Cauliflower mushroom (Sparassis crispa) is an edible mushroom that shows remarkably high contents of $1,3-{\beta}-D-glucan$ compared to other edible mushrooms. The mushroom is known to give high antitumor and immunologic activities, thus the mushroom is recently cultivated in Japan and Korea. However, cultivation methods are being kept in secret or under patents by some companies with complicated procedures. This study was conducted to solve the problem by providing a simple method for the cultivation of cauliflower mushroom. We could produce the mushroom using sawdust-based media of Larix leptolepis, Pinus densiflora and P. koraiensis with addition of barley flours and sugar. The ratio of the sawdust : barley flours : sugar used for this study was 80:20:3 in dry-weight base, and the water content was adjusted as 65%. The productivity was somewhat different among the species of sawdust media and the strains of Sparassis crispa for the cultivation, and the mushroom production reached up to 177g from 650g base(productivity : 27%). Thus, this medium is strongly recommended as a practical cultivation method for cauliflower mushroom. By the way, further study for the determination of adequate concentration of barley flour and the selection of proper strain for each sawdust species is needed.

The biological carbon dioxide fixation using microalgae and photo-bioreactor has been known as an effective carbon dioxide reduction technology. As algae has many other environmental factor for its growth, the desirable cultivation factors were investigated using a green alga, Euglena gracilis Z. In this study, Euglena gracilis Z showed good $CO_2$ fixation ability in high $CO_2$ concentration of 10-20% and it contained the high protein and vitamin E enough to be used as fodder. For the mass cultivation, the continuous and semi-continuous cultivation methods were employed. The optimum hydraulic retention time (HRT) for the continuous cultivation was 4 days at carbon dioxide concentration of 10%. In this condition, the final cell number was $3.57{\times}10^6/m{\ell}$. The growth of Euglena gracilis Z increased according to the light intensity.
BACKGROUND: In order to cultivate upland crops in reclaimed land, economically appropriate crops should be selected. Jerusalem artichoke (Helianthus tuberosus L.) is one of the primary sources for inulin in higher plants. The objective of this study was to evaluate and compare growth and inulin of H. tuberosus L. according to the cultivation region in Korea. METHODS AND RESULTS: 21 varieties of H. tuberosus L. were collected in Korea, and then 7 varieties of H. tuberosus L. were selected for this study. To compare growth and inulin, those varieties were cultivated in reclaimed land and upland soils. The growth of H. tuberosus L. significantly decreased when cultivated in reclaimed land. There was an increas at 17.8% in the content of white H. tuberosus L. tuber inulin in reclaimed land. CONCLUSION: The content of inulin in H. tuberosus L. tubers was found to be dependent on cultivation region and tuber color. There was a difference of the inulin content between the tubers with different colors in reclaimed land. Purple tubers of P20 were considered suitable varieties for cultivation in reclaimed land. White tubers of W1 and W8 were also considered suitable varieties for cultivation in reclaimed land.
BACKGROUND: Biochar-based fertilizers delay the nutrient release and feature a slow release effect for agricultural and environmental advantages. This experiment was conducted to evaluate agro-environmental effects of different application ratios of modified biochar pellets supplemented. METHODS AND RESULTS: The treatments consisted of the control, 40% N, 60% N and 60% N (0.07M MgO) of modified supplemented biochar pellets (MSBP), which were based on recommended ratio of nitrogen for rice cultivation. For the paddy water, the NH4-N and NO3-N concentrations in whole treatments rapidly increased at 84 days and 40 days after transplanting, respectively. The PO4-P concentrations in the MSBP were generally lower than those of the control. For the paddy soil, NH4-N concentrations in the MSBP were higher than those of the control at 5 days after transplanting, while NO3-N concentrations were not significantly different in the treatments through rice cultivation. P2O5 concentrations in the control were higher than those of the MSBP until 40 days after transplanting while K2O concentrations were not significantly different among the treatment. The highest carbon sequestration was 970 kg ha-1 in the 60% N (0.07M MgO), and the potential carbon storage in the 60% N (0.07M MgO) was higher at 222 kg ha-1 than the control during rice cultivation. It shown that the rice yield in the control was not significantly different from the 40% N and 60% N (0.07M MgO) application plots. CONCLUSION: Application of MSBP for rice cultivation was effective for carbon sequestration and agro-environmental effects even though nitrogen application ratio was reduced at 40% based on recommended application ratio of fertilizer.
Purpose : This study was conducted to investigate the inhibitory effects of Cortex ulmi pumilae on cell proliferation in HeLa cell. Methods : Human uterine cervical carcinoma HeLa cells were cultured in the 1%, 5% and 10% concentration of Cortex ulmi pumilae solution for 24 hours, 48 hours and 72 hours for the direct inhibitory effects of Cortex ulmi pumilae. Afterwards, we executed the analysis of the effect of Cortex ulmi pumilae solution on cell proliferation inhibition using XTT assay, DNA fragmentation, molecular biological method through MAP kinase activity and FACS analysis of caspase activity in the HeLa cells. Results : After 48 and 72 hours cultivation, the HeLa cells showed the concentration-dependently significant increase in all Cortex ulmi pumilae solution containing groups compared to the control. In the FACS analysis, all Cortex ulmi pumilae solution containing groups showed concentration-dependent increase compared to the control after 24 hours cultivation and the caspase-3 activities were decreased in all Cortex ulmi pumilae solution containing groups compared to the control after 24, 48 and 72 hours cultivation. After 48 and 72 hours cultivation, we could examined the apparent DNA fragmentation in all Cortex ulmi pumilae solution containing groups. In the XTT study, all Cortex ulmi pumilae solution containing groups showed concentration-dependent decrease compared to the control after 24 and 72 hours cultivation but 10% group after 48 hours and 5% and 10% groups after 72hours were presumed statistically significant differences. The expressions of MAP kinase were decreased in all Cortex ulmi pumilae solution containing groups compared to the control after 24, 48 and 72 hours cultivation. Conclusion : From this study we could suggest that Cortex ulmi pumilae be available to the inhibition of apoptosis of human cervical carcinoma cell line in vitro.
Background: Ginsenosides are not only the principal bioactive components but also the important indexes to the quality assessment of Panax ginseng Meyer. Their contents in cultivated ginseng vary with the growth environment and age. The present study aimed at evaluating the significant difference between 36 cultivated ginseng of different cultivation areas and ages based on the simultaneously determined contents of 14 ginsenosides. Methods: A high-performance liquid chromatography (HPLC) coupled with triple quadrupole mass spectrometer (MS) method was developed and used in the multiple reaction-monitoring (MRM) mode (HPLC-MRM/MS) for the quantitative analysis of ginsenosides. Multivariate statistical analysis, such as principal component analysis and partial least squares-discriminant analysis, was applied to discriminate ginseng samples of various cultivation areas and ages and to discover the differentially accumulated ginsenoside markers. Results: The developed HPLC-MRM/MS method was validated to be precise, accurate, stable, sensitive, and repeatable for the simultaneous determination of 14 ginsenosides. It was found that the 3- and 5-yr-old ginseng samples were differentiated distinctly by all means of multivariate statistical analysis, whereas the 4-yr-old samples exhibited similarity to either 3- or 5-yr-old samples in the contents of ginsenosides. Among the 14 detected ginsenosides, Rg1, Rb1, Rb2, Rc, 20(S)-Rf, 20(S)-Rh1, and Rb3 were identified as potential markers for the differentiation of cultivation ages. In addition, the 5-yr-old samples were able to be classified in cultivation area based on the contents of ginsenosides, whereas the 3- and 4-yr-old samples showed little differences in cultivation area. Conclusion: This study demonstrated that the HPLC-MRM/MS method combined with multivariate statistical analysis provides deep insight into the accumulation characteristics of ginsenosides and could be used to differentiate ginseng that are cultivated in different areas and ages.

