Fluorescent and Luminescent Proteins Derived from Marine Organisms: Functions and Applications

  • Organisms constituting a large proportion of marine ecosystems, ranging from bacteria to fish, exhibit fluorescence and bioluminescence. A variety of marine organisms utilize these biochemically generated light sources for feeding, reproduction, communication, and defense. Since the discovery of green fluorescent protein and the luciferin-luciferase system more than a century ago, numerous studies have been conducted to characterize their function and regulatory mechanism. The unique properties of fluorescent and bioluminescent proteins offer great potential for their use in a broad range of applications. This short review briefly describes the functions and characteristics of fluorescent and bioluminescent proteins, in addition to summarizing the recent status of their applications.

  • In power cables as one of the important power applications adopting HTS tapes, a good insulation should be kept at its optimum performance. As an insulation material for superconducting device applications, polypropylene laminated paper (PPLP) is now widely used instead of the conventional Kraft paper. In addition to its dielectric property, the insulation material should also possess superior mechanical property at cryogenic temperatures and operability that is necessary for the insulation winding process. This study aims to evaluate the mechanical property of the PPLP material at ambient and cryogenic temperatures. At cryogenic temperature, the failure stress of PPLP increased significantly as compared with that at ambient temperature. The failure stress at both temperatures depended upon the sample orientation to the load application.

A study on the fracture toughness degradation in cryogenic structural material using single-specimen method (단일 시험편법에 의한 극저온용 구조재료의 파괴인성 저하에 관한 연구)

  • This paper was investigated degradation of the fracture toughness caused by sensitizing heat-treatment of the cryogenic structural material JN1 base metal using unloading compliance method reported as useful a method in evaluating the elastic-plastic fracture toughness at cryogenic temperature. The specimens used in this paper were 20% side-grooved 0.5T-CT specimens which were machined in the JN1 base metal. Also, to investigate cryogenic fracture toughness of the fusion line region in the JN1 GTA weldments, it was also used 20% side-grooved 0.5T-CT specimens that was machined fusion line to located in the middle of the specimen. The cryogenic fracture toughness values of the JN1 base metal were significantly decreased with increasing the time and temperature of the heat treatment. The fracture toughness value obtained from the fusion line specimen was invalid, but it was lower value than that of the JN1 base metal. Especially, this value was approximately equal with that obtained from the JN1 650.deg. C-5h heat-treated material.

  • The cryogenic fracture behaviors of austenitic stainless steel HN2 developed for nuclear fusion reactor were evaluated quantitatively by using the small punch(SP) test. The electrochemical polarization test was applied to study thermal aging degradation of HN2 steel. The X-ray diffraction(XRD) analysis was conducted to detect carbides and nitrides precipitated on the grain boundary of the heat treated HN2 steel. The mechanical properties of the HN2 steel significantly decreased with increasing time and temperature of heat treatment or with decreasing testing temperature. The integrated charge(Q) obtained from electrochemical polarization test showed a good correlation with the SP energy(ESP) obtained by means of SP tests. From the results observed in the x-ray diffraction and anodic polarization curve, it was known that the material the grain boundary. Combining SP test and electrochemical polarization test, it could be useful tools to non-destructively evaluate the cryogenic fracture behaviors and the aging degradation for cryogenic structural material.

  • Dielectric and piezoelectric properties of perovskite materials such as La modified $Pb(Zr,Ti)O_3$ ceramics and $Pb(Zn_{1/3}Nb_{2/3})O_3-PbTiO_3$ single crystals were investigated for cryogenic capacitor and actuator applications. Enhanced extrinsic contributions resulted in piezoelectric coefficient (d33) as high as 250 pC/N at 30 K, superior to that of PZT ($d_{33}$ ~ 100 pC/N). This cryogenic property enhancement was associated with retuning the MPB (or cryogenic temperatures. PZN-PT single crystals exhibited dramatic property improvements such as $d_{33}$ > 500 pC/N at 30 K as a result of an engineered domain state.

  • Austenitic stainless steels of 300 series are widely used as the structural material due to excellent their cryogenic mechanical properties at cryogenic temperature. There are 316 steel which molybdenum is added to improve the austenitic stability, 316L which carbon contents is reduced to decrease the grain boundary precipitation during welding process, and 316LN which nitrogen is added to improve the austenitic stability and the mechanical strength. But material researches for the welding conditions and mechanical properties at the cryogenic temperature were insufficient so far. In this paper, the characteristics of mechanical properties considering the effect of welding conditions and cryogenic temperature are studied.

Temperature Prediction of Al6061 Tube in Cryogenic Heat Treatment by CFD Analysis and Experimental Verification (CFD 해석을 이용한 Al6061 튜브의 극저온 열처리 시 소재의 온도 예측 및 실험적 검증)

  • The purpose of this study is to establish the analysis method for prediction of temperature during cryogenic heat treatment. Experimental cryogenic heat treatment is conducted to observe the phenomena such as boiling of fluid, ice layer on the material surface and to measure the temperature distribution of Al6061 tube. The CFD analysis considering the observed phenomena in the experiment is performed to predict the temperature distribution and convection heat transfer coefficient at each stage of cryogenic heat treatment, in which the boiling of fluid is considered as the multi-phase condition of vapour and liquid. The formation of ice layer on the tube surface is also modeled between material and fluid. The predicted results are in good agreement with the experimental ones. From the results, it is shown that the analysis method can predict the temperature distribution and convection heat transfer coefficient during cryogenic heat treatment.

  • Recently, superconducting cable has attracted a great deal of interest from the viewpoint of energy saving, one of the most important and difficult problems to be solved to assure the high reliability, safety and economy of cable is breakdown is cryogenic temperature. In this paper, we will report on the characteristics and mechanisms of the breakdown and creeping discharge in cryogenic liquids, dual role of those as the cooling and insulating material, and composited system of cryogenic liquid - gas taking into consideration for application.

  • This paper was undertaken to develop the fracture toughness testing method using small and single specimen compared to the conventional method in evaluating elastic-plastic fracture toughness of the superconducting magnet structural material at cryogenic temperature. The elastic-plastic fracture toughness test was conducted by using the unloading compliance method recommended by ASTM E813-89 to accomplish the above purpose. And, the 20% side-grooved 0.5TCT and 1TCT specimens were used to evaluate the fracture toughness by using as possible as miniaturized CT specimen. The unloading compliance method was a very useful method in evaluating elastic-plastic fracture toughness at cryogenic temperature. It could be taken valid fracture toughness values by using 20% side-grooved 0.5TCT specimen recommended by ASTM E813-89.
