Geochemistry and Genetic Environments of the Daejang Vein Deposits (대장광상(大藏鑛床)의 지화학(地化學) 및 생성환경(生成環境) 연구(硏究))

  • The Daejang mine is one of the representatives of Cu-Pb-Zn-(Ag) vein deposit related genetically to late Cretaceous granitoid in Korea. Sericite from an alteration halo of the mine yielded a K-Ar date of $95{\pm}3.5Ma$. Based on macrostructures of vein filling, three major mineralization stages (I, II and III) are distinguished by tectonic breaks. Major ore constituents are arsenopyrite, pyrite, pyrrhotite, sphalerite, chalcopyrite, galena, boulangerite, with small amounts of Ag-bearing tetrahedrite, pyrargyrite, native bismuth, marcasite, siderite, ankerite, gudmundite and calcite. Characteristic feature of each mineralization stage and compositional variation of sphalerite and arsenopyrite are discussed in relation to the genetic environments. The FeS contents of sphalerites are 20.5~14.9 mole % in stage I, 17.9~11.9 mole % in stage IIA, 17.0~9.2 mole % in stage IIB, and 6.9~4.7 mole % in stage III. Their results are indicative of decreasing FeS contents during mineralization process in sphalerite coexisting with sulfur-rich sulfide assemblages, such as monoclinic pyrrhotite and pyrite, and is agreement with the conclusions shown by Scott and Kissin(1973). The composition of arsenopyrite decrease also in As content from stage I to stage III, and the compositional variation correlate with position of the associated minerals in the paragenesis. Temperature and pressure of the mineralization are determined as $250{\sim}430^{\circ}C$ and 4.0~0.3kb respectively, based on the chemistry of the minerals.

  • This study is concerned with quality and analysis of nutritive value of commercially available honey and pollen load. Of thirteen kind of commercially available honeys, acid levels were less than KP IV standard. Color reaction for ammonia, resorcine and varium chloride was negative and trace of starch and dextrine was not detected. There were no extraneous materials in honey samples. Specific gravity of sample was slightly higher than KP IV standard. Total ash lied between 0.01 and 0.15% of honey weight which was less than KP IV standard, except 0.56% of Castanea Honey. Studies on mineral compositions (AAS) for honeys and pollen loads showed that $Na^+,\;K^+,\;Ca^{2+},\;Fe^{2+}\;Cu^{2+}\;and\;CO^{2+}$ were the most commonly occuring minerals. Pollen loads showed higher levels of mineral contents than honeys. Castanea Honey revealed rich in minerals. $Cd^{2+}\;and\;Pb^{2+}$ were found relatively higher level in Robinia Honey. Reducing sugar level showed $60{\sim}70$ in honeys and $25{\sim}30$ in pollen loads. Non reducing sugar varied between 2 to 7% in pollen loads.

Statistical Optimization of Medium Components for the Production of Biosurfactant by Bacillus licheniformis K51

  • The nutritional medium requirement for biosurfactant production by Bacillus licheniformis K51 was optimized. The important medium components, identified by the initial screening method of Plackett-Burman, were $H_3PO_4,\;CaCl_2,H_3BO_3$, and Na-EDTA. Box-Behnken response surface methodology was applied to further optimize biosurfactant production. The optimal concentrations for higher production of biosurfactants were (g/l): glucose, $1.1;NaNO_3,\;4.4;MgSO_4{\cdot}7H_2O,\;0.8;KCl,\;0.4;CaCl_2,\;0.27;H_3PO_4,\;1.0ml/l;\;and\;trace elements\;(mg/l):H_3BO_3,\;0.25;CuSO_4,\;0.6;MnSO_4,\;2.2;Na_{2}MoO_4,\;0.5;ZnSO_4,\;6.0;FeSO_4,\;8.0;CoCL_2,\;1.0;$ and Na-EDTA, 30.0. Using this statistical optimization method, the relative biosurfactant yield as critical micelle dilution (CMD) was increased from $10{\times}\;to\;105{\times}$, which is ten times higher than the non-optimized rich medium.

