Covalent Interactions of Toluenediisocyanate with DNA and Proteins

  • The covalent interactions of toluenediisocyanate (TDI) with macromolecules were investigated both in vitro and in vivo. In vitro incubations of 2,4- and 2,6-TDI with DNA or proteins resulted in dose-dependent formation of TDI-protein and TDI-DNA adducts. TDI-treated DNA was highly resistant to enzymatic digestion and thermal hydrolysis, but was readily hydrolyzed under acidic conditions by releasing its corresponding toluenediamine (TDA), suggesting that TDI caused the crosslinking of DNA. Reaction of TDI with albumin and globin resulted in the formation of several adducts, and some adducts were formed in blood of TDI-treated rats in a dose-dependent fashion. Administration of TDI to rats resulted also in a dose-dependent binding of TDI to hepatic tissue. Levels of TDI-albumin adducts were 10 times higher than those of TDI-globin adducts; the biological half lives of TDI-albumin and TDI-globin adducts were 1.2 and 12.5 days, respectively. Globin adducts were detected up to 28 days after the treatment. Hepatic TDI protein adducts were persistent for a substantial period whereas the levels of hepatic TDI-DNA adduct were decreased rapidly. These results indicate that the isocyanato group of TDI is not readily hydrolyzed under physiological conditions, is transported to other organs, and is bound to DNA and/or proteins without further metabolic activation. As the adducted products degrade in the body, TDA is released and introduced to the liver. TDA may additionally bind to hepatic tissue after metabolic activation. Thus, the toxic effect of TDI exposure is considered to persist during the lifetime of the adducted biological macromolecules.

Effects of heat and gamma radiation on the degradation behaviour of fluoroelastomer in a simulated severe accident environment

  • In this study, the effects of heat and radiation on the degradation behaviour of fluoroelastomer under simulated normal operation and a severe accident environment were investigated using sequential testing of gamma irradiation and thermal degradation. Tensile properties and Shore A hardness were measured, and thermogravimetric analysis was used to evaluate the degradation behaviour of fluoroelastomer. Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy were used to characterize the structural changes of the fluoroelastomer. Heat and radiation generated in nuclear power plant break and deform the chemical bonds, and fluoroelastomer exposed to these environments have decreased C-H and functional groups that contain oxygen and double bonds such as C-O, C=O and C=C were generated. These functional groups were formed by auto oxidation by reacting free radicals generated from the cleaved bond with oxygen in the atmosphere. In this auto oxidation reaction, crosslinks were generated where bonded to each other, and the mobility of molecules was decreased, and as a result, the fluoroelastomer was hardened. This hardening behaviour occurred more significantly in the severe accident environment than in the normal operation condition, and it was found that thermal stability decreased with the generation of unstable structures by crosslinking.

Adsorption of uranium(VI) ion on the nitrogen-donor macrocyclic synthetic resin adsorbent (질소-주게 거대고리 합성수지 흡착제에 의한 우라늄(VI) 이온의 흡착)

  • Resins were synthesized by mixing 1-aza-18-crown-6 macrocyclic ligand into styrene(dangerous matter) divinylbenzene(DVB) copolymer with crosslink of 1%, 2%, 6% and 12% by substitution reaction. The synthesis of these resins was confirmed by content of chlorine, elemental analysis, thermogravimetric analysis, electron microscopy, and IR. The effects of pH, time, crosslink of resins and dielectric constant of solvent on adsorption of uranium ion by resin adsorbent were investigated. Uranium ion showed a great adsorption above pH 3 and adsorption equilibrium of metal ions was established in about two hours. In addition, adsorptive selectivity of resin in ethanol solvent was $UO{_2}^{2+}$ > $Zn^{2+}$ > $Lu^{3+}$ ion and adsorption of uranium ion increased with the increase of the degree of crosslinking (1%~12%) and was inversely in proportional to the order of dielectric constant of solvents.

