Bifidobacterium sp. Int-57 ${\alpha}-galactosidase$의 효소 특성 (Characterization of ${\alpha}-galactosidase$ from Bifidobacterium sp. Int-57)

  • ${\alpha}-galactosidase$는 raffinose와 stachyose 등의 ${\alpha}-galactoside$ 결합을 분해하는 효소이다. Raffinose와 stachyose는 특히 두류에 다량 존재하는데 이들 당은 섭취후 복부팽만감의 원인 물질로 발표되었는가 하면 한편으로는 비피더스균 증식인자로 최근에 보고된 바 있기 때문에 이들 당의 인체 건강에 대한 재평가가 요구되고 있다. 본 연구에서는 이에 대한 기초 자료로 사용하기 위하여 한국인으로부터 분리된 Bifidobacterium sp. Int-57이 생산하는 ${\alpha}-galactosidase$의 효소 특성을 조사하였다. Bifidobacterium sp. Int-57은 여러 종류의 다른 장내 세균에 비하여 높은 ${\alpha}-galactosidase$ 활성을-보유하고 있고 raffinose에 의하여 효소 생산이 증진되었다. 세포의 음파 파쇄후 DEAE-celluose와 Sepharose 6B gell filtration을 거쳐 부분정제한 효소를 사용하여 PNP-${\alpha}-galactosidase$에 대한 작용을 분석한 결과 최적 pH는 7.0, 최적온도는 $40^{\circ}C$ 였고 금속이온을 5mM 농도로 처리하였을 때 효소활성을 $CoCl_{2}$는 33%, $CuCl_{2}$는 21% 저해하였다. 또한 얻어진 효소는 실제 기질로써 raffinose와 stachyose를 잘 분해하는 것으로 나타났다.

제주지역 목장 토양 및 조사료 자원의 미량광물질 함량 평가 (The Evaluation of Trace Minerals Levels of Pasture Soils and Forages in Jeju)

  • 본 연구는 제주지역 목장 토양 및 조사료 자원의 미량광물질 함량을 평가하기 위해 수행되었다. 제주지역 98 목장에서 187 토양시료(농암갈색 76, 흑색 111) 및 말 농가에서 활용되고 있는 이탈리안라이그라스 건초, 혼합목초 건초, 수입 알팔파 건초 각 60시료를 채취해서 분석했다. 토양시료는 화학특성(pH, 유효인산, 치환성 양이온 함량)과 미량광물질(Cu, Fe, Cd, Zn, Mo, Se, Mn, Cr, Co, Ni) 총 함량 및 가용성 함량이 분석되었고, 조사료 시료는 미량광물질 총 함량이 분석되었다. 토양의 pH는 전체적으로 산성을 띄고 있었다(pH 5.1). 목장 토양 유효인산은 20.5 mg/kg, 치환성 Ca, Mg, K, Na은 각각 2.6, 0.9, 0.5, 0.2 cmol+/kg으로 낮게 나타나고 있다. 목장 토양 미량광물질 중 총 함량이나 가용성 함량 모두에서 검출되지 않은 원소는 없었다. 토양 미량광물질 총 함량에 비해 가용성 함량은 매우 낮은 수준을 나타냈다. 토양 미량광물질 중 Fe 총 함량이 가장 높고, Mn, Ni, Zn 총 함량도 다소 높게 나타났다. 부분적으로 토양 무기물 간 상관이 높았다. 조사료 자원에서 Cr은 검출되지 않았다. 기타 미량광물질 함량은 수입 알팔파 건초와 제주지역에서 생산된 건초 모두 일반적인 범위를 보여주었다. 그러나 제주지역에서 생산된 건초가 수입 알팔파 건초보다 Mn 함량(105 vs 23 mg/kg)은 높고 Mo 함량(2.7 vs 4.9 mg/kg)은 낮은 수준을 보였다. 본 연구결과 제주지역 목장 토양은 대부분 산성을 나타내고 양분 함량도 낮게 나타나고 있다. 제주지역 목장 토양 및 조사료에서 특정 미량광물질이 결핍되어 있는 것으로 보이지 않는다. 따라서 가축에게 미량광물질 첨가제의 보충급여는 사양형태나 사료 미량광물질의 함량 및 이용도 등을 고려하여 결정해야 할 것으로 사료된다.

