A green microalga, Chlorella vulgaris UTX 259, was cultivated in a bench-scale raceway pond. During the culture, 15%(v/v) $CO_2$ was supplied and industrial wastewater discharged from a steel-making plant was used as a culture medium. In a small scale culture bottle, the microalga grew up to 1.8 g $dm^{-3}$ of cell concentration and ammonia was completely removed from the wastewater with an yield coefficient of 25.7 g dry cell weight $g^{-1}\;NH_3-N$. During the bottle-culture, microalga was dominant over heterotrophic microorganisms in the culture medium. Therefore, the amount of carbon dioxide fixation could be estimated from the change of dry cell weight. In a semi-continuous operation of raceway pond with intermittent lighting (12 h light and 12 h dark), increase of dilution rate resulted in increase of the ammonia removal rate as well as the $CO_2$ fixation rate but the ammonia removal efficiency decreased. Ammonia was not completely removed from the medium (wastewater) of raceway pond which was operated in a batch mode under a light intensity up to 20 klux. The incomplete removal of ammonia was believed due to insufficient light supply. A mathematical model, capable of predicting experimental data, was developed in order to simulate the performance of the raceway pond under the light intensity of sun during a bright daytime. Simulation results showed that the rates of $CO_2$ fixation and ammonia removal could be enhanced by increasing light intensity. According to the simulation, 80 mg $dm^{-3}$ of ammonia in the medium could be completely removed if the light intensity was over 60 klux with a continuous lighting. Under the optimal operating condition determined by the simulation, the rates of carbon dioxide fixation and ammonia removal in the outdoor operation of raceway pond were estimated as high as $24.7 g m^{-2} day^{-1}$ and $0.52 g NH_3-N m^{-2} day^{-1}$, respectively.

This study was carried out to investigate the amounts of vitamin C in 22 sweet potato cultivars cultivated in Korea as well as evaluate the effects of cooking methods on vitamin C contents. Methods for determining vitamin C was validated by determining linearity, specificity, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy using HPLC. Results showed high linearity in the calibration curve with a coefficient of correlation ($R^2$) of 0.9999. The LOD and LOQ values for ascorbic acid (AA) were 0.03 and $0.10{\mu}g/mL$, respectively. The relative standard deviations (RSDs) for intra- and inter-day precision of AA were less than 5%. The recovery rates of AA and dehydroascorbic acid (DHA) were in the range from 98.21~98.64 and 98.28~100.68%, respectively. Depending on cultivar, contents of AA, DHA, and total ascorbic acid (TA) in sweet potatoes varied in the range from 37.76 (Sinyulmi)~89.25 (Juhwangmin), 23.37 (Sinjami)~63.94 (Sinyulmi), and 68.52 (Sinjami)~115.95 (Juhwangmin) mg/100 g, respectively, and their average levels were $56.98{\pm}12.53$, $36.46{\pm}9.03$, and $93.44{\pm}12.00mg/100g$, respectively. The average TA levels were also dependent on flesh color, whish was significantly higher in general sweet potato and orange sweet potato than in purple sweet potato. Steaming, baking, and frying processes significantly reduced AA (10.61~58.41%), DHA (2.57~52.81%), and TA (14.54~49.92%) contents in sweet potatoes. The highest reduction of AA, DHA, and TA contents was observed after baking, followed by steaming and frying. We expect that the basic information provided by this study will be useful to plant breeders and food scientists.