  • Unsaturated polyester(UP) resin is one of the major thermosetting resins. It is very useful as the matrix resin of the composite material because of its low viscosity. The polymer resin, however, has several drawbacks; The volume shrinkage occurs during the crosslinking reaction of the UP resin with styrene monomer and the resulting polymer is weak to the alkali and also brittle. The mechanical properties of UP resin can be improved by blending various materials. In this study, polyurethane(PU) was used as a modifier in order to enhance the toughness of the UP resin. The goal of the research is to study the effect of the polyol molecular weight as a PU soft segment and the PU contents on the toughness of PU-modified UP resins. UP/PU polymer network may occur through the reaction between isocyanate group in the methyldiisocyanate(MDI) and hydroxyl group in the UP molecules. The maximum toughness value was shown at 2 wt% of the PU content. This effect results from the incorporation of the PU segment into the UP resin.

Electron Microscopical Property of Transglutaminase Added Milk (트랜스글루타미나제를 첨가한 우유의 전자현미경적 특성)

  • Raw skim milk and colloidal calcium phosphate-free skim milk were treated with microbial transglutaminase (TGase), ultracentrifuged at varying rates and were observed to contain textural properties using a scanning electron microscope (SEM). Skim milk showed irregular signs of conformation at lower centrifugal rate, and associated regular (10,000 ${\times}$g) and thin with broad holes (20,000 ${\times}$g). The associated texture became thick and irregular (40,000 ${\times}$g), and fine particles were regularly associated (100,000 ${\times}$g). When skim milk was incubated for 1 hr with TGase, casein micelles aggregated and broadened as centrifugation rate increased. When skim milk was incubated for 8 hrs with TGase, casein micelles associated to large widened aggregates, and were associated regularly which then became irregular (100,000 ${\times}$g). When colloidal calcium phosphate-free skim milk incubated for 1 hr with TGase showed no sediment, the milk incubated for 8 hrs with TGase associated together, yielding broadened and regular layers as the centrifugation rate increased. It is assumed that such phenomena could be caused by protein crosslinking reaction with TGase and conformational change of casein molecules, as well as dependencies on reaction time, temperature and ultracentrifugation rate.

Preparation and Adhesion of One Part Room Temperature Curable Alkoxy Type Silicone Sealant (일액형 알코올형 실리콘 실란트의 제조 및 접착 물성)

  • Silicone sealants are composed of polymer, plasticizer, crosslinker, catalyst and filler. Types and compositions of components are effected on sealant performances. In recent, use of alkoxy type silicone sealant increased due to environmental advantage. In this study, we investigated effects of component types and ratios on one-part room temperature curable alkoxy type silicone sealant preparation and adhesion properties. Alkoxy type silicone sealants were prepared with various PDMS (polydimethylsiloxane) viscosities. In addition, the effect of plasticizer, crosslinkers, and catalyst on sealant obtained from by mixture of PDMS viscosities of 20000 and 80000 was investigated. Reaction temperature on change of mixing time was observed, and then proper crosslinking systems were found. Adhesion (properties) of silicone sealants were measured. In the sealants preparation, stable reaction was achieved by adjusting composition variance ratio in the sealant mixture temperature below $40^{\circ}C$. The adhesion properties of sealant differ from substrate composition. The order of adhesion strength was glass/glass > glass/aluminum > aluminum/aluminum system. The elongation of sealant was increased as polymer viscosity and plasticizer content increased. The strength was increased as crosslinker and plasticizer decreased, while catalyst increased.

A Basic Study on the Generation of Tire & Road Wear Particles by Differences in Tire Wear Performance (타이어 마모성능 차이에 의한 타이어 마모입자 생성에 관한 기초 연구)

  • In this study, in order to observe the change in the amount of Tire and Road Wear Particles and the ratio of tire components in it according to the tire wear resistance performance, carried out the evaluation by varying the vulcanization reaction design of the tire tread rubber. In addition, in order to improve the reliability of the evaluation of Tire and Road Wear Particles, the evaluation was performed indoor laboratory test equipment that simulates the condition on real driving to exclude various environmental influences including minerals, driver's habits, road surface, weather, tire structure and pattern designs. After the evaluation in closed space, it is estimated that the amount of collected Tire and Road Wear Particles is 84% compared to 100% of the tire and road wear loss weight, of which 96.4~97.7% was around the road and 2.3~3.6% was in the air. As a result of analy sis of the collected Tire and Road Wear particles, the tire component existed 63~75% in the Tire and Road Wear Particles depending on the wear resistance performance of the tire.

