The Differences of Temperatures, Growth and Crown Gall Occurrence in Young 'Kyoho' Grapevines According to Heat Conservation Materials during Winter (보온피복재 종류가 '거봉' 포도 유목의 월동시 온도 차이, 발아 및 근두암종병 발생에 미치는 영향)

  • This experiment was carried out to find out the effects of heat conservation materials (burying in soil, lagging, lagging +straw, nonwoven fabric, nonwoven fabric+straw) on freezing damage, labor saving, and crown gall occurrence of 'Kyoho' grapes. Temperature differences in burying in soil and lagging with $2.8^{\circ}C$ and $6.4^{\circ}C$, respectively and were considered favorable for over-wintering of grapevines. Heat conservation index in lagging +straw and burying in soil calculated from degree-hours below $-10^{\circ}C$ was 5 to 7 times higher than that of open field. Budbreak started earlier in lagging with+straw and nonwoven fabric+straw covering, and percent budbreak was increased by 22% and 7%, respectively, as well as higher than burying in soil. Diameter of bearing mother branch and length of internode and daughter branch were gross or long with soil and lagging straw and nonwoven fabric+straw. Cane growth was enhanced by burying in soil and lagging with+straw treatment. Crown gall occured higher in soil covered grape vines Labor saving was obtained in lagging with as much as 44% compared to burying in soil.

  • Artemisia annua contains the antimalarial principle, artemisinin. The possibility of the production of this compound through tissue culture technique was studied. The optimum combinations of hormones for the induction of callus were p-chlorophenoxyacetic acid(pcPA) and 6-benzylaminopurine(BAP) or pcPA and N-isopentenylaminopurine(2iP) in 0.05 mg/l each. For the growth of callus, the same combination of pcPA and BAP was optimum in concentrations of $1.0\;{\mu}M\;and\;0.5\;{\mu}M$, respectively, and the optimal concentration of sucrose was also found to be 2%(w/v). Tissue culture from the crown gall grew faster than normal callus. In the suspension culture broth and the cells of normal callus or Agrobacterium-transformed tumors, arteannuic acid and 11,12-dihydroarteannuic acid were found together with common phytosterols, whereas artemisinin was not found.

  • Agrobacterium vitis is a causal agent of crown-gall disease on grapevine. Bacterial strains were isolated from crown galls of different grapevine varieties in grapevine farms of Kyungbuk(Kimcheon), Chungbuk(Okcheon), Chungnam(Daejon, Choenan) and Kyeonggi(Suwon, Ansung) areas in Korea from 2002 to 2005. Primer sets, Phe A and VirA, which were derived from pectate lysase gene and virA gene of Ti-plasmid in A. vitis were used to detect A. vitis strains from crown galls. PheA and VirA primers amplified DNA fragments of 0.25 kb and 0.5 kb from fifty-one bacterialstrains. They formed crown galls on grapevine variety, Kyoho, or carrot disks with variable pathgenecity It was confirmed that the biochemical characteristics of 10 bacterial strains that was strong pathogene city on grapevine were mostly in agreement with type culture strains of A. vitis, showing growth in the presence of 2% NaCl, non-production of acid from melezitose and negative response in production of 3-Ketolactose.

  • The cells of Rubia cordifolia var. pratensis were transformed by Agrobactrium tumefaciens strain 11157. Surface-sterilized young leaves and stems of the plants were cocultivated with bacterial suspensions. Crown galls induced from stems were cultured with variation of culturing conditions and compared with untransformed cells. The growth rates and production of anthraquinone pigments of cells were remarkably improved by transformation. Furthermore, hairy roots were induced by inoculation or cocultivation with Agrobacterium rhizogenes strains.

Effective Combination of Agrobacterium tumefaciens Strains and Ti Plasmids for the Construction of Plant Vector System

  • The purpose of this study is to obtain the most efficient combination of Agrobacterium tumefaciens strains and Ti plasmids for the construction of dicotyledonous plant vector system. Ti plasmid-curing A. tumefaciens A136 and KU12C3 were transformed with four kinds of Ti plasmids, pTiBo542, pTiA6, pTiKU12 and pTiAch5, respectively. The stems of 28 species of dicotyledonous plants were then inoculated with these transformants and examined for crown gall formation. The different combination of A. tumefaciens strains and Ti plasmids showed quite a difference in terms of the crown gall formation. Agrobacterium strins A136 and KU12C3 have a same plant host range in case that both strains harour the same kind of Ti plasmid, pTiBo542 or pTiAch5. However, the above-mentioned both strains have quite different host range in the event of containing the same Ti plasmid, pTiKU12 or pTiA6. In case that KU12C3 contains pTiA6 or pTiKU12, this strain has a wider plant host range than A136. The plant host range of pTiBo542 is the widest, followed by pTiA6, pTiKU12 and pTiAch5. Twelve plants among 28 tested plants are not transformed by any virulent Agrobacterium strains used in this study. In conclusion, A. tumefaciens KU12C3 and A136 harboring pTiBo542 showed the widest host range for transforming dicotyledonous plants. Also, it was acertained that the host range of Ti plasmids is affected by chromosomal level.

