Recovery of Sodium Sulfate from Farm Dyainage Salt and Using It in Directive Dyeing of Cotton

  • Agricultural drainage salt generated during irrigation of crops in San Joaquin Valley, California exceeds 600,000 tons annually and cumulates in the field in a rapid rate. As a result, the waste is taking out more farmlands for salt storage and disposal, imposing serious concerns to environment and local agricultural industry. in searching for a potential solution to reduce or eliminate the waste, this research explored feasibility of producing a value-added product, sodium sulfate, from the waste and utilizing the product in textile dyeing. The result indicated that sodium sulfate could be produced the salt and could be purified by a recrystalization method in a temperature range within the highest and lowest daily temperatures in summer in the valley. Re recovered sodium sulfate samples, with purifies ranging from 67% to 99.91, were compard with commercially available sodium sulfate in directive dyeing of cotton fabrics. Direct Yellow 27 and direct Blue 1 had similar exhaustions among Na₂So₄Ⅰ, Na₂So₄Ⅱ, Na₂So₄Ⅲ and V which had similar ratios of sodium sulfate and sodium chloride in recovered salts. Na₂So₄Ⅳ had high exhaustion despite low ratios of sodium sulfate and sodium chloride. In direct Red 80, exhaustion depends more on the ratios of sodium sulfate and sodium chloride than sodium chloride. Na₂SO₄Ⅳ and Na₂SO₄V with high ratios of sodium chloride had more exhaustion than Na₂So₄and Na₂So₄Ⅲ with low ratios of sodium chloride. Generally, directive dyeing using recovered salts from farm drainage has similar or more excellent exhaustion than directive dyeing using commercial sodium sulfate.

A Novel Transglutaminase Substrate from Streptomyces mobaraensis Inhibiting Papain-Like Cysteine Proteases

  • Transglutaminase from Streptomyces mobaraensis is an enzyme of unknown function that cross-links proteins to high molecular weight aggregates. Previously, we characterized two intrinsic transglutaminase substrates with inactivating activities against subtilisin and dispase. This report now describes a novel substrate that inhibits papain, bromelain, and trypsin. Papain was the most sensitive protease; thus, the protein was designated Streptomyces papain inhibitor (SPI). To avoid transglutaminase-mediated glutamine deamidation during culture, SPI was produced by Streptomyces mobaraensis at various growth temperatures. The best results were achieved by culturing for 30-50 h at $42^{\circ}C$, which yielded high SPI concentrations and negligibly small amounts of mature transglutaminase. Transglutaminasespecific biotinylation displayed largely unmodified glutamine and lysine residues. In contrast, purified SPI from the $28^{\circ}C$ culture lost the potential to be cross-linked, but exhibited higher inhibitory activity as indicated by a significantly lower $K_i$ (60 nM vs. 140 nM). Despite similarities in molecular mass (12 kDa) and high thermostability, SPI exhibits clear differences in comparison with all members of the wellknown family of Streptomyces subtilisin inhibitors. The neutral protein (pI of 7.3) shares sequence homology with a putative protein from Streptomyces lavendulae, whose conformation is most likely stabilized by two disulfide bridges. However, cysteine residues are not localized in the typical regions of subtilisin inhibitors. SPI and the formerly characterized dispase-inactivating substrate are unique proteins of distinct Streptomycetes such as Streptomyces mobaraensis. Along with the subtilisin inhibitory protein, they could play a crucial role in the defense of vulnerable protein layers that are solidified by transglutaminase.

  • Sargassum fulvellum (Turner) C. Agardh, an edible brown alga is farmed commercially by sexual reproduction and vegetative regeneration. Investigations were made on the phenology, abundance and maturity of reproductive structures in mature fronds, egg release and young germling development under different light conditions (20, 50, 80 and $100{\mu}mol/m^2/s$) and temperatures (5, 10, 15, 20 and $25^{\circ}C$). Monthly sampling was carried out by SCUBA diving at Chungsando on the southwestern coast of Korea from September 2002 to August 2003. The Maximum length of thalli was $104.6{\pm}20.7{\cal}cm$ in March 2003 when the water temperature was $9.0^{\circ}C$ and minimum was $0.8{\pm}0.5{\cal}cm$ in June when the water temperature was $19.5^{\circ}C$. Receptacle formation was observed from February to April. The peak period of egg release for this alga was in April when the water temperature was about $10^{\circ}C$ in nature. In the culture regimes of temperature and irradiance, the egg release of the excised female receptacle was highly affected by temperature. The maximum rate of egg release was $96.7{\pm}5.8{\%}$ under $20^{\circ}C$ and $80{\mu}mol/m^2/s$. The maximum length of young germlings was $3.9{\pm}0.2{\cal}mm$ after 35 days culture under $15^{\circ}C$ and $80{\mu}mol/m^2/s$.