Biodiesel production using lipase producing bacteria isolated from button mushroom bed (양송이 배지에서 유래한 Lipase 생산균을 이용한 바이오디젤 생산)

  • A lipase producing bacterium was isolated from button mushroom bed, which showing high clear zone on agar media containing Tributyrin as the substrate. The strain was identified as Burkholderia cepacia by analysis of 16S rDNA gene sequence. Crude lipase (CL) was partially purified from 70% ammonium sulfate precipitation using the culture filtrate of B. cepacia. Immobilized lipases were prepared by cross-linking method with CL from B. cepacia and Novozyme lipase (NL) onto silanized Silica-gel as support. Residual activitiy of the immobilized CL (ICL) and immobilized NL (INL) was maintained upto 61% and 72%, respectively. Biodiesel (Fatty acid methyl ester, FAME) was recovered by transesterification and methanolysis of Canola oil using NaOH, CL and ICL as the catalysts to compare the composition of fatty acids and the yield of FAME. Total FAME content was NaOH $781mg\;L^{-1}$, CL $681mg\;L^{-1}$ and ICL $596mg\;L^{-1}$, in which the highest levels of FAME was observed to 50% oleic acid (C18:1) and 22% stearic acid (C18:0). In addition, the unsaturated FAME (C18:1, C18:2) decreased, while saturated FAME (C16:0, C18:0) increased according to increasing the reaction times with both CL and ICL, supporting CL possess both transesterification and interesterification activity. When reusability of ICL and INL was estimated by using the continuous reaction of 4 cycles, the activity of ICL and INL was respectively maintained 66% and 79% until the fourth reaction.

Effects of Heat Treatment on Protein Quality as Lysine Damage (열처리가 Lysine 손상에 의한 단백질의 품질에 미치는 영향)

  • During the industrial preparation and the storage of foods, the side chain of some protein-bound amino acids can react chemically each other or with other molecules present in the food. The following reactions have been described : destruction of amino acids, racemization, protein-protein interactions, reactions of proteins with reducing sugars, oxidizing agents, or polyphenols. Apart from total destruction, the main reacitons are the forming of Maillard reactions products(e.g. fructoselysine) and the crosslinking with other amino acids in the same or in another protein molecule(e.g. lysinoalanine). The most often involved amino acid is lysine because of its free functional ${\varepsilon}-amino$ acid group. Generally derivatives of amino acids or crosslinks in polypeptides influence the bioavailability and the overall digestibility of the protein. This work reviews the technological, analytical, nutritional, and physiological problems related to the formation of fructoselysine and lysinolalnine in human foods, and evaluates the possible health risk for humans. A summary of the available information is of help in considering whether or not the presence of fructoselysine/lysinoalanine in foods represents a danger to man. The reduction in protein quality through these reactions is not a problem for the general population, but it is extremely important in infant foods, since infants are often nourished with a limited number of food product(e.g. formular foods) which are sensitive to the Mailard reaction.

Changes in the Dyeing Property of Tencel Blended Fabrics as Susceptible Functional Fiber through Chitosan Finishing (키토산 가공에 의한 감성기능소재로서의 텐셀 혼방직물의 염색성 변화)

  • Cationization is effective to complement the defects of Tencel blended fabrics by introducing new functions. For this purpose, we used chitosan, which is congenial to the human body, free of pollution, and easily reacted. Then, we compared it to the Tencel single fabrics. To perform such effective cationization, the fabrics were treated with chitosan after NaOH pretreatment and enzyme treatment thereof. After that, the fabrics were treated with a crosslinking agent and a softner. The dyeing property of the cationized Tencel blended fabrics and reactive dye, which is a type of anionic dye, show a high concentration in neutral salt and excellent repulsive power between the fabrics and the decreased dyes. The dyeing property of the chitosan treated fabrics represented better performances than that of untreated fabric in the lower concentration of neutral salt. Meanwhile, when it was dyed with certain acid dyes, the dyeing property of the chitosan treated fabrics showed better results due to the reaction of an amine group, which was introduced by chitosan treatment. Thus, the verification of the cationization of the Tencel blended fabrics was performed. The washing fastness of the Tencel blended fabrics showed a little bit better than that of the Tencel single fabric, and it represented a better performance in the dye with a reactive dye than that of an acid dye.