Pleurotus ostreatus, the oyster mushroom, is one of the most widely cultivated and important edible mushrooms in the world. In order to study the developmental process of P. ostreatus and its regulatory mechanism, a new culturing method needs to be established for inducing the fruiting body and sporulation in the laboratory. In this study, we have examined whether the fruiting body of P. ostreatus can be formed on the plastic petri dish which are commonly used for cell culture in the laboratory. The strain was cultured on $60{\times}15mm$ plastic petri dish with potato dextrose agar media at $28^{\circ}C$ for mycelial growth and then at $18^{\circ}C$ for the formation of primordia and fruiting bodies within plant growth chamber. The development of primordia into fruiting bodies was achieved on cultured dishes under air ventilation. At the primordia stage, the normal formation of fruiting body was blocked by sealing the plastic dish with parafilm. The periods requiring for the formation of primordia and fruiting bodies were examined on the dish culture. About 96% and 76% of cultured samples formed primordia and fruiting bodies under the optimal conditions during ten weeks of culture, respectively. These culturing periods, however, were changed by the mechanical injury treatment to mycelia. As other factors affecting the fruiting body formation, the effects of light and cold shock have been tested. No fruiting formation was observed on the cultured dishes under the dark. The cold shock treatment by storing cultured dishes for one day at $4^{\circ}C$ did not have any significant effects in the fruiting body formation. Spores of fruiting bodies acquired from the petri dishes could be germinated on culture media at $28^{\circ}C$. These results suggest that the fruiting bodies of P. ostreatus can be formed on the experimental petri dish and this dish-culturing method is useful for understanding of the developmental process of P. ostreatus in the laboratory. Furthermore, the dish-culturing method is able to shorten the life cycle of P. ostreatus without requiring large area and expensive device.
To determine the possibility of solidified se wage sludge for use as a soil cover material in reclaimed land, the growth of energy crops and soil chemical properties investigated in each experimental plots during 2 years (2010 and 2011). The experimental plots consisted of the mixing with solidified sewage sludge plot (SS50), the covering with solidified sewage sludge plot (SS100), and the original reclaimed land plot (ORL) on reclaimed land for the intended landfill in Sudokwon Landfill Site Management Corporation (SLC). Plant height, measured in the second year (2011), was highest in the Geodae 1 grown at plots treated with solidified sewage sludge. The growth of energy crops cultivated in both SS50 and SS100 were better than in ORL. The contents of organic matter (OM) and total nitrogen (T-N) at both SS50 and SS100 were considerably higher than that of the ORL over 2 years. However, the soil from ORL showed higher salinity with high contents of exchangeable $Na^+$ cation than that of SS50 and SS100 over 2 years. We consider that soil chemical and physical properties on reclaimed land used in this study could be improved by the application of solidified sewage sludge due to following reasons. Firstly, the application of solidified sewage sludge may provide soil nutrients on reclaimed land i.e. the growth of energy crops better than in ORL, resulted in more OM and T-N contents in SS50 and SS100. Secondly, the top layers mixed or covered with solidified sewage sludge on reclaimed land may be prevented the salinity accumulation due to capillary rise to surface soil, and improved the cultivation layer for effectively propagating the rhizomes of energy crops. Thus the solidified sewage sludge may be a great soil cover materials for cultivation of bioenergy crops in reclaimed land.
The object of this stduy was to investigate the pollution by Pb in rice plants which were cultivated in paddy field soils located near roadside areas and to discover the relationship between lead content in rice plants and soils. Samples of soils and rice plants were collected directly from paddy fields at various distances from highways and expressways located in Kyungpook province. The results obtained were as follows : 1. The average content of Pb in each part of the rice plant was 12.8ppm in roots, 4.8ppm in leaves, 4.3ppm in stems, and 0.4ppm in brown rice. 2. The content of Pb in roots showed highly positive correlation with the Pb content of the soil. 3. The Pb content in stems, leaves, and brown rice showed positive correlation with the Pb content in roots. 4. The content of organically bounded, carbonate, and sulfide Pb in soils showed highly positive correlation with the Pb content in roots. 5. The absorption ratio showed positive correlation with soil pH and negative correlation with the content of Fe in soils.

