Effects of Feeding the Whole Crop Rice Silage on the Milking Performance of Holstein and Profitability (총체벼사일리지 급여가 Holstein 착유우의 산유성적과 수익성에 미치는 영향)

  • This study was conducted to evaluate the effect of feeding the whole crop rice silage (WCRS) which partially replaces the imported grass hay crop on the milking performance of Holstein and profitability. Two experimental diets were used according to the traditional practice of dairy farms. The control diet (C) was composed of 13kg/day of mixed hay, 6.8kg/day concentrate, and orchardgrass/bermudagrass hay (1.8kg/day). The treatment diet (T) was composed of 9.6kg/day of mixed hay, 6.8kg/day of concentrate and 2.2kg/day of WCRS. The CP content of the WCRS (4.6%) was lower than orchardgrass (11.3%) and bermudagrass hay (8.4%) due to the harvest being 30days later than the optimum harvest date of rice. The lower intake was observed in T during the entire experiment period which seems to be related to the higher proportion of fiber in rice (p<0.05). No significant difference was found in milk production between C (26.9kg) and T (26.3kg) during the entire experiment period (p>0.05). Also, no difference in milk fat, milk protein, total solids, and milk composition was observed between the two diets. Higher income per head was resulted in T (21,141won) compared to C (21,915 won). These results indicate that the partial replacement of the grass hay by the WCRS can bring the profit to the dairy farm due to the no difference between the control diet and the treatment diet.

Isoflavone Composition and Estrogenic Activity of Germinated Soybeans (Glycine max) according to Variety (품종별 발아 콩(Glycine max)의 아이소플라본 조성 및 In Vitro 에스트로겐 유사활성)

  • This study was performed to investigate the changes in isoflavone composition, estrogenic activity and antiestrogenic activity of soybean cultivars of different isoflavone content (Aga 8, Uram, Cheongja 3, and Dawon) with germination. Total isoflavone contents of Aga 8, Dawon and Cheongja 3 was increased from 2,671.74, 261.08 and 2,240.08 to 2,977.50, 966.13 and $2,354.11{\mu}g/g$, respectively after germination except for Uram cultivars, and highest contents of total isoflavone showed $2,977.50{\mu}g/g$ and $2,354.11{\mu}g/g$, respectively in Aga 8 and Cheongja 3 after germination. MTT cell proliferation assay using MCF-7 cells revealed that germinated soybean of Aga 8 and Cheonja 3 obtained not only contained a high content of isoflavone but also had estrogenic activity. Estrogenic activity of Aga 8 and Cheongja 3 soybean extracts increased from 116.21% and 101.60% to 135.34% and 121.05% after germination. These results suggest that germinated soybean of Aga 8 and Cheongja 3 might have a potential preventive effect on estrogen-deficient diseases.

Changes of Seed Germination Rate and Lipid Components in Different Brown Rices during Ageing (저장에 따른 생태형별 현미의 발아율과 지질 성분의 변화)

  • The objective of this study was to determine quality changes in seed germination rates and lipid components in Japonica and Tongil (indica/japonica) type brown rices with different tocol composition during storage. Moisture content, seed germination rate, crude lipid content, fat acidity, tocol content, free fatty acid composition of brown rice and pH of milled rice were measured to evaluate their quality after storage. Seed germination rates of stored Japonica and Tongil type brown rices were decreased by 22.4% and 59.7%, respectively, after 8 weeks of storage. The fat acidity of Japonica rice was significantly (p<0.05) higher than that of Tongil type brown rices after storage. The major tocol homologue of Tongil type was ${\gamma}-tocotrienol$, whereas, major tocol homologues of Japonica brown rices were ${\alpha}-tocopherol$ and ${\alpha}-tocotrienol$ throughout the entire storage period.

Species Composition and Seasonal Variations of Fishes in Eelgrass (Zostera marina) Bed in Kwangyang Bay (광양만 잘피밭에 서식하는 어류의 종조성 및 계절 변동)

  • A total of 57 species of fish species was collected by a trawl from the eelgrass bed in Kwangyang Bay. The dominant species were Pholis nebulosa, Syngnathus schlegeli, Leiognathus nuchalis, Pseudoblennius cottoides, Sebastes inermis, Favonigobius gymnauchen, which accounted for 69.9% of the total numbers of fish collected. Fish collected in the study area were primarily small fish species or early juveniles of large fish species. Only about 10% of fishes which were collected in the eelgrass bed exceeded 15cm in standard length. Seasonal variations in both species composition and abundance were major characteristics in the study area. The peak abundance occurred in spring, while the number of species was the highest in fall. However, both the number of species and abundance of fishes showed the lowest values in winter. High species diversity indices were observed in fall. Temperature, eelgrass standing crop and abundance of food organisms influenced seasonal changes of the fish community in the study area. More abundant and more diverse fishes were collected during nighttime than daytime.

