Botrytis cinerea에 의한 들깨 잿빛곰팡이병의 발생 (Occurrence of Gray Mold Rot of Perilla Caused by Botrytis cinerea)

  • Gray mold rot of perilla was epidemic at Kangdong, Pusan and Miryang, Kyungnam in 1997 and 1998. The incidence of this disease ranged form 21.3 to 68.1% at Kangdong area. Leaf necrosis initially appeared on the edge of the infected leaves, and it was developed to the center of the leaves forming typical V-shaped brown necrotic lesions. Under high moisture condition, abundant mycelia of the pathogen was formed on the surface of the lesions. Infected stems became slender and were completely blighted up to the top of the plant. Two isolates, LVF12 and SD7, were isolated from diseased lesions showing typical symptoms, and the pathogenicity was tested using mycelial disks and conidial suspension inoculation. The developed symptoms were same as the naturally produced ones. These two pathogenic fungi were identified as Botrytis cinerea based on the morphological characteristics using a microscope and a scanning electron microscope, and cultural characteristics. This is the first report of gray mold or perilla in Korea.

Fusarium oxysporum f. sp. eustomae에 의한 꽃도라지 시들음병(가칭) 발생 (Occurrence of Fusarium Wilt on Lisianthus (Eustoma grandiflorum) Caused by Fusarium oxysporum f. sp. eustomae)

  • Fusarium wilt of lisianthus occurred severely throughout all cultivation areas, especially in alpoine areas during summer season and the disease incidence was 5 to 30 percent in Korea. The major symptoms of the disease were wilt with chlorosis and water deficiency, stunted plants and scorched leaves. Severe symptoms appeared just after high temperature period around late August in Daekwallyong area. Whit and pale red colored mycelia were developed on stems of infected plants near the soil surface. The causal organism of Fusarium wilt of lisianthus was isolated and identified as Fusarium oxysporum f. sp. eustomae on the basis of pathogenic and cultural characteristics. The causal organism was reisolated from all infected and inoculated stems of plants. This is the first report referring to F. oxysporum f. sp. eustomae of lisianthus in Korea.

Cladophialophora lanosa sp. nov., a New Species Isolated from Soil

  • Cladophialophora is characterized by simple hyphomycetes with brown hyphae that give rise to branched chains of pale brown conidia and shows affinities with the Herpotrichiellaceae. A fungal strain belonging to the genus Cladophialophora was isolated from soil in Daegu, Korea. This strain produces numerous greenish to dark black lanose aerial mycelia with hair like structures. It is morphological similar to C. chaetospira, C. inabaensis, and C. multiseptata; however, the conidiophores and conidia sizes of the newly isolated strain (KNU16-032) are clearly different from them. The novelty of the strain was also confirmed based on phylogenetic analysis using the data sets of the internal transcribed spacer region of and the partial sequence of 28S ribosomal DNA region along with the cultural characteristics. Because morphological observations and phylogenetic analysis indicated that the strain is distinct from previously known Cladophialophora species, we propose this species as a new species Cladophialophora lanosa sp. nov., and provide the detailed descriptions in this study.

좀나무싸리버섯(Clavicorona pyxidata) '왕관' 수집균주의 배양적 특성조사 (Cultural characteristics survey of Clavicorona pyxidata (WANG GWAN) strains collected from Korea)

  • This study was carried out to investigate the characteristics survey of Clavicorona pyxidata strains. C. pyxidata are selected 3 kinds of strains collected in GWANG DEOCK Mt., DO BONG Mt., and BUK HAN Mt., Mycelial growth of C. pyxidata collected from GWANG DEOCK and BUK HAN Mt. is 64mm, 60mm for 18days respectively. DO BONG Mt strain is 85mm for 9 days. Transparent crystal body of C. pyxidata mycelia were appeared by microscopic examination of 100 magnification. Optimal temperature of mycelial growth in C. pyxidata strains are 25~30℃ respectively.

  • 검은비늘버섯의 균사체와 기능성 물질 대량생산을 목적으로 배지의 종류, 배양온도, 배양중의 진탕 유무가 균사생장에 미치는 영향을 조사한 결과 균사생장 최적배지는 MCM, 최적온도는 25℃로 밝혀졌고 진탕배양을 하였을 경우 정치배양보다 약 2~3배의 균사생장을 보였다.

Zoysiagrass에 Waitea circinata에 의한 Waitea Ring Patch 발생 (First Report of Waitea Ring Patch caused by Waitea circinata on Zoysiagrass)

  • Zoysiagrass는 대표적인 난지형 잔디로 Curvularia leaf spot, Large patch, Rhizoctonia blight, Spring dead spot, 그리고 Dollar spot 등의 다양한 병에 직면해 있으며 최근에는 많은 병원균들의 복합 감염에 의한 잔디병 양상을 보이고 있다. 최근에 패취형태의 병반을 가진 zoysiagrass 엽조직으로 부터 Rhizoctonia 속과 유사한 곰팡이를 분리하였다. 리보소옴 DNA의 염기서열을 분석하여 Waitea circinata 균주들과 98% 이상의 상동성을 보였고, Koch's 법칙에 따라 zoysiagrass에서 병원성을 확인하여 이 병의 주 원인균은 Waitea circinata로 판명되었다. 따라서 본 병은 zoysiagrass에서 Waitea circinata에 의해 최초로 국내에서 발생한 병으로 사료되며, 기본 병명과 동일하게 Waitea ring patch로 사용하기를 제안하고자 한다.

