An experiment was conducted to investigate the effects of three water temperatures (15, 20 and $25^{\circ}C$) in combination with three salinities (0, 15 and 30 psu) on the oxygen consumption rate of juvenile spotted sea bass, Lateolabrax maculatus (mean body weight $5.5{\pm}0.3g$). The oxygen consumption rates of L. maculatus were measured in triplicate for 24 hours using a continuous flow-through respirometer. Water temperature resulted in significant differences in the mean oxygen consumption rate of L. maculatus (p<0.001), but salinity and combinations of salinity and water temperature did not have (p>0.05). The oxygen consumption increased with increasing water temperatures in all experimental salinity regimes (p<001). Mean oxygen consumption rates at 15, 20 and $25^{\circ}C$ ranged 328.8~342.3, 433.9~441.0 and 651.5~659.9 mg $O_2\;kg^{-1}\;h^{-1}$, respectively. $Q_{10}$ values did not vary with salinity, bud varied with water temperature. $Q_{10}$ values ranged 1.63~1.75 between 15 and $20^{\circ}C$, 2.24~2.26 between 20 and $25^{\circ}C$, and 1.92~1.98 over the full temperature range. The energy loss by metabolic cost increased with increasing water temperatures in all experimental salinity regimes (p<0.001) Mean energy loss rates at 15, 20 and $25^{\circ}C$ ranged 224.6~233.8, 296.3~301.2 and $444.9{\sim}450.7kJ\;kg^{-1}\;d^{-1}$, respectively. These data suggest that the culture of juvenile spotted sea bass is possible without energy loss by salinity difference in freshwater as well as seawater after salinity acclimation. Thus, this result has an application for culture management and bioenergetic model for growth of this species.
Cremated archaeological bones provide crucial information to unravel the details of ancient cremation events and funeral culture. The research on cremated bones to date has been mainly focused on extracting archaeological information in Korea. Recently, the techniques of physical anthropology have been applied to obtain biological information and cremation temperatures for individuals. This study analyzed human remains excavated from Seoul Seokchon-dong Ancient Tombs and determined whether there were human cremations as well as the estimated cremation temperatures. There was no trace of fire in the pits where cremated bones were found; therefore, it is assumed that they were deposited with the artifacts. In this study, four samples were selected according to the surface color of the bones, and X-ray diffraction analysis (XRD) and Fourier-transformed infrared spectroscopy (FT-IR) were used for analysis. All four of the Seokchon-dong cremated bones were found, based on the crystallization index of X-ray diffraction analysis, to have experienced cremation above the temperature of 700℃. Infrared spectroscopy results indicated that the four bones from Seokchon-dong were cremated at temperatures above 700℃ and below 1,000℃. IR peaks were observed at 700℃, whereas no changes were found when bones experienced more than 1000℃. We assume that the cremated people in the Seokchon-dong Tombs were people of high-status because cremation at such a high temperature at that period required much effort. Here we present significant evidence of the cremation status and temperature of archaeological human bones on the basis of XRD and FT-IR, allowing for the restoration of the cremation events and funeral culture in archaeological sites.
Upper temperature tolerance of the turbot Scophthalmus maxim us, one of the popular aquaculture species in European community and China, was evaluated in terms of survival and growth at $20^{\circ}C$, $23^{\circ}C$, $26^{\circ}C$, or $29^{\circ}C$. Best growth was achieved at temperature $20^{\circ}C$ in this experiments. The fish exposed to $20^{\circ}C$ or $23^{\circ}C$ were comparable in survival, condition factor and feed conversion efficiency reminiscent of the latter temperature to be agreeable for the fish. The temperature over $23^{\circ}C$ appeared to be the temperatures beyond the fish can tolerate. For instance, the fish exposed to 26 showed mortality of 60.9% by day 60; none of the fish exposed to $29^{\circ}C$ survived beyond day 7. Culture densities between 80 and $200\;fish/m^2$ did not influence the survival, growth, condition factor and specific growth rate of the fish. The final production of the culture density experiment was $10\;kg/m^2$ on average. These results imply that the location where water temperature remains lower than $25^{\circ}C$ during summer can be a candidate site for the turbot aquaculture.
