  • A method is described for the rapid and simple isolation of genomic DNA from 3 ml culture of Bifidobacterium. The method is expected to be used in gene manipulation of Bifidobacterium spp. The isolated DNA using this method is shown to be an excellent substrate for restriction endonuclease digestion and ligation with T4 DNA ligase.

  • An adaptive on-line optimization method has been applied to test the ability to maximize the cellular productivity of a continuous methylotroph culture system which was simulated by a variable yield Monod-type model. Optimum dilution rate and productivity were successively obtained and maintained at all times by the algorithm that utilizes steepest descent technique as optimization method and recursive least-square method with forgetting factor as dynamic model identification.

Quantitative analysis of oral disease-causing bacteria in saliva among bacterial culture, SYBRgreen qPCR and MRT-PCR method (타액내 구강질환 원인 균의 세균배양법, SYBR green qPCR법, MRT-PCR법 간의 정량분석)

  • Objectives: The purpose of this study was to compare SYBR Green qPCR, TaqMan, and bacterial selective medium cultures for accurate quantitative analysis of oral microorganisms. Methods: The SYBR Green method is widely used to analyze the total amount of oral microorganisms in oral saliva. However, in this study, MTR-PCR method based on TaqMan method was performed using newly developed primers and probes. In addition, it was designed to confirm the detection agreement of bacteria among bacteria detection method. Results: As a result of MRT-PCR and SYBR Green qPCR analysis, more than 40 times (0.9-362.9 times) bacterium was detected by MRT-PCR. In addition, more bacteria were detected in saliva in the order of MRT-PCR, SYBR Green qPCR, and bacterium culture, and the results of MRB-PCR and SYBR Green qPCR showed the highest agreement. The agreement between the three methods for detecting P. intermedia was similar between 71.4 and 88.6%, but the agreement between MRT-PCR and SYBR Green qPCR was 80% for S. mutans. Among them, the number of total bacteria, P. intermedia and S. mutans bacteria in saliva was higher than that of SYBR Green qPCR method, and bacterium culture method by MRT-PCR method. P. intermedia and S. mutans in saliva were detected by MRT-PCR and MRT-PCR in 88.6% of cases, followed by the SYBR Green qPCR method (80.0%). Conclusions: The SYBR Green qPCR method is the same molecular biology method, but it can not analyze the germs at the same time. Bacterial culturing takes a lot of time if there is no selective culture medium. Therefore, the MRT-PCR method using newly developed primers and probes is considered to be the best method.

Effect of Nursing Organizational Culture on Job Satisfaction of Preceptors (간호조직문화가 프리셉터의 직무만족에 미치는 영향)

  • Purpose: This study was to identify effect of nursing organizational culture on job satisfaction of preceptors. Method: The subjects were 73 preceptors who were working for the 4 of university hospitals in Seoul, Daejeon and Kyong Ki. This study was conducted from Aug to Oct 2005. The data was collected by questionnaires and were analyzed using descriptive statistics, pearson correlation coefficient, stepwise multiple regression. Result: Job satisfaction of preceptors were significantly correlated among nursing organizational culture. The stepwise multiple regression analysis on job satisfaction revealed that nursing organizational culture was a powerful predictor of job satisfaction. Conclusion: The findings indicate that nursing organizational culture is important for improving job satisfaction of preceptors. Therefore it is necessary to develop of good nursing organizational culture for successful preceptorship.

  • Work and family life affect each other. Especailly in a corporate-centered society the impact of work on the family is very strong. Organigational sexrole culture is used not only as a method of business management, but also as a style of family living. The Purpose of this study was to analyze an effect of occupation and organizational sexrole culture of husband on the family life. Data for 214 husbands were gathered from the structured questionnaire and time diary. The major findings of this study were as follows; (1) According to the occupation, organizational sexrole culture of husbands is very different. (2) The occupation and organizational sexrole culture of husbands have an important effect on their life time structure. (3) The occupation and organizational sexrole culture of husbands effect on their participation rate of household labor. These results suggested that husbands’ occupation and organization sexrole culture have a significant effect on the family life.

