Quality Monitoring of Specification of Crataegi Fructus in the Korean Pharmacopoeia and Studies HPLC Standard Chromatogram (산사(山楂)의 규격 기준 모니터링 및 HPLC 표준크로마토그램 연구)

  • Objectives : Crataegi Fructus has been used as traditional medicines for more digestion action, amenorrhea due to blood stasis, and hyperlipemia. The aim of this study was to compare of Crataegi Fructus in South Korea collected during three years according to the standards in monographs of the Korean Pharmacopoeia Eleventh edition (KP 11). Methods : Crataegi Fructus was carried out identification test (Qualitative reaction, Thin layer chromatography), heavy metal test, and total ash registered at KP. Add to we tested loss on dry, contents of ethanol-soluble extracts, and HPLC profiling. Results : Identification test (TLC) was on comparing with ursolic acid standard solution in $R_f$ value, all samples showed red purple spot ($R_f$ value 0.9). Ursolic acid spot in $R_f$ value 0.35 showed by changing mobile phase condition. Heavy metals showed contents for Pb, Cd, As, and Hg range of 0.0 ~ 0.5 ppm, 0.0 ~ 0.2 ppm, 0.0 ~ 0.3 ppm, and 0.0 ~ 0.1 ppm. Loss on drying was ranged from 5.5 to 11.9 %, total ash was between the range 2.7 ~ 4.0 %. Contents of ethanol-soluble extracts was ranged from 17.8 to 44.9 %. The content of chlorogenic acid was ranged from 0.0 to 0.1 % based on the chlorogenic acid standard curve. Conclusion : We have verified the current specification standard of Crataegi Fructus and standard that is not set. We hope that it will help the standardization of Crataegi Fructus.

Component Analysis and Digestive Enzyme Activities of Fermented Crataegi Fructus Extracts (산사 발효액의 함유 성분 분석 및 소화 활성)

  • Currently many studies aimed at enhancing efficacy of medicinal food on biological activity using bioconversion technology including fermentation process. In this study, the quality characteristics and antioxidative activity of fermented Crataegi fructus was investigated. The antioxidant activity of fermented Crataegi Fructus was assessed by various radical scavenging assays using DPPH (2,2-Diphenyl-1-picrylhydrazyl), FRAP (Ferric ion reducing antioxidant power), Reducing power and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)). Moisture content of fermented Crataegi Fructus was $39.3{\pm}0.06%$. Contents of crude ash, crude protein, and crude fat were $0.20{\pm}0.01$, $1.77{\pm}0.04$, and $1.40{\pm}0.59%$, respectively. Moreover, the hunter's color values of fermented Crataegi Fructus were 79.24 (lightnees), 1.58 (redness), and 31.25 (yellowness), respectively. Total phenolic contents of fermented Crataegi Fructus were $3,015{\pm}250$ GAE ${\mu}g/g$. The antioxidative activities of fermented Crataegi Fructus significantly increased in a dose dependent manner. In addition, fermented Crataegi Fructus slightly (10.4%) inhibited ${\alpha}$-glucosidase activity; however, there was no inhibitory activity against ${\alpha}$-amylase. In terms of proteolytic activity, fermented Crataegi Fructus showed a strong activity than pancreatin (used as a positive control). These results indicate that fermented Crataegi Fructus can be used as a natural resource for material aiding digestion.

Physiological Functionalities of Solvent Fractions Isolated from Crataegi Fructus

  • In this study we investigated the biological activities of Crataegi Fructus, including antioxidative, fibrinolytic, $\alpha$-glucosidase inhibitory, and thrombin inhibitory activities. Crataegi Fructus, hot water extract was fractionated into hexane, $CHCLl_3$, ethyl acetate, butanol, and water fractions, and each of these was assayed individually. The water fraction showed the highest extraction yield at 4.08% (w/w). The antioxidative activities of the water, ethyl acetate, and butanol fractions were 31.07%, 45.87%, 50.28%, and 91.74%, respectively. Assays for fibrinolytic activity indicated that only the butanol fraction has significant efficacy at 1.93 plasmin units/ml. Thrombin inhibitory assays indicated that the 10-fold dilutions of the $CHCLl_3$, ethyl acetate, and butanol fractions had inhibitory activities of 34.97%, 41.43%, and 58.10%, respectively. The 10-fold dilutions of the only ethyl acetate fraction demonstrated $\alpha$-glucosidase inhibitory activities of 75.07%. From the above results, we propose that extracts of Crataegi Fructus can be used as a material for the development of biofunctional foods.

Nutritional Characteristics and Screening of Biological Activity of Crataegi fructus (산사 영양성분 분석 및 생리활성 탐색)

  • The purpose of this study is to determine the possibility of using Crataegi fructus as a natural food source. To accomplish this purpose, the contents of general and biological activities were measured. The contents of carbohydrate, crude protein, crude lipid and ash are 85.6%, 2.4%, 1.9% and 0.4%, respeectively. Further, the calories of Crataegi fructus was 369.1 kcal. The contents of essential and non-essential amino acids were 852.26 mg and 1,178.29 mg, respeectively. The K was the largest mineral followed by Ca, P, Mg, which means Crataegi fructus is an alkali material. Crataegi fructus extracts slightly(17.6~32.8) inhibited ${\alpha}$-glucosidase activity. However, there is no inhibitory activity against ${\alpha}$-amylase. In terms of proteslytic activity, Crataegi fructus extracts showed a strong activity than pancreatin(used as a positive control). These results indicate that Crataegi fructus can be used as a natural resource for material aiding digestion.