Bacitlus sphaericus ts-Dl290 was characterized comparatively with the wild type strain 1593 by themeasurements of the biosynthesis of total DNA, RNA and protein on the temperature-shift culturesat permissive temperature of $30^{\circ}C$ and at nonpermissive temperature of $42^{\circ}C$. The growth patterns of the wild type strain and ts-Dl290 were similar at $30^{\circ}C$, but at 4Z C the mutant almost did not grow (temperature-sensitivity). When the growth temperatures of both stains were shifted-up from $30^{\circ}C$ to $42^{\circ}C$ after a 4 hour culture, their growths were normal, but when shifted-down from $42^{\circ}C$ to $30^{\circ}C$ after a 4 h culture, the mutant did not grow. When shifted up from $30^{\circ}C$ to $42^{\circ}C$ after a 4 hculture, the DNA syntheses of the two strains were at a normal rate for 1 h, but after 1 h the biosynthesesdecreased. The rate of DNA synthesis of the wild type strain at the nonpermissive temperature was about 93%, and that of the mutant was about 50% of the ratio of the wild type strain, and the RNA synthesis of the wild type strain was maintained for 3 h, and that of the mutant for 2 h. Thereafter the RNA synthesis decreased, and the synthesis of proteins in the both strains were similarlykept high for 8 h. The reversibility of the DNA synthesis of the mutant at $42^{\circ}C$ was lessened whenthe culture times were increased.re times were increased.
A field study was carried out to compare the performances of mulching with recycled paper, oil-treated paper and urea-coated paper in spring season potato culture. Soil temperature, weed occurrence, and the growth and yield of potato (cv. Sumi) were compared with unmulched control. The average soil temperatures at 5cm soil depth during the first 10 days after planting were 18.9$^{\circ}C$, under paper mulch and 20.6$^{\circ}C$, under oil-treated paper mulch, being lower 0.8$^{\circ}C$ and higher 1.2$^{\circ}C$, respectively, than unmulched control. Paper mulch lowered maximum soil temperature by 4.6$^{\circ}C$, on a fine day, while oil-treated paper mulch elevated it by over 6.6$^{\circ}C$, Urea-coated paper was decomposed fester than the mulch paper without urea coating. The former was decomposed 50% by 80days after mulching, but the later only 20%. In all mulch treatments, weeds were effectively controled throughout the potato growing season. The percent emergence of potato was not different significantly among treatments. However, a little lower emergence and poor early growth were observed under oil-treated paper mulch because of higher soil temperature. Except oil-treated paper mulch, the paper mulches with and without urea coating showed no difference in growth and yield of potato from the unmulched control. The growth and yield performances were poorest under oil-treated paper mulch. Oil-treated paper mulch would result in difference growth and yield performances if potato is planted earlier than April IS in the present experiment. Further researches are needed for this aspect.
This study, collaborated Gifu University, Japan, was performed to analyze chemical pollutants and microorganism and to clarify the distribution of sulfate-reducing bacteria and their insolubilization of heavy metal ions in leachates sampled seasonally between 1994 and 1996 from Nanjido waste landfill site, sampled 4 times between 1995 and 1996 from Pusan and Daejeon waste landfill site, and sampled 1 time between 1992 and 1994 from Hokkaido, Nagoya, Osaka and Hukuoka waste landfill site in Japan. The results were as follows: 1. The temperatures of internal leachate and leachate effluent were 40$\circ$C and 30$\circ$C, respectively, and the pH values of both leachates were about 8.0 at Nanjido waste landfill site. The concentration of SO$_4^{-2}$ gradually increased with the degree of stabilization and that of NO$_3$-N was detected in a part of sampling sites at one and half years, and in all sampling sites at 3 years after completion of landfill. 2. The organic substances in leachate of Nanjido waste landfill site decreased with the degree of stabilization and they were very fluctuated with measuring point and time. The concentration of organic substance and heavy metals in internal leachate were higher than in leachate effluent and those of Cd, Hg, and Pb were lower than detection limit except a part of samples in 1996. 3. APCs in internal leachate and leachate effluent were not much different and the minimum of APCs in internal leachate and leachate effluent were $1.0\times 10^4$/ml and $4.0\times 10^1$/ml, respectively. 4. The maximums of SRBs in Nanjido, Pusan, and Daejeon waste landfill site were 9180 MPN/ml, 24000 MPN/ml, and 348 MPN/ml, respectively and the maximum of SRBs in Japan waste landfill site was 9300 MPN/ml. 5. During 2-week-SRB culture, the values of MPN were high at 50$\circ$C for initial culture period and at 30$\circ$C for last culture period. MPN started to appear at first day and rapidly increased between 7th day and 9th day. 6. Cadmium and copper were insolubilized by SRB within 6 hr and iron and zinc were done within 48 hr. The rates of insolubilization of Cd, Cu, Fe, Zn, T-Cr were 100%, 99.5%, 95.0%, 99.8%, 16.1% after 48 hr treatment with SRB, respectively.
This study was carried out to investigate the growth response of Pinus densiflora seedling to relatively high day temperature during the container culture in summer season. The experiment was performed with three maximum day temperature regimes, 30~35$^{\circ}C$, 35~40$^{\circ}C$ and above 40$^{\circ}C$ in the PE house. The reduction in plant height was observed since 6 weeks after germination in the seedlings grown at relatively high day temperatures, especially the seedlings at above 40$^{\circ}C$. High temperature also reduced the root collar diameter during growth of the seedlings, therefore 17.3% of the reduction was observed in the seedlings grown at above 40$^{\circ}C$ compared with the seedlings at 30~35$^{\circ}C$. The dry weight of the seedlings grown for 16 weeks was apparently reduced by elevated dat temperature in the PE house, and the seedlings at relatively high day temperature resulted in increased T/R ratio.