Background: The ginseng ginsenosides, which have various physiological activities, are known to be more abundant in the leaves than in the roots, and the consumers' interest in ginseng sprout as a functional vegetable has been increasing. Methods and Results: The aim of this study was to investigate the effects of growth period on growth properties, active ingredients and rheology of ginseng sprouts cultivated in a non-heated greenhouse equipped with a shade net for 60 days, starting from the end of May to the middle of July. The chlorophyll content of the leaves decreased, but their length and width increased with increasing cultivation days. In particular, growth increased significantly until 40 days, but only slightly after 50 days. The stem length did not increase greatly from the 20 th to the 30 th day of cultivation, but increased significantly from the 30 th to the 40 th day, and then further increased gradually. The weight of the leaves, stems, and roots increased slightly, but not change significantly. After 40 days of cultivation, the total ginsenoside content increased by 1.07 times in the leaves and decreased by 0.80 times in the roots with increasing cultivation days. The leaf contents of ginsenosides $Rg_1$, Re, $Rb_1$, Rc, $F_3$ and $F_4$ increased with increasing cultivation days. The rheological properties of ginseng sprout showed the greatest influence on stem hardening with increasing cultivation days. Conclusions: Therefore, based on the growth characteristics, active ingredients and physical properties, 40 days after sowing was considered to be an appropriate harvesting time for ginseng sprouts.

Background : In order to determine the effects of planting methods on root growth of ginseng varieties, two different methods, direct seed sowing and transplanting were compared in terms of their effects on different root growth characteristics. Methods and Results : Higher fresh root weight was observed in ginseng grown by direct seed sowing. Direct seed sowing of three cultivars (Sunhyang, Chungsun and K-1) resulted in higher yield, whereas no difference was observed in the yield of one cultivar (Chungsun). Gumpoong was highly tolerant to physiological stress, as it showed fewer symptoms of rusty and rough skin root diseases in both direct seed sowing and transplanting. The average main root length per total root length of ginseng grown by direct seed sowing was 33.6%, whereas that of ginseng grown by the average of those by transplanting was 22.4%. Other root growth characteristics, including root length, main root diameter, and number of side roots, improved when the direct seed sowing method was used. Conclusions : To our knowledge, this is the first study reporting the differences in root growth parameters of ginseng varieties grown by direct seed sowing or transplanting at the same planting density. Because of the advantages of direct sowing during ginseng planting, developing new varieties and improving cultivation methods are imperative.

As the low labor-productivity is being floated up to be the most limiting factor in barley cropping, the endeavor of many researchers concerned have been concentrated on recovering from that. For this reason, in this experiment 8 kinds of sowing methods were employed to survey the yield productivity and the labor-requirement of each methods. And with promising the possibility of consistent mechanization, both prediction of the assurable requirement of labor investment and calculation of the labor productivity in each method were estimated so that the possibility of labor-saving in barley cultivation investigated. Also sowing methods for labor-saving cultivation were presented according to the labor-structure of farm as divided with the phases.
