반추수의 혈청과 장기조직의 Creatine Phosphokinase(CPK) 총활성 및 CPK Isoenzyme 분획에 관한 연구 (Studies on the Total Creatine Phosphokinase (CPK) Activities and CPK Isoenzymes Fractions of the Sera and Organ Tissues in Ruminant)

  • Total CPK activities and CPK isoenzymes fractions of the sera and tissues were examined to obtain the physiological basic data of ruminant available in veterinary clinical practice. For the sera total CPK activities and CPK isoenzymes fractions, total 39 clinically healthy Korean native goats (3 to 10 months old, IS of female and 18 of male) and 6 of Korean native goats (1 to 2 years old, 3 of female and 3 of male) were used. Seventeen Korean native cattle (3 to 6 years old, 10 of female and 7 of male) and 27 Holstein-Friesian cattle (2 to 8 months old, 7 of female and 3 to 12 years old, 20 of female) were also examined for the sera total CPK activities and CPK isoenzymes fractions. For the total CPK activities and CPK isoenzyme fractions, 3 of female Korean native goats (7 months old), 3 of female Korean native cattle (2 years old) and 3 of dairy cattle (2 years old, 2 of female and 1 of male) were used. The tissues examined were the cerebrum (2 of Korean native cattle), spinal cord (1 of Korean native cattle), heart, lung, diaphragm, reticulum, liver, spleen, kidney, jejunum. colon and femoral muscle. The results obtained were as follows : 1. In Korean native goats less than 1-year-old. serum total CPK activities were 67.8${\pm}$17.7(39.0~96.5) IU/$\ell$ in female and 63.4${\pm}$19.0(28.7~94.4) IU/$\ell$ in male. Further they were 67.0${\pm}$5.3(59.5~70.7) IU/$\ell$ and 54.5${\pm}$11.1(39.1~69.4) IU/$\ell$ in female and male Korean native goats over 1-year-old, respectively. Serum total CPK activities of female were slightly higher than those of male. Significance between age and sex was not found. 2. Serum total CPK activities were 56.8${\pm}$19.7(27.6~90.5) IU/$\ell$ and 65.6${\pm}$10.8(52.8~78.0) IU/$\ell$ in female and male Korean native adult cattle, respectively, Serum total CPK activities of male were slightly higher than those of female, but they were not significant 3. Serum total CPK activities we,e 72.5${\pm}$8.2(57.2~83.2) IU/$\ell$ and 60.8${\pm}$12.5(42.7~80.6) IU/$\ell$ in calves and adult of dairy acttle, respectively. Serum total CPK activities of calves were significantly higher than those of adult(p<0.05). 4. In Korean native goats less than 1-year-old, serum CPK isoenzymes fractions were high with decreasing order of MM>MB>BB and MM>BB>MB in female and male, respectively. Further they were high with decreasing order of MM>MB>BB and MM>B8>MB in female and male Korean native goats over 1-year-old, respectively. The main fractions of CPK isoenzymes were MM in sera of Korean native goats. 5. Serum CPK Isoenzyme fractions were high with decreasing order of MM>MB>BB In both female and male of Korean native cattle. The main fraction among them was MM. 6. Serum CPK isoenzymes fractions were high with decreasing order of MM>BB>MB in both calves and adult of dairy cattle. The main fraction among them was MM. 7. Total CPK activities were high with decreasing order of the femoral muscle>kidney>reticulum>diaphragm>liver>spleen>heart>colon>lung>jejunum in Korean native goats. 8. Total CPK activities were high with decreasing order of the spinal cord >cerebrum>femoral muscle>reticulum>kidney>liver>spleen>diaphragm>lung>colon>heart>jejunum in Korean native cattle. 9. Total CPK activities were high with decreasing order. of the femoral muscle >liver>retoculum>kidney>heart>colon>lung>spleen>jejunum>diaphrasm in dairy cattle. 10. The pattern of the cardiac CPK isoenzymes fractions was identical in Korean native goats, Korean native cattle and dairy cattle. They were high in the order of MM>MB without BB fractions and the main fraction was MM. 11. The pattern of the pulmonary CPK isoenzymes fractions was the same Korean native goats, Korean native cattle and dairy cattle. They were high with decreasing order of MM>MB>BB and the main fraction among them was MM. 12. The pattern of CPK isoenzymes fractions of the diaphragm was Identical in Korean native goats and Korean native cattle. They were high with decreasing order of MM >BB >MB except dairy cattle (MM>MB>BB) but the main fraction among them was MM. 13. The pattern of the reticular CPK isoenzymes fractions was identical in Korean native cattle and dairy cattle. They were high with decreasing order of BB >MM >MB except Korean native goats(BB>MB>MM) but the main, fraction among them was BB 14. The pattern of the hepatic CPK isoenzymrs fractions was identical in Korean native cattle and dairy cattle. They were high with decreasing order of MB >BB >MM except Korean native goats(MB>MM>BB)but the main fraction was MB. 15. The splenic CPK isoenzymes fractions showed different pattern. They were high with decreasing order of MB>BB>MM, MM>BB>MB and BB>MB>MM in Korean native goats, Korean native cattle and dairy cattle, respectively. The main fraction among them was different from each other. 16. The pattern of the renal CPK isoenzymes fractions was identical in Korean native cattle and dairy cattle. They were high with decreasing order of MM >MB>BB except Korean native goats(BB>MB>MM). 17. The CPK isoenzymes fractions of the Jejunums showed different pattern. They were high with decreasing order MM>MB>BB, MM>BB>MB and BB>MM>MB in Korean native goats, Korean native cattle and dairy cattle, respectively. The main fractions were MM In Korean native goats and Korean native cattle, and BB in dairy cattle. 18. The colonic CPK isoenzymes fractions showed different pattern. They were high with decreasing order of MM>MB>BB, MM>BB>MB and BB>rrfB>MM in Korean native goats, Korean native cattle and dairy cattle, respectively. The main fractions were MM in Korean native goats and Korean . native cattle, and BB in dairy cattle. 19. The cerebral CPK isoenzymes fractions were high with decreasing order of BB >MM without MB detected in Korean native cattle and those of spinal cord were high with decreasing order of BB >MM >MB. The main fractions in both cerebrum and spinal cord were BB.