Molecular Modification of Perilla Lipid Composition

  • In order to modify lipid production of Perilla qualitatively as well as quantitatively by genetic engineering, genes involved in carbon metabolism were isolated and characterized. These include acyl-ACP thioesterases from Perilla frutescens and Iris sp., four different $\beta$-ketoacyl- ACP synthases from Perilla frutescens, and two $\Delta$15 a-cyl-ACP desaturases(Pffad7, pffad3). Δ15 acyl-ACP desa turase (Δ15-DES) is responsible for the conversion of linoleic acid (18:2) to $\alpha$-linolenic acid (ALA, 18:3). pffad 3 encodes Δ15 acyl-desaturase which is localized in ER membrane. On the other hand, Pffad7 encodes a 50 kD plastid protein (438 residues), which showed highest sequence similarity to Sesamum indicum fad7 protein. Northern blot analysis revealed that the Pffad7 is highly expressed in leaves but not in roots and seeds. And Pffad3 is expressed throughout the seed developmental stage except very early and fully mature stage. We constructed Pffad7 gene under 355 promoter and Pffad3 gene under seed specific vicillin promoter. Using Pffad7 construct, Perilla, an oil seed crop in Korea, was transformed by Agrobacterium leaf disc method. $\alpha$-linolenic acid contents increased in leaves but decreased in seeds of transgenic Perilla. Currently, we are transforming Perilla with Pffad3 construct to change Perilla seed oil composition. We isolated three ADP-glucose pyrophosphorylase (AGP) genes from Perilla immature seed specific cDNA library. Nucleotide sequence analysis showed that two of three AGP (Psagpl, Psagp2) genes encode AGP small subunit polypeptides and the remaining (Plagp) encodes an AGP large subunit. PSAGPs, AGP small subunit peptide, form active heterotetramers with potato AGP large subunit in E. coli expressing plant AGP genes.

Growth, Flowering, and Nutrient Composition of Salvia Grown in Peat moss Media Containing Pellets Processed with Poultry Feather Fibers at Different Mixing Ratios

  • The objective of this study was to determine the effect of replacing perlite (PL) with pellets processed with poultry feather fiber as an inert material to prepare growing medium. The growth and flowering of Salvia splendens 'Vista Red' grown in individual growing medium $Biosangto^{(R)}$, peat moss (PM), PL, and two pellets (P45-1 and P45-2) were evaluated. Peat moss was mixed with PL, P45-1, or P45-2 at various ratios (1:0 to 1:3 or 3:1 by volume) to investigate the feasibility of replacing PL with pellets. Nutrient composition of the growing medium and leaf tissues was analyzed. The number of florets, inflorescence length, plant height, and fresh weight of plants grown in media containing P45-1 or P45-2 were reduced compared to those grown in individual growing medium PM or PL. As the mixing ratio of P45-1 or P45-2 to PM was higher, the growth of salvia, such as inflorescence length, plant height, number of leaves, and fresh weight was inhibited. Our results indicate that mixing three parts PM with one part of P45-1 (PM/P45-1/3:1) or P45-2 (PM/P45-2/3:1) accelerated flowering and increased the number of florets and leaves compared to other mixing ratios of PM and pellets media. The concentrations of phosphorus (P), calcium (Ca), boron (B), iron (Fe), and copper (Cu) in individual growing medium PL, P45-1, and P45-2 were significantly lower than those in PM. The concentration of N was the highest in leaves of plants grown in P45-1 or P45-2 amended media, and the concentrations of P, Ca, and zinc (Zn) in leaves were lower in individual growing medium P45-1 or P45-2 than in PM and PL. The pH of PM/P45-1/3:1 or PM/P45-2/3:1 media was maintained at optimal level (5.8-5.9) and the concentrations of macro- and micro-elements in the media and leaves were considered to be optimal levels. Therefore, mixing three parts PM with one part P45-1(PM:P45-1/3:1) or P45-2 (PM:P45-2/3:1) is recommended for improved growth and flowering in salvia. This suggests that P45-1 or P45-2 can replace PL as an inert material to prepare growing medium.

Chemical Composition of Barley Leaves from Different Varieties (품종별 보리잎의 이화학적 특성)

  • Chemical composition of barley leaves of four varieties (Olbori, Suwon 298, Suwon 311 and Milyang 60) grown under the same environmental conditions at the same location was investigated. Barley leaf samples were collected on two weeks before heading period. The barley leaves contained $25{\sim}29%$ crude protein, $9.3{\sim}9.9%$ crude lipid, $9.3{\sim}11.2%$ ash, $1.3{\sim}1.8%$ ${\beta}$-glucan on dry matter basis. Chlorophyll contents of Olbori, Suwon 298, Suwon 311 and Milyang 60 were 951 mg%, 885 mg%, 866 mg%, 826 mg%, respectively. Mineral contents of barley leaves were not significantly different each other and potassium content was found to be the highest among all the minerals observed. Vitamin C contents of Olbori, Suwon 298, Suwon 311 and Milyang 60 were 328 mg%, 266 mg%, 278 mg% and 269 mg%, respectively. Free sugar contents of barley leaves were somewhat different and the glucose content was the highest among the free sugars, ranging from 5.70% to 8.35%. Besides glucose, fructose, sucrose and raffinose contents were also relatively higher than the other free sugars. In other oligosaccharides, 1-kestose($GF_2$) content was between 0.26% and 0.39% and a slight amount of 1-nystose($GF_3$) was also detected. Although aspartic acid, glutamic acid and serine were relatively higher content than the other free amino acids, the values were not consistent in different varieties.