PMF 모델을 이용한 대기 중 PM-10 오염원의 확인 (Source Identification of Ambient PM-10 Using the PMF Model)

  • The objective of this study was to extensively estimate the air quality trends of the study area by surveying con-centration trends in months or seasons, after analyzing the mass concentration of PM-10 samples and the inorganic lements, ion, and total carbon in PM-10. Also, the study introduced to apply the PMF (Positive Matrix Factoriza-tion) model that is useful when absence of the source profile. Thus the model was thought to be suitable in Korea that often has few information about pollution sources. After obtaining results from the PMF modeling, the existing sources at the study area were qualitatively identified The PM-10 particles collected on quartz fiber filters by a PM-10 high-vol air sampler for 3 years (Mar. 1999∼Dec.2001) in Kyung Hee University. The 25 chemical species (Al, Mn, Ti, V, Cr, Fe, Ni, Cu, Zn, As, Se, Cd, Ba, Ce, Pb, Si, N $a^{#}$, N $H_4$$^{+}$, $K^{+}$, $Mg^{2+}$, $Ca^{2+}$, C $l^{[-10]}$ , N $O_3$$^{[-10]}$ , S $O_4$$^{2-}$, TC) were analyzed by ICP-AES, IC, and EA after executing proper pre - treatments of each sample filter. The PMF model was intensively applied to estimate the quantitative contribution of air pollution sources based on the chemical information (128 samples and 25 chemical species). Through a case study of the PMF modeling for the PM-10 aerosols. the total of 11 factors were determined. The multiple linear regression analysis between the observed PM-10 mass concentration and the estimated G matrix had been performed following the FPEAK test. Finally the regression analysis provided source profiles (scaled F matrix). So, 11 sources were qualitatively identified, such as secondary aerosol related source, soil related source, waste incineration source, field burning source, fossil fuel combustion source, industry related source, motor vehicle source, oil/coal combustion source, non-ferrous metal source, and aged sea- salt source, respectively.ively.y.

의성지역(義城地域)의 금(金)-은(銀) 광화작용(鑛化作用) (Gold-Silver Mineralization of the Euiseong Area)

  • The Au-Ag deposits of the Euiseong area occurred in quartz veins which filled fissures in Cretaceous sedimentary and volcanic rocks. These ore veins can be classified in two types of deposits based on metallic mineral assemblages as follow: a pyrite type gold-silver deposit (Hoedong mine), characterized by Cu sulfides with Au-Ag alloy, and a Sb-rich silver deposit (Keumdongdo mine), characterized by base metal with Ag-bearing sulfosalts. Mineralogic and fluid inclusion evidences suggest that the ore minerals of these deposits was deposited from initial high temperatures (near $350^{\circ}C$) to later lower temperatures ($200^{\circ}C$) with moderate salinity fluids ranging from 5.8 to 3.8 eq. wt. % NaCl. The gold-silver mineralization of the Hoedong mine occurred at temperatures between 300 and $200^{\circ}C$ from fluids with log $f_{s_2}$ of -10 ~ -16 atm. The antimony - silver mineralization of the Keumdongdo mine were deposited at the higher temperatures (350 to $250^{\circ}C$) and $f_{S_2}$ (-10 ~ -13 atm) than gold mineralization of the Hoedong mine. The calculated log f02 of fluids at $250^{\circ}C$ in two deposits are -32 to -34 atm and -36.5 to -38.5 atm, respectively. Boiling evidences indicate that the ore mineralization of the Hoedong mine occurred at more shallow depth (0.5km) than that (1km) of the Keumdongdo mine. The above differences of depositional environments between two deposits caused the compositional changes of ore minerals such as electrum and sphalerite.