흰쥐의 적출된 심장에서 Verapamil이 허혈성 심근에 미치는 효과 (Effects of Verapamil in Cardioplegic Perfusates on the Ischemic Myocardium in Isolated Rat Heart)

  • 저자들은 흰쥐의 적출된 심장을 이용하여 Verapamil이 허혈성 심근에 대하여 심근보호효과가 있는지를 관찰하였다 흰쥐의 적출된 심장은 허혈상태에서 심근온도가 $25^{\circ}C$가 되게 유지하였다. 24마리의 흰쥐로부터 적출된 심장을 Krebs-Henseleit완충액을 사용하여 30분간의 비작업성 역관류로 안정시킨 후 $25^{\circ}C$의 심정지액 (St. Thomas' Hospital Cardioplegic Solution)에 60분 동안 저장하였다. 허혈성 심정지를 유도하기 전에 적출된 심장을 저온의 심정지액으로 처리한 군을 대조군(n=12)으로 하고, Verapamil 이 첨가된 저온의 심정지액으로 처리한 군을 실험군(n=12)으로 하였다. 60분 동안의 허혈성 심정지후 심정지전에 측정했던 혈역학적 및 생화학적 지표인 심박동수, 좌심실압, +dpfdt max, 관상관류량과 CPK치를 재관류후 30분에 재측정하여 심정지전과 비교하여 심기능 회복 정도를 관찰하였다. Verapamil이 첨가된 저온의 심정지액을 사용한 실험군이 심박동수, 좌심실압, +dp/dt max, 관상관류량과 CPK치에서 대조군에 비하여 유의하게 높은 회복율을 나타내었다(p <0.05).

