STUDIES ON BIOCHEMICAL POLYMORPHISM OF MILK PROTEIN AS GENETIC MARKERS IN PIGS

  • Biochemical polymorphisms of sow's milk proteins, $\beta$-casein ($\beta$-CN), $\beta$-lactoglobulin ($\beta$-LG), post-lactoglobulin (post-LG), $\alpha$-lactalbumin ($\alpha$-LA) and X-protein, as genetic markers for major pig breeds (Landrace, Yorkshire, Duroc, Hampshire and cross bred) in Korea were determined by starch gel electrophoresis. Phenotype and gene frequencies at all marker loci were estimated and genetic differences among breed populations were analyzed. Three $\beta$-CN phenotypes (AA, AB and BB) controlled by two codominant alleles (${\beta}-CN^A$ and ${\beta}-CN^B$), four $\beta$-LG phenotypes (AA, AC, $AC^{\pm}$ and CC) controlled by two codominant alleles (${\beta}-LG^A$ and ${\beta}-LG^C$) and ten X-protein phenotypes (AA, BB, CC, DD, AB, AC, AD, BC, BD and CD) controlled by four codominant alleles ($X^A,\;X^B,\;X^C\;and\;X^D$) were identified. In addition, a genetically controlled polymorphism of post-LG was found for the first time in sow's milk protein. Three different phenotypes (AA, AB and BB) were designated $post-LG^A$ and $post-LG^B$. Of the five marker loci examined, $\alpha$-LA locus was observed to lack any individual variation in all breeds studied. All populations were in Hardy-Weinberg equilibrium for all loci. There were marked breed differences for phenotype and gene frequencies in the post-LG and X-protein marker loci. However, there were little differences between breeds in the gene frequencies at the $\beta$-CN and $\beta$-LG marker loci.

Effects of Fermented Soy Protein on Growth Performance and Blood Protein Contents in Nursery Pigs

  • Fifty-four cross-bred ((Landrace${\times}$Yorkshire)${\times}$Duroc) pigs (13.47${\pm}$0.03 kg average initial BW) were evaluated in a 42 d growth assay to determine the effects of the fermented soy product (FSP). The dietary treatments were: FSP 0 (corn-soybean basal diet), FSP 2.5 (FSP 0 amended with 2.5% FSP), and FSP 5 (FSP 0 amended with 5% FSP). The body weight at the end of the experiment increased linearly (p = 0.05) as the FSP levels in the diets increased. In addition, the ADG and G/F ratio also increased (linear effect, p = 0.06) as the levels of FSP increased. However, there was no effect of FSP on ADFI or DM digestibility (p>0.05). Furthermore, the N digestibility increased as the FSP levels increased (linear effect, p = 0.003), although the total protein concentration in the blood was not affected by FSP (p>0.05). Additionally, the albumin concentration was higher in pigs fed diets that contained 2.5% FSP than in pigs in the control group or the FSP 5 group (quadratic effect, p = 0.07). The creatinine concentrations were also evaluated at d 42 and found to be greater in pigs that received the FSP 2.5 diet (quadratic effect, p = 0.09). Moreover, the creatinine concentration increased linearly in response to FSP treatment (p = 0.09). Finally, although the BUN concentration on the final day of the experiment was greater in pigs that received the FSP 2.5 diet (quadratic effect, p = 0.10), there were no incremental differences in BUN concentrations among groups (p>0.05). Taken together, the results of this study indicate that feeding FSP to pigs during the late nursery phase improves growth performance and N digestibility.

