The Purification Characteristics of Reactive Soil-Bentonite Landfill Liner (혼합반응 차수재의 오염정화특성)

  • The purpose of this paper is to investigate purification characteristic of soil-bentonite landfill liner and to develop a desirable liner system. In order to clarify the purification characteristics, high pressure column tests using soil-bentonite, reactive soil-bentonite and reactive bentomat were carried out in the presence of water and leachate. The test results indicated that the significant amount of NH$_3$-N, Pb and Cu was removed through the reactive soil-bentonite liner system.

  • The objective of this study is to examine a stabilized efficiency of heavy metals including Cd, Cu, Pb and Zn using slaked lime. Tailings from the Janggun Pb-Zn mine, the second Yeonhwa Pb-Zn mine, the Jisi Au-Ag mine and the Sangdong W mine were sampled and measured heavy metal concentrations contents using AAS as various extraction methods. During 156 hours, column test were undertaken to evaluate the possibility of stabilization by slaked lime. The result shows that $Ca(OH)_2$ has a good efficiency in heavy metal stabilization, especially at the Jisi mine with stabilized efficiencies of 97%(Cd), 99%(Cu), 86%(Pb) and 99%(Zn), respectively.

Micro-Determination of D-Amino Acids in Milk by using Column Switching System (Column-Switching System을 이용한 우유속의 D-아미노산의 미량정량)

  • Free amino acids were isolated from milk and their absolute amounts were determined by reversed phase high performance liquid chromatography after derivatization with dansyl chloride. The determination of D- and L-amino acids was based on achiral separation on a C18 column. It was found that milk contained totally 41.00 mg DL-amino acids in 100 mL milk. The level of D-amino acids to L-amino acids was determined by a column-switching system combining an achiral reversed phase separation and chiral chelate additive. The chiral separation was carried out with addition of the chiral Cu(N-benzyl-L-proline)2 chelate to the mobile phase in reversed phase liquid chromatography. It was found that the determination of 16 different amino acids is feasible in the milk sample with a C18 column separation and 12 D-amino acids out of the 16 amino acids can be determined via the column-switching system with chiral separation. 2.05% of D-glutamic acid and 2.93% of D-alanine were found in milk.

Competitive Adsorption Characteristics of Cupper and Cadmium Using Biochar Derived from Phragmites communis (갈대 biochar의 구리 및 카드뮴 경쟁흡착특성)

  • BACKGROUND: Heavy metal adsorptionnot only depends on biochar characteristics but also on the nature of the metals involved and on their competitive behavior for biochar adsorption sites. The goal of this study was to investigate the competitive absorption characteristics of Cu and Cd in mono-metal and binary-metal forms by biochar derived from Phragmites communis. METHODS AND RESULTS: Batch and column experiments were conducted to evaluate the competitive adsorption characteristics of the biocharfor Cu and Cd. In the batch experiments, the maximum adsorption capacity of Cd(63 mg/g) by biochar was higher than that for Cu (55 mg/g) in the mono-metal adsorption isotherm. On the other hand, the maximum Cu adsorption capacity (40 mg/g) by biochar was higher than that for Cd(25 mg/g) in the binary-metal adsorption isotherm. Cu was the most retained cations. Cd could be easily exchanged and substituted by Cu. The amounts of adsorbed metals in the column experiments were in the order of Cd (121 mg/g) > Cu (96 mg/g) in mono-metal conditions, and Cu (72 mg/g) > Cd (29 mg/g) in binary-metal conditions. CONCLUSION: Overall, the results demonstrated that competitive adsorption among metals increased the mobility of these metals. Particularly, Cd in binary-metal conditions lost its adsorption capacity most significantly.

Optical Assembly and Fabrication of a Micro-electron Column (마이크로 전자렌즈의 광학적 정렬과 조립)

  • A silicon lens and an insulator of pyrex, components of a micro-electron column, should be assembled by aligning and stacking simultaneously. An optical alignment of a diffraction beam and a laser welding were employed for the assembly of a source lens and an Einzel lens with precision within $\pm$4% for the maximum aperture size. The experimental condition for optical alignment and laser welding are explained. Anodic bonding was used to assist in stacking lenses. A micro-electron column of smaller apertures assembled with precise alignment and fabrication was tested with a current image of a Cu grid of 9$\mu$m in linewidth, and showed a higher resolution in acceleration mode.

A study on the fixation of heavy metals with modified soils in the landfill liner (개량혼합토를 이용한 폐기물 매립지 차수층의 중금속 고정능력에 관한 연구)