도계처리 전후의 환경온도가 계육의 생화학적 대사 및 연도에 미치는 영향 (Effect of Antemortem and Postmortem Environmental Temperatures on Biochemical Metabolism and Tenderness in Chicken Muscels)

  • 1. 도살전의 환경온도(38$^{\circ}C$, 2$0^{\circ}C$, 4$^{\circ}C$, -2$0^{\circ}C$)가 닭 가슴고기의 연도와 근육내 해당작용에 미치는 영향을 연구하였다. 고온구는 현저하게(p<0.05) 계육의 연도를 저하시켰고 저온구도 다소 연도의 저하를 초래하였다. 고온구는 대조구보다 근육내 glycogen의 함량과 최초 Ph가 높았고 24시간후의 최종 PH는 현저하게 (P<0.05) 낮았다. 저온구는 대조구와 고온구 사이의 중간수치를 보여주었다. 고기의 절단력(즉 연도)와 근육내 glycogen 함량, 최초 및 최종 근육 산도(PH) 간에는 현저하게 (p<0.01) 높은 상관관계를 나타냈고 사후 해당속도와 가열처리후의 고기의 수화도 역시 유의한(p<0.05) 상관관계를 보여 주었다. 근육 혹은 혈청내의 효소(LDH, CPK)의 활력과 고기 연도와는 아무런 상관관계가 없었다. 2. 도살후 가슴고기와 다리고기를 절취하여 각각 다른 보존온도에 방치하여 근섬유의 단축도를 조사하였던 바 4-1$0^{\circ}C$ 사이에서 가장 낮은 단축도를 보여주었다. $0^{\circ}C$에서 보존시 약간의 단축이 있었으나 현저하지 않았고 보존온도가 2$0^{\circ}C$ 이상일 때 온도상승에 따라 비례적으로 단축도의 급격한 증가를 보여주었다. 따라서 계내에서는 저온보다는 고온에서의 근섬유단축이 더 실제적인 중요성을 가지며 도계후 가급적 속히 15$^{\circ}C$이하로 냉각하므로 심한 연도의 저하를 방지할 수 있다.

寒冷環境에 순화시킨 흰쥐의 血淸酵素 및 同位酵素의 活性 (Serum Enzyme and Isozyme Activities of Rats Acclimated to Cold Environment)

  • 한냉환경에 Sprague-Dawley계 웅성흰쥐를 순화시켜 SLDH, SLDH 동위효소, SALP, SALP 동위효소 및 SCPK 등의 활성치의 보상 변화에 따른 변화를 관찰 검토하였다. 한냉순화는 $4\\pm1^\\circC$(상대습도 70$\\sim$80%)에서 안정상태로 36일간 순화처리 하였으며 SLDH의 활성치는 한냉순화 초기에 유의성 있게 증가하였으나 곧 감소되어 정상치로 접근하며 36일구 까지 항정상태를 지속하였으며, 대조군의 SLDH 동위효소는 주로 3개의 전기영동분획 즉 $LDH_1, LDH_2, LDH_4 및 LDH_5$를 나타냈으나 한냉순화 기간중에는 3개의 분획 즉 $LDH_1, LDH_2 및 LDH_3$가 나타났으며, 한냉순화 시의 SLDH의 상승은 $SLDH_1, SLDH_2 및 SLDH_3$의 반영이라고 생각되며 SALP의 활성치는 한냉순화 초기에 유의적으로 증가하였으나 곧 감소되어 정상치로 접근하여 36일구까지 항정상태를 지속하였고, 대조군의 SALP동위효소는 2개의 전기영동분획을 지속하고 있으며, 한냉순화군 초기의 SALP 활성치의 증가는 $SALP_1$ 분획의 즉각적인 증가의 반영임을 알 수 있고, 한냉순화로 인하여 SCPK의 활성치는 순화초기에 유의적으로 상승하였으나 곧이어 안정치로 접근하여 순화 말기까지 항정상태를 지속하였다. 한냉순화$(4\\pm1^\\circC)$처리는 한냉 초기에 SLDH, SLDH 동위효소, SALP, SALP 동위효소 및 SCPK 활성치 모두에 영향을 끼치는 것으로 생각되며 따라서 상기 각종 효소가 직접 또는 간접적으로 열적조절에 관여하는 것으로 생각된다.