Production and Rheological Properties of Bioflocculant Produced by Bacillus sp. DP-152

  • The culture conditions for Bacillus sp. DP-152 in the flask were investigated for the production of polysaccharide locculant, DP-152. The optimum pH and temperature for the locculant production were 8.0 and $30^{\circ}C$, respectively. The avorable substrates for flocculant production were soluble tarch and ammonium nitrate. The medium composition was optimized as follows: 30 g soluble starch, 0.75 g $NH_4NO_3,\; 2.0g\; K_2\;HPO_4,\; 0.1\; g KH_2PO_4,\; 0.2g\; MgSO_4.\; 7H_2O,\; and\; 0.2g\; MnSO_4~5H_2O$ in 11 of distilled water. Under this optimized condition, flocculating activity has been improved 4-fold compared with that of the basal medium. In the culture flask, the highest flocculating activity was obtained after 70 h of cultivation and the amount of bioflocculant DP-152 yielded was 12.4 g/$\ell$. The solution of bioflocculant DP-152 showed non-Newtonian characteristics. Bioflocculant DP-152 exhibited apparently higher viscosity at all concentrations compared to that of zooglan (from Zoogloea ramigera), and it was stable over a wide range of temperatures and pHs.

생쥐배의 동결보존 (The Freezing of Mouse Embryos)

  • These experiments were carried out to examine the effect of rapid thawing (500$^{\circ}C$/min) on the survival of 8-cell mouse embryos cooled slowly (0.5-1$^{\circ}C$/min) to precooling temperatures between -10 and 070$^{\circ}C$ before direct transfer ofembryos to -196$^{\circ}C$, and to compare the survival of embroys thawed slowly (20$^{\circ}C$/min) and rapidly (500$^{\circ}C$/min) after in vitro culture. In addition, the sensitivity of 8-cell mouse embroys to the rate of addition and removal of cryoprotectant at room temperauture, and the effect of developing stages on the survival of embryos frozen-thawed slowly were investigated. The results obtained were summarized as follows: 1. Embryos were survived in rapid thawing (500$^{\circ}C$/min) only when slow cooling was terminated at relatively high subzero teperaure (-20 to -60$^{\circ}C$). The highest survival rate(77.0%) in in vitro culture of embryos thawed rapidly was obtaeined after transfer to -196$^{\circ}C$ from -40$^{\circ}C$. 2. For the survival of embryos in slow thawing (20$^{\circ}C$/min.), slow cooling to lower subzero temperature (-50$^{\circ}C$ and below) was required before transfer of embryos to -196$^{\circ}C$. These results indicate that embryos transferred to -196$^{\circ}C$ from high subzero temprature contain much interacellular ice to damage them during slow warming but to permit survival of embryos after rapid warming. 3. The Survival rate (72.7%) of 8-cell mouse embryos after rapid addition and removal of cryoprotectant, DMSO at room temperature was similar to that (83.9%) after slow addition and removal of cryoprotectant at same temperature. 4. The survival rates of 1-, 2-, 4- and 8-cell embryos frozen-thawed slowly were 26.7, 76.4, 70.0 and 83.9%, respectively.

Na₂SO₄ Ⅳ had low exhaustion which had low ratios of sodium sulfate and sodium chloride. In nylon/wool fabrics, C.I. Acid Blue 158 had similar exhaustions among Na₂SO₄ I, Na₂SO₄ II, Na2₂SO₄ III, Na₂SO₄ IV and Na₂SO₄ Ⅴ despite of Na₂SO₄ Ⅳ had low ratios of sodium sulfate and sodium chloride Generally, the dyeing of levelling dyes using recovered salts from farm drainage has similar or low exhaustion than the dyeing of levelling dyes using commercial sodium sulfate.

옥덩굴(CAULERPA OKAMURAE WEBER VAN BOSSE)의 생장과 성숙 (Growth and Maturation of a Green Alga, Caulerpa okamurae Weber van Bosse)

  • Seasonal growth and maturation process of Caulerpa okamurae were investigated in natural populations and in culture. Monthly sampling was carried out by SCUBA diving at Baekunpo, Busan, southeastern coast of Korea from November 1999 to October 2001. Growth of erect branches depended mainly on the water temperature in the natural habitat. Maximum length of erect branches was 14.0 $\pm$ 1.4 cm in June 2001 when the water temperature was 19.7$^{\circ}C$ and minimum was 2.8$\pm$0.2 cm in April when the water temperature was 14.7$^{\circ}C$. Fresh weight of erect branches was 3.9 $\pm$ 0.5 g in June 2001 and 0.2 $\pm$ 0.04 g in September 2000. Biomass of the population was maximum of 922.5 g dw${\cdot}m^{-2}$ in July and minimum of 125.6 g dw${\cdot}m^{-2}$ in April. Gametangial network was observed on the ramuli when the water temperature was over 19$^{\circ}C$ in August 2000 and June 2001. In the culture regime of 4 temperatures (15, 20, 25 and 30$^{\circ}C$) and 3 irradiances (20, 50 and 80 $\mu$mol${\cdot}m^{-2}{\cdot}s^{-1}$) combination, the maturation of excised erect branches was mainly affected by temperature. Maturation was induced under all irradiance conditions at 20 and 25$^{\circ}C$; under 80 $\mu$mol${\cdot}m^{-2}{\cdot}s^{-1}$ at 15$^{\circ}C$; and under 20 $\mu$mol${\cdot}m^{-2}{\cdot}s^{-1}$ at 30$^{\circ}C$. The highest rate of maturation was 64% under 20 $\mu$mol${\cdot}m^{-2}{\cdot}s^{-1}$ at 25$^{\circ}C$. These results suggested that developmental initiations of C. okamurae occurred at higher than 13$^{\circ}C$ and maturation took about 270 degree-days.

