Molecular differentiation of Russian wild ginseng using mitochondrial nad7 intron 3 region

  • Background: Cultivated ginseng is often introduced as a substitute and adulterant of Russian wild ginseng due to its lower cost or misidentification caused by similarity in appearance with wild ginseng. The aim of this study is to develop a simple and reliable method to differentiate Russian wild ginseng from cultivated ginseng. Methods: The mitochondrial NADH dehydrogenase subunit 7 (nad7) intron 3 regions of Russian wild ginseng and Chinese cultivated ginseng were analyzed. Based on the multiple sequence alignment result, a specific primer for Russian wild ginseng was designed by introducing additional mismatch and allele-specific polymerase chain reaction (PCR) was performed for identification of wild ginseng. Real-time allele-specific PCR with endpoint analysis was used for validation of the developed Russian wild ginseng single nucleotide polymorphism (SNP) marker. Results: An SNP site specific to Russian wild ginseng was exploited by multiple alignments of mitochondrial nad7 intron 3 regions of different ginseng samples. With the SNP-based specific primer, Russian wild ginseng was successfully discriminated from Chinese and Korean cultivated ginseng samples by allele-specific PCR. The reliability and specificity of the SNP marker was validated by checking 20 individuals of Russian wild ginseng samples with real-time allele-specific PCR assay. Conclusion: An effective DNA method for molecular discrimination of Russian wild ginseng from Chinese and Korean cultivated ginseng was developed. The established real-time allele-specific PCR was simple and reliable, and the present method should be a crucial complement of chemical analysis for authentication of Russian wild ginseng.

HPLC를 이용한 인삼, 홍삼, 산양산삼 및 홍산삼의 성분 비교 분석 (Component Analysis of Cultivated Ginseng, Red Ginseng, Cultivated Wild Ginseng, and Red Wild Ginseng Using HPLC Method)

  • Objectives The aim of this experiment is to provide an differentiation of ginseng, red ginseng, cultivated wild ginseng(CWG), and red wild ginseng(RWG) through component analysis using HPLC(High Performance Liquid Chromatography, hereafter HPLC). Methods Comparative analyses of ginsenoside $Rg_3$, ginsenoside $Rh_2$, and ginsenosides $Rb_1$ and $Rg_1$ of various ginsengs were conducted using HPLC. Results 1. CWG was relatively heat-resistant and showed slow change in color during the process of steaming and drying, compared to cultivated ginseng. 2. Ginsenoside $Rg_3$ was not detected in cultivated ginseng and CWG, whereas it was high in red ginseng and RWG. Ginsenoside $Rg_3$ was more generated in red ginseng than in RWG. 3. Ginsenoside $Rh_2$ appreared during steaming and drying of cultivated ginseng, whereas it was more increased during steaming and drying of CWG. 4. Ginsenoside $Rg_1$ content was more increased during steaming and drying of cultivated ginseng, whereas it was more decreased during steaming and drying of CWG. 5. Ginsenoside $Rb_1$ content was increased about 500% during steaming and drying of cultivated ginseng, whereas it was increased about 30% during steaming and drying of CWG, indicating that ginsenoside $Rb_1$ was more generated in red ginseng than in RWG. 6. Ginsenoside $Rg_3$ content was higher, whereas ginsenoside $Rg_1$ content was lower in 11th RWG than in 9th RWG, indicating that ginsenoside $Rg_3$ content was increased and $Rg_1$ content was decreased as steaming and drying continued to proceed. Ginsenoside $Rh_2$ and $Rb_1$ contents began to be increased, followed by decreased after 9th steaming and drying process. Conclusions Above experiment data can be an important indicator for the dentification of ginseng, red ginseng, CWG, and RWG. And the following studies will be need for making good product using CWG.

국내 유기와 관행 재배 인삼의 토양화학성과 생육 특성 조사 (Research of Chemical Properties of Soil and Growth Characteristics of Panax ginseng under Organic and Conventional Cultivation Systems in Korea)

