Epidemiology and Control of Strawberry Bacterial Angular Leaf Spot Disease Caused by Xanthomonas fragariae

  • Strawberry bacterial angular leaf spot (ALS) disease, caused by Xanthomonas fragariae has become increasingly problematic in the strawberry agro-industry. ALS causes small angular water-soaked lesions to develop on the abaxial leaf surface. Studies reported optimum temperature conditions for X. fragariae are $20^{\circ}C$ and the pathogen suffers mortality above $32^{\circ}C$. However, at the nursery stage, disease symptoms have been observed under high temperature conditions. In the present study, results showed X. fragariae transmission was via infected maternal plants, precipitation, and sprinkler irrigation systems. Systemic infections were detected using X. fragariae specific primers 245A/B and 295A/B, where 300-bp and 615-bp were respectively amplified. During the nursery stage (from May to August), the pathogen was PCR detected only in maternal plants, but not in soil or irrigation water through the nursery stage. During the cultivation period, from September to March, the pathogen was detected in maternal plants, progeny, and soil, but not in water. Additionally, un-infected plants, when planted with infected plants were positive for X. fragariae via PCR at the late cultivation stage. Chemical control for X. fragariae with oxolinic acid showed 87% control effects against the disease during the nursery period, in contrast to validamycin-A, which exhibited increased efficacy against the disease during the cultivation stage (control effect 95%). To our knowledge, this is the first epidemiological study of X. fragariae in Korean strawberry fields.

Utilizing the grazing effect of fresh water clams (Unio douglasiae) for the remediation of algal bloom during summer

  • The occurrence of 'algal bloom', caused by the mass proliferation of phytoplankton, causes serious problems in streams and lakes in Korea. Therefore, in this study, the phytoplankton filter-feeding trait of Unio douglasiae, a type of freshwater clam, was used to reduce the algal bloom in outdoor water tanks during the summer. This involved the construction of a U. douglasiae cultivation apparatus, wherein 1,000 clams were divided into 8 rectangular baskets arranged in the shape of an empty square. The control tank was manufactured in exactly the same shape within the water tank, but without the addition of clams. The algal bloom-reducing effect of U. douglasiae was confirmed by the measurement of (and comparing between) the water quality at the center and periphery of the test and control cultivation apparatus. Water quality measurements included the measurement of water temperature, pH, turbidity, dissolved oxygen (DO) content, and chlorophyll-${\alpha}$ concentrations; the water quality was measured twice a month between June and November 2014.The results of these analyses did not show a significant difference in water quality (temperature, pH, turbidity, DO) between the center and periphery of the test and control tanks. However, the chlorophyll-${\alpha}$ concentration was observed to be much lower at the center of the test tank compared to that at the center and periphery of the control tank, as well as at the periphery of the test tank. This was believed to be a result of the U. douglasiae surrounding the center of the test tank, which prevented the influx of plankton from the periphery. Accordingly, the results of these analyses suggest the possibility that U. douglasiae cultivation could reduce the proliferation of algal blooms in lakes and streams during the summer. In particular, these results indicate possible improvements in U. douglasiae activity (reduction in algal blooms) by their effective arrangement in the water bodies.

당귀작약산(當歸芍藥散)이 HeLa Cell의 증식억제(增殖抑制)와 사멸(死滅)에 미치는 영향(影響) (Inhibitory Effects of Dangguijakyaksan on Cell Proliferation in HeLa Cells)

  • Purpose : This study was conducted to investigate the inhibitory effects of Dangguijakyaksan on cell proliferation in HeLa cells. Methods : Human uterine cervical carcinoma HeLa cells were cultured in the 1%, 5% and 10% concentration of Dangguijakyaksan extract solution for 24 hours, 48 hours and 72 hours for the direct inhibitory effects of Dangguijakyaksan. Afterwards, we executed the analysis of the effect of Dangguijakyaksan extract solution on cell proliferation inhibition using XTT assay, molecular biological method through MAP kinase activity and FACS analysis of caspase activity in the HeLa cells. Results : After 24, 48 and 72 hours cultivation, Dangguijakyaksan extract solution group showed significant decrease of HeLa cells except 1% solution after 24 hours compared with the control group. In the FACS analysis, Dangguijakyaksan extract solution groups showed increase of caspase activity except 1% solution after 48 hours compared with the control group. In the XTT assay, the caspase-3 activities were increased in Dangguijakyaksan extract solution groups except 1% solution after 24 hours in a dose-dependent manner. In the XTT study, cell activities were significantly decreased in 10% Dangguijakyaksan extract solution groups after 48 and 72 hours cultivation compared with the control group. In all Dangguijakyaksan extract solution groups, The activities of MAP kinase were decreased after 24, 48 and 72 hours cultivation compared with the control group. Conclusion : It could be concluded that Dangguijakyaksan is available to the inhibition of proliferation of human cervical carcinoma cell line in vitro.

