Effect of Ethanol Extracts of Youngia denticulata and Youngia sonchifolia on the Serum and Hepatic Lipids and Activities of Ethanol Metabolizing Enzymes in Acute Ethanol-Treated Rats (이고들빼기 및 고들빼기 에탄올 추출물 첨가식이가 급성 에탄올 투여 흰쥐의 혈청과 간지질 및 알코올 대사 효소활성 변동에 미치는 영향)

  • This study examined the protective effects of an ethanol extract of Youngia denticulata leaf (YDL) and Youngia denticulata root (YDR), and Youngia sonchifolia leaf (YSL) and Youngia sonchifolia root (YSR) on acute ethanol-intoxicated rat. The rats were pretreated with an ethanol extract of YDL, YDR, YSL and YSR for 4 weeks before being exposed to ethanol (5 g ethanol, po/kg BW). The biochemical indices (hepatic alcohol metabolic enzymes and serum ALT activities, and hepatic and serum lipid profiles) were examined to evaluate the protective effects. The hepatic ADH activities in all experimental groups were not changed significantly by acute ethanol after a pretreatment with the YS and YD ethanol extracts. In contrast, the ALDH activity in EC (ethanol control) was higher than that of NC (normal control); these activities in the YDL and YSL groups were significantly higher than that of the EC group. On the other hand, acute ethanol exposure resulted in a significant increase in the serum TG, total cholesterol, LDL-cholesterol, hepatic TG, total lipid and cholesterol levels, and serum ALT activity, and a decrease in the serum HDL-cholesterol. A pretreatment with the YS and YD ethanol extracts dramatically attenuated these adverse effects. In particular, the YDL pretreatment markedly suppressed the ethanol-induced increase in the serum and hepatic TG and total cholesterol levels. Furthermore, serum ethanol was decreased by a pretreatment with YSL, YSR, YDL, or YDR. Overall, YD and YS ethanol extracts attenuate acute ethanol-induced hyperlipidemia and fatty liver significantly. Nevertheless, further study will be needed.

The Microbiological Assessment and Identification of Food Utensils and Food Service Facilities in School (학교 급식설비 및 집기류의 미생물학적 위해요소 분석)

  • This study was conducted to evaluate microorganism contamination of food utensils and service facilities in school and to prevent hazards by food poisoning occurrence. As a result, the highest number of microorganism growth plate ($12.3{\pm}2.6$) was detected in total bacteria test plate, and also observed $10.3{\pm}3.9$ growth plates in Staphylococcus aureus test plate and $9.5{\pm}3.9$ growth plates in E. coli and coliform bacteria test plate. But we could detect to the lowest number of growth plates ($1.5{\pm}1.0$) in Vibrio test plate. We also assessed that floors were appeared to the highest microorganism contamination rate in food utensils and service facilities. Therefore, $4.5{\pm}0.6$ growth plates was detected in pre-operation floor and $4.3{\pm}1.0$ growth plates in floor. And high level of microorganism contamination also observed in tables as $3.3{\pm}1.0$ growth plates in cooking table and $3.0{\pm}0.0$ growth plates in dining table. The level of microorganism contamination of food utensils such as kitchen knife, cutting board, and food tray were lower than that in food service facilities. We analysed microorganism contamination according to purpose of use in kitchen knifes and cutting boards. The microorganism contamination rate in fish kitchen knife ($2.0{\pm}0.8$) and fish cutting board ($1.3{\pm}1.5$) were slightly higher than that of others purpose of use. As a result of microorganism identification, various strains of microorganism were contaminated in food service facilities and some strains could detected more than two times. Especially, Staphylococcus aureus was repeatedly identified in cooking table, trench, and kitchen knife. Bacillus cereus was identified in kitchen knife, and then Pseudomonas fluorescens and Pseudomonas aeruginosa were also detected in food utensils and service facilities as known to food spoilage microorganisms. Klebsiella pneumoniae was detected four times repeat, which widely distribute natural environment as normal bacterial flora but sometimes cause acute pneumonia. These results suggest that food utensils and service facilities are contaminated with not only major food poisoning microorganisms such as Staphylococcus aureus, but also food spoilage microorganisms. Taken together, strict personal hygiene control and efficient food service facilities management will be needed to enhance food safety in school feeding and to improve student health.