A study on the characteristics of low Pb Sn-5%Pb-1.5%Ag-x%Bi solder alloys (저Pb Sn-5%Pb-1.5%Ag-x%Bi계 솔더 합금의 특성에 관한 연구)

  • Recently as environmental pollution caused by Pb has posed a serious threat to the global environment, the trend to regulate the usage of Pb in electronic industry is one the rise. Currently, the solder alloy with high Pb content, Sn-37%Pb, is utilized in the electronic assembly therefore, the objective of this study is to develop an alternative solder alloy for the existing Sn-37%Pb solder alloy. First thing, this work choosed Sn-5%Pb-1.5%Ag, Sn-5%Pb-1.5%Ag-x%Bi(x=1~5%) for candidate solder alloys, and examined their properties such as melting range, wettability, microhardness, tensile property, oxidation behavior and microstructure. Wettability was on the same level of Sn-37%Pb. Dissolution of Pb ion in Sn-5%Pb solder was 0.46ppm. This solder alloy revealed cellular dendrite microstructure $\beta$-Sn matrix, Pb-rich phase, Ag/Sn, and Cu/Sn Intermetallic compounds. The range of solidification temperature was within 3$0^{\circ}C$. Also these alloy displayed higher tensile strength and lower elongation than Sn-37%Pb. The resistance of oxidation in Sn-5%Pb-1.5%Ag solder alloy was superior to that of Sn-37%Pb solder alloy. But that of Sn-5%Pb-1.5%Ag-5%Bi solder alloy was equal to that of Sn-37%Pb solder alloy.

  • Ore criteria at the Sheba Deposit indicate orogenic mineralization type. Rocks and mineral assemblages suggest low formation-temperature of green-schist facies. Pyrite found in two generations; Type1 is irregular grains, contains higher arsenic and gold contents, compared to the relatively younger phase Type2 pyrite, which is composed of euhedral grains, found adjacent to late quartz-carbonate veins or at rims of type1 pyrite. Two gold generations were identified; type1 found included in sulphides (mainly pyrite). The second gold type was remobilized (secondary) into free-lodes within silicates (mainly quartz). Gold fineness is high, as gold contains up to 95 wt. % Au, Ag up to 3.5 wt. %, and traces of Cu, Ni, and Fe. Pyrite type2 contains tiny mineral domains (rich in Al, Mn, Hg, Se, Ti, V, and Cr). Zoning, and replacement textures are common, suggesting multiple mineralization stages. The distribution and relationships of trace elements in pyrite type2 indicate three formation patterns: (1) Al, Mn, Hg, Se, Ti, V, Cr, and Sn are homogeneously distributed in pyrite, reflecting a synchronous formation. (2) As, Ni, Co, Zn, and Sb display heterogeneous distribution pattern in pyrite, which may indicate post-formation existence due to other activities. (3) Au and Ag show both distribution patterns within pyrite, suggesting that gold is found both in microscopic phases and as chemically bounded phase.

Sedimentary Geochemical Characteristics and Environmental Impact of Sediments in Tamjin River and Doam Bay (탐진강 및 도암만 지역 퇴적물의 퇴적지구화학적 특성과 환경영향)

  • To examine the sedimentary geochemical characteristics of sediment in the Tamjin River and Doam bay, the analysis was conducted, using the sample obtained in February 2000, on the grain size and the contents of metallic elements and organic carbon. The factors that influence the geochemical behavior of metallic elements in the surface sediment are grain size, organism, surrounding soil and $CaCO_3$. To find out the pollution level of metallic elements, the enrichment factor (EF) and the index of geoaccumulation ($I_{geo}$) were researched. The majority of metallic elements sustain their values in natural state. The elements such as K, Ba, Zr, etc. appear to be rich in some places. The EF and $I_{geo}$ of P, Cu, Zn, and Pb, which belong to toxic heavy metals, are partly related with man-made pollution. P and Cu have a high EF, Pb has a high $I_{geo}$ and Zn is high in both EF and $I_{geo}$. The low contents of P and Cu are not likely to be related with the pollution of water environment. However, given the development of relative pollution, the research and the management regarding the pollutants are needed. Because Pb, naturally enriched by geological characteristics, has a large influence on water environment along with Zn, the adequate measures against man-made pollution should be worked out.