말초신경 손상 백서모델에서 초음파 치료의 신경 및 근육 재생효과 (The Effect of Ultrasound Treatment on Regeneration Nerve and Muscle on Injured Peripheral Nerve of Rats Model)

  • The purposes of this study is to discuss and analyze the effect on the recovery from cut in sciatic nerve. This study used 9 weeks male rats of Sprague-Dawley family. Rat in groups 4 were treated with pulsed therapeutic ultrasound for 3 minutes. 3 times weekly at 3MHz respectively (intensity; $0.2W/cm^2,\;0.5W/Cm^2,\;10W/cm^2$); rat in group 1 received placebo ultrasound. In addition, changes of serum aspartate amino-transferase(AST) and creatine phosphokinase(CPK) levels were also demonstrated with diameter of individual muscle fasciculate and number of muscle fiber in each of three types of muscles located in gastrocnemius, soles. The results of comparing the changes in groups are as follows; 1. We found out that hypertrophic epineurium was present in sciatic nerve injured ultrasound treatment of groups. 2. In the gastrocnemius morphological investigation of the group I (control group), severe muscle atrophy were observed at the 7th days of the sciatic nerve injury. however, muscle atrophy of the group IV ($1.0W/cm^2$) were slightly recovered at the 14th days after treatment ultrasound. At the 28th days, muscular fibers were formed in polygon and were significantly recovered. 3. C-fos immunoreactive of the group II ($0.2W/cm^2$), III ($0.5W/cm^2$) were remarkably increased at the 1th day after treatment of ultrasound. Group IV were markedly deceased. 4. Brain-Derived Neurotrophic Factor(BDNF) immunoreactive of the group II, III were increased after 7 days of the sciatic nerve injury. Group IV were markedly increased from 14th days to 28th days after treatment of ultrasound. 5. A significant increase of serum AST levels were demonstrated in control group. However, serum AST levels of massage groups were significantly decreased compared to that of control group in followed order ; ($0.2W/cm^2<0.5W/cm^2<1.0W/cm^2$). 6. A significant increase of serum CK levels were demonstrated in control of group. However, serum CK levels of massage groups were significantly decreased compared to that of control group in followed order ; ($0.2W/cm^2<0.5W/cm^2<1.0W/cm^2$). The above results suggest that ultrasound treatment after peripheral nerve injury might reduce noxious stimuli, facilitate nerve recovery and effective in the functional improvement delaying muscle atrophy or degeneration.

마우스 및 랫드에서 botulinum toxin type A의 단회 및 28일 반복투여 독성시험 (Single and 28-day repeated dose toxicity studies of botulinum toxin type A in mice and rats)

  • Single and 28-day repeated dose toxicity studies of botulimnn toxin type A were carried out in ICR mice and Sprague-Dawley rats, respectively. In the single dose toxicity study, botulinwn toxin was injected intraperitoneally to male and female mice at a single dose of 40, 59, 89 133 and 200 ng/10 ml saline/kg. All animals died from 59 ng/kg group. Some clinical signs, such as decrease in locomotor activity, dyspnea, prone position and ptosis, were observed in most of both sexes from 59 ng/kg group, but no signs were seen in all animals at 40 ng/kg group. The results showed that the median lethal dose of botulinum toxin might be in the range of 40-59 ng/kg in both sexes. In the repeated dose toxicity study, the test material was administered intradermally for 28 days at doses of 0 (vehicle-treated control), 1.25, 2.5, 5.0 and $10.0ng/head/50{\mu}{\ell}$ saline in male and female rats. No test material-related changes were noted in survivals, clinical signs, food and water consumptions and gross finding in any group. Botulinum toxin treatment significantly decreased the body weight gain rate in male of 5.0 ng/head group and over and in female of 10.0 ng/head group compared to vehicle-treated control. One or more relative organ weights (i.e., spleen, thymus, liver and kidney) were increased significantly from 5.0 ng/head group compared to vehicle-treated control in both sexes. Serum biochemistry revealed increases in aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatine phosphokinase, total protein and albumin in male, and increases in AST and ALT and decreases in $K^+$ and $Cl^-$ in female without dose-pendent manners. In the histopathological study, physical stimulation by needle caused slight inflammations of dennis. In addition, botulinum toxin treatment induced denervation of nerve cell and disuse of muscle, resulting in atrophy of skeletal muscle in both sexes from 2.5 ng/head group. When the antibodies to toxin were determined in all animals, a significant increase in serum antibodies was observed from 5.0 ng/head group. The results showed that the NOAEL of botulinum toxin might be 1.25 ng/head for 28-day repeated dose toxicity in rats.