  • The authors selected the modified soil method, and then performed the geotechnical and environmental laboratory test, and evaluated whether the modified soil liner could be accepted as a barrier layer in landfill. Unlike the results of the natural soil(CL), those of the hydraulic conductivity test of stabilized soil met the standard value. According to these results, the optimal mixing ratio of a mixture(cement : bentonite : stabilizing agent) was 90 : 60 : 1 with mass ratio(kg) for 1㎥ with soil, and it was possible to use poor quality bentonite. B\circled2 because of a little difference from results with high quality bentonite. B\circled1. The Cation Exchange Capacity(CEC) of the modified soil was increased about 1.5 times compared with the natural soil; however. the change of CEC with a sort of additives was not detected. In order to observe the change of the chemical components and crystal structures, the natural and the modified soils with the sorts of additives were measured by the XRF(X-Ray Flourescence Spectrometer) and SEM, but there was no significant change. The artificial leachate with the heavy meals ($Pb^{2+}$ , $Cu^{2+}$, $Cd^{2+}$ Zn$^{2+}$ 100mg/L) was passed through the natural soil and modified soils in columns. In the natural soil, Cd$^{2+}$ and $Zn^{2+}$ were identified, simultaneously the pH of outflow was lower, and then came to the breakthrough point. The removal efficiency of the natural soil was showed in order of following : $Pb^{2+}$$Cu^{2+}$ > $Zn^{2+}$ > $Cd^{2+}$ On the other hand, modified soils were not showed the breakthrough condition like the result of the natural soil. The modified soil with the lower quality bentonite, B\circled2(column3) was more stable with respect to chemical attack than that with the higher bentonite, B\circled1(column2) because the change range of outflow pH in columns was less than that of outflow pH in column2. In addition, the case of adding the stabilizing agent(column4) was markedly showed the phenomena.ena.

Evaluation of Removal Efficiencies of Heavy Metals Using Brown Seaweed Biosorbent Under Different Biosorption Systems (폐미역을 이용한 생물흡착 시스템별 중금속 제거 효율 평가)

  • BACKGROUND: Heavy-metal pollution represents an important environmental problem due to the toxic effects of metals, and their accumulation throughout the food chain leads to serious ecological and health problems. METHODS AND RESULTS: Optimum conditions in continuous-flow stirred tank reactor (CSTR) and packedbed column contactor (PBCC) using brown seaweed biosorbent were investigated. Under optimum conditions from both lab-scale biosorbent systems, removal efficiency of copper (Cu) in a large-scale PBCC system was investigated. Removal capacity of Cu using brown seaweed biosorbent in a lab-scale CSTR system was higher than that in a lab-scale PBCC system. On the other hand, over 48 L/day of flow rate in Cu solution, removal efficiency of Cu in a lab-scale PBCC system was higher than that in a lab-scale CSTR system. Optimum flow rate of Cu was 24 L/day, optimum Cu solution concentration was 100 mg/L. Removal capacity of Cu at different stages was higher in the order of double column biosorption system > single column biosorption system. Under different heavy metals, removal capacities of heavy metal were higher in the order of Pb > Cr > Ni > Mn ${\geq}$ Cu ${\geq}$ Cd ${\fallingdotseq}$ Zn ${\geq}$ Co. Removal capacity of Cu was 138 L in a large-scale PBCC system. Removal capacity of Cu a large-scale PBCC system was similar with in a lab-scale PBCC system. CONCLUSION(s): Therefore, PBCC system using brown seaweed biosorbent was suitable for treating heavy metal wastewater.

Purification and Characterization of Cholesterol Oxidase Produced by Streptomyces sp. No.4 (방선균 Streptomyces sp. No.4가 생산하는 Cholesterol Oxidase의 정제 및 특성)

  • The cholesterol oxidase(EC.1.1.3.6) produced from Streptomyces sp. No.4 which isolated from soil was purified and investigated for the enzymatic properties. The enzyme was purified specifically by cholesterol affinity column chromatography with a yield of 28.3%. The purified enzyme showed a single polypeptide on SDS-PAGE and the molecular weight was estimated to be 60,000 daltons. The enzyme activity was strongly inhibited by metal ions such as $HgCl_2$ and $CuSO_4$. Dithiothreitol and mercaptoethanol inhibited the enzyme activity at concentration of 1mM. The Michaelis constant(Km) for cholesterol was found to be 1.38mM by Lineweaver-Burk plot analysis. Amino acid analysis showed that the enzyme protein was composed of 416 amino acid residues including 52moles of glycine and 19moles of tryptophane.

  • Ion exchange is the most reliable process to remove the ionic impurities and the economic operation. ion exchange is widely used in water and wastewater treatment, especially softening and demineralization. ion selectivity depends on the hydrated radius, charge of ions and concentration. The objective of this study was to determine the selectivity order of cations with equilibrium and column ion exchanges and to investigate the effect of the background anion on selectivity. Cation selectivity increases with decreasing concentration and increasing charge ( $H^+$ < $K^+$ << $Cu^{2+}$ < $Co^{2+}$ < TEX>$Ca^{2+}$ << $Ce^{3+}$)in equilibrium and column cation adsorptions.

  • Fe(III)-Coated Star Fish (ICSF) was prepared by reaction of calcined Star Fish (SF) with Fe(III) solution at an elevated temperature. To investigate the stability of ICSF at acidic condition, dissolution of Fe was studied at pH 2 as a function of time. Extracted iron was negligible over the entire reaction time. This stability test suggests the applicability of ICSF in the treatment of wastewater even at low pH. Adsorption capacity of Cu(II) onto SF and ICSF was investigated in a batch and a column test. In the pH-edge adsorption, adsorption of copper onto SF and ICSF was quite similar over the entire pH range due to the presence of an important amount of Fe in SF itself. From the adsorption isotherm obtained with variation of the concentration of Cu(II), ICSF showed 1.6 times greater adsorption capacity than SF. Also, ICSF showed a greater removal capacity of Cu(II) in the column test.