A Study of Antimicrobial Activity of Herbal Extracts on Clostridium difficile (Clostridium difficile에 대한 항균 한약재 탐색 연구)

  • Objectives This study was conducted to confirm the possibility of Clostridium difficile infection (CDI) treatment through natural herbal medicines. Methods After screening a total of 77 herbal medicines through the paper disc agar diffusion method, we selected the herbal medicines that showed a effectiveness compared to the positive control vancomycin. Afterwards, drugs that showed inhibitory effects compared to C. difficile without inhibition of Bifidobacterium bifidum and Lactobacillus plantarum, known as beneficial bacteria, were selected and minimal inhibitory concentration (MIC) was confirmed by applying the Broth microdilution method. Results The Coptidis Rhizoma, well known for its antimicrobial effect, was found to have antimicrobial effects on C. difficile, but also had inhibitory effects on the beneficial bacterium B. bifidum. 30% ethanol extraction Crataegi fructus, Corni fructus and Mume fructus had antimicrobial effects on C. difficile without inhibiting the beneficial bacteria B. bifidum and L. plantarum. The MIC values of 30% ethanol extraction Crataegi fructus, Corni fructus and Mume fructus were found to be 10 mg/mL, 20 mg/mL and 5 mg/mL, respectively. Conclusions Crataegi fructus, Corni fructus and Mume fructus were identified as candidate medicines for C. difficile. Further researchs will need to be done in vivo, and to find an optimal extraction method accompanied by economic evaluation.

Quality Characteristics of the Yanggeng made by Crataegi fructus Extracts (산사추출액 첨가 양갱의 품질특성)

  • The objective of this study is to investigate the quality characteristics of yanggeng prepared with different amounts of Crataegi fructus extracts(CFE; 0, 5, 10, 15, 20). Increasing the amount of CFE in the yanggeng tended to increased the moisture and acidity, while it decreased the level of pH. In chromaticity determination, the values of lightness(L) and yellowness(b) showed a decrease. However, the value of redness(a) increased by increasing levels of CFE. In addition, hardness increased by increasing levels of CFE. Although regarding texture profile analysis, hardness and springiness of yanggeng were increased, adhesiveness and cohesiveness of yanggeng were decreased. Cohesiveness shown similar result compared to the control and CFE added treatments. The sensory evaluation indicated that CFE 15 showed the best preference in color, taste, texture, flavor, and overall acceptance. Based on these results, 15% should be recommended as a optimum level of CFE to be added for the preparation of yanggeng. And these results suggest that CFE may be a useful ingredient in yanggeng for improvement of the quality.

A Study on The Natural Dyeing of Crataegi fructus Extracts (산사 추출물을 이용한 천연염색 연구)

  • In this study, the dyeing properties and functionalities of Crataegi fructus extract were investigated for the purpose of application to new natural dye resources. The effects of dyeing conditions(concentration of dye, dye bath temperature, dyeing duration and dye bath pH) and mordanting on dye uptake and color changes were also examined. The study also estimated the colorfastness, antibacterial properties, and ultraviolet protection properties of the dyed and mordanted silk fabrics. The dye uptake increased in tandem with the dye concentration and dye bath temperature. The highest K/S values were obtained at the following conditions: a dye concentration of 100%(v/v), a dyeing period of 60 minutes, a dye bath temperature of $90^{\circ}C$, and a dyeing pH of 3. The colorfastness of the material when dry cleaned or rubbed ranged from good to excellent and fastness to light was rated at grade 2-3. The change in the color grade when exposed to washing was not good; however, the stain of washing fastness was good at a range of 4-5. In regards to the functional property aspects, the dyed and mordanted fabrics exhibited excellent results with a 99.9% reduction rate, and excellent ultraviolet protection factors.

The Effects of an Extract of Artemisiae Capillaris, Curcumae Longae, and Crataegi Fructus (IUS) on Anti-hyperlipidemia and Anti-oxidation in db/db Mouse Model (db/db mouse에서 인진(茵蔯), 울금(鬱金), 산사(山査) 복합추출물의 항고지혈 및 항산화 효과)

