담금액 온도에 따른 히카마 피클의 저장 중 품질변화 (Quality Changes in Jicama (Pachyrhizus erosus) Pickle Due to Variations in Soaking Solution Temperature During Storage)

  • This study was carried out to determine the optimum temperature of the jicama pickle solution by evaluating physicochemical and sensory characteristics at various temperatures. The soaking solution of the jicama pickle was prepared at different temperatures (95, 75, 50, and 25℃). During storage, the titratable acidity, Brix, and the salinity of the jicama pickles increased, and these values increased with increasing temperature of the soaking solution. The higher the temperature of the soaking solution, the lower the L and b values. In all samples, the hardness increased rapidly until the 5th day of storage, and then gradually decreased. During storage, the jicama pickles showed a high tendency to harden at soaking solution temperatures of 75 and 95℃. Sensory evaluation indicated that the taste, flavor, crispness, and overall preference of the jicama pickle were higher at a soaking solution temperature of 75℃ and above. The higher the temperature of the soaking solution, the higher the acceptability. Based on these results, we conclude that soaking solution temperature of 75°¦95℃ in the manufacture of jicama pickles had a positive effect on the reduction in tissue softening and the sensory properties of the jicama pickle.

Thin-layer Drying Kinetics of Robusta Coffee

  • This paper was aimed to study the drying kinetics of coffee and to investigate the thin-layer drying kinetics of coffee by using a convective air dryer. The coffee was dried for the temperatures of 40, 50 and $60^{\circ}C$ with relative humidity in the range of 14-25% the airflow rate fixed at 1 m/s. According to the experiment result, the drying rate curve showed that drying process took place only in the falling rate period. Seven thin layer drying models (Newton, Page, Henderson and Pabis, Logarithmic, Wang and Singh, Two terms, Modified Henderson and Pabis) were fitted to the experimental moisture content data. The Two-trem model was found to be a better model for describing the characteristics of coffee for the temperatures of 40, 50 and $60^{\circ}C$. The effective moisture diffusivity of coffee increased when the drying temperature increased. The value was in the range of $4.5028{\times}10^{-11}$ to $6.4803{\times}10^{-11}m^2/s$.

울릉도 수집 균주의 배양조건에 따른 균사 생장 특성 비교 (Comparison of Mycelial Growth Characteristics According to Culture Conditions of Ulleungdo Collection Strains)

  • 본 연구에서는 울릉도에서 수집한 자생 버섯 균주 5종의 평판배지 배양 특성과 액체배양에서의 특성을 확인하여 자생 균류의 이용을 위한 기초 정보를 확보하였다. 5종의 야생 균주의 최적 배양온도는 25-30℃이고, pH는 4.0-5.0의 산성임을 확인하였다. 특히P. brumalis는 35℃에서 생장 속도가 가장 빠른 것으로 보아 고온성 버섯으로 판단된다. 실험에 사용한 상용 배지 중 최적 배지는 F. punctata의 경우 MEPA 배지, P. ulleungus는 MMNA 배지, G. subnudus는 MEA 배지, T. kmetii는 MMNA 배지, 마지막으로P. brumalis는 모든 배지에서 빠른 생장 속도를 보였으며 6종류의 배지 중 MEA 배지는 모든 균주의 균사 밀도가 낮아 배양이 적합하지 않은 조성임을 확인하였다. P. brumalis는 5종의 균주중 가장 빠른 생육 속도를 보였으며, 반면에P. ulleungus는 가장 저조한 생장을 보여 같은 속(genus)의 종이지만, 생육특성의 차이가 극명함을 보였다. 액체배양을 통해 배양 기간에 따른 배양여액의 건조 중량을 비교한 결과 배양 기간이 길수록 건조량이 감소하는 양상을 보였으며, 특히 6개월 이전까지 큰 폭으로 감소하였다. 이 결과로 정치배양 조건에서의 액체배양은 한달 이상의 배양 시간이 주어져야만 배지 성분을 충분히 이용할 수 있다는 것을 확인하였다. 본 실험의 결과로 5종의 균주에 대한 최적 배양조건을 확립하고 향후 응용 연구에 기초 자료로 활용할 수 있을 것으로 기대한다.

고환 내 정자의 체외배양 중 운동성에 미치는 인간 난포액과 온도의 영향 (Effect of Human Follicular Fluid (hFF) and Temperatures on the Motility of Testicular Sperm Cultured In Vitro)