Effect of Arsenic on Immunity, Oxidative Enzyme and Various Hematological Parameters in Cross Bred Calves

  • An experiment was conducted on crossbred male calves to study the effect of arsenic (As) on immunity status and certain hematological parameters. Ten crossbred male calves of 3-4 months of age were distributed into two equal groups. Group I was kept as control, whereas, group II was supplemented daily with 50 ppm As (as $As_sO_3$) up to 90 days, in the diet. Calves of both groups were fed as per ICAR standards and their requirements were fulfilled by feeding concentrate mixture and green oats. All calves were kept under similar managemental conditions. Blood samples were collected at fortnightly intervals to estimate various haematological parameters and superoxide dismutase (SOD) enzyme activity. Serum Ig and serum glutamic pyruvate transaminase (SGPT) were also measured. Cell-mediated immune responses of the calves were monitored at 0, 45 and 90 of experimental feeding, through lymphocyte proliferation. No change in blood total leukocyte counts (TLC), differential leukocyte counts (DLC), packed cell volume (PCV), haemoglobin (Hb) and SGPT was observed with As supplementation. A decrease in SOD activity was noticed in group II calves. Stimulation index (SI) for lymphocyte proliferation decreased from 1.14 to 0.79 in group II calves during 90 days experimental feeding, whereas, there was no change in SI values in group I indicating significant decrease in immune response of As supplemented calves. Blood As concentration increased in group II calves with the decrease in immune response. Short term supplementation of As to growing calves suggested suppressive effects on cell-mediated immunity. However, long term experiments are required to demonstrate clearly the efects of this toxic metal in calves.

Response of Feeding Amino Acids Supplemented Cottonseed Meal on Growth Performance and Digestibility of Early Weaned Cow Calves

  • This study was designed to examine the response of feeding early weaning diets without and with amino acids supplemented cottonseed meal on growth performance and digestibility of early weaned cow calves. Fifteen 14-21 days old cross bred cow calves were randomly allotted to three experimental diets. Diet A comprised of milk replacer and concentrate feed whereas two isocaloric and iso-nitrogenous early weaning diets viz., B and C were prepared without and with lysine and methionine supplemented cottonseed meal, respectively. These early weaning diets were used as substitute of milk replacer for calves. Calves were placed in individual cages and fed twice daily for a period of 60 days. Daily feed intake, feed refused and weekly weight gain was recorded. Two digestibility trials I and II were performed at 5th and 9th week of the experiment, respectively. During the digestibility trial I, calves were fed on ad libitum basis whereas in trial II, calves were fed at 90% of their voluntary DMI. Feed, orts and feces samples were collected, weighed daily, composited, subsampled and analysed for DM and CP. Results indicated that weight gain of calves was (p<0.05) higher on diets A (0.63 kg/d/calf) and C (0.64 kg/d/calf) compared to calves on diet B (0.57 kg/d/calf). Significantly (p<0.05) less daily DMI was observed on diet A (1.48 kg) compared to early weaning diets B (1.70 kg) and C (1.72 kg). The feed efficiency was (p<0.05) better on diet A (2.33) compared to diets B (2.95) and C (2.65). The economic efficiency was noticed to be better on diets B and C compared to diet A. In trial I, digestibility of DM and CP of diet A was (p<0.05) higher than diets B and C. Whereas in trial II, digestibility of DM and CP of diet A was (p<0.05) less than diets B and C. It was concluded that early weaning diet based on lysine and methionine supplemented cottonseed meal produced better weight gain and feed efficiency compared to non-supplemented cottonseed meal based diet.

Effects of Replacing Corn with Chinese Brown Rice on Growth Performance and Apparent Fecal Digestibility of Nutrients in Weanling Pigs