Removal Effects of Bifenthrin and Metalaxyl Pesticides during Preparation and Fermentation of Baechu Kimchi (배추김치의 제조 및 발효과정 중 Bifenthrin과 Metalaxyl 농약의 제거 효과)

  • The removal effects of two commonly used pesticides of Bifenthrin and Metalaxyl during preparation and fermentation of kimchi were studied. The two pesticides were applied to Baechu cabbages intentionally for 20 seconds; the applied amounts of Bifenthrin and Metalaxyl were 3.02$\pm$0.09 ppm and 6.79$\pm$0.17 ppm, respectively. The Baechu cabbages were washed by water 3 times. Then the residual amounts of the two pesticides of Bifenthrin and Metalaxyl were measured and the removal rates were 21.7% and 16.1%, respectively. When Baechu cabbages were brined with 10% salt solution for 12 hours after the application of the two pesticides, the removal rates significantly increased to 98.7% and 60.8%, and when brined and washed 3 times by water, the removal rates even more increased to 99.7% and 73.4% respectively. Then we made kimchi and investigated the quantities of the residual pesticides during the fermentation at $4^{\circ}C$ for 3 weeks. The residual amounts of the pesticides in kimchi decreased in a time dependent manner, finally the amounts of the pesticides to 0.35$\pm$0.04 ppm and 0.48$\pm$0.06 ppm while the removal rates of the two pesticides were 57.8% and 81.0%, respectively. When the kimchi was fermented at $4^{\circ}C$ and $10^{\circ}C$ for 3 weeks, the removal rates of Bifenthrin were 57.8% and 72.2% and those of Metalaxyl were 81.0% and 85.6%, respectively. Consequently, it appeared that the residual pesticides can be removed during preparation, especially brining; the fermentation process of kimchi also removed the residual pesticides effectively, especially at higher temperature and long period.

Quality Characteristics of Yulmoo Mul-kimchi Containing Saltwort (Salicornia herbacea L.) (함초 분말 첨가 열무 물김치의 품질특성)

  • Saltwort (Salicornia herbacea L.), as a natural additive for regulating Mul-kimchi fermentation, was assessed for physicochemical and sensory characteristics of Yulmoo Mul-kimchi during storage. Saltwort in the form powder was directly added to the Yulmoo Mul-kimchi preparation at 0 (control), 1, 3, 5, and 7% (w/v) per weight of salt to evaluate their physicochemical, sensory, and microbiological characteristics in storage at $10^{\circ}C$ for 30 days. The pH values of all treatments were high, indicating a less acidic environment in all treatments compared to those of the control sample throughout the preservation period. Total acidity increased with storage time as is usually seen with normal kimchi fermentations, whereas the increases were more gradual in the 3 and 5% treatments. The increases in total vitamin C continued until days 6 (control) to 13 (7%), and were different according to the amount of added saltwort and then they decreased after each peak. The fluctuation in reducing sugars fol owed a similar trends of total vitamin C content by showing an initial increase, followed by a decrease based on the saltwort concentration and storage day. The 5% treatment was most effective for suppressing the increase in turbidity among all treatments. The 3 and 5% treatments showed the highest lactic acid bacteria counts during the entire preservation period. In the sensory evaluation results, adding saltwort at more than 3% concentration attained an overall higher scores of acceptability with respect to color, smell, taste, fresh taste, sour taste, crisp, and overall acceptability characteristics. In conclusion, adding saltwort, particularly at concentrations of 3 and 5% extended the preservation period of Yulmoo Mul-kimchi by retarding fermentation effectively.

Healthy Functional Food Properties of Phenolic Compounds Isolated from Ulmus pumila (유근피(Ulmus pumila)로부터 분리한 phenol성 물질의 건강기능식품 활성)

  • The phenolic compounds which were extracted with 70% ethanol from Ulmus pumila for 12 hr were the highest as $17.9{\pm}1.0\;mg/g$. DPPH scavenging activity of 70% ethanol extracts was also the highest as $89.5{\pm}1.9%$ and it was confirmed to be high as 80% over in both of water and 70% ethanol extracts containing $50{\mu}g/mL$ over phenolic concentration. ABTS radical cation decolorization activities of water and 70% ethanol extracts were higher as $96.8{\pm}2.9%$, antioxidant protection factor (PF) was 2.0 PF in 70% ethanol and showed higher activities in both of water and 70% ethanol extracts containing $200{\mu}g/mL$ phenolic concentration as 2.5 PF than BHA. TBARs of 70% ethanol extracts was $86.5{\pm}4.6%$, it showed high anti-oxidative activity in $50{\sim}200{\mu}g/mL$ phenolic concentrations of water and 70% ethanol extracts as 80% over. The angiotensin converting enzyme (ACE) inhibitory activity of Ulmus pumila extracts against hypertension was 77.4% and 90.6% in water and 70% ethanol extracts of $200{\mu}g/mL$ phenolic concentration. Xanthine oxidase inhibitory activity of Ulmus pumila extracts for anti-gout effect was not observed in water extracts, but it showed 30% inhibitory activity in 70% ethanol extracts, and 48.1% at $200{\mu}g/mL$ phenolics concentration.