  • The aim of this study was to evaluate the improvement of testicular sperm motility following different culture conditions such as human follicular fluid (hFF) and temperature. Testicular tissues obtained from azoospermia (n=21) were minced into small pieces by blade and recovered sperm suspension were cultured in Ham's F10 with or without 40% hFF at different temperatures (Group I: 37$^{\circ}C$/with hFF, Group II: 32$^{\circ}C$/withGroup III: 37$^{\circ}C$/without, Group IV:32$^{\circ}C$ /without The motility and viability of sperm were monitored during culture for 48 hours. Initial motility of testicular sperm was 10.9$\pm$1.9%. After 24 hours culture, sperm motility was 23.5$\pm$2.1% (Group I), 8.1$\pm$1.1% (Group II), 10.4$\pm$ 1.4% (Group III) and 4.0$\pm$0.8% (Group IV), respectively. After 48 hours, the motility had been changed as 32$\pm$2.3% (Group I), 14.3$\pm$1.7% (Group II), 5.3 $\pm$1.4% (Group III) and 4.3$\pm$0.9% (Group IV). In hFF group (I and II), sperm motility of group I cultured at 37$^{\circ}C$ was higher than those of group II at 32$^{\circ}C$. But, sperm viability of group I cultured at 37$^{\circ}C$ was lower than those of group II at 32$^{\circ}C$ (54.4$\pm$4.1% vs. 59.4$\pm$3.7%) after cultured for 48 hours. We acquired the best motility of testicular sperm when performed in vitro culture for 48 hours in hFF supplemented medium at 37$^{\circ}C$. Increase of sperm motility by in vitro culture could be useful tool fur human TESE-ICSI program.

온도 및 생육시기에 따른 수경재배 인삼의 생장특성과 수량에 관한 연구 (Studies on Growth Responses and Yields of Panax ginseng C. A. Meyer Grown under Hydroponic Culture with different Temperatures and Growth Stages)

  • This study was carried out to have the basic and applied informations relating to increase the productivity and quality of ginseng. 2 years-old ginseng was cultivated under hydroponic culture with the controlled environment conditions in a greenhouse. Major growth characters and yields were investigated with two different temperatures and several growth stages. The plant height and stem diameter were higher at low temperature than those at high temperature. They were not clearly different with six different growth stages. The root length was not clearly different between two temperatures; however it was continuously grown from June until August. The root diameter was higher at low temperature than that at high temperature. It was rapidly increased from June until August. The length, width, and area of leaf were higher at low temperature than those at high temperature. The fresh and dry weights of different plant tissues were also heavier at low temperature than those at high temperature. The moisture content of ginseng root was continuously decreased from June until August. The yield of ginseng was higher at low temperature compared to that at high temperature. The cultivating conditions in hydroponic culture of ginseng, especially temperature, would be an important factor to have better growth and production.

Influence of Isolation Temperature on Isolating Diverse Lactic Acid Bacteria from Kimchi and Cultural Characteristics of Psychrotrophs

  • Kimchi is a traditional Korean fermented vegetable that is stored and fermented at low temperatures. However, kimchi lactic acid bacteria (LAB) are typically isolated under mesophilic conditions, which may be inappropriate for isolating the diverse LAB. Therefore, this study investigated the suitable conditions for isolating various LAB from kimchi. Here, LAB were isolated from four kimchi samples using MRS, PES, and LBS media and varying isolation temperatures (30, 20, 10, and 5℃). Then, MRS was selected as the suitable medium for LAB isolation. A comparison of culture-dependent and culture-independent approaches indicated that 5℃ was not a suitable isolation temperature. Thus, the number and diversity of LAB were determined at 30, 20, and 10℃ using 12 additional kimchi samples to elucidate the effect of isolation temperature. With the exception of two samples, most samples did not substantially differ in LAB number. However, Leuconostoc gelidum, Leuconostoc gasicomitatum, Leuconostoc inhae, Dellaglioa algida, Companilactobacillus kimchiensis, Leuconostoc miyukkimchii, Leuconostoc holzapfelii, and Leuconostoc carnosum were isolated only at 10 and 20℃. The growth curves of these isolates, except Leu. holzapfelii and Leu. carnosum, showed poor growth at 30℃. This confirmed their psychrotrophic characteristics. In Weissella koreensis, which was isolated at all isolation temperatures, there was a difference in the fatty acid composition of membranes between strains that could grow well at 30℃ and those that could not. These findings can contribute to the isolation of more diverse psychrotrophic strains that were not well isolated under mesophilic temperatures.

Effect of Environmental Factors on Flavonol Glycoside Production and Phenylalanine Ammonia-lyase Activity in Cell Suspension Cultures of Ginkgo biloba

  • A study was carried out to elucidate the relation between the production of flavonol glycosides and the change of phenylalanine ammonia-lyase activity in cell suspension cultures of Ginkgo biloba by the unassisted and synergistic effects of various factors. The quercetin production showed a mixed-growth-associated pattern in cell suspension cultures. Fluorescent light and UV radiation increased phenylalanine ammonia-lyase (PAL) activity, and resulted in the increase of the production of quercetin and kaempferol ten- and four-fold, respectively, as compared to that obtained in the normal culture condition. The cell growth of Ginkgo biloba was enhanced .at higher temperatures whereas the quercetin production was at its maximum at low temperatures. Moreover, the quercetin production was increased by temperature change during the culture period. In particular, the quercetin production was at the highest level when the culture temperature was elevated from $10^{\circ}C\;to\;30^{\circ}C$. The addition of phenylalanine as a precursor in the culture medium stimulated an 8-fold increase in the production of quercetin; the addition of naringenin caused a l0-fold increase. The quercetin production was also greatly increased by feeding enzyme cofactors such as 2-ketoglutarate and ascorbic acid in the culture medium, but specific PAL activity was not increased except with phenylalanine feeding. The synergistic effect of UV radiation and naringenin feeding was observed, resulting in the increase of flavonol glycoside production at a rate higher than in any other case investigated.