Composting of livestock feces is economic and safe process to decrease the possibility of direct leakage of organic pollutants to ecosystem from commercial and environmental point of view. This study was conducted with three different experiments related to composting of livestock feces. The purpose of experiment 1 was to investigate changes of characteristic of compost pile during composting period by low temperature in cold season. To compare composting effect of experimental compost pile and control pile exposed in cold air, experimental compost piles were warmed up by hot air until their temperatures were reached at $35^{\circ}C$. Sawdust, Ricehull and Ricestraw were mixed with livestock feces as bulking agent. The highest temperatures of compost pile during composting period were in sawdust, rice hull, rice straw, and control were $75^{\circ}C$, $76^{\circ}C$, $68^{\circ}C$, $45^{\circ}C$ respectively. Moisture content, pH, C/N and volume of compost were decreased during composting period. Experiment 2 was carried out to study utilization effect of compost by plant. A corn was cultivated for 3 years on fertilized land with compost and chemical fertilizer. The amount of harvest and nutrition value of corn were analyzed. In first year of trial, the amount of harvest of corn on land treated with compost was lower by 20% than that of land treated with chemical fertilizer. In second year, there was no difference in yield of com between compost and chemical fertilizer. In third year, the yield of com on land fertilized with compost was much more than that of land fertilized with chemical fertilizer. The purpose of experiment 3 was to estimate the decrease of malodorous gas originating from livestock feces by bio-filter. Four types of bio-filters filled with saw dust, night soil, fermented compost and leaf mold were manufactured and tested. Each bio-filter achieved 87-95% $NH_3$ removal efficiency. This performance was maintained for 10 days. The highest $NH_3$ removal efficiency was achieved by leaf mold on the first day of operation period. It reduced the concentration of $NH_3$ by about 95%. Night soil and fermented compost showed nearly equal performance of 93 to 94% for 10 days from the beginning of operation. The concentration of hydrogen sulfide and methyl mercaptan originating for compost were equal to or less than $3mg/{\ell}$ and $2mg/{\ell}$, respectively. After passing throughout the bio-filter, hydrogen sulfide and methyl mercaptan were not detected.

The aim of this study was to evaluate key properties of the prickly pear cactus (Opuntia ficus-indica var. saboten (OFI) ie, levels of key chemicals (carotenoids, flavonoids and polyphenolic compounds as well as phenolic acid) and its antioxidative potential, depending on where the plant had been cultivated in Korea. The levels of flavonoids and polyphenolic compounds in OFI were 55.45~65.32 mg (+)-catechin/g and 149.00~181.15 mg gallic acid/g, respectively. Protocatechuic acid was the most abundant phenolic acid in the ON1 and ON2 (161.90 and $196.25{\mu}g/g$ DW (dry weight)). Nineteen flavonoids were identified and analyzed by LC-ESI-MS in cladodes from OFI. Narcissin was the most abundant flavonoid in all of the samples ($1,241.89{\sim}1,775.10{\mu}g/g$ DW). Capxanthin and zeaxanthin were the most abundant carotenoids in OFI (64.88~128.08 and $48.10{\sim}93.82{\mu}g/g$ DW). The level of DPPH radical and ABTS radical scavenging activities in OFI were 10.78~25.35 and 16.85~34.16 mg AA eq/100 g, respectively. OFI by cultivar has different kind of phenolic acid, flavonoids, and carotenoids. Therefore, dietary intake of cladodes from OFI may be helpful for improving human health.