Electrical, Optical, and Electrochemical Corrosion Resistance Properties of Aluminum-Doped Zinc Oxide Films Depending on the Hydrogen Content

  • Aluminum-doped zinc oxide (AZO) is a commonly used material for the front contact layer of chalcopyrite $CuInGaSe_2$ (CIGS) based thin film solar cells since it satisfies the requisite optical and electrical properties with low cost and abundant elemental availability. Low-resistivity and high-transmission front contacts have been developed for high-performance CIGS solar cells, and nearly meet the required performance. However, the durability of the cell especially for the corrosion resistance of AZO films has not been studied intensively. In this work, AZO films were prepared on Corning glass 7059 substrates by radio frequency magnetron sputtering depending on the hydrogen content. The electrical and optical properties and electrochemical corrosion resistance of the AZO films were evaluated as a function of the hydrogen content. With increasing hydrogen content to 6 wt%, the crystallinity, crystal size, and surface roughness of the films increased, and the resistivity decreased with increased carrier concentration, Hall mobility, oxygen vacancies, and $Zn(OH)_2$ binding on the AZO surface. At a hydrogen content of 6 wt%, the corrosion resistance was also relatively high with less columnar morphology, shallow pore channels, and lower grain boundary angles.

Thermal Degradation of BZO Layer on the CIGS Solar Cells

  • We investigated a study on the thermal degradation of boron doped zinc-oxide (BZO) layer which used as a transparent conducting layer on the Cu (In1-xGax) Se2 (CIGS) based thin film solar cells. Devices were annealed under the temperature of $100^{\circ}C$ or 100 hours and then Hall measurement was carried out to characterize the parameters of mobility (${\mu}Hall$), resistivity (${\rho}$), conductivity (${\sigma}$) and sheet resistance (Rsh). The initial values of ${\mu}Hall$, ${\rho}$, ${\sigma}$ and Rsh were $29.3cm^2$/$V{\cdot}s$, $2.1{\times}10^{-3}{\Omega}{\cdot}cm$, $476.4{\Omega}^{-1}{\cdot}cm^{-1}$ and $19.1{\Omega}$/${\Box}$ respectively. After the annealing process, the values were $4.5cm^2$/$V{\cdot}s$, $12.8{\times}10^{-3}{\Omega}{\cdot}cm$, $77.9{\Omega}^{-1}{\cdot}cm^{-1}$ and $116.6{\Omega}$/${\Box}$ respectively. We observed that ${\mu}Hall$ and ${\sigma}$ were decreased, and ${\rho}$ and Rsh were increased. In this study, BZO layer plays an important role of conducting path for electrons generated by incident light onthe CIGS absorption layer. Therefore, the degradation of BZO layer characterized by the parameters of ${\mu}Hall$, ${\rho}$, ${\sigma}$ and Rsh, affect to the cell efficiency.

유기 태양전지의 개발 현황과 기술 과제 (Technical Tasks and Development Current Status of Organic Solar Cells)

  • Serious environmental problems have been caused by the greenhouse effect due to carbon dioxide($CO_2$) or nitrogen oxides($NO_x$) generated by the use of fossil fuels, including oil and liquefied natural gas. Many countries, including our own, the United States, those of the European Union and other developed countries around the world; have shown growing interest in clean energy, and have been concentrating on the development of new energy-saving materials and devices. Typical non-fossil-fuel sources include solar cells, wind power, tidal power, nuclear power, and fuel cells. In particular, organic solar cells(OSCs) have relatively low power-conversion efficiency(PCE) in comparison with inorganic(silicon) based solar cells, compound semiconductor solar cells and the CIGS [$Cu(In_{1-x}Ga_x)Se_2$] thin film solar cells. Recently, organic cell efficiencies greater than 10 % have been obtained by means of the development of new organic semiconducting materials, which feature improvements in crystalline properties, as well as in the quantum-dot nano-structure of the active layers. In this paper, a brief overview of solar cells in general is presented. In particular, the current development status of the next-generation OSCs including their operation principle, device-manufacturing processes, and improvements in the PCE are described.