  • 본 연구는 유기재배와 관행재배지의 토양화학성 및 인삼의 생육특성을 비교 분석하기 위하여 수행하였다. 유기재배 인삼과 관행재배 인삼 시료 채취는 전국 14개 지역 52개 포장에서 4, 5, 6년근을 대상으로 2013년부터 2015년까지 3년에 걸쳐 10월부터 11월까지 2개월 동안 이루어졌다. 인삼 유기재배에서는 수량을 높이기 위해 관행재배지와 비슷하게 양분의 함량이 높아지고 있어 토양화학성기준에 맞는 적합한 관리가 요구된다. 유기재배 인삼은 관행재배 인삼보다 생육기간이 60일 이상 짧은 특징을 보였으며 1 ha당 평균 수량도 관행 수량의 60% 수준이었다. 근중은 유기재배 인삼이 관행재배 인삼의 54% 정도였다. 동직경 및 체형계수도 유기재배 인삼이 관행재배보다 더 작아 유기재배 인삼이 가늘고 긴 특징을 보였다. 근장은 답전윤환지의 비율이 낮은 관행재배 인삼 5년근에서 길었다. 지근의 수는 직파재배의 비율이 높은 유기재배 인삼 5, 6년근에서 적었다. 건물중은 관행재배보다 조기낙엽이 발생된 유기재배 인삼의 5, 6년근에서 뚜렷하게 낮았다. 동할과 뿌리썩음병은 관행재배에서 많은 경향이었는데 동할은 4, 6년근에서 뚜렷하게 많았으며 뿌리썩음병은 5, 6년근에서 많이 발생되었다. 토양수분의 영향을 많이 받는 적변과 선충의 피해는 유기재배 4년근에서 많았다. 즉 유기재배 인삼은 관행재배보다 조기낙엽 현상이 뚜렷하여 수량 및 근중이 낮은 반면 생리장해는 적었다. 유기재배 인삼의 수량을 높이기 위해서는 조기낙엽의 문제를 극복할 수 있는 재배 기술적인 연구가 필요하며 생리장해를 줄일 수 있는 토양 수분관리가 필요하다.

처치온도에 따른 초미분화 산양삼의 홍삼화 연구

  • Objectives : The aim of this study was to find optimal conditions for producing red ginseng from cultivated wild ginseng using the Turbo Mill. Methods : Characteristics of powdered cultivated wild ginseng based on various temperature settings of the Turbo Mill were observed, and changes in the content was measured by HPLC for various ginsenosides. Results : 1. The diameter of cultivated wild ginseng powder ground by the Turbo Mill was around 10${\mu}m$. 2. As the temperature rose, pressure, Specific Mechanical Energy(SME), and density decreased, whileas Water Solubility Index(WSI) increased. 3. As the temperature rose, super fine powder showed tendency to turn into dark brown. 4. Measuring content changes by HPLC, there was no detection of ginsenoside Rg3 and ginsenosideRg1, Rb1, and Rh2 concentrations decreased with increase in temperature. Conclusions : Super fine powder of cultivated wild ginseng produced by the Turbo Mill promotes easy absorption of effective ingredients by breaking the cell walls. Using this mechanism to produce red ginseng from cultivated wild ginseng, it yielded less than satisfactory results under the current experiment setup. Further researches are needed to verify more suitable condition for the production of red ginseng.

수삼의 지역별 연근별 인삼 비사포닌 성분 함량 비교 (Comparison of Non-saponin Composition and Contents in Fresh Ginseng Roots Cultivated in Different Areas and at Various Ages)

  • This study was carried out to obtain the basic information for non-saponin contents that can be used to index fresh ginseng roots (Panax ginseng C. A. Meyer) cultivated in the Republic of Korea and China. Non-saponin components in fresh gingeng roots which were cultivated in various areas and ages in Korea were determined. Acidic polysaccharide, total polysaccharide, crude polyacetylene were quantitatively analyzed by using the method of spectrophotometric determination, while the total protein was analyzed by using Lowry method. The results show that there were no statistically significant differences for the average contents of four non-saponins among 4-years-old, 5-years-old, and 6-years-old fresh ginseng roots. Additionally, this study assessed the average contents of non-saponin components in 4-years-old fresh ginseng roots (Panax ginseng C. A. Meyer) which were cultivated in Korea and China. The result showed that the average contents of crude polyacetylene and acidic polysaccharide were statistically significant. Four-years-old fresh ginseng roots cultivated in Korea had the higher average contents of crude polyacetylene and acidic polysaccharide than those cultivated in China. However the average contents of total polysaccharide and total protein had no statistically significant difference.

산양산삼 추출물이 지방세포의 분화에 미치는 영향 (The Effect of Cultivated Wild Ginseng Extract on Preadipocyte Proliferation)

  • Objectives The purpose of this study is to investigate the effects of cultivated wild ginseng extract on primary cultured preadipocyte and adipocytes. Methods Diminish preadipocyte proliferation does primary role to reduce obesity. So, preadipocytes and adipocytes were performed on cell cultures with using Sprague-Dawley rats and treated with 0.01-1mg/ml cultivated wild ginseng extract. Result At all concentrations, cultivated wild ginseng extract wasn't show the suppress proliferation of preadipocytes significantly and failed to show effects on decomposition of adipocytes except high dosage. Conclusion Based on these findings, cultivated wild ginseng is not a suitable choice for the treatment of localized obesity.