남북극 유래 저온성 박테리아 Culture Collection에서 저온활성 프로테아제 생산균주의 스크리닝과 효소 특성 (Screening for Cold-Active Protease-Producing Bacteria from the Culture Collection of Polar Microorganisms and Characterization of Proteolytic Activities)

  • 극지연구소(KOPRI)는 국내외적으로 유일하게 남북극 지역에서 분리한 저온적응성 박테리아 균주를 대상으로 culture collection(약 6,300균주)을 구축하여 운영하고 있다. 보유 중인 프로테아제(protease) 생산 균주들(총 874균주) 중에서 활성이 높은 프로테아제를 생산하는 78개의 균주들을 1차 선발한 후, 1% skim milk가 포함된 0.1${\times}$ ZoBell 고체배지에 접종하고 다양한 온도($5-30^{\circ}C$)에서 배양하면서 세포외분비성 프로테아제의 활성을 비교하였다. 위의 신속하고 직접적인 균주 스크리닝 방법을 통해서, 최종적으로 저온활성 프로테아제를 생산하는 15개의 저온적응성 균주들을 선발하였다. 최종 선발된 균주들은 16S rRNA 유전자의 분석결과 Pseudoalteromonas (13균주)와 Flavobacterium (2균주) 속(genus)으로 분류되었고, $5-15^{\circ}C$ 저온에서도 활성을 나타내는 저온성 프로테아제를 생산하였다. 15개 균주들이 생산하는 각각의 프로테아제는 특이적 화합물에 의한 효소활성 억제 정도에 따라 5개의 그룹(serine protease, aspartic protease, cysteine protease, metalloprotease, 그리고 미분류 프로테아제)으로 분류되었다. 본 실험을 통해서 선발한 남북극 유래 박테리아 균주들은 새로운 저온활성 프로테아제를 발굴하기 위한 유용한 생물자원으로서의 가치를 가지고 있다.

육묘시의 주야간 기온이 서광 토마토의 생육 및 초기 착화 절위에 미치는 영향 (Effect of Day/Night Temperatures during Seedling Culture on the Growth and Nodes of Early Flower Cluster Set of 'Seokwang' Tomato (Lycopersicum esculentum Mill.))

  • 육묘시 주간/야간 온도 처리가 서광 토마토 묘의 정식후 초기결실 절위 및 생육에 미치는 영향을 알아보고자 실험을 수행하였다. 2차에 걸쳐 환경조절실 내에서 육묘하여 유리온실 내에서 양액재배 하였다. 서광 토마토 종자를 상토를 채운 128구 플러그 트레이에 파종하여 생육상에서 3일간 발아시킨 후 주간/야간 온도가 $25^{\circ}C$/$25^{\circ}C$, 16$^{\circ}C$/16$^{\circ}C$, 16$^{\circ}C$/$25^{\circ}C$ 또는 $25^{\circ}C$/16$^{\circ}C$로 각각 조절되는 4개의 생육실에서 1차 실험은 33일, 2차 실험은 35일간 육묘하였다. 일일 12시간씩 3파장 cool-white 형광램프로 140$\mu$mol.m$^{-2}$ .s$^{-1}$의 광을 공급하고, 2차 실험에서는 1000$\mu$mol.mol$^{-1}$의 CO$_{2}$를 공급하였으며, 공기 유속은 0.3m.s$^{-1}$, 그리고 공기 중 습도는 80%를 유지하였다. 육묘한 토마토 묘를 자연 일장조건인 유리온실의 암면 슬레브에 이식하여 1차 실험은 37일간, 그리고 2차 실험은 35일간 재배하면서 육묘시에 사용했던 양액과 동일한 양액을 공급하였다. 초장은 주간과 야간 온도의 상호작용보다는 일평균 온도의 영향을 더 많이 받았으며, 16/16$^{\circ}C$ 처리구에서 가장 컸다. 엽구는 주간과 야간의 기온의 영향을 받지 않았고, 엽록소 농도는 16/$25^{\circ}C$ 처리구에서 가장 높았다. 그리고 줄기의 생체중과 건물중은 주야 항온 처리구에서 타처리에 비해 다소 큰 경향이었다. 제 1화방과 2화방의 절위는 25/$25^{\circ}C$구에서 1화방의 착생 절위가 유의성 있게 높았고, 나머지 처리들간에는 차이가 없었다. 1화방 1번화의 개화 소요일수는 25/$25^{\circ}C$구에서 가장 컸고 16/$25^{\circ}C$구에서 낮았다. 토마토의 생육과 1, 2화방 절위에 육묘시의 일평균 기온이 큰 영향을 미치는 것으로 나타났으나 토마토의 2화방 비절현상은 나타나지 않았다.