  • Long-term repeated culturing of biocontrol agents on a certain medium often results in reduced biocontrol efficacy and altered physiology. Effect of culturing media on biocontrol ability and physiological state of Burkholderia gladioli strain B543 was investigated. Over 20 times repeated cultivation of B. giadioli strain B543 on Kings B medium or nutrient agar medium showed improved biological control of cucumber damping-off caused by Pythium ultimum, while one time cultivation on KB or NA did not. The repeated cultivation also induced the physiological changes of the biocontrol agent such as antifungal activity and the production of protease and siderophore. Our result indicates that adaptation to proper culturing medium can alter biocontrol ability and must consider in optimizing the use of biocontrol agents.

  • A novel integrated method for increasing dissolved oxygen concentration in culture media has been developed. It involves adding hydrogen peroxide to the medium, which is then decomposed to oxygen and water by catalase and adding vegetable oil to the medium as antifoam agent and oxygen vector. A new apparatus for automated addition of hydrogen peroxide to the bioreactor to keep the dissolved oxygen concentration constant over the range $10-100%\;{\pm}\;5%$ was tested. A significant increase (over threefold) of cultivation time was obtained while the dissolved oxygen concentration remained stable ($30%\;{\pm}\;5%$). Therefore, use of corn oil mixed with Ca-stearate as oxygen vector and antifoam and hydrogen peroxide as oxygen source to control excessive foam that was generated by microorganism biosurfactant, GB16-BS produced at Bacillus sp. GB16 cultivation was appropriate for stable cultivation.

  • A precision planting system using computer controlled technology for mulching cultivation was developed and tested . The system consisting of a micro-computer, several optical fiber sensors and control actuators realized the mechanization of the precision planting operation. The film hole positions, existences of a seed on shutter were detected and the planting speed was measured. The shutter opening mechanism and a seed metering device driven by a stepping motor were controlled, automatically . The planting timing of the shutter opening mechanism were analyzed from a video camera motion analysis, theoretically. The results showed a sufficient accuracy of a seed planted into the center a film hole with a variety of planting speeded. The gravity point positions in film hole of seeds planted by the system just were within the area of +-5mm of the hole center when the hole diameter was 40mm.

  • Culture conductivity and on-line NADH fluorescence were used to measure cellular growth in plant cell suspension cultures of Podophyllum hexandrum. An inverse correlation between dry cell weight and medium conductivity was observed during shake flask cultivation. A linear relationship between dry cell weight and culture NADH fluorescence was obtained during the exponential phase of batch cultivation In a bioreactor under the pH stat (pH 6) conditions. It was observed that conductivity measurement were suitable for biomass characterisation under highly dynamic uncontrolled shake flask cultivation conditions. However, if the acid/alkali feeding is done for pH control the conductivity measurement could not be applied. On the other hand the NADH fluorescence measurement allowed online-in situ biomass monitoring of rather heterogenous plant cell suspension cultures in bioreactor even under the most desirable pH stat conditions.

  • An Aero-NFT nutriculture system using microcomputer for cultivation of greenhouse melon was developed and the performance of the system was evaluated through experiments. The system could control temperature, EC and pH of the nutrient solution within the error ranges of $\pm$ 0.2$^{\circ}C$, $\pm$ 0.2 mS/cm, $\pm$ 0.1 pH, respectively. The results of cultivation experiment showed that temperature, EC and pH of the nutrient solution were generally controlled within the setting ranges during cultivation period. The growth results were good until pinching, but the fruit quality of melons was not high except sweetness and shape. To optimize performance of the system, more techlical information for nutriculture of greenhouse melon was needed.