  • This experiment was conducted to study the effects of replacing corn with Chinese brown rice on performance and apparent fecal digestibility of nutrients in weanling pigs. Seventy-two cross-bred (Duroc${\times}$Landrace${\times}$Large White) weanling pigs, weighing an average of $7.38{\pm}0.06kg$, were randomly assigned to either a corn-soybean meal diet (treatment 1), a corn-brown ricesoybean meal diet (treatment 2) and a brown rice-soybean meal diet (treatment 3). The brown rice used in this experiment was husked from one kind of early, long grain, non-glutinous rice (ELGNR, indica rice). The growth performance of weanling pigs in the experiment was similar during weeks 1 and 2, but feed conversion ratio (FCR) was significantly better in treatment 2 during weeks 3 and 4. During week 2, only the digestibility of organic matter in treatments 2 and 3 was significantly higher than that in treatment 1. During week 4, the digestibility of gross energy, ether extract, organic matter and dry matter in brown rice treatment, especially in treatment 3, were significantly higher than in treatment 1. At the end of the experiment, the concentration of serum urea nitrogen of pigs in treatment 2 was lower than that in treatment 1 and 3 and no difference was found in serum glucose and phosphorus. It could be concluded from this experiment that replacing 50% of corn with brown rice in the corn-soybean meal diet caused no negative effects to weanling pig performance, and apparent fecal digestibility was unaffected. It is feasible to replace corn with brown rice in weanling pig diets in southern China.

  • The purpose of this study was to investigate the effect of manganese (Mn) supplementation on bone status and calcium balance in ovariectomized rats according to the calcium intake levels. Total of 50 Sprague Dawley female rats (6 weeks) were divided into 5 groups and bred for 12 weeks: sham operated control group (SACa), OVX Ca deficiency group (OLCa) with Ca deficiency diet (0.1% Ca modified AIN-93N diet), OVX Ca deficiency & Mn supplement group (OLCaMn), OVX adequate Ca group (OACa; 0.5% Ca AIN-93N diet) and OVX adequate Ca & Mn supplement group (OACaMn). BMD (bone mineral density) of the femur was increased by Mn supplementation in OVX adequate Ca group. However, BMDs of spine, femur and tibia were lowered in OLCa compared to the OLCaMn group. Bone strength of tibia in OLCaMn group was significantly lower than OLCa group. Serum ALP (alkaline phosphatase) and CTx (C-telopeptide of collagen cross-links) levels were significantly higher in ovariectomized rats than those in the sham group, but they were not changed by Mn supplementation. Ca retention rate and Ca absorption rate did not differ among the experimental groups. Urinary Ca excretion was increased by Mn supplementation in Ca deficiency rats. In summary, Mn supplementation resulted in positive effects on bone mineral density ovariectomized rats with which intake adequate Ca. However, Mn supplementation on Ca deficiency ovariectomized rats resulted in decrement of BMO and bone strength by increasing Ca excretion. Therefore, it is encouraged to consider calcium intake levels in supplementation of manganese in order to prevent postmenopausal osteoporosis and to keep bone healthy. 백·분홍색 절화장미 '수려' 육성 (A New Rose Cultivar 'Suryeo' with White-pink Flower Color)

  • 충북농업기술원에서 육성한 장미 '수려'는 절화용 장미로 황적색 스탠다드 계통의 장미 품종인 'Konfetti' 품종을 모친으로, 분홍색의 화색을 가진 스탠다드 계통의 'Laser' 품종을 화분친으로 하여 2003년에 인공 교배한 후 계통선발 및 3차 특성 검정을 거쳐 2007년에 최종 선발하여 '수려(Suryeo)'로 명명하였으며 2009년에 품종보호권이 등록되었다. 수려는 백색과 분홍색(Red-purple Group N57B)이 혼합된 스탠다드형 장미로 연간 절화수량은 149(본/$m^2$/년)이고 꽃잎수는 64매, 개화소요일수는 48일, 절화수명은 12일 정도이다. 절화장은 78cm이며 꽃무게는 50g이었다. 특히 수세가 강건하며 흰가루병에 강한 특성이 있다.