Real-Time PCR Detection of 16S rRNA Novel Mutations Associated with Helicobacter pylori Tetracycline Resistance in Iran

  • Background: Tetracycline is an antibiotic widely used for the treatment of Helicobacter pylori infection, but its effectiveness is decreasing due to increasing bacterial resistance. The aim of this study was to investigate the occurrence of 16S rRNA mutations associated with resistance or reduced susceptibility to tetracycline ofHelicobacter pylori by real-time PCR (RT-PCR) assays from culture. Materials and Methods: Tetracycline susceptibility and minimal inhibition concentration (MIC) was determined by the Epsilometer test (Etest) method. A LightCycler assay developed to detect these mutations was applied to DNA extracted from culture. The 16S rRNA of these isolates was sequenced and resistance-associated mutations were identified. From 104 isolates of H. pylori examined, 11 showed resistance to tetracycline. Results: LightCycler assay was applied to DNA extracted from 11 tetracycline-susceptible and 11 tetracycline resistance H. pylori isolates. In our study the sequencing of the H. pylori wild types in 16 s rRNA gene were AGA 926-928 with MIC (0.016 to $0.5{\mu}g/ml$), while the sequencing and MIC for resistant were GGA and AGC, (0.75 to $1.5{\mu}g/ml$), respectively. Also we found a novel mutation in 2 strains with $84^{\circ}C$ as their melting temperatures and exhibition of an A939C mutation. Conclusions: We conclude that real-time PCR is an excellent method for determination of H. pylori tetracycline resistance related mutations that could be used directly on biopsy specimens.

Identification and Characterization of Bacteriocin-Producing Lactic Acid Bacteria Isolated from Kimchi

  • Lactic acid bacteria were isolated from Kimchi and screened for bacteriocin. A total of 99 strains showed antimicrobial activity when grown on solid media, yet only 10 showed antimicrobial activity in liquid media. Strain H-559, identified as Lactococcus lactis subsp. lactis, exhibited the strongest inhibitory activity and was active against pathogenic bacteria including Listeria monocytogenes, Staphylococcus aureus, and Bacillus cereus as well as other lactic acid bacteria. The antimicrobial substance produced by L. lactis subsp. lactis H-559 was confirmed to be a bacteriocin by the treatment of $\alpha$-chymotrypsin, and protease type Ⅸ and ⅩIV. The bacteriocin activity remained stable between pH 2.0 and pH 11.0 and during heating for 10 min at $100^{\circ}C$. The bacteriocin production started in the exponential phase and stopped in the stationary phase. L. lactis subsp. lactis H-559 showed the highest bacteriocin activity at a culture temperature of $25^{\circ}C$, and an inverse relationship between the bacteriocin productivity and mean growth rate at different culture temperatures was observed. The mean growth rate and bacteriocin productivity of L. lactis subsp. lactis H-559 increased as the initial pH of the media increased.

절임 고추장 종류가 다른 박대장아찌의 저온 저장 중 품질특성 (Quality Characteristics of Tongue Sole (Cynoglossus semilaevis) Jangajji with Different Types of Gochujang during Low Temperature Storage)

  • In this study, pH, $^{\circ}Brix$, salt concentration, sodium content, color, hardness, and sensory evaluations were measured during storage at low temperatures for 21 days after making tongue sole Jangajji with different types of Gochujang sauces. The pH was higher than those of samples not containing Maesil fermented liquor at all storage periods. The $^{\circ}Brix$ of the Maesil fermented liquor added samples was higher than that of the samples not added at the early stages of storage. On the other hand, there was no significant difference between the samples with and without Maesil fermented liquor at the later stages of storage. The sodium content was lower than that of the samples not containing Maesil fermented liquor. The lightness (L) was lower in the early stages of storage than those of the Maesil fermented liquor. In the late stage of storage, however, the lightness was higher than those of the samples not containing Maesil fermented liquor. The redness (a) of the GRWO sample group showed the highest value at all storage periods compared to the other sample groups. The yellowness (b) of the GRWO sample group was high at the early stages of storage. On the $21^{st}$ day of storage, however, the GRW sample group was significantly higher (p<0.01). The hardness results showed that the value of the later storage period was lower than that at the initial storage stage. The sensory evaluation showed that the Maesil fermented liquor reduced the spicy, salty, bitter, and unpleasant taste. As a result of the acceptance test, the GRWO and GRW samples were evaluated as the acceptance group with color, aroma, texture and overall acceptability compared to the brown rice Gochujang sample group.
