빨간집모기 유충 내에 있는 직장 상피세포들의 미세구조 (Ultrastructure of the Rectum Epithelial Cells in the Mosquito Larvae, Culex pipiens pallens)

  • 본 연구는 경기도 양평에서 채집한 빨간집모기(Culex pipiens pallens: Culicidae)를 사육하여 유충 시기에 있는 직장 조직을 구성하는 세포들의 미세구조를 관찰하였다. 직장조직은 점막상피조직, 결합조직, 그리고 근육조직 등으로 구성되었으며, 상피 내면에는 않은 큐티클 내막이 덮고 있었다. 점막 상피조직은 단층의 편평상피세포들로 이루어졌으며, 첨단원형질막이 약 $5\sim8{\mu}m$ 길이의 막주름이 잡혀 있었고 그 사이에는 크게 발달한 사립체가 관찰되었다. 측부원형질막의 첨단 쪽에서는 격막연접부위가 길게 관찰되었으며 중간부위는 심하게 주름이 접혀 물질의 수송통로인 세포간극이 불규칙하게 관찰되었고 이 주변에서도 크게 발달한 사립체가 산재해 있었다. 그리고 상피세포의 기저막주름은 세포 내로 짧고 불규칙하게 함입되어 미로 형태로 관찰되었다. 기점막 바깥쪽에는 신경 및 기관소지가 분포하고 결합조직과 전형적인 골격근의 형태를 하고 있는 환상근관 종주근이 장외면에 부착되었다.

중금속이 빨간집모기(Culex pipiens)의 치사율과 대사물질의 변화에 미치는 영향 (Effects of Heavy Metals on Mortality and Metabolite Changes in Mosquitoes, Culex pipiens pallens (Diptera: Culicidae))

  • 중금속이 곤충에 미치는 영향을 조사하기 위하여 빨간집모기(Culex pipiens pallens) 금속용액(Cu, Cd, Hg, Pb, Zn)에서 사육한 후 50% 치사농도($LC_{50}$) 용화시기, 총 지질함량, 지질조성, 총 단백질 함량변화를 측정하였다. 50% 치사농도($LC_{50}$)는 3령 유충으로 24시간 처리한 Hg 처리군에서 $0.45mg\;kg^{-1}$으로 나타나 다른 금속에 비해 독성이 강한 것으로 나타났고, 용화 시기는 처리군이 대조군(129시간)에 비해 지연되었으며 Cd가 처리군 중 273시간으로 가장 늦게 용화되었다. 중금속에 노출된 모기시료에서 총 여섯 종류의 지질 band가 분리되었으며, 이 중 5종의 지질이 동정되었다(phospholipid, cholesterol, fatty acid, triglyceride, cholesterol ester). 모든 처리군의 총 지질함량은 대조군에 비해 감소하였다. 또한, 대조군의 단백질 함량($0.51mg\;ind.^{-1}$)이 처리군의 단백질 함량에 비해 높게 나타났으며, 처리군 중 Hg과 Cd을 처리한 유충의 단백질 함량이 매우 낮게 나타났다. 결론적으로 중금속이 처리된 용액에서 사육된 모기유충은 오염된 환경에 의한 먹이 섭취의 장애 및 중금속의 독성으로 인해 단백질 함량과 지질의 감소가 일어나 발육이 느려지고 용화시기가 길어지는 것으로 생각되며 이는 중금속에 의한 체내 물질대사의 변화를 의미한다. 이러한 결과는 중금속 오염이 모기의 생존과 성장을 억제시켜 모기 개체군 크기에 영향을 줄 것을 나타내며, 추후 연구를 통해 모기의 생육정도를 이용한 오염도 측정에 활용이 가능할 것으로 기대된다.

