We examined the species diversity of Herposiphonia on Korean coasts, based on a combination of morphology and molecular analyses of the mitochondrial COI-5P DNA barcode marker and plastid rbcL gene. We report the presence of eight species including three novel species: H. donghaensis sp. nov., H. jejuinsula sp. nov., H. sparsa sp. nov., H. caespitosa, H. fissidentoides, H. insidiosa, H. parca, and H. subdisticha. Specimens were separated into eight clades in both the COI-5P and rbcL gene analyses, with 1.3-19.6 and 6.6-15% interspecific sequence divergence, respectively. These eight species are also distinguishable by several morphological characteristics such as: branching pattern (d/i pattern in H. donghaensis sp. nov. and H. sparsa sp. nov.; d/d/d/i pattern in others), shape of determinate branch (ligulate in H. fissidentoides; terete in others), number of vegetative trichoblasts (1-2 in H. insidiosa and H. sparsa sp. nov.; 3-4 in H. caespitosa; absent in others), and number of segments and pericentral cells in determinate branches. About three novel species revealed by our analyses, H. donghaensis sp. nov. is newly discovered, and H. jejuinsula sp. nov. and H. sparsa sp. nov. were previously reported in Korea as H. nuda and H. secunda, respectively. Our results show that DNA barcoding and rbcL analyses are useful for delimiting species boundaries and discovering cryptic species diversity in the genus Herposiphonia.
Eleven Gonioctena species have been recorded in the Korean Peninsula. Most species in the genus have very similar coloration and morphology. The male genitalia is generally used as the only reliable diagnostic character differentiating closely related species, but it is still difficult to identify females to species accurately. Here, we determined the COI barcodes of two Gonioctena species described from Korea and give morphological descriptions. The interspecific genetic divergence (Kimura 2-parameter) among the Gonioctena ranged from 0.073 to 0.138 and averaged 0.108. The pairwise genetic distance between the two Korean species(0.102) is similar to the average interspecific divergence. We predict that these analyses will help to resolve taxonomic issues and find cryptic species.
Although widely studied, the natural diversity of the hard tick is not well known. In this study, we collected 194 sequences from 67 species, covering 7 genera of hard tick. The 5' region of the mitochondrial cytochrome c oxidase subunit 1 region (586 bp) has been used to investigate intra- and inter-species variation and the phylogenetic tree of neighbor joining method has been used for assessment. As a result, by comparing the K2P-distance of intra- and interspecies, 30 samples (15.2%) shown that interspecies distance was larger than the minimum interspecfic distance. From the phylogenetic analysis, 86.8% (49) of the species were identified correctly at the genus level. On deeper analysis on these species suggested the possibility of presence cryptic species. Therefore, further work is required to delineate species boundaries and to develop a more complete understanding of hard tick diversity over larger scale.
Ishizakiella supralittoralis (Schornikov, 1974) was collected from rock pools near Busan, South Korea, and is redescribed herein. It is the first record of this species from Korea, and also the first finding of a living population of Ishizakiella McKenzie & Sudijono, 1981 from the continental margins. Here we also comment on the diversity of Ishizakiella in Japan, possible existence of cryptic species, and provide a key to all six currently known species of this genus, including the two fossil ones.

Cryptic species complexes are increasingly recognized in phycological research, obscuring taxonomy and raising questions about factors influencing speciation. A recent exploration of kelp genetic diversity on Haida Gwaii, British Columbia revealed the existence of a new species, Saccharina druehlii, which is cryptic with Saccharina sessilis. This suggests that molecular investigations further north may be required to elucidate the taxonomy and evolutionary history of this lineage. Although, for several decades, S. sessilis was considered a single highly variable species, its taxonomy has been far from straightforward. In particular, Hedophyllum subsessile (Areschoug) Setchell is now recognized as a synonym of S. sessilis in North America, but as a growth form of Saccharina bongardiana in Far East Russia. To resolve this taxonomic confusion, we sequenced mitochondrial (CO1-5P) and nuclear (internal transcribed spacer) markers of S. sessilis populations from the Aleutian Islands, Alaska, USA. Interestingly, none of our sequences matched S. sessilis sensu stricto. Instead, CO1-5P sequences from populations in the central and eastern Aleutians matched exactly S. druehlii with increasing sequence divergence occurring westward. Samples from Attu, the western-most island, composed a genetic group that clearly represents Kjellman's concept of Hafgygia bongardiana f. subsessilis and is distinct enough from S. druehlii and S. sessilis to potentially constitute a distinct species. Therefore, Saccharina subsessilis comb. nov. is proposed for this entity. Our results suggest the existence of a species complex at the crown node of S. sessilis and thus further investigation of Saccharina in Alaskan waters should be conducted to reconstruct the evolutionary history of this fascinating lineage.
Gobiids are hyperdiverse compared with other teleost groups, with about 2,000 species occurring in marine, freshwater, and blackish habitats, and they show a remarkable variety of morphologies and ecology. Testing the effectiveness of DNA barcodes on species that have emerged as a result of radiation remains a major challenge in evolutionary biology. Here, we used the cytochrome c oxidase subunit 1 (COI) sequences from 144 species of gobies and related species to evaluate the performance of distance-based DNA barcoding and to conduct a phylogenetic analysis. The average intra-genus genetic distance was considerably higher than that obtained in previous studies. Additionally, the interspecific divergence at higher taxonomic levels was not significantly different from that at the intragenus level, suggesting that congeneric gobies possess substantial interspecific sequence divergence in their COI gene. However, levels of intragenus divergence varied greatly among genera, and we do not provide sufficient evidence for using COI for cryptic species delimitation. Significantly more nucleotide changes were observed at the third codon position than that at the first and the second codons, revealing that extensive variation in COI reflects synonymous changes and little protein level variation. Despite clear signatures in several genera, the COI sequences did resolve genealogical relationships in the phylogenetic analysis well. Our results support the validity of COI barcoding for gobiid species identification, but the utilization of more gene regions will assist to offer a more robust gobiid species phylogeny.