Characteristics of Geochemical Behaviors of Trace Metals in Drainage from Abandoned Sechang Mine (세창 폐금속광산 수계에서 미량원소의 지구화학적 거동특성 규명)

  • The geochemical evolution of mine drainage and leachate from waste rock dumps and stream water in Pb-As-rich abandoned Sechang mine area was investigated to elucidate mechanisms of trace metals. Total and sequential extractions were applied to estimate the distribution of trace metals in constituent phases of the waste rocks and to assess the mobility of trace metals according to physicochemical conditions. These discharged waters varied largely in chemical composition both spatially and temporally, and included cases with significant]y low pH (in the range 2.1-3.3), and extremely sulphate (up to 661 mg/l and metal contents (e.g. up to 169 mg/l for Zn, 27 mg/l for As, 3.97 mg/l for Pb, 2.99 mg/l for Cu, and 1.88 mg/l for Cd). Arsenic and heavy metal concentrations at the down-stream of Sechang mine have been decreased nearly to the background level in downstream sites (sites 8 and 16) without any artificial treatments. The oxidation of Fe-sulfides and the subsequent hydrolysis, of Fe(II), with precipitation of poorly crystallized minerals, constituted an efficient mechanism of natural attenuation which reduces considerably the transference of trace metals (i.e. Fe and As) to rivers. The dilution of drainage by mixing with pristine waters provoked an additional decrease of trace metal concentrations and a progressive pH increase. On the other hand, the most soluble cations (i.e. Zn) remained significantly as dissolved solutes until the pH was raised to approximately neutral values. With respect to ecotoxicity, it is likely that the Zn pollution is of particular concern in Sechang mine area. This was confirmed by the sequential extraction experiment, where Zn in wet waste-rock samples occurred predominantly in the exchangeable fraction (65-89% of total), while Pb was the highest in the reducible and carbonate fractions, and Cd, Cu and As in the residual fraction. Pb concentration in the readily available exchangeable fraction (34-48% of total) was dominated for dried waste rock samples. Considering the proportion of metals bound to the exchangeable and carbonate fractions, the comparative mobility of metals probably decreased in the order of Zn>Pb>Cd>As=Cu.

Effects of Dietary Levels of Corn and Tuna Oils on the Formation of Preneoplastic Lesions in Rat Hepatocellular Carcinogenesis (쥐간세포암화과정에서 옥수수기름과 참치기름의 수준에 따른 전암성 병변의 변화)

  • This study is conducted to determine the effects of dietary levels of corn and tuna oils on the formation of preneoplastic lesions in die-thylnitrosamine (DEN) induced rat hepatocarcinogenesis. Weanling male Sprague-Dawley rats were fed 2.5, 5, 15, 25% (w/w) corn or tuna oils. Hepatocellular carcinogenesis was induced by DEN (200 mg/kg body weight) and two-thirds partial hepactectomy was carried out 3 weeks later and were sacrificed 8 weeks after DEN initiation. Tuna oil group showed smaller area of placental glutathione S-transferase (GST-P) positive foci than com oil group. Com oil group of 25% (w/w) showed the widest area of GST -P positive foci, and tuna oil group showed significantly smaller area of GST-P positive foci than com oil in 25% (w/w) level but had no differences between oil levels. Thio-barbituric acid reactive substances (TBARS) content was the highest in 25% (w/w) level of tuna oil group fed long chain and highly polyunsaturated fatty acids. Also serum ${\gamma}$ -glutamyltranspeptidase (GGT) activities in 25% level of tuna oil group were significantly higher than by other levels. As oil contents increased, glucose 6-phosphatase (G6Pase) seems to decrease in com oil groups but remained the same in tuna oil groups. Glutathione reductase (GR) activities were significantly higher in tuna oil group, and the higher the level of tuna oil, the higher GR activities. But Cu/Zn superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities didn't seem to be influenced by levels and kind of dietary fats. Therefore, as oil levels increased, com oil rich in n-6 fatty acids promoted carcinogenesis but tuna oil rich in eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) of n-3 fatty acids suppressed. Although lipid peroxidation products were elevated in 25% (w/w) tuna oil group, GST-P positive foci didn't increase. Therefore pre-neoplastic lesions might be reduced through mediation of a lipid peroxidation process in tuna oil. As fat contents of tuna oil increased, elevated GR activities may give a rise to produce more reduced glutathione in order to protect against free radical attack, and high G6Pase activities remained the same and they contributed to membrane stability. So tuna oil diet seems to protect hepatocarcinogenesis.