Analysis of Ginsenoside Composition of Ginseng Berry and Seed

  • This study was performed to provide basic information that can be used to differentiate Korean ginseng (Panax ginseng CA. Meyer) berry and seed from American ginseng (Panax quinquefolium L.) seed. Total ginsenoside contents of Korean ginseng berry, Korean ginseng seed, and American ginseng seed were 9.09, 3.30, and 4.06%, respectively. Total ginsenoside content of Korean ginseng berry was about 2.2 to 2.7 times higher than those of Korean ginseng seed and American ginseng seed. Particularly ginsenoside Re content of 4-year cultivated Korean ginseng berry (5.99%) was about 3.6 to 5.4 times higher than that of 4-year cultivated Korean ginseng seed (1.65%) and 4-year cultivated American ginseng seed (1.10%). The contents of total ginsenoside and ginsenoside Re of Korean ginseng berry were about 4.8 and 28 times higher, respectively, than those of 4-year cultivated Korean ginseng root. In general the contents of total ginsenoside and ginsenoside Re of Korean ginseng berry were significantly higher than those of Korean ginseng seed and American ginseng seed.

  • This study was carried out to obtain basic informations that can be used in index for Korea ginseng (Panax ginseng C.A. Meyer) cultivated in East Asia (Geumsan. Ganghwa. Punggi. Umsong, Jinan, Hongchon. Jilin. Nagano). Ginsenosides composition in various Panax ginseng roots cultivated in different places and years were carried by the Shibata method. The average about total saponins and each ginsenosides content of four year-age Ginseng Radix aquosa cultivated in Korea were higher than those of ginseng cultivated for its longer period. (omitted)

인삼, 산양삼 및 산삼의 부위별 Proteome분석 (Proteome Analysis of various types of Panax ginseng using 2-Dimensional Electrophoresis)

  • Objectives : The purpose of this study was to obtain an objective differentiating method for various types of Panax ginseng: ginseng, cultivated wild ginseng, and natural wild ginseng which are distinctive according to their growing environment. Methods : The roots, stem, and leaves of several types of ginseng were collected and comparative analysis of proteome was conducted on each part using 2-DE and the results examined. Results : 1. Proteome images of the respective parts within the samples showed spot-matching in most cases, suggesting that they are genetically identical panax ginseng. 2. Similar distribution patters were seen within the different parts of the Panax ginseng: ginseng, Chinese cultivated wild ginseng, and the 5 and 10 years old Korean cultivated wild ginseng. 3. For a quantitative evaluation of spots showing differences among the samples, 102 spots from the roots, 109 spots from the stems, and 132 spots form the leaves which showed a difference were selected and centrifugal identification was conducted. 4. Peculiar proteins from each respective part of the Panax ginseng were identified and the top 20 spots with significant differences were selected and analyzed in order to provide a differentiation rate among the samples. The accuracy rate ranged between 23.0-38.8%. 5. Differentiation rate of the top 10 spots with significant differences showed a 50-85% accuracy rate, and the differentiation rate was especially high for the stem of Chinese cultivated wild ginseng and Korean cultivated wild ginseng.

Colon26-L5 대장암 세포를 이용한 간전이 모델에 산삼약침 처치가 혈중 cytokine에 미치는 영향 (Effects of Cultivated Wild Ginseng Herbal Acupuncture to the serum cytokine on Hepatic Metastatic Model using Colon26-L5 Carcinoma Cells)

  • Objective : This experiment was conducted to evaluate inhibitory effects against hepatic metastasis by cultivated wild ginseng Herbal Acupuncture. Methods : Colon26-L5 carcinoma cells were injected through hepatic portal vein to induce hepatic metastatic cancer. After treated cultivated wild ginseng Herbal Acupuncture and investigated various kinds of cytokine level using cytokine chip. Results : 1. Mice treated with cultivated wild ginseng Herbal Acupuncture reduced the level of $IL-l{\alpha}$, $IL-{\beta}$, and $TNF-{\alpha}$ compared to the control group. 2. Mice treated with cultivated wild ginseng Herbal Acupuncture was not showed significant change in the level of IL-4, IL-l0, IL-12 and $INF-{\gamma}$ compared to the control group. 3. Observing the level of various kinds of cytokine, cultivated wild ginseng Herbal Acupuncture was suppressed pro-inflammatory cytokine. These findings indicate cultivated wild ginseng Herbal Acupuncture is possible to use the inflammatory disease and futher studies carry out for the explanation of anticancer mechanism.
