쑥갓 및 참나물 중 Lufenuron의 생산단계 잔류허용기준 설정을 위한 잔류 특성 연구 (Residual Characteristics of Lufenuron in Crown Daisy and Chamnamul for Establishing Pre-Harvest Residue Limit)

  • Pre-harvest residue limits (PHRLs) have been proposed as criteria for a proactive role to exceed the maximum residue limit (MRL) of pesticides in agricultural products at harvest. However, PHRL numbers are significantly less than those of established MRLs. This study was performed to determine the dissipation constants and residual concentrations of lufenuron after application on crown daisy and chamnamul under green house conditions. Two residue field trials for each crown daisy and chamnamul were selected to consider a different geographical site at least 20 km far from one another. The pesticide was treated according to critical GAP. After samples were sprayed with lufenuron, they were collected at 0, 1, 3, 5, 7, 10, and 14 days and analyzed using HPLC-DAD. The mean recoveries of crown daisy and chamnamul were within the range of 70-120% with below 20% coefficient variation, which is within the acceptable limits specified by the manual of pre-harvest residue study for pesticides (MFDS, 2014). The biological half-lives in field I and field II were 7.0 and 4.6 days for crown daisy and 2.7 and 2.8 days for chamnamul, respectively. The lower bounds of 95% confidence intervals of dissipation rate constants of lufenuron in crown daisy were determined to be 0.0692 and 0.1298 for field I and field II, respectively, and in chamnamul were 0.2067 for both field I and field II. After applying lufenuron 5% EC, the lufenuron residues on crown daisy and chamnamul at the pre-harvest intervals (14 days for crown daisy and 7 days for chamnamul) were below the safe levels. The dissipation rates of lufenuron in crown daisy and chamnamul were evaluated for similarities with leafy vegetables based on a 95% confidence interval.

저온처리 전해산화수로 세정한 쑥갓과 케일의 저장중 품질변화 (Changes in Quality of Crown Daisy and Kale Washed with Cooled Electrolyzed Acid Water during Storage)

  • 쑥갓과 케일을 시료 중량 50배의 5$^{\circ}C$로 냉각한 전해산화수에 2분 3회 다단침지 처리한 후 저장중의 품질변화를 조사한 결과, 쑥갓의 경우 총균수는 무처리 쑥갓에 비해 평균 1/130, 대장균군수는 평균 1/1,170, 케일의 경우 총균수는 무처리 케일에 비해 평균 1/870, 대장균구수는 평균 1/470 수준으로 감소하였으나 1$0^{\circ}C$ 저장 6일후 부터는 무처리, 수도수 침지 처리구와 유사한 수준으로 증가하였다. 저장중 중량감소율은 쑥갓과 케일 모두 저장 3일까지는 무처리구에 비해 낮은 감소율을, 그리고 저장 3일 이후부터 무처리구에 비해 다소 높은 중량감소율을 나타내었다. 한편 폐기율은 전해산화수로 침지한 쑥갓과 케일이 저장 6일까지는 무처리와 수도수 침지구에 비해 낮은 값을 나타내었다. 케일의 경우 rupture srenth 는 저장 초기에는 무처리와 수도수 침지 처리구에 비해 높은 값을 나타내었으나 저장기간 동안에는 유사한 경향을 나타내었다. 저장중 색도는 쑥갓과 케일 모두 저장 초기부터 무처리구보다는 수도수 침지구가, 수도수 침지구보다는 전해산화수 처리구가 다소 높은 L, b값과 낮은 a값을 나타내었으며, 클로로필 함량 변화도 쑥갓과 케일 모두 침지 직후에는 전해 산화수로 침지한 처리구가 다소 적은 값을 나타내었으나 저장 3일부터 무처리구, 수도수처리구, 전해산화수 처리구의 순으로 감소속도가 빨랐다. 저장기간 중 관능적인 특성도 저장기간 전반에 걸쳐 쑥갓과 케일 모두 변색, 조위, 폐기율, 종합적 기호도에서 무처리와 수도수 처리구에 비해 다소 높은 점수를 나타내었다.

  • 다양한 아미노산과 당을 $90^{\circ}C$에서 반응시켜 생성한 마이얄반응 생성물(Maillard reaction product)은 쑥갓에서 추출한 폴리페놀산화효소(polyphenol Oxidase, PPO)를 강하게 저해하였다. 글리신(glycine)과 포도당을 함유한 반응물으로부터 MRP의 생성은 $90^{\circ}C$에서 반응시간에 따라 직선적으로 증가하였으나 글리신과 포도당의 양은 상대적으로 감소하였다. 반응액에서 MRP의 증가함에 따라 반응액은 쑥갓 PPO활성을 비례적으로 강하게 저해하였다. 실험에 사용한 아미노산과 당중에서 글루타민과 자일로스를 아미노산과 당으로 이용했을 경우에 생성된 MRP가 쑥갓 PPO의 활성에 강한 저해제로 작용하였다. 카테콜을 기질로 사용한 쑥갓 PPO의 마이클레스 상수(Michaelis Menten constant)는 22.0 mM이었으며, MRP는 쑥갓 PPO의 비경쟁적 저해제로 작용하였다.

