흰쥐 간조직의 활성산소 및 제거효소에 미치는 목초액의 영향 (Effect of Pyroligneous Liquor on Oxygen Radicals and Their Scavenger Enzymes in Liver of CD Rats)

  • 시판 목초액 A (원액 35%)을 사용하여 음용수에 0%, 1.0%, 25.0%, 50.0%, 75.0%가 되도록 조제한 다음, CD계 수컷 흰쥐 (150 ${\pm}$ 10 g)에 2개월 동안 조제사료와 함께 자유 음용케 하여 간조직 중의 활성산소로서 수퍼옥시드 라디칼(O$_{2}\;^-$), 히디록시 라디칼, 과산화수소 및 제거효소로서 수퍼옥시드 디스무타아제 (SOD), 글루타치온 퍼옥시다아제 (GPx) 및 카탈라아제 (CAT)에 미치는 영향을 평가하였다. 목초액 PL-25 및 PL-50 투여군의 간조직의 미토콘드리아 획분에서는 O$_{2}\;^-$의 생성량은 대조군 대비 12${\sim}$14%의 O$_{2}\;^-$의 생성량 억제효과가 나타났고, 간조직의 마이크로솜획분에서는 대조군 대비 각각 11.0${\sim}$15.0의 O$_{2}\;^-$의 생성량 억제효과가 나타났다. 간조직 미토콘드리아획분에서 ${\cdot}$OH 생성량이 목초액 PL-25 및 PL-50 투여군에서 대조군 대비 12${\sim}$20%의 ${\cdot}$OH 생성 억제효과가 나타났고, 간조직의 마이크로솜획분에서는 대조군 대비 17${\sim}$20%의 ${\cdot}$OH 생성 억제효과가 나타났다. 목초액 PL-25 및 PL-50 투여군의 간조직의 미토콘드리아획분에서는 대조군 대비 12${\sim}$15%의 유의적인 H$_{2}$O$_{2}$의 생성 억제효과가 나타났고, 간조직의 마이크로솜획분에서는 대조군 대비 20${\sim}$22%의 상당히 유의적인 H$_{2}$O$_{2}$의 생성 억제효과가 나타났다. 목초액 PL-25 및 PL-50 투여군의 간조직의 Mn-SOD 활성은 대조군 대비 15${\sim}$25%나 유의적인 활성 증가효과가 인정되었고, 간조직의 Cu/Zn-SOD 활성은 대조군 대비 11${\sim}$16%의 유의적인 SOD활성 증가효과가 인정되었다. 목초액 PL-25 및 PL-50 투여군의 간조직의 미토콘드리아획분의 GPx 활성은 대조군 대비 10${\sim}$17%의 GPx 활성 증가효과가 인정되었고, 간조직의 마이크로솜획분의 GPx 활성 증가효과가 인정되었다. 목초액 PL-25 및 PL-50 투여군의 간조직의 미트콘드리아획분의 CAT 활성은 대조군 대비 12${\sim}$14%의 유의적인 CAT 활성 증가효과가 나타났고, 간조직의 시토졸 1획분에서는 대조군 대비 15${\sim}$27%의 CAT 활성 증가효과가 인정되었다. 이상의 결과에서 목초액의 장기간 투여는 간조직 중의 활성산소의 억제효과뿐만 아니라 방어시스템으로서 활성산소 제거효소의 역할도 충실히 수행하여 노화를 효과적으로 예방하고 억제할 수 있을 것으로 기대된다.

생식제품 급여가 영양불균형식이를 섭취하는 흰쥐의 항산화체계 및 혈청 무기질 농도에 미치는 영향 (Effects of Uncooked Powdered Food on Antioxidative System and Serum Mineral Concentrations in Rats Fed Unbalanced Diet)

