CDDO-Me alleviates oxidative stress in human mesenchymal stem cells

  • Mesenchymal stem cells (MSCs) have been recognized as a therapeutic tool for various diseases due to its unique ability for tissue regeneration and immune regulation. However, poor survival during in vitro expansion and after being administrated in vivo limits its clinical uses. Accordingly, protocols for enhancing cell survivability is critical for establishing an efficient cell therapy is needed. CDDO-Me is a synthetic C-28 methyl ester of 2-cyano-3,12-dioxoolean-1,9-dien-28-oic acid, which is known to stimulate nuclear factor erythroid 2-related factor 2 (Nrf2)-antioxidant response element (ARE) pathway. Herein, report that CDDO-Me promoted the proliferation of MSCs and increased colony forming units (CFU) numbers. No alteration in differentiation into tri-lineage mesodermal cells was found after CDDO-Me treatment. We observed that CDDO-Me treatment reduced the cell death induced by oxidative stress, demonstrated by the augment in the expression of Nrf2-downstream genes. Lastly, CDDO-Me led to the nuclear translocation of NRF2. Our data indicate that CDDO-Me can enhance the functionality of MSCs by stimulating cell survival and increasing viability under oxidative stress.

  • This study intend to analysis the main issue of deterioration of old housing complex for reproduction and derive the solution. Most of existing reproduction of hosing complex pushed to improve residential environment or expected rise real estate in value. For that reason, there was a limit to improve social problem of urban area behind hosing complex and surrounding area. Because government propel to make brand of city, have parking facilities and rise floor space index rather than established reproduction plan based on the aging problem of housing complex. Therefore, city need to deduct major reproduction issue to improve aging problem and prepare effective counterplan in complex and surround area by reproduction. This study progress through three steps of research. First, arrange problem of aging complex and way of response sustainably. Second, understand the importance by group toward improvement and aging problem that appear aging apartment complex through survey and opinion of expert. Third, comprehend major reproduction issue over this.

Effect of the Mechanical Properties of Cell-Interactive Hydrogels on a Control of Cell Phenotype (세포친화적 하이드로젤의 기계적 물성이 세포 표현형 제어에 미치는 영향)

  • A critical element in tissue engineering approaches is a control of the mechanical properties of polymer scaffolds to regulate cell phenotype, which may lead to clinically successful tissue regeneration. In this study, we hypothesized that gel stiffness could be a key factor to manipulate adhesion and proliferation of different types of cells. RGD-modified alginate gels with various mechanical properties were prepared and used as a substrate for MC3T3-E1 and H9C2 cells. Adhesion and growth rate of MC3T3-E1 cells in vitro were increased in parallel with an increase of gel stiffness. In contrast, those of H9C2 cells were decreased. This approach to control the mechanical properties of polymer scaffolds depending on the cell types may find useful applications in the tissue engineering.

Prevalence and risk factors of peri-implantitis: A retrospective study (임플란트 주위염의 유병률 및 위험요소분석에 관한 후향적 연구)

  • Purpose: The study analyzed the prevalence of peri-implantitis and factors which may have affected the disease. Materials and methods: This study based on medical records and radiographs of 422 patients (853 implant cases) who visited Ewha Womans University Mokdong Hospital Dental Center from January 1, 2012 to December 31, 2016. Generalized estimation equations (GEE) was utilized to determine the statistical relationship between peri-implantitis and each element, and the cumulative prevalence of peri-implantitis during the observation period was obtained by using the Kaplan Meier Method. Results: The prevalence rate of peri-implantitis at the patient level resulted in 7.3% (31 patients out of a total of 422 patients), and at the implant level 5.5% (47 implants out of a total of 853 implants). Sex, GBR, guided bone regeneration (GBR) and functional loading periods had statistical significance with the occurrence of peri-implantitis. Upon analysis of the cumulative prevalence of peri-implantitis in terms of implant follow-up period, the first case of peri-implantitis occurred at 9 months after the placement of an implant, and the prevalence of peri-implantitis showed a non-linear rise over time without a hint of a critical point. Conclusion: The prevalence of peri-implantitis at the patient level and the implant were 7.3% and 5.5%, respectively. Male, implant installed with GBR and longer Functional Loading Periods were related with the risk of peri-implantitis.