Screening for Korean Vegetables with Anticarcinogenic Enzyme Inducing Activity Using Cell Culture System

  • There is extensive evidence suggesting the protective role of fruits and vegetables against chemically induced carcinogenesis. We have tested the ability of a representative range of Korean vegetables to act as blocking agents against neoplastic initiation by determining the induction level of quinone reductase , an anticarcinogenci marker enzyme, in hepalclc 7 cells exposed to vegetable extracts. Among thirty vegetables tested, Arcitum lappa(Burdock), Brassica juncea (Mustard leaf), Pteridium aguilinum (Bracken) and Chrysanthemum cornoratium(Crown daisy) caused a significant induction of quinone teductase activity with a limited increase in arylhdrocarbon hydroxylase activity. Combination of crown daisy with burdock had synergistic effect on quinone reductase induction. Quinone reductase-inducing activity was found mostly in hesane and ehtylactate fractions of MeOH extract of crown daisy while it ws not quinone reductase activity in liver, kideny, lung, and small intestine, confirming the presence of potent QR inducer (s) in crown daisy. These sata suggest that some vegetables including crown daisy induced QR merits further investigation as a potential cancer preventive agent in human.

쑥갓과 머위의 잎과 중기의 구조탄수화물의 변화 (The Changes in Structural Carbohydrate on Crown Daisy and Butterbur)

  • Change on the structural carbohydrate(several fiberous components) was determined by vegetables(crown daisy and butterbur)-cultivated in Ulsan, Kyungnam, Korea-as its stage of maturity developed. Samples were separated into leaf and stem, which were dried at 7$0^{\circ}C$ for 24hr, and ground to pass a 1mm screen. They were subjected to moisture, crude protein, crude fat and several dietary fiber-DF(dietary fiber, include unavaible components), NDF(neutral detergent fiber), ADF(acid detergent fiber), lignin, hemicellulose, cellulose and protein corrected NDF(c-NDF), IDF(indigestible fiber, include lignin, hemicellulose and cellulose). In general, structural carbohytrate(several dietary fiber) of vegetable was affected by the growth stage. In case of crown daisy and butterbur, dietary fiber in leaf was higher than DF in stem.

시설재배 쑥갓 및 피망의 작물특성에 따른 Boscalid 및 Spinetoram의 잔류량 감소추이 (Residual Dissipation based on Crop Commodities Classification of Boscalid and Spinetoram on Crown Daisy and Sweet Pepper under Green Houses)

  • BACKGROUND: This study was performed to evaluate the residue dissipation of boscalid and spinetoram on crown daisy and sweet pepper affected by the morphology of the crop. The half-lives and dissipation rate constants for boscalid and spinetoram on crown daisy and sweet pepper were calculated. And then lower limit of 95% confidence interval for dissipation rate constant could be used to propose the pre-harvest residue limit. METHODS AND RESULTS: The pesticide products diluted according to the pesticide label were applied one time on crown daisy and sweet pepper at 2 field sites, respectively. Initial concentration of boscalid and spinetoram on crown daisy after application were in the range of 72.80~117.15 mg/kg and 2.82~4.67 mg/kg, respectively. And Initial concentration of boscalid and spinetoram on sweet pepper were in the range of 1.58~1.62 mg/kg and 0.10~0.21 mg/kg, respectively. Boscalid and spinetoram for crown daisy dissipted below the maximum residue limit(MRL) at 10 and 2 days after application, respectively. All residues concentration of boscalid and spinetoram for sweet pepper below the MRL at 0 day after application. The half-lives based on dissipation rate constant for boscalid and spinetoram on crown daisy were 4.2~4.9 days and 3.0~2.4 days respectively. And the half-lives for boscalid and spinetoram on sweet pepper were 6.7~7.0 days and 2.8~4.0 days respectively. CONCLUSION: The difference in initial concentration of boscalid and spinetoram among crop commodities were due to different crop morphology with larger surface areas. This study was suggested that pre-harvest residue limit would be calculated from lower limit of 95% confidence interval for dissipation rate constant and would be useful to protect consumers by controlling the pesticide residues in crop.

줄기 엽채소의 기계적 파지시 리올로지 특성 (Rheological Properties of Bundled Leaf Vegetables Held and Picked-up by Machine)

  • This study was carried out as basic researches to develop the leaf vegetable harvester. This study was conducted to investigate physical and rheological properties of bundled leaf vegetables with stem (Chinese leek, Crown daisy and Chamnamul) as test materials held and picked-up by a machine. Stress-strain behavior, stress relaxation, and strain recovery for the bundled materials were analyzed using simple Maxwell model. Physical and rheological properties of the materials were investigated by measuring rupture load, deformation and stress experimentally. Also, strain recovery time when unloading was measured using super high speed camera. Recorded recovery time for stress-strain behavior was0.026 s for Chinese leek with liner strain recovery, 0.046 s for Crown daisy and 0.05 s for Chamnamul with non-linear strain recovery. Furthermore, the strain recovery time for permanent deformation was 0.026 s, 0.046 s, and 0.05 s for Chinese Leek, Crown daisy and Chamnamul, respectively. Finally, strain recovery time and strain recovery ratio for the test materials were 0.17 s, 60.4% in Chinese leek, 0.12 s, 55.3% in Crown daisy, 0.15 s, 58.7% in Chamnamul. Here strain recovery time means that how fast the test materials are recovered from initial deformation and strain recovery ratio means how much the test materials are recovered from initial deformation. The above results show that the test materials can be held enough and moved by the belts.