빨간집모기와 점박이응애에 대한 관동화(Tussilago farfara) 추출물의 살충효과 (Insecticidal Activities of Tussilago farfara Extracts against Culex pipiens pallens and Tetranychus urticae)

  • Aspiring to the new raw materials of insecticides is one of the plant extracts. The material structure of a variety of plant extracts have because the material to the defense of the plant itself, the case of insecticides, using plant extracts, safe, has low toxicity and has the advantage of highly distinctive and fall. Coltsfoot (Tussilago farfara L.) is belonging to the family Compositae, and distributed in all parts of China as medicinal plants and indigenous plants used. Coltsfoot is known that it is effect to respiratory disease and has an antiviral effect. However, the reported insecticidal activity of coltsfoot could be not found. Fortunately, I found insecticidal activities when I was screening the bioassay against several insects with a lot of plant extracts. Using the ethanol extract of the Tussilago farfara, there were insecticidal activities against Culex pipiens pallens and Tetranychus urticae. There were several fractions in ethanol extract of coltsfoot by using various organic solvents. Hexane fraction showed a higher insecticidal activity than any other fraction. It is confirmed that hexane fraction contained pyrethrin by using HPLC analysis. So, it might be suggested that extract of coltsfoot has an insecticidal activity and its effect due to a ingredient of pyrethrin.

Monitoring Culicine Mosquitoes (Diptera: Culicidae) as a Vector of Flavivirus in Incheon Metropolitan City and Hwaseong-Si, Gyeonggi-Do, Korea, during 2019

  • The flaviviruses are small single-stranded RNA viruses that are typically transmitted by mosquitoes or tick vectors and are etiological agents of acute zoonotic infections. The viruses are found around the world and account for significant cases of human diseases. We investigated population of culicine mosquitoes in central region of Korean Peninsula, Incheon Metropolitan City and Hwaseong-si. Aedes vexans nipponii was the most frequently collected mosquitoes (56.5%), followed by Ochlerotatus dorsalis (23.6%), Anopheles spp. (10.9%), and Culex pipiens complex (5.9%). In rural regions of Hwaseong, Aedes vexans nipponii was the highest population (62.9%), followed by Ochlerotatus dorsalis (23.9%) and Anopheles spp. (12.0%). In another rural region of Incheon (habitat of migratory birds), Culex pipiens complex was the highest population (31.4%), followed by Ochlerotatus dorsalis (30.5%), and Aedes vexans vexans (27.5%). Culex pipiens complex was the predominant species in the urban region (84.7%). Culicine mosquitoes were identified at the species level, pooled up to 30 mosquitoes each, and tested for flaviviral RNA using the SYBR Green-based RT-PCR and confirmed by cDNA sequencing. Three of the assayed 2,683 pools (989 pools without Anopheles spp.) were positive for Culex flaviviruses, an insect-specific virus, from Culex pipiens pallens collected at the habitats for migratory birds in Incheon. The maximum likelihood estimation (the estimated number) for Culex pipiens pallens positive for Culex flavivirus was 25. Although viruses responsible for mosquito-borne diseases were not identified, we encourage intensified monitoring and long-term surveillance of both vector and viruses in the interest of global public health.

Pesticidal Constituents Derived from Piperaceae Fruits

  • Fungicidal, insecticidal, and mosquito larvicidal activities of piperidine alkaloids, pipernonaline and piperoctadecalidine, and isobutylamide alkaloids, pellitorine, guineensine, pipercide, and retrofractaminde A, derived from Piperaceae fruits were studied. Pipernonaline and piperoctadecalidine showed potent fungicidal activities against Puccinia recondita with 91 and 80% control values at 500 ppm. Against Phytophthora infestans, pipernonaline showed strong fungicidal activity with 91 and 80% control values at 1,000 and 500 ppm. $LD_{50}$ values of pipernonaline and piperoctadecalidine against Plutella xylostella were 125 and 95.5 ppm, respectively, and that of piperoctadecalidine against Tetranychus urticae was 246 ppm. Against larvae of Aedes aegypti and Culex pipiens pallens, $LD_{50}$ values of pipernonaline were 0.35 and 0.21 ppm, respectively. Highest larvicidal activities of pipercide and retrofractamide A were found against A. aegypti, A. togoi, and C. pipiens pallens. $LD_{50}$ values of pipercide and retrofractamide A were 0.10 and 0.039 ppm against A. aegypti, 0.26 and 0.01 ppm against A. togoi, and 0.004 and 0.028 ppm against C. pipiens pallens, respectively. Based upon these results and earlier findings, bioactive components derived from Piperaceae fruits may be valuable for development of useful lead product of possibly safer fungicidal, insecticidal, and mosquito larvicidal agents.