신규 목질진흙버섯 KACC93057P의 배양적, 형태적 특성 (Morphological and cultural characteristics of a novel Phellinus linteus KACC93057P)

  • 국내에서 분리한 신규 P linteus KACC93057P의 자실체의 형태적 특성을 육안적 으로 관찰한 결과 대가없이 기주부착형으로 자실체표면은 단단한 목질형태 (hardy woody) 로 동심원으로 윤층대 (zonate)와 홈을 형성하고 담갈색을 띠고 있었다. 자실체 뒷면 테두리는 진황색, 내부는 담황갈색을 띠며 현미경 미세구조를 관찰한 결과 관공은 원형또는 각진형이며, 관공구는 5-7 per mm크기로 나타났다. hyphal system은 2분지형이며 포자는 $4.5-6{\times}4-5{\mu}m$크기로 난형또는 유구형 담갈색을 띠어 P linteus와 유사한 특성을 보였다. YGMA배지상에서 KACC93057P는 $25-30^{\circ}C$가 최적온도로 균사체 성장률이 장수상황과 고려상황에 비하여 10-25% 높았으며 균사밀도가 다른 균주보다 현저히 높게 나타났다. ITS-rDNA염기서열비교결과 96% 이상의 상동성으로 I. lonicericola, I. linteus, I. baumii와 근연관계를 형성 하였다.

Cultural characteristics of Cordyceps militaris strain 'Yedang 3' on various media and nutritional conditions

  • The effects of environmental and nutritional requirement such as temperature, pH, different media, carbon, nitrogen, and carbon-to-nitrogen ratio on the mycelial growth of Cordyceps militaris strain 'Yedang 3' were studied. The optimum temperature and pH for the growth of mycelium were $20-25^{\circ}C$ and pH 6-7, respectively. Out of ten media tested, mushroom complete media (MCM) was the best medium for fast mycelial growth, and Sabouraud's dextrose agar yeast extract (SDAY), malt extract yeast extract agar (YMA) also were favored. The color and shape of colonies varied in different media. The best carbon sources for mycelial growth were fructose, mannitol, and sucrose, whereas the best nitrogen sources were tryptone and peptone. However, mycelia grew slowly in inorganic nitrogen compounds such as $NH_4Cl$, $(NH_4)_2SO_4$, $NH_4NO_3$, and $NaNO_3$. The optimum C:N ratio observed on the culture media was 30-40 range. These results provided basic information on cultural characteristics of vegetative growth and might be useful for spawn production in Cordyceps militaris.

Biocontrol of root diseases of fruit trees with fungal viruses

  • Helicobasidium mompa Tanaka and Resellinia necatrix Prillieux cause violet root rot and white root rot of various crops, respectively. Intensive cultural practices, such as the use of dwarf stock, glasshouse cultivation, etc., predispose plants to the diseases. The diseases can be controlled only by biennial drench of 50100L of chemicals for each tree. Biocontrol with soil microorganisms proved ineffective under field conditions. Long-term control may be hampered by the perennial growth of hosts and by the difficulty in the establishment of antagonists in soil. Crop rotation or soil amendment is not applicable, either. Fungal viruses with dsRNA genome (Buck 1986) are promising against root diseases of fruit trees since they exist within the cytoplasm of fungal hyphae and need no effort to help them persist in the field. The viruses are considered to spread though the network of fungal mycelia in the soil once they enter the fungal cytoplasm. Here, we present preliminary results from a project to control the root diseases of fruit trees with dsRNA.(중략)

Verticillium Wilt of Potato Caused by Verticillium albo-atrum in Daegwallyong Area in Korea

  • Verticillium wilt was first observed in 2001 on potatoes (Solanum tuberosum) cv. Superior at Daegwallyong area, one of the major seed potato producing areas in Korea. The wilted potato plants showed typical symptoms including gradual yellowing and interveinal necrosis. There was discoloration in the vascular tissues of the infected stems which turned light brown. Fungal isolates from discolored vascular tissues were whitish to creamy with folding on potato dextrose agar medium, where they used to produce resting dark mycelia but no micro-sclerotia. Conidiophores were septate with side branches, swelled at the base, and arranged in a whorl. Conidia were 2.5-11.2$\times$2.0-4.5 $\mu\textrm{m}$ um in size and were borne in small clusters at the tips of phialides. Optimal temperature range for mycelial growth was $25-30^{\circ}C$. Based on these cultural and morphological characteristics, the fungus was identified as Verticillium albo-atrum Reink & Berth. Pathogenicity tests by root dipping method revealed that the fungus caused the same symptoms as observed in naturally infected potato plants. This is the first report of Verticillium wilt on potato caused by Verticillium albo-atrum in Korea.