Even though the microalgal species of Isochrysis and Pavlova are widely used as live food in bivalve hatcheries, they are difficult to culture in mass during the summer season. Therefore, the present study was conducted to determine the optimum species or strains of Isochrysis and Pavlova to produce good growth and high contents of fatty acids at temperatures over $30^{\circ}C$. Four species of Isochrysis (I. galbana KMMCC12, I. galbana KMMCC214, I. aff. galbana, and Isochrysis sp.) and four of Pavlova (P. lutheri, P. gyrans, P. viridis, and Pavlova sp.) were cultured at $25^{\circ}C$, $29^{\circ}C$, and $33^{\circ}C$, and then analyzed for specific growth rate and fatty acid composition. Microalgae were cultured in f/2 medium at 23 psu and continuous light of $80{\mu}mol$ photons $m^{-2}s^{-1}$. For the I. galbana, growth rates were highest at $29^{\circ}C$ and decreased at $33^{\circ}C$ to the level observed at $25^{\circ}C$. I. galbana (KMMCC12) and Isochrysis sp. cultured at $29^{\circ}C$ and $33^{\circ}C$, respectively, exhibited the highest growth rates of all Isochrysis species. In terms of fatty acids, I. galbana (KMMCC12) contained higher contents of PUFA and n-3 HUFA at $33^{\circ}C$ than did Isochrysis sp. For species of Pavlova, growth rates of P. gyrans and P. viridis at $29^{\circ}C$ and $33^{\circ}C$, respectively, were higher than those of the other Pavlova species. In particular, P. viridis grew as well at $33^{\circ}C$ as it did at $29^{\circ}C$. However, P. lutheri and Pavlova sp. did not grow at $33^{\circ}C$. In terms of fatty acids, P. viridis cultured at $33^{\circ}C$ also exhibited higher contents of PUFA and n-3 HUFA, as compared to P. gyrans. Based on these results, we suggest that I. galbana (KMMCC12) and P. viridis are suitable species for mass culture during the high temperature season.
The characteristics of growth and esterase activity of bacterial strains isolated from acid hydrolysed soybean protein were examined. All the isolated strains having decomposition activity of p-hydroxybenzoic acid butyl ester and esterase producing activity were identified as Bacillus sp. by morphological and biochemical methods. The specific growth rates, esterase activities and p-hydroxybenzoic acid butyl ester decomposition activities of isolated strains were $0.844{\sim}1.213\;h^{-1}$, $21{\sim}222\;mU/ml$ and $5.4{\sim}8.1\;mU/ml$, respectively. In the fermentation of Bacillus sp. KB8 strain which had the highest esterase producing activity, growth, extracellular excretion and intracellular synthesis of esterase were inhibited by adding NaCl in the culture broth. Esterase producing activity gradually increased after late exponential growth phase, until maximum value of 420 mU/ml reached after 64 hours culture period. Esterase of Bacillus sp. KB8 strain was stable up to $50^{\circ}C$ for 30 minutes, but was inactivated by heating for 30 minutes at $70^{\circ}C$. The enzyme activity exponentially decreased during the incubation time at the temperatures of $60^{\circ}C$ and $65^{\circ}C$.
In order to investigate the live food value of microalgae for efficacious rearing of larvae and spats of bivalve, we studied growth rates of four microalgal species (Isochrysis galbana, Pavlova lutheri, Chaetoceros simplex, Tetraselmis tetrathele) cultured in different environmental conditions. These include changes in temperatures (20, 25, 30 and $35^{\circ}C$), salinities (20, 25, 30 and 33 psu) and light intensities (60, 100 and $140{\mu}mol\;m^{-2}s^{-1}$). The growth rate of I. galbana was faster at $25^{\circ}C$ than that of $20^{\circ}C$. At $25^{\circ}C$ the highest growth rate of I. galbana was observed at 33 psu (0.413) and the lowest at 20 psu (0.368) in 10 days of culture (P < 0.05). The growth rate of I. galbana was lower at 25 psu (0.383) than that of 30 psu and higher than that of 20 psu (P < 0.05). Similar temperature and salinity-dependent changes were also found in P. lutheri and T. tetrathele. C. simplex showed faster growth rate at $30^{\circ}C$ than that of $25^{\circ}C$. The highest growth rate of C. simplex was observed at 33 psu (0.428) and the lowest at 20 psu (0.389) in 10 days of culture (P < 0.05). Upon exposure to the light with different intensities, all four microalgal species showed a significantly faster growth rate at $140{\mu}mol\;m^{-2}s^{-1}$ than at $100{\mu}mol\;m^{-2}s^{-1}$ (P < 0.05).