The highland area remained bare for the winter and spring seasons and this condition has resulted in soil erosion. In this areas, crop rotation by legume has not been commonly practiced. Thus, this study was carried out to evaluate the effects of the growth characteristics and nitrogen yield as a green manure crop according to sowing seasons of hairy vetch and woolly pod vetch. In this experiment, we was investigated the growth characteristics, winter tolerance, soil coverage and productivities of hairy vetch and woolly pod vetch. We cultivated two hairy vetch(Madison, H1) and two woolly pod vetch(Naomi, Haymaker) varieties in the highland area from 2006 to 2007. The result are summarized as follows; The plant height of hairy vetch increased with the progress of growth stage until flowering stage. The wintering rate of hairy vetch was $79{\sim}83%$ in highland area. But, the wintering rate of woolly pod vetch was $29.5{\sim}39.2%$. The 100% soil coverage of hairy vetch was reached between end of May and beginning of June under autumn sowing, and between end of June and beginning of July under spring sowing. The weeds density of hairy vetch field under autumn sowing was lower than that under spring sowing. Dry matter yield of hairy vetch was significantly higher than that of woolly pod vetch under autumn sowing. The dry matter yield of hairy vetch and woolly pod vetch were $5,255{\sim}5,405kg/ha$ and $3,520{\sim}3,640kg/ha$, respectively. But, hairy vetch in spring sowing was not significantly higher fresh matter yield than that of woolly pod vetch. The nitrogen content in hairy vetch and woolly pod vetch was $2.98{\sim}3.08$ and $2.74{\sim}3.21%$, respectively. There were no significant difference in nitrogen content of the hairy vetch and woolly pod vetch under the spring sowing. N-uptake of hairy vetch was significantly higher than that of woolly pod vetch in autumn sowing. The N uptake was $156{\sim}164\;kg/ha$ in hairy vetch and $96{\sim}101\;kg/ha$ in woolly pod vetch under autumn sowing. It was thought that hairy vetch was better than woolly pod vetch as a green manure crop in autumn sowing considering wintering rate and yield of dry matter in highland area. And woolly pod vetch could use as a green manure crop under spring sowing in highland area.

Scutellaria baicalensis G. was cultivated in plots of different soil textures which were sand loam(SL), loam(L) and clay loam(CL). Also three fertilizer conditions were used; non application (NA)treatment, rice straw manure(RSM) treatment and N-P-K, chemical fertilization(CF) treatment. The chemical-physical properties, such as organic matter, available phosphate, $K_2O$, CaO, clay contents and porosity ratio in CL plot with RSM application were the most proper in CL plot and RSM application for the culture of S. baicalensis plants. RSM had very high contents of total nitrogen, 2.25% and C/N ratios, 21.4. Thus the growth of shoot and root in loam plot with RSM treatment were greater compared to that of CL plot with NA treatment. Whereas, The highest baicalin, baicalein and wogonin contents in roots were found in CL plot with RSM treatment. There was significantly positive correlation between aerial and underground parts of plant, yield and contents of T-N, $K_2O$ but negatively correlated with the contents of baicalin, baicalein and wogonin in S. baicalensis roots.
This study was carried out to investigate the difference of growth characteristics and the content of root chemical components in four years old ginseng by paddy and upland cultivation at farmers' field in Korea. Proportions of silt, clay, liquid phase and porosity were higher in paddy soil than upland soil. The range of liquid phase was $17.5{\sim}19.5%$ in paddy and $7.0{\sim}12.8%$ in upland during growth period. EC and the other contents of OM, $NO_3^-,\;K_2O$, and Mg in paddy soil were higher than those of upland soil, while the contents of $P_2O_5$ and Ca were less than those of upland soil. The levels of chemical components of tested soil exceeded recommended range in EC, $NO_3^-$ and Ca of paddy soil, and in $P_2O_5$ and Ca of upland soil. Stem length, fresh root weight and total dry weight per plant in paddy were greater than those of upland. Root weight in paddy-ginseng showed a great increase on September, while it was not increased in upland because of early defoliation. Net assimilation rate and crop growth rate by paddy and upland cultivation showed distinct differences on May and September, and those of paddy-ginseng were higher than those of upland-ginseng. Yield and ratio of red-colored root showed no significant difference by paddy and upland cultivation, while significant differences were observed in diameter and length of primary root, contents of crude saponin and 50% ethanol extracts of primary root, and water content of root. Hardness of primary root showed no significant difference by paddy and upland cultivation until August, but it showed distinct difference on September, at which the hardness in upland cultivation was drastically decreased.