Antioxidative and Antimicrobial Activities of Monascus pilosus(Corn Silage Mold) Mycelial Extract and Its Culture Filtrate (Monascus pilosus 균사체 및 배양여액의 항산화 및 항균활성)

  • We evaluated the nutritional value of a Monascus pilosus mycelial ethanolic extract (MEM) and culture filtrate (CFM) by determining the contents of monacolin K and citrinin, and by measuring antioxidant and antimicrobial activities. The yields of freeze-dried MEM and CFM powder were 4.02% and 3.35% of wet weight, respectively. Pigment content ($OD_{500}$ value) of MEM (0.79) and CFM (0.63) were lower than those of commercial rice beni-koji ethanolic extracts (EERB) (0.87), but were in good agreement with the L*, a*, and b* values and the hue angles of the products. The total monacolin K content of MEM (24.91 mg%) was higher than those of CFM (1.27 mg%) and EERB (14.65 mg%). However, the active monacolin K content of EERB (5.48 mg%) was higher than those of MEM (3.35 mg%) and CFM (0.4 mg%). Citrinin was not detected in any sample. The total polyphenol content of MEM (4.68%, w/w) was similar to that of CFM (4.29%, w/w), thus 13.75.20.94% higher than that of EERB. The total flavonoid content of EERB was 6.8.7.0-fold higher than those of MEM (0.64%, w/w) and CFM (0.66%, w/w). The total antioxidant capacity of CFM (3.51%, w/w) was 1.62.2.08-fold higher than those of MEM (2.74%, w/w) and EERB (1.69%, w/w). The electron-donating capacities of 1% (w/v) solutions of CFM, MEM, BHT, and EERB were 86.20%, 77.25%, 77.25%, and 44.82%, respectively, and the corresponding reducing powers ($OD_{700}$ values) were 2.1, 2.4, 1.1, and 1.6, respectively. SOD(superoxide dismutase)-like activities were in the order MEM (39.85%) > BHT (37.68%) > EERB (26.70%) > CFM (21.5%). Although the TBARS (% value) of MEM was a little lower than that of BHT, it was higher than those of CFM and EERB. The antibacterial activities of CFM acting on Bacillus brevis and Escherichia coli were somewhat higher than those of MEM, whereas the activities of MEM on Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Staphylococcus epidermidis, and Salmonella enteritidis were higher than those of CFM. However, the antibacterial activities of MEM and CFM were less than those of EERB and BHT. In conclusion, although further studies are needed, we offer experimental evidence that the by-products of M. pilosus MEM and CFM contain significant antioxidant and antimicrobial activities that may be useful in the development of healthy foods.

The work performance and the factor contributing to the work performance of nutrition teacher & school dieticians (영양교사 (정규직)와 학교영양사 (비정규직)의 직무수행도 및 직무수행에 영향을 미치는 요인)

  • Purpose: The objective of this study is to investigate the work performances and the factors contributing to the work performance of nutrition teachers and school dieticians according to employment status in Daejeon and Chungcheongnamdo. Methods: A survey was conducted among 415 school dieticians from selected elementary, middle, and high schools in the area. This survey was used for analysis of the general characteristics and the status of working and foodservice environments. The frequency of occurrence was measured using the ${\chi}^2$-test while analysis of differences in work performance of 57 variables grouped according to 12 categories by mean ${\pm}$ SD was performed using student t-test. In addition, a regression analysis was performed to assess the influence of occupational (working and foodservice) environment on work performances. Results: All 14 environmental factors including general characteristics were different between nutrition teachers and school dieticians. In 12 of 57 variables there were significant differences in work performance between the two groups. When compared using average work performance, 5 domains showed significant difference in work performance between the two groups. These 5 domains were other works (T = 6.050), dietary habit guide (T = 4.789), menu (nutrition) management (T = 3.524), foodservice administration and evaluation (T = 2.757), and production and service management (T = 2.588). Overall, the work performances of nutrition teachers were comparably higher than those of school dietitians. A correlation analysis between occupational (working or foodservice) environments and work performances of the two groups showed that the work performances of nutrition teachers were influenced by the number of employees, number of students, meal frequency a day, annual salary, age, foodservice years of school, and school type. Conclusion: Meanwhile, the work performances of school dieticians were influenced by the number of students, number of employees, meal frequency a day, work hour, education level, age, school type, and service years as a dietitian.