  • 본 연구에서는 비타민과 무기질을 sub-optimal 수준으로 섭취시키고 고콜레스테롤식이와 에탄올을 병행 섭취시키는 흰쥐를 대상으로 5주간 생식제품을 20% (생식20군) 및 40% (생식40군) 수준으로 보충 섭취시킨 후, 체내 항산화체계에 미치는 영향을 평가하여 다음과 같은 결과를 얻었다. 일일 식이섭취량, 체중증가량 및 식이효율은 일반대조군과 영양불균형대조군 간에 유의적인 차이가 관찰되지 않았고, 생식40군의 경우 영양불균형대조군에 비해 식이효율이 유의하게 향상되었다 (p < 0.05) . 간조직의 지질과 산화물농도는 일반대조군에 비해 영양불균형대조군에서 유의하게 더 낮았으며 (p < 0.05), 식이내 생식첨가에 의해 영향을 받지 않았다. 간조직의 SOD 활성은 일반대조군에 비해 영양불균형대조군에서 유의적으로 증가하였고, 생식 섭취군의 경우 영양불균형대조군에 비해 유의하게 감소하였다 (p < 0.05), 간조직의 catalase 활성은 영양불균형대조군에서 일반대조군에 비해 유의하게 더 낮았으며 (p <0.05), 생식섭취에 의해 영향을 받지 않았다. 간조직의 (GSH-Px + catalase)/SOD 비율은 영양불균형대조군에서 일반대조군보다 유의하게 더 낮았으며, 영양불균형대조식이에 생식을 20% 수준으로 첨가시킨 결과 유의하게 증가하였다 (P < 0.05). 영양불균형대조식이는 일반대조식이에 비해 혈중 Cu 농도를 유의하게 증가시켰다. 생식분말 섭취군의 경우 영양불균형대조군에 비해 Fe, Se, Mn, Zn 및 Cu 등의 무기질 섭취량이 더 많음에도 불구하고 혈중 농도는 오히려 더 낮게 나타났으며, 특히 Fe 및 Cu농도는 유의적으로 감소하였다. 이는 생식제품이 phytate 및 섬유질이 풍부한 전곡류 및 식물성식품들로 구성되어 있어 무기질의 생체이용률을 저하시키기 때문으로 사료되며, 생식제품의 장기 섭취시 이들 무기질 영양상태가 저하되지 않도록 주의가 필요할 것으로 생각된다.

  • Several micronutrients (vitamins and minerals) are required as co-factors in DNA synthesis, DNA repair, DNA methylation and apoptosis. Some notable examples include (a) folic acid and vitamin B12 required for maintenance methylation of DNA and the synthesis of dTTP from dUTP, thus prevent the misincorporation of uracil into DNA, a highly mutagenic and chromosome-breaking event, (b) niacin, is essential in the form of the coenzymes NAD and NADP which act as a substrate for polyADPribose polymerase (PARP), an enzyme thought to facilitate efficient DNA repair and telomere length regulation and (c) zinc, apart from its antioxidant role as a co-factor in Cu/Zn SOD, it is required in its stabilizing role of the DNA-binding domain of p53 (residues 102-292) and thus is essential for apoptotic response to DNA damage. 과산화지질의 투여가 흰쥐 간의 산화와 항산화계에 미치는 영향 (The Degree of Lipid Oxidation of Rat Liver Fed Peroxidized Lipid and Its Effects on Anti-Oxidative System)

  • Accumulation of peroxidized lipid, fed or injected in the body of rats was investigated and the effect of peroxidized lipid on the antioxidative system was studied also. Three groups each having six of Sprague-dawley rats were raised for 8 weeks. the peroxide value(POV) of diet fed to the control and the peroxidized group was 5.47 and 22.14meq/kg , respectively. Injected group was given the control diet and peroxidized linoleic acid(POV 31.81meq/kg) was injected into the peritoneal area three times a week. The POV, MDA, and protein carbonyl values of the peroxidized and the injected group (experimental groups) were significantly higher (p<0.05) than those of the control group. Cu, Zn-SOD and M-SOD activity of the experimentla groups increased 1.6 times that of control group at 4 th week. and decreased by 60% of their activityafter 8 weeks of feeding (p<0.05) . Catalase activity, glutathione and Vt, E contents of the experimental groups were significantly lower (p<0.05) than those of the control group during 8 weeks. The accumulation of peroxidcized lipid in liver were ovserved both in the fed or the injected group. The increased of enzyme activity of the experimental group during 4 weeks suggests ianadaptation of antioxidative system to get rid of the peroxidized lipid. Decrease of enzyme activity and glutathione observed as the peroxidized lipid lipid accumulation proceeded further, however, seems to indicate the oxdiative damage of enzyme and protein . Determination of the protein carbonyl content may be used as a method for measuring the oxidative damaging effect of peroxidized lipid.