음주남성의 나이대별 생리학적 변수들의 비교 (Comparison of Physiological Variables by Age Group in Drinking Men)

  • 본 연구는 남성 음주자의 나이대별 생리학적 변수들에 어떠한 차이가 있는지를 연구하였다. 30대 남성들은 waist-hip ratio, 좌 우 심장-발목 혈관지수, 좌 우 발목-상완지수, 아밀라제, hs-CRP, CPK가 가장 낮았고 체중, IL-6, 좌 우 안와혈압, 혈색소, 적혈구수, 적혈구 분포도, ALT, 총 콜레스테롤, 중성지방, 페르틴 농도는 높게 나타났다. hs-CRP, CPK는 40대 음주남성에서 높았지만 페리틴 농도는 반대였다. 50대 음주남성에서는 WHR이 높았으며 이완기 혈압, 좌 우 안와혈압, 호산구와 아밀라제 농도는 높게 나타냈다. 60대 음주남성에서는 좌 우 심장-발목 혈관지수, 평균 혈색소 농도, $\gamma$-GTP, RF, AFP, PSA의 농도는 높았으며, 체중, 이완기 혈압, 좌 우 안와혈압, 혈색소, 혈소판, 백혈구, 적혈구 분포지수, 호산구, 단구, 아밀라제 중성지방, CPK 농도 변화에서는 높게 나타났다. 이러한 결과를 볼 때 음주는 남성들에 있어 나이대별로 다양한 생리학적 변화를 유도 할 것으로 사료된다. 향후 음주가 성별, 즉 남녀 간에 미치는 생리학적 영향의 차이에 대한 연구 역시 더 필요할 것으로 생각된다.

운동 유형이 노인 여성의 근손상 지표 및 혈중지질에 미치는 영향 (The Effects of Exercise Type in Elderly Women on the Muscle Damaged and Blood lipid)

  • 본 연구는 12주간의 저항성운동 유형이 여성 노인의 근손상 지표 및 혈중지질에 미치는 영향을 규명하고자 하였다. 연구대상은 여성노인 30명을 통제집단 10명, 아쿠아로빅운동 집단 10명, 순환운동 집단 10명을 무선으로 배정한 후 주당 3회 1회 운동시 60분을 적용하여 각 집단별 운동을 12주간 실시하였다. Total Cholesterol(TC), Low Density Lipoprotein Cholesterol(LDL-C), High Density Lipoprotein Cholesterol(HDL-C)은 교호작용효과가 나타나지 않았으나, 체중과 체지방량은 교호작용효과가 나타났다. 체중, 체지방량, TC, LDL-C은 통제집단에 비해 수중운동과 순환운동 후 유의한 감소를 나타냈으며, HDL-C은 통제집단에 비해 수중운동과 순환운동 후 유의한 향상을 나타냈다. Triglyceride(TG), Creatine Phosphokinase(CPK), Lactate Dehydrogenase(LDH)는 유의한 차이를 나타내지 못하였다. 이를 통하여 수중운동과 순환운동은 여성노인에게 있어 체중 및 혈중지질 감소와 HDL-C의 증가에 있어 효율적인 운동이라 제시 할 수 있으며 여성노인에게 있어 근감소예방과 개선에 효과적인 운동으로 적용될 수 있을 것이라 사료된다.