Inclusion of Dietary Palm Oil and Soy Oil on Growth Performance and Nutrition Digestibility in Broiler Chickens

  • The effects of palm oil and soy oil supplementation in broiler chickens on growth performance and nutrient digestibility of dry matter, nitrogen, and energy was evaluated. In this experiment, 702 one-day-old Ross 308 cross bred chicks with an initial body weight of 42.23±0.05 g (mean±SD) were utilized in a completely randomized treatment design (13 replicates/treatment, 18 chickens/cage) and fed for 35 days with CON (basal diet), T1 (Basal diet + 1.3 % palm oil), and T2 (Basal diet + 1.3% soy oil). From days 1 to 7, the body weight gain (BWG), feed intake (FI), and feed conversion ratio (FCR) were not significantly affected by the supplementation with palm oil or soy oil. However, from days 7 to 21, BWG showed a tendency to increase compared to that with the control treatment. Moreover, BWG was significantly improved (P<0.05) with palm and soy oil supplementation during the overall experiment in comparison to that with the control diet. In addition, the palm oil-supplemented group had higher BWG than the soy oil treatment group (P<0.05). During the overall experiment, FI tended to increase in the T1 group compared to that in the other groups. There was no significant effect on FCR and nutrient digestibility of dry matter, nitrogen, and energy during the overall trial with the supplementation of palm or soy oil. In summary, the inclusion of dietary palm and soy oil has a beneficial effect on the growth performance of broiler chickens.

돼지에서 Medetomidine-ketamine 마취에 대한 Atipamezole과 Yohimbine의 길항효과 (Antagonistic Effects of Atipamezole and Yohimbine against Anesthesia with Medetomidine and Ketamine Combination in Pigs)

  • 돼지에서 medetomidine-ketamine (MK) 합제에 대한 마취효과와 이 합제에 대한 atipamezole (MKA) 과 yohimbine (MKY)의 길항효과를 비교하였다. 24 마리 Landrace - Yorkshire 혼혈 종 돼지를 사용하였다. Medetomidineketamine 는 한 주사기로 근육주사 하였고 atipamezole 과 yohimbine 은 마취 후 20 분에 정맥 주사 하였다. 평균마취시간, 평균흉와시간, 평균기립시간 및 평균보행시간은 MKA와 MKY군에서 MK군보다 유의적으로 짧았다. 그러나 MKA군과 MKY군간의 유의적인 차이는 없었다. 평균혈압은 MKA와 MKY군에서 MK군보다 유의적으로 낮았다. 결론적으로 Medetomidine-ketamine 에 의한 마취 및 혈압 상승 효과는 atipamezole과 yohimbine 에 의해 안전하고 빠르게 길항되었다. 따라서 atipamezole과 yohimbine은 돼지에서 Medetomidine-ketamine 마취를 길항하는 데 안전하고 효과적으로 사용될 수 있다.

Seroprevalence of brucellosis in cattle in selected area of Bangladesh and comparison between Rose Bengal test and i-ELISA used for the screening of brucellosis

  • Brucellosis, a bacterial zoonoses caused by the genus Brucella is responsible for abortion and infertility in cow. Brucellosis is causing economic loss in dairy industries and prevalent worldwide including Bangladesh but limited studies are devoted to determine the prevalence and its association with reproductive factors of dairy cows in Bangladesh. Therefore, the present study was conducted to determine the seroprevalence of brucellosis in dairy cattle using screening test Rose Bengal test (RBT) and the positive sera were further confirmed by indirect- ELISA. For this purpose, a total of 400 serum samples from dairy cows with history of abortion and various reproductive disorders were collected from the Kurigram district of Bangladesh for the detection of Brucella antibody. The overall prevalence of brucellosis in dairy cattle was 2.25%. Brucellosis in cases of abortion and repeat breeding was 8.3% and 2.8%, respectively. The results shows higher prevalence of brucellosis in cases of abortion followed by repeat breeding, while there was no seropositive cases from other reproductive disorders. Age-wise sero-prevalence was found 3.0% in 2~3 years age group and 2.0% in 4~8 years age group. The prevalence of brucellosis in indigenous and cross-bred cattle was 3.6% and 1.7%, respectively. All the animals detected positive to brucellosis by RBT were not found to be positive by i-ELISA. However, the RBT might be a suitable screening test for the diagnosis of Brucella infection in field condition in Bangladesh. These data will help to develop effective disease prevention strategies.