Fluxless Bonding Method between Sn and In Bumps Using Ag Capping Layer (Ag층을 이용한 Sn과 In의 무 플럭스 접합)

  • We utilized Ag capping layer for fluxless bonding. To investigate the effect of Ag capping layer, two sets of sample were used. One set was bare In and Sn solders. The other set was In and Sn solders with Ag capping layer. In ($10{\mu}m$) and Sn ($10{\mu}m$) solders were deposited on Cu/Ti/Si substrate using thermal-evaporation, and Ag ($0.1{\mu}m$) capping layers were deposited on In and Sn solders. Solder joints were made by joining two In and Sn deposited specimens at $130^{\circ}C$ for 30 s under 0.8, 1.6, 3.2 MPa using thermal compression bonder. The contact resistance was measured using four-point probe method. The shear strength of the solder joints was measured by the shear test of cross-bar sample in the direction. The microstructure of the solder joints was characterized with SEM and EDS. In and Sn solders without Ag capping layers were only bonded at $130^{\circ}C$ under high bonding pressure. Also the shear strength of the In-Sn solder joints under was lower than that of the Ag/In-Ag/Sn solder joints. The resistance of the solder joints was $2-4\;m{\Omega}$ The solder joints consisted of In-rich phase and Sn-rich phase and the intermixed compounds were found at the interface. As bonding pressure increased, the intermixed compounds formed more.

Molecular Characterization of Metallothionein Gene of the Korean Bitterling Acheilognathus signifer (Cyprinidae) (묵납자루 (Acheilognathus signifer; Cyprinidae) metallothionein 유전자의 클로닝 및 특징 분석)

  • Genetic determinant for metallothionein (MT), a cysteine-rich protein playing essential roles in metal detoxification and homeostasis, was characterized in the Korean bitterling (Acheilognathus signifer, Cyprinidae), an endemic fish species. The full-length A. signifer MT (AsMT) cDNA (551 bp) is composed of a single open-reading frame (ORF) to encode a polypeptide of 60 amino acids containing 20 cysteine residues whose positions are conserved in most cypriniform MTs. At the genomic level, the AsMT (2,593 bp spanning the 5'-flanking region to the 3'-untranslated region) represented a conserved tripartite (three exons interrupted by two introns) structure with AT-rich introns. The upstream regulatory region (-1,914 bp from the ATG initiation codon) of AsMT displayed various sites and motifs for transcription factors involved in the metal-mediated regulation and stress/immune responses. The AsMT transcript was ubiquitously detected in various organs with variable expression levels, where the ovary and intestine showed the highest expression, while the heart and skeletal muscle represented the lowest level. During an exposure to copper (immersion in $0.5\;{\mu}M$ Cu for 48 h), the levels of AsMT transcripts were significantly elevated in the liver (more than 3.5-fold), moderately in the gill, kidney, and spleen (ranging from 1.5- to 2.5-fold), and barely in the brain and intestine. Results of this study could form a useful basis to explore the metal-related stress physiology of this endangered fish species.