산수유(山茱萸)와 보골지(補骨脂) 복합추출물의 Sprague-Dawley 랫드를 이용한 13 주 반복경구투여 독성시험 및 4 주 회복시험 (A 13-Week Repeated Oral Dose Toxicity Test and a 4-Week Recovery Test of Standardized Cornus officinalis and Psoralea corylifolia L . in Sprague-Dawley Rats)

  • Objectives : In the current study, we performed the 13-week repeated oral dose toxicity test and a 4-week recovery test of standardized Cornus officinalis Sieb. et Zucc. and Psoralea corylifolia L. 30 % ethanol extract (SCP) in Sprague-Dawley (SD) rats owing to aims for verifying no observed adverse effect level (NOAEL). Methods : The animal study was performed according to OECD guidelines for the testing of chemicals section 4 health effects test No.408 repeated dose 90-day oral toxicity study in rodents (03 October 2008). In the repeated dose toxicity study, SCP was orally administered to female and male rats at dose levels of 1,000, 2,000, and 4,000 mg/kg/day for 13-week. The control group and high dose (4,000 mg/kg/day) group were then monitored for 4 extra weeks to determine recovery time after the study period. 1) Results : Compared with the control group, there were no treatment-related adverse effects in clinical signs, body weight, hematology, serum biochemistry (Aspartate aminotransferase, Alanine aminotransferase, Alkaline phosphatase, 𝛾-Glutamyl transpeptidase, Blood urea nitrogen, Creatinine, Glucose, Total cholesterol, Total protein, Creatine phosphokinase, Albumin, Total bilirubin, Triglyceride, Inorganic phosphorus, Albumin/Globulin ratio, Calcium ion, Sodium ion, Potassium ion, Chloride ion), necropsy findings and organ weight (Ovary, Adrenal gland, Pituitary, Thymus, Prostate, Testis, Epididymis, Spleen, Kidney, Heart, Lung, Brain, Liver) at any dose tested. Conclusions : Taken together, these results suggest that the NOAEL of SCP in both genders was considered as over 4,000 mg/kg. Results from this study provide scientific evidence for the safety of SCP.

마사지가 좌골신경 절단 마우스의 골격근에 미치는 영향 (Effect of Massages on The Skeletal Muscles of Sciatic Nerve Neurectomized ddY Mice)

  • To demonstrate the effect of massages on the skeletal muscles of immobilization ddY mice models induced by right side sciatic nerve neurectomy, the cross sectional histological profiles of the muscularis (M) gastrocnemius, M. tibialis cranialis and M. tibialis caudalis were observed after 28 days of treatment of massages with the changes of body weight, thickness of hind limb and individual muscle weights. In addition, changes of serum aspartate amino- transferase (AST) and creatine phosphokinase (CK) levels were also demonstrated with diameter of individual muscle fiber and muscle fasciculata, and number of muscle fiber in each of three types of muscles located in the calf. The experimental groups were divided into five groups, 1) Sham-operated group (Sham), 2) Neurectomized but not massage control group (Control), 3) Neurectomized and massage at knee joint regions (T1), 4) Neurectomized and massage at calf regions (T2), and 5) Neurectomized and massages at achilles tendon regions (T3). The experimental animals were used 5 per groups. The results are as follow: 1. No significant changes of body weight were demonstrated in this groups during whole experimental periods compared to that of Sham group. In addition, no significant changes of body weight gains during experimental periods (Day 0 - Day 27: 28 days of treatment) were also observed. 2. The thickness of intact left side hind limb was not changes in all tested groups compared to that of Sham group. A significant increase of serum AST levels were demonstrated in Control group. However, serum AST levels of massage groups were significantly decreased compared to that of Control group in followed order: T3 > T1 > T2. In case of 73 group, the values decreased into similar levels compared to that of Sham group. 7. A significant increase of serum CK levels were demonstrated in Control group. However, serum CK levels of massage groups were significantly decreased compared to that of Control group in followed order: T3 > T1 > T2.