실내종 Chironomus riparious와 야외종 Chironomus plumosus의 중금속 노출에 따른 분자지표 유전자 발현 (The Molecular Biomarker Genes Expressions of Rearing Species Chironomus riparious and Field Species Chironomus plumosus Exposure to Heavy Metals)

  • Chironomous is aquatic insect belonging to order Diptera, family Chironomidae. Their larval stage can be found mainly in aquatic benthic environment, hence good model organism to study environmental toxicology assessments and consider as useful bio indicators of contamination of the aquatic environment. In this study, Chironomus Heat Shock Proteins, Cytochrome 450, Glutathione S-transferase, Serine-type endopeptidase gene expressions were compared between polluted field areas (Chironomus plumosus) and under laboratory conditions (Chironomus riparious) to investigate molecular indicators for environmental contaminant stress assessment. Heavy metal (Al, Fe, Mn, Cu, Cr, Zn, Se, Pb, As, Cd) concentrations in sediments collected from three study areas exceeded the reference values. Moreover, HSPs, CYP450 and GST gene expression except SP for C. plumosus showed higher expression than C. riparious gene expression. Similar gene expression pattern was observed in C. riparious that exposed environment waters up to 96 h when compared to C. plumosus exposed to waters that grown in lab conditions. In summary, this comparative gene expression analysis in Chironomous between field and laboratory condition gave useful information to select candidate molecular indicators in heavy metal contaminations in the environment.

다이캐스팅용 고열전도도 Al-Zn-Mg-Fe 합금의 특성에 미치는 Zn 및 Mg 첨가량의 영향 (Effects of Zn and Mg Amounts on the Properties of High Thermal Conductivity Al-Zn-Mg-Fe Alloys for Die Casting)

  • The effects of Zn and Mg amounts on the solidification characteristics, microstructure, thermal conductivity and tensile strength of Al-Zn-Mg-Fe alloys were investigated for the development of high thermal conductivity aluminium alloys for die casting. Zn and Mg amounts in Al-Zn-Mg-Fe alloys had a little effect on the liquidus / solidus temperature, the latent heat for solidification and the fluidity of Al-Zn-Mg-Fe alloys. Thermo-physical modelling of Al-Zn-Mg-Fe alloys by JMatPro program showed $MgZn_2$, AlCuMgZn and Al3Fe phases on microstructure of their alloys. Increase of Zn and Mg amounts in Al-Zn-Mg-Fe alloys resulted in gradual reduction of the thermal conductivity of their alloys. Increase of Mg amounts in Al-2%Zn-Mg-Fe alloys had little effect on the tensile strength of their alloys, but increase of Mg amounts in Al-4%Zn-Mg-Fe alloys resulted in steep increase of the tensile strength of their alloys.

중금속의 토양 오염: 한국중부지역의 농경지를 중심으로 (The Concentration of Heavy Metals in Some Agricultural Soils of Central Korea)

  • To assess soil distribution patterns of heavy metals in agricultural environments, we have measured the concentrations of six metals from surficial soils of three different groups divided on the basis of the surrounding environment: (1) soil group I-cultivated soils within the purely agricultural regions, (2) soil group II-both cultivated and uncultivated soils near various livelihood facilities, and (3) soil group III-mainly cultivated soils near major polluting sources. The mean concentrations for the three soil groups were found in the range as follows: 0.12~0.15 (Cd), 4.94~6.08 (Pb), 0.05~0.11 (Hg), 2.82~3.50 (Cu), 4.69~7.82 (Zn), and 0.36~0.78 (As) ppm. Examination of data distribution trends indicates that the concentrations determined from the relatively unpolluted soil environs (groups I and II) were comparable not only between each other but also with those reported previously in background soil environs of Korea. The concentration data for the soil group III were however found to be much higher than the rest two groups. Unlike the direct comparison of the magnitudes of concentrations, results of a regression analysis exhibited much different patterns: it was seen that the correlation patterns for soil group I were rather analogous to those of soil group III. The similarities in correlation patterns between groups I and III along with the lack of correlations in soil group II suggest that soil characteristics such as whether being cultivated or not are important factors affecting soil distributions of heavy metals.