DNA barcoding of Bryozoa or "moss animals" has hardly advanced and lacks reference sequences for correct species identification. To date only a small number of cytochrome c oxidase subunit I (COI) sequences from 82 bryozoan species have been deposited in the National Center for Biotechnology Information (NCBI) GenBank and Barcode of Life Data Systems (BOLD). We here report COI data from 53 individual samples of 29 bryozoan species collected from Korean seawater. To our knowledge this is the single largest gathering of COI barcode data of bryozoans to date. The average genetic divergence was estimated as 23.3% among species of the same genus, 25% among genera of the same family, and 1.7% at intraspecific level with a few rare exceptions having a large difference, indicating a possibility of presence of cryptic species. Our data show that COI is a very appropriate marker for species identification of bryozoans, but does not provide enough phylogenetic information at higher taxonomic ranks. Greater effort involving larger taxon sampling for the barcode analyses is needed for bryozoan taxonomy.

Freshwater red algae of the order Batrachospermales are poorly studied in India and Nepal, especially on a molecular basis. During a survey in northeast India and east Nepal, six populations of the genus Sheathia were found and analyzed using molecular and morphological evidence. Phylogenetic analyses based on the rbcL gene sequences grouped all populations in a large clade including our S. arcuata specimens and others from several regions. Sheathia arcuata represents a species complex with a high sequence divergence and several smaller clades. Samples from India and Nepal were grouped in three distinct clades with high support and representing new cryptic species: a clade formed by two samples from India, which was named Sheathia assamica sp. nov.; one sample from India and one from Nepal formed another clade, named Sheathia indonepalensis sp. nov.; two samples from Nepal grouped with sequences from Hawaii and Indonesia (only 'Chantransia' stages) and gametophytes from Taiwan, named Sheathia dispersa sp. nov. Morphological characters of the specimens from these three species overlap one another and with the general circumscription of S. arcuata, which lacks the heterocortication (presence of bulbous cells in the cortical filaments) present in other species of the genus Sheathia. Although the region sampled is relatively restricted, the genetic diversity among specimens of these three groups was high and not closely related in the phylogenetic relationship with the other clades of S. arcuata. These data corroborate information from other groups of organisms (e.g., land and aquatic plants) that indicates this region (Eastern Himalaya) as a hotspot of biodiversity.
Studies of the red algal genus Bostrychia over the last 15 years have made it a model system for many evolutionary processes within red algal species. The combination of newly developed, or first employed methods, in red algal species studies has made Bostrychia a pioneer genus in intraspecific studies. Bostrychia was the first genus in which a mitochondrial marker was used for intraspecific red algal phylogeny, and the first for which a 3-genome phylogeny was undertaken. The genus was the first red alga used to genetically show maternal plastid and mitochondria inheritance, and also to show correlation between cryptic species (genetically divergent intraspecific lineages) and reproductive incompatibility. The chemotaxonomic use, and physiological function of osmolytes, has also been extensively studied in Bostrychia. Our continuous studies of Bostrychia also highlight important aspects in algal species studies. Our worldwide sampling, and resampling in certain areas, show that intensive sampling is needed to accurately assess the genetic diversity and therefore phylogeographic history of algal species, with increased sampling altering evolutionary hypotheses. Our studies have also shown that long-term morphological character stability (stasis) and character convergence can only be correctly assessed with wide geographic sampling of morphological species. While reproductive incompatibility of divergent lineages supports the biological species nature of these lineages, reproductive incompatibility is also seen between isolates with little genetic divergence. It seems that reproductive incompatibility may evolve quickly in red algae and the unique early stages of fertilization (e.g., gametes covered by walls, active movement of spermatium nuclei to the distant egg nucleus), also well investigated in Bostrychia,. may be key to our understanding of this process.
Morphological and isozyme analyses were used to examine closely related species of Tridentiger obscurus and \ulcorner: brevispinis, a pair of cryptic species which has intricate problem with regard to species boundary, and to clarify the taxonomic position. In this study, it was revealed from their 17 allopatric streams that there are differences of band ornamentation on the lateral side and three genetic markers between two species. However, genetic relationship between them (S = 0.813, D = 0.192) was within the intraspecific level of similarity. However, at the sympatry of two species no hybrid was found except at Bangjuk stream where the electrophoretic hybrids with slight deficiency of heterozygosity were observed. It is shown that at sympatry there is a finer microenvironmental subdivision between two species at all populations including Bangjuk stream. To manifest the extent of assortative mating, four syrnpatric subpopulations at Bangjuk stream were analyzed for departure from Hardy-Weinberg equillibrium by F-statistics. All subpopulations were deviated significantly from the expectation under random mating. The evidences presented here supports the recognition of Tridentiger obscurus and T. brevispinis as semispecies in the late stage of reproductive isolation