  • A novel estimation method was investigated for determining the concentrations of residual biomass, poly-3-hydroxybutyrate (PHB), and main nutrients including carbon and nitrogen sources, phosphate, and mineral ions from the supplied amount of ammonia solution used for a pH-control solution and nitrogen source in a PHB fermentation. The estimation equations for a batch culture and a fed-batch culture were derived from the relationship between the growth rate of residual biomass and the feed rate of the pH-control solution, and then were applied to the batch culture and the fed-batch cultures of Alcaligenes latus. This method was successfully applied to estimate the concentrations of residual biomass, PHB, and nutrients.

  • Aminoacyl tRNA synthetases of bacteria are known as potential targets for new anti-microbial agents. To isolate new inhibitors of bacterial methionyl-tRNA synthetases from natural sources, a new target-oriented screening system using whole cells which are over-expressing a target enzyme was developed. Approximately 8,000 culture broths of microorganisms from soils were tested by this screening system. Among them, ten culture broths was found to contain inhibitory activity against methionyl -tRNA synthetases of Escherichia coli. For the validation of the screening system, this new method was compared with in vitro enzymatic method. Seven out of 10 culture broths showed inhibitory activity against methionyl-tRNA synthetases of E. coli. This result showed that the new screening system was comparable to the enzyme assay. Thus we believe that our screening system as a new method can be applied for the screening of new antibiotics inhibiting bacterial methionyl-tRNA synthetases from natural products.

  • As of today-half a century has passed after 1945, the liberation from Japanese imperialism-it's significant to introspect our culture. On the one hand our culture has succeeded due to the peculiar history, but on the other hand it hasn't. So to speak, it has gained in Quantity but lost in Quality. There are keen, international competition and exchange in the modern culture. So the Qualitive success of modern design depends on if it had an original character or not. According to Korean rapid industrialization, technological extence, and economic growth, the export has increased greatly, but package design hasn't been match for them. Korean expoters have been meeting with cutthroat competition in world market. Under these circumstances it's very important that we have a real understand and interest in our culture, we strengthen the competitive power of home products with good package design. This is our age of not home market but world market. We live in an age when design wins, so that we have to develop the good design native to Korea. Many designers have proposed their opinions with the characteristic designs, but it has been only a fragmentary and passive method. We must seek for the new method more positively. The effective and proper method resolves itself into the following four points. First, to understand Korean history and culture. Second, to have the exact grip of this economic situation. Third, to promote the national emotion. Fourth, in conclusion, to accord the package design with them. We cannot put off this task to overcome the cultural subordination, to place Korea on a firm cultural basis in the world, and to contribute to the cultural exchange of package design. Production of Haploid and Doubled Haploid Plants from Isolated Microspore Culture of Hot Pepper (Capsicum annuum L.) (고추 소포자를 이용한 반수체 및 배가반수체 생산)

  • Haploid/double haploid plants developed from isolated microspores can significantly accelerate plant breeding. Haploid plants can naturally double their chromosomes to create a pure homozygous line of diploid plants. We present a method for producing embryos from isolated microspores of hot peppers (Capsicum annuumL.). We analyzed the polyploidization levels of the regenerated plants. The donor plants produced the optimal stage of microspores following short-term growth under low-intensity light, which resulted in high rates of embryogenesis and cotyledonary embryogenesis. To find an efficient culture method, liquid, doubled-layer, and 2-step cultures were tested. Liquid culture yielded the highest number of embryos, whereas the highest efficiency for cotyledonary embryogenesis was afforded by the doubled-layer culture. When normal cotyledonary embryos were transplanted onto a regeneration medium, they developed into complete plants. From these, 208 plants were tested via flow cytometric analysis, and 35.6% and 72.7% of the chromosomes from the Milyang-jare and LV2319 genotypes, respectively, were found to be spontaneous double haploids. These results are the same as those obtained on analyzing horticultural characteristics, including the size of leaves and the size and shape of fruits. The present study provides information on the practical application of isolated microspore culture of hot peppers, factors that affect embryogenesis, and methods for polyploidy testing.