Isolation of a Gibberellin-producing fungus (Penicillium sp. MH7) and Growth Promotion of Crown Daisy (Chrysanthemum coronarium)

  • Plant growth promoting fungi (PGPF) are well known for the production of useful secondary metabolites. However, limited information is available on the gibberellin (GA) production capacity of PGPF of endophytic origin. In the current study, 15 fungal endophytes were isolated from the roots of Crown daisy, and then screened on Waito-c rice, in order to identify plant growth promoting fungi. The fungal isolate MH7 significantly increased the shoot length (12.1 cm) of Waito-c in comparison with control treatment (7.9 cm). In a separate experiment, the culture filtrate (CF) of MH7 significantly promoted the growth attributes of Crown daisy. The MH7 CF was analyzed for gibberellins and it contained all physiologically active gibberellins ($GA_1$, 1.37 ng/ml; $GA_3$, 5.88 ng/ml; $GA_4$, 8.62 ng/ml; and $GA_7$, 2.05 ng/ml) in conjunction with physiologically inactive $GA_9$ (0.83 ng/ml), $GA_{12}$ (0.44 ng/ml), $GA_{15}$ (0.74 ng/ml), $GA_{19}$ (1.16 ng/ml), and $GA_{20}$ (0.98 ng/ml). The CF of MH7 produced higher amounts of $GA_3$, $GA_4$, $GA_7$, $GA_9$, and $GA_{12}$ than wild-type Fusarium fujikuroi, which was used as a control for GA production. The fungal isolate MH7 was later identified as a new strain of Penicillium on the basis of its morphological characteristics and phylogenetic analysis of the 188 rDNA sequence.

생산단계에서 소비단계 생식 쑥갓의 azoxystrobin 잔류량에 따른 위해성 평가 (Risk Assessment of Azoxystrobin Residues in Fresh Crown Daisy from Farm to Fork)

  • The biological half-life and dissipation rate of azoxystrobin in crown daisy were calculated to establish the pre-harvest residue limits (PHRLs). The pesticide residues were calculated after washing with five different processes to propose an effective process in the household and conducted a risk assessment to confirm dietary safety. Azoxystrobin was sprayed according to the critical good agricultural practices (cGAP) in two different field trials, and the samples were harvested 7 times. The limit of quantitation was 0.02 mg/kg, and the mean recoveries of azoxystrobin were within the range of 70~120% with below 20% coefficient variation at the concentration of 0.02 and 0.2 mg/kg . The biological half-lives were 7.4 and 4.7 days, and the dissipation rate constants were 0.0872 and 0.1217 in fields 1 and 2, respectively. The average removal rates were 58.13~78.13% by the different washing processes, and there were significant differences between the washing processes (one-way ANOVA analysis and post-hoc Duncan test, p-value<0.05). The residues of azoxystrobin in crown daisy were safe levels from farm to fork after application with the critical good agricultural practice (cGAP) registered in Korea.

Metalaxyl-M 및 dinotefuran 입제의 쑥갓 중 잔류 특성 및 위해성 평가 (Residual Characteristics and Risk Assessments of Metalaxyl-M and Dinotefuran in Crown Daisy)

  • BACKGROUND: This study was performed to determine residual characteristics of soil-treated metalaxyl-M and dinotefuran in crown daisy and to evaluate the risks from intake of the residual pesticides in the crop. METHODS AND RESULTS: The pesticide granules were treated in soil on two levels, and the plants samples were collected 51 days after seeding. The analytes were extracted and partitioned using the QuEChERS extraction packet (MgSO4 4 g, NaCl 1 g). The quantitative methods for metalaxyl-M and dinotefuran were validated in linearity, accuracy, and precision. Risk assessments of the pesticides were performed using Korea national nutrition statistics 2019. CONCLUSION(S): The residual concentrations of metalaxyl-M in crown daisy were 0.09-0.10 mg/kg (for the treatment at 6 kg/10 a) and 0.17-0.19 mg/kg (12 kg/10 a), respectively. The residual concentrations of dinotefuran in the crop were 0.53-0.75 mg/kg (3 kg/10 a) and 1.17-1.26 mg/kg (6 kg/10 a). The amounts of pesticides were less than MRL (Maximum Residue Limits) according to the Korean MFDS (Ministry of Food and Drug Safety). The HI (Hazard Index) of metalaxyl-M and dinotefuran for consumers was 0.0075% and 0.2250%, respectively. For females in the age between 50-64, the major consumer group, the HIs of the pesticides were <3%. Considering the consumption of crown daisy, they are not considered to be of toxicological concern.
