until pH 7 through pH 12, and water until pH 3 through pH 7. 3. In the method of crushing and granule extractions, crushing extraction produced the one of higher activity than granule. 4. The highest activity showed up when Aspergillus sojae took 5 hours (Aspergillus flavus 4 hours) in the time of extracting enzyme solution. 5. The highest activity showed up when both Aspergillu sojae and Aspergillus flavus reacted and indicated $37.60^{\circ}C$ in the reaction temperature and activity.

Bridging the Connectivity Gap Within a PLC-Wi-Fi Hybrid Networks

  • The implementation of a hybrid network utilizing Power Line Communication (PLC) and Wi-Fi technologies has been demonstrated to improve signal strength and coverage in areas with poor connectivity due to internet shadow areas. In this study we strategically positioned Wi-Fi relays and utilized the capabilities of PLC technology to significantly improve signal strength and coverage in areas with poor connectivity. We also analyzed the effects of metallic obstacles on Wi-Fi signal propagation and proposed a solution to strengthen the signal enough to pass through them. Our experiment demonstrated the feasibility and potential of using this hybrid network in industrial scenarios for real-time data transmission. Overall, the results suggest that the use of PLC and Wi-Fi hybrid networks can be a cost-effective and efficient solution for overcoming internet connectivity challenges and has the potential to provide high-speed internet access to areas with unreliable signals.

고정상세포분리기의 개발 및 Cyclosporin A 생산을 위한 고정화 연속배양공정에의 적용

  • We have developed an efficient immobilized cell separator for continuous operation of immobilized fungal cell cultures, and applied this separator to actual fermentation process for the production of cyclosporin A (CyA), a powerful immunosuppressant. In the experiments employing highly viscous polymer (carboxymethyl cellulose) solution, the decantor showed good separating performances at high solution viscosites and fast dilution rates. Air duct and cylindrical separator installed inside the decantor turned out to play key roles for the efficient separation of the immobilized cells. By installing the decantor in an immobilized perfusion reactor system (IPRS), continuous immobilized culture was stably carried out even at high dilution rate for a long period, leading to high productivities of free cells and CyA. Almost no immobilized biomass existed in effuluent stream of the IPRS, demonstrating the effectiveness of the decan- tor system for a long-term continuous fermentation. It was noteworthy that we could obtain these results despite of the unfavorable fermentation conditions, i.e., reduced density of the biosupports caused by overgrowth of cells inside the bead particles and existence of high density of suspended fungal cells (10g/l) in the fermentation broth.

Influence of pretreatment medium, fresh medium addition, and culture plate size on the production of embryos in isolated microspore culture of hot pepper (Capsicum annuum L.) (고추의 소포자 배양 시 전처리 배지, 새 배지의 첨가, 및 배양 용기의 크기가 배의 생산에 미치는 영향)

  • The influences of pretreatment medium, the addition of fresh medium, and the size of culture plate on the production of embryos were investigated in isolated microspore culture of hot pepper (Capsicum annuum L.). Among the media used for heat shock pretreatment ($32{\pm}1{^{\circ}C}$), high frequency embryo production was obtained when the sucrosestarvation medium A was used. On the other hand, neither 0.37 M mannitol solution nor NLNS medium supplemented with sucrose was not efficient for embryo production. The addition of culture medium to pretreatment media considerably decreased the embryo production even though embryo development proceeded further. The embryo production was not improved by the addition of fresh medium after 2 or 3 weeks from starting culture. Increase in the size of the culture plate from $3.5{\times}1.0$ cm to $6.0{\times}1.5$ cm improved embryo quality. These results will provide valuable information for developing an efficient microspore culture system of hot pepper for high frequency embryo production.