  • Objective: This study was performed to investigate the effect of IUS (Inulsan, an extract of Artemisiae capillaris (茵蔯), Curcumae longae (鬱金), and Crataegi fructus (山査)) on anti-hyperlipidemia, anti-oxidation, and anti-inflammation.Method: We administered water extracts of Artemisiae capillaris, Curcumae longae, and Crataegi fructus for three weeks to db/db mice (C57BL/Ks), animal models induced with type 2 diabetes mellitus. Mice were divided into three groups: normal (C57BL/6J mice group), control group (db/db mice without administration of IUS) and IUS group (db/db mice treated with IUS). Then we measured total cholesterol, LDL cholesterol, HDL cholesterol, and triglyceride in the serum after the oral administration of IUS.Results: 1. IUS did not show any cytotoxicity in RAW 264.7 cells. 2. IUS decreased AST, ALP, and creatinine levelsand did not show any liver or renal toxicity in the db/db mice. 3. IUS increased DPPH and ABTS radical scavenging activity and decreased ROS production in RAW 264.7 cells. 4. IUS significantly decreased IL-1β, IL-6, and TNF-α production in RAW 264.7 cells. 5. IUS increased HDL cholesterol and significantly decreased total cholesterol and triglyceride in db/db mice. 6. IUS significantly decreased the atherogenic index and cardiac risk factor. 7. In contrast with the control group, fat infiltration in the liver and aorta decreased in IUS treated mice. The cell nucleus was located in the central area in H&E staining of liver. And endomembranes also were more thinner than the control group in H&E staining of aorta.Conclusions: These results suggest that IUS might be effective in the prevention and treatment of dyslipidemia.

The Effects of Saengkankunbi-tang and Its Composition on Free Fatty Acid-Induced Lipotoxicity in HepG2 Cell (HepG2 cell에서 유리지방산 유발 지방독성에 대한 생간건비탕(生肝健脾湯)과 일부 조성 한약물의 효과)

  • Objectives : The aim of this study was to investigate whether the effects of extract from Saengkankunbi-tang and its composition that Artimisiae capillaris Herba, Crataegi Fructus, Alismatis Rhizoma, Hoelen and Raphani Semen could protect HepG2 cells from palmitic acid-induced lipotoxicity through lysosomal and mitochondrial pathways in an in vitro model. Methods : To examine the effects of the extracts from Saengkankunbi-tang and its composition that Artimisiae capillaris Herba, Crataegi Fructus, Alismatis Rhizoma, Hoelen and Raphani Semen on palmitic acid-induced lipotoxicity in HepG2 cells, we measured the contents of cell viability, cytotoxicity. Then to investigate the effects of the extract from Saengkankunbi-tang, Artimisiae capillaris Herba and Raphani Semen, we measured that triglyceride, reactive oxygen species, ATP levels, glutathione levels, cytochrome c and cathepsin B. Results : The extracts from Saengkankunbi-tang and its composition had a cell-protective function. The extracts from Saengkankunbi-tang, Artimisiae capillaris Herba and Raphani Semen controlled triglyceride over-accumulation in cells and reduced overproduction of reactive oxygen species. The extracts from Saengkankunbi-tang and Raphani Semen increased ATP and glutathione levels which had been decreased by lipotoxicity. The extracts from Saengkankunbi-tang, Artimisiae capillaris Herba and Raphani Semen reduced leakage of cytochrome c and the extracts from Saengkankunbi-tang and Raphani Semen reduced leakage of cathepsin B in lipotoxicity. Conclusions : These results show that the extracts from Saengkankunbi-tang and its composition that Artimisiae capillaris Herba and Raphani Semen have cell protective effects on palmitic acid-induced lipotoxicity through lysosomal and mitochondrial pathways.

Comparison of the Therapeutic Efficacy of Rhizoma Alismatis, Fructus Crataegi, Fructus Lycii, Radix Curcumae, Radix Salviae Miltiorrhizae, Herba Artemisiae Scopariae on the Experimental Cellular Model of Nonalcoholic Fatty Liver Disease (비알코올성 지방간 세포 모델에 대한 택사, 산사, 구기자, 울금, 단삼, 인진의 효능 비교)

  • Objectives : We try to compared the efficacy of six herbal medicines, Rhizoma Alismatis (RA), Fructus Crataegi (FC), Fructus Lycii (FL), Radix Curcumae (RC), Radix Salviae Miltiorrhizae (RSM), and Herba Artemisiae Scopariae (HAS), constituting KHchunggan-tang which was previously proven to be hepatoprotective on non-alcoholic fatty liver disease with combined properties of cellular steatosis, ROS production, and cytoprotection. Methods : HepG2 cells were pretreated with aqueous extracts of the six herb medicines at concentrations of 1, 10, 50 and 100 ${\mu}g/ml$ each, and treated with 0.5 mM palmitate consecutively. After 21 hrs, cell viability was assessed using MTT assay, and the percentage of cells with sub-G1 DNA content was measured using fluorescence-activated cell sorting after propidium iodide staining. Results : The first three extracts, RA, FC, and FL restored cell viability reduced by palmitate in MTT assay, and RA, FC, FL and RC inhibited palmitate-induced apoptosis in sub-G1 analysis. FL showed relatively weak potential only at tested maximal dose, and RA showed the greatest higher efficacy on this experimental cellular model of nonalcoholic fatty liver disease. Conclusions : According to this comparative experiment, Rhizoma Alismatis seems to have the most powerful potential among the six herbs constituting KHchunggan-tang, and consecutive further study seems to be required for more standardized and effective clinical application of KHchunggan-tang for treatment of non-alcoholic fatty liver disease.