뇌조직의 활성산소 및 그 제거효소에 미치는 뽕잎 추출물의 영향 (Effects of Mulberry (Morus alba L.) Leaf Extract on Oxygen Radicals and Their Scavenger Enzymes in Brain of SD Rats)

  • This study was designed to investigate the effects of mulberry (Morus alba L.) leaf extract (MLE) on oxygen radicals and their scavenger enzymes in brain membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (MLE-100 and MLE-300 groups) added 100 and 300 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) lecels resulted in significant decreases (13.4% and 21.1%, 12.0% and 13.4%, respectively) in brain mitochondria and microsome of MLE-100 and MLE-300 groups compared with control group. Superoxide radical ($O_2$) levels were significantly decreased about 12% in brain cytosol of MLE-300 group compared with control group. Lipid peroxide (LPO) levels were effectively inhibited (18.1% and 12.3%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Oxidized protein (OP) levels were significantly decreased (14.2%, and 10.9%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Mn-SOD activities in brain mitochondria were significantly increased (13.5% and 18.6%, respectively) in MLE-100 and MLE-300 groups, and Cu/Zn-SOD activities in brain cytosol were also effectively increased (about 17.7%) in MLE-300 groups compared with control group. GSHPx activities in brain cytosol were remarkably increased (17.2% and 23.9%, respectively) in MLE-100 and MLE-300 groups compared with control group. These results suggest that anti-aging effect of mulberry leaf extract (MLE) may play a pivotal role in attenuating a various age-related changes in brain.

  • This study was designed to investigate the effects of silk fibroin(Mw 500) powder (SFP) on oxygen radicals and the scavenger enzymes in brain membranes of rats. Spragu-Dawley(SD) male rats(160${\pm}$10g) were fed basic diet(control group), and experimental diets(SFP-2.5 and SFP-5.0 groups) added 2.5 and 5.0g/kg BW/day for 6 weeks. Hydroxyl radical($.$OH) levels resulted in a decreases(6.6% and 9.7%, 2.8% and 11.9%, respectively) in brain mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group, but were significantly decreased in these membrances of SFP-5.0 group only. Superoxide radical (O2) levels were a slightly decreased (2.0% and 9.1%, respectively) in brain cytosol of SFP-2.5 and SFP-5.0 groups compared with control group. Lipid peroxide(LPO) levels were significantly decreased (12.9% and 21.9%, 13.2% and 22.5%, respectively) in brain mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. Oxidized protein (OP) levels were significantly decreased (16.7% and 15.7%, respectively) in brain microsomes of SFP-2.5 and SFP-5.0 group compared with control group, but significantly difference between in brain mitochondria of these two groups could not be obtained. Mn-SOD activities were remarkably increased (11.2% and 24.2%, respectively) in mitochodria of SFP-2.5 and SFP-5.0 groups. CuZn-SOD activities were effectively increased (7.7% and 19.6%, respectively) in brain cytosol of SFP-2.5 and SFP-5.0 groups, but significant difference between control and SFP-2.5 groups could be not obtained. GSHPx activities were considerably increased (5.3% and 11.7%, respectively) in brain cytosol of SFP-2.0 and SFP-5.0 groups compared with control group. There results suggest that anti-aging effect of silk fibroin may play an effective learning and memory role in a attenuating a oxidative stress and increasing a scavenger enzyme activity in brain membranes.

Lipid peroxide (LPO) levels were significantly decreased (8.3% and 18.0%, 13.4% and 18.4%, respectively) in liver mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. Oxidized protein (OP) levels were remarkably decreased about 11.6% in liver mitochondria of SFP-5.0 group compared with control group. Mn-SOD activities were remarkably in creased (17.6% and 28.8%, respectively) in mitochondria of SFP-2.5 and SFP-5.0 groups, and Cu/Zn-SOD activities were also effectively in creased (about 14.4%) in liver cytosol of SFP-5.0 groups, but significant difference between GSHPx activity in liver cytosol of these two groups could be not obtained. These results suggest that anti-aging effect of silk fibroin may play an effective anti-aging role in a aattenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.