Spatio-temporal Distribution of Phytoplankton Community in the Jangsu Bay and Adjoining Sea of South Sea, Korea (장수만 식물플랑크톤 군집의 시.공간적 분포 특성)

  • This study describes about the spatio-temporal distributions in phytoplankton community such as species composition, standing crop and dominant species from May 2006 to February 2007 in the Jangsu bay and the northwestern parts of Gamak bay. Based on the principal component analysis (PCA) of the environmental factors as well as biological parameters, the bio-oceanographical characteristics were analysed. A total of 83 species of phytoplankton belonging to 47 genera were identified. Whereas diatoms and dinoflagellates occupied more than 65% and 30% of total species, respectively. The annual dominant species were Chaetoceros affinis, Paralia sulcata and Bacillaria paxillifera in spring, Chaetoceros didymus, Ch. affinis and Octactis octonaria in summer, Skeletonema costatum-like species and B. paxillifera in autumn. Moreover phytoplankton cell density was ranged between 3.1 $cells{\cdot}mL^{-1}$ in spring and 521.0 $cells{\cdot}mL^{-1}$ in winter. It fluctuated with an annual mean of 76.0 $cells{\cdot}mL^{-1}$ between the lowest value of 7.6 $cells{\cdot}mL^{-1}$ in spring and the highest value of 220.2 $cells{\cdot}mL^{-1}$ by Skeletonema costatum-like species in winter. Briefly, the phytoplankton cell density in the mixing seasons was higher in comparison with the other seasons. According to the PCA, the biological oceanographic characteristics of the Jangsu bay was affected by the introduction of outside seawater particularly in temperature increasing seasons, and the other seasons, it may be described the light intensity, and mix between inner and outer bay sea waters.

Characteristics of Ionic Composition of Rainwater in Suwon (수원지역 빗물의 이온 조성)

  • The issue of acid precipitation and related environmental problems in East Asia have been emerging. To evaluate the acidity and chemical characteristics of rainwater in Korea, its chemical properties during crop cultivation season from April to October were investigated at Suwon, Korea. Also, to estimate the contribution of ions on its acidity, ion composition characteristics and neutralization effects by cation ions were determined. Ion balance and electrical conductivity balance between the measured and estimated values showed high correlation. Rainwater had distributed highly in the range of pH 4.5~5.6. The pH of rainwater was relatively high at June as compared with other monitoring periods. $Na^+$ was the main cation followed by $NH_4{^+}$, $Ca^{2+}$, $H^+$ > $K^+$ > $Mg^{2+}$. Among these, $Na^+$, $NH_4{^+}$, $Ca^{2+}$ and $H^+$ covered over 93% of total cations. About 86% of anion in rainwater was composed of $SO{_4}^{2-}$ and $NO_3{^-}$. In rainwater samples, $NH_4{^+}$ and $Ca^{2+}$ contributed greatly to neutralization of the rain acidity. Also, 88% of soluble sulfate in rainwater was nss-$SO{_4}^{2-}$(non-sea salt sulfate).

Characteristice Study of Ancient Northeast Asian Lead Glass and Green Glaze Based on Analysis Results (분석자료를 기초한 고대 납유리와 녹유의 특성 연구)

  • This study examines the results of analyses of the lead isotope ratio and chemical composition of lead glass and green glaze from ancient Northeast Asia in order to suggest their production sites and reveal further characteristics. The comparison of the lead isotope ratio of lead glass and green glaze from two Baekje remains in Iksan-the Wanggung-ri Site and Mireuksa Temple Site-suggests that they were produced to the west of the South Gyeonggi Massif (Zone 4) using lead extracted from the same area. With a few exceptions, it has proved difficult to identify the production sites of most of the green-glazed roof tiles from Unified Silla-period Buddhist temples across Northeast Asia. The major component of the lead glass from Baekje, Silla, China, and Japan during the seventh century is PbO, SiO2, Al2O3, CuO, and Fe2O3, with a ratio of PbO and SiO2 of 70 and 30 wt.%, respectively. The green-glazed roof tiles excavated from a temple from the Unified Silla period have a high proportion of lead, ranging from 64 to 90 wt.%. Green-glazed lozenge tiles excavated from the Sacheonwangsa Temple site in Gyeongju were shown to contain PbO, SiO2, Al2O3, and CuO, a similar composition with lead glass. An experiment was conducted to reproduce a glaze according to the production method mentioned in the Zō hotokesho sakumotsu-chō (Buddhist statue workshop crop book) in the Shosoin Repository. In this experiment, an identical ratio of PbO was observed for Japanese green-glazed ceramics from the eighth to eleventh century as that found in Chinese lead-glazed ceramics excavated from kilns operated from the seventh to tenth century in Henan. This indicates that production methods for lead glass and glaze were shared across Northeast Asia.