  • Bradyrhizobium japonicum spheroplasts were prepared by culturing cells in the presence of glycine, follwed by treatment with lysozyme. The cells were examined by electron microscopy during the formation of spheroplast. Then Ti plasmid from Agrobacterium tumefaciens 15955 was introduced into Bradyrhizobium japonicum by glycine-lysozyme induced spheroplast transformation. After cell wall regeneration, transformants were selected by the ability of utilization of octopine. Transformation were received at a frequency of $1{\times}10^{-7}$. The transformants obtained from spheroplast transformation harbored the introduced Ti plasmid, which was identified by agarose gel electrophoresis. Furthermore, the differences in their gene products were observed between the transformant and the recipient cell by two-dimensional polyacrylamide gel electrophoresis. The transformants which still possessed the same ability nodulate soybean (Glycine max.) as that of the original host strain, acquired the ability to induce tumor on Petunia hybrida like Agrobacterium, but formed the small crown galls in size compared to those of Agrobacterium tumefaciens.

Genetic Diversity of Agrobacterium vitis Strains in Korea (국내 포도나무 혹병(Agrobacterium vitis) 균주의 유전적 다양성)

  • Fifty nine strains of Agrobacterium vitis, the causal agent of crown-gall disease on grapevine, originating from different geographical regions and 16 grapevine cultivars including 35 Kyoho cultivar of Korea, were characterized by PCR polymorphic analysis using Universal Rice Primer(URP). Of 12 URP primers, primers URP1F, URP2R, URP2F, and URP4R, URP17R were available for detecting PCR polymorphic bands among the A. vitis strains. PCR polymorphic bands produced by primers URP2F and URP17R were profiled to 12 strain types. A. vitis strains originated from Kyoho cultivar of grapevine showed relatively simple genetic diversify of the four PCR types, while the A. vitis strains originated from other grapevine cultivars and type culture strains showed various genetic diversity with 8 types. Unweighted Pair-Group Method with Arithmetic mean(UPGMA) cluster analysis using the URP-PCR polymorphic bands showed 59.4. vitis strains are genetically clustered into large seven groups.

  • Locust gall midge (LGM), Obolodiplosis robiniae (Haldeman) (Diptera: Cecidomyiidae), is a cecidomyiid insect forming roll-up galls on leaves of Robinia pseudoacacia Linnaeus (Fabaceae). LGM, known as native to North America, was reported from Korea and Japan in 2002. LGM was observed weekly or biweekly to clarify their voltinism and distribution within the crown of the host tree in two sites of Osan and Siheung in Korea from May to August, 2007. Density of LGM was investigated based on the number of larvae per leaf. Two generations of LGM were observed in Siheung site whereas three generations in Osan site during the present study. The result indicated that LGM had at maximum three generations per year. The density of LGM in Osan was higher in the upper crown of the host trees than middle or lower part. In Siheung, LGMs were distributed more on the exterior of the lower crown than the interior. The average number of larvae per gall was $3.3{\pm}0.1$ and $2.8{\pm}0.1$ individuals per leaf in Osan and Siheung, respectively.

Isolation and identification of Agrobacterium spp. using carrot disc and transformation of Nicotiana gluca by selected strains (당근 절편을 이용한 Agrobacterium spp. 분리, 동정과 선발균주에 의한 연초의 형질전환)

  • These studies were carred out to obtain the transformant from tobacco cells by Agrobacterium spp. from crown gall and soil at the natural field in Korea, and identified their virulence. Kodo's and Clark's selective media were used for isolation of Agrobacterium spp. In these media, total of 99 strains were characterized based on the morphological characteristics of colonies. Among them 34 strains were able to induce on carrot discs. And hypervirulent strains C23-1 and K29-1 were identified as Agrobacterium tumefaciens biotype 1 and biotype 2, respectively. These strains formed fast growing, larger gall as compared to those induced by other strains on the carrot discs. Transformed tobacco callus was initiated on the phytohormone free MS medium with 250$\mu\textrm{g}$/ml carbenicillin after co-cultivation of tobacco stem explants and Agrobacteria. On the phytohormone free media, shoot was rarely formed from transformed callus. However, these shoot were teratoma shoots which were not grown as normal shoot, and teratoma shoot from transformant by C23-1 was smaller than that of K29-1.

First Description of Crown Gall Disease on Ginseng

  • In March of 2003, tumors (galls) were observed on ginseng seedling roots in ginseng seedbeds at Yeoju, Gyeonggi province, Korea. Symptoms were spherical or galls with about 0.5-1.0cm in diameter formed on the upper through middle parts of the primary roots. Bacterial isolates obtained from the root galls were Gram-negative, rod-shaped with peritrichous flagella, aerobic, not forming yellow or orange colonies on nutrient glucose agar, yeast extract-dextrose $CaCO_3$ agar and nutrient-broth yeast extract agar, non-fluorescent on King's B agar, and non-spore forming, which were identical to characteristics of the genus Agrobacterium. They were identified as Agrobacterium tumefaciens with 0.732-0.993 similarities in 100% probability by the Biolog analyses. The 16S rRNA gene partial sequences of the six isolates tested (Genbank Accession EF486308-EF486313) were 100% homologous to those of other A. tumefaciens strains (GenBank accession AF501343, AY701900, AY701898, AY701899). The above results confirmed that this bacterium is A. tumefaciens. Pathogenicity of the bacteria was proved by the inoculation test on carrot root discs and tomato seedlings. This is the first description of A. tumefaciens causing root gall in ginseng seedling. The disease occurred locally and sparsely, but considering its appearances in seedbeds suggests that the ginseng root gall may become a threat to ginseng in Korea.
