Acute Toxicity Study on Gumiganghwal-tang and Fermented Gumiganghwal-tang Extracts (구미강활탕 및 발효 구미강활탕 추출물의 급성독성 연구)

  • Objectives : Traditional medicine Gumiganghwal-tang (GT) has been used in Asia to treat inflammatory diseases including common cold, pain, fever, and algor. In this study we investigated the acute toxicity and safety of GT and fermented GT (FGT). Methods : Acute toxicity and safety were evaluated in male and female ICR mice orally administered 0 (control) and 2,000 mg/kg of GT and FGT. After the administration of GT and FGT, we observed mortality, body weight, clinical symptoms. After necropsy, organ weights were measured and blood analysis was performed. Results : There was no mortality and clinical symptoms according to the administration of GT and FGT. Comparing with control group, there were no significant alterations on the organ weight, complete blood cell count and biochemical parameters. Conclusions : Median lethal dose of GT and FGT considered to be over 2,000 mg/kg in both male and female mice, and recognized as safe with no toxicity.

Growth rate, carcass characteristics and meat quality of growing lambs fed buckwheat or maize silage

  • Objective: This study evaluated inclusion of buckwheat silage to the diet of growing lambs in terms of meat quality as compared to maize silage. Methods: Buckwheat, rich in total phenols (TP, 33 g/kg dry matter [DM]), was harvested at the end of the milk stage and ensiled in 40 kg plastic bags after wilting (294 g/kg silage DM). A total of 18 growing lambs ($21.6{\pm}1.2$) were individually fed isonitrogenous and isoenergetic total mixed rations (TMR) for 75 d that either contained buckwheat or maize silage at DM proportions of 0.50. At the end of feeding trail all lambs were slaughtered to assess carcass characteristics and meat quality. Results: Buckwheat silage increased (p<0.01) the DM intake of lambs as compared to maize silage, but had no effects (p>0.05) on live weight gain and feed efficiency. Carcass weight, dressing percentage, meat pH, water holding capacity, cooking loss, shear force ($kg/cm^2$), and total viable bacteria count of meat did not differ (p>0.05) between the treatments. However, TP content of meat increased (p<0.001) by feeding buckwheat TMR. Feeding buckwheat TMR also decreased (p<0.05) the b* values of meat. Conclusion: The results provide that buckwheat silage is palatable and could successfully include TMR of growing lambs with no adverse effects on performance, carcass and meat quality. Additionally, feeding buckwheat silage to lambs offers increased TP in meat.

Comparative study of herbal-acupuncture using Ursi Fel·Bovis Calculus and acupuncture at ST36 and GB39 effect on Adjuvant Arthritis in rats (자침(刺鍼)과 우황(牛黃)·웅담(熊膽) 약침(藥鍼)이 백서(白鼠)의 Adjuvant Arthritis에 미치는 영향에 대한 비교 연구)

  • Objective : This research carried out to find out the effective efficiency of acupuncture at ST36 GB39 and herbal-acupuncture using different concentrational Uris Fel Bovis Calculus on adjuvant arthritis in rats. Method : In this study, I do acupuncture treatment and herbal-acupuncture in ST36 GB39 by solution that diluted to fivefold and tenfold with combined solution of Bovis Calcuclus and Ursi Fel at 10:1. And then I observed the change of inhibitory rate of paw edema, body weight, hematologic assay, histologic assay. Result : 1. In Adjuvant Arthritis in rats, the herbal-acupuncture group 1 is more efficienct then acupuncture group. 2. In histologic assay of knee joint, synovial cell and fibroblast markedly activated in herbal-acupuncture group. 3. The albumin level in serum more markedly increased in herbal-acupuncture group 2 than the control group. 4. The paw edema significantly decreased in acupuncture group and herbal-acupuncture group 2 compared with the control group. 5. The body weight significantly increased in acupuncture group and herbal-acupuncture group 2 compared with the control group. Conclusion : In these results, The herbal-acupuncture using Ursi Fel, Bovis Calculus, suppress inflammation of Adjuvant Arthritis, and decrease markedly the count of WBC, and increase the albumin level in serum, and have an influence on Adjuvant Arthritis with activating of synovial cell, fibroblast. And these were more effective than the others, and then needed consistent study of its mechanism.

High Vitamin E Supplement is Needed to Have an Anticarcinogenic Effect of Fish Oil (어유와 비타민 E 보강 수준이 쥐간의 전암성 병변에 미치는 영향)

  • The influences of fish oil and different levels of vitamin I supplement on hepatocellular chemical carcinogenesis have been studied. Male Sprague-Dawley rats received diethylnitrosamine (DEN)(200mg/kg body weight) and were subjected to two-thirds partial hepatectomy to induce murine chemical hepatocarcinogenic procedure. Placental glutathione S-transferase(GST-P) positive foci area, antioxidant enzymes(Cu/Zn-superoxide dismutase(SOD), catalase, glutathione reductase (GR), total- glutathione peroxidase (TGPx), glutathione S -transferase (GST)), glucose 6-phosphatase (G6Pase) activities, and lipid peroxidation of microsomes(thiobarbituric acid reactive substances (TBARS)) were measured. Experimental animals were fed 15% corn or fish oil with 0, 40, 1,000, 10,000IU vitamin E /kg diet for 8 weeks. Vitamin E supplements decreased the area of GST-P positive foci in both groups. The higher the vitamin E levels, the smaller the area of GST-P positive foci were noticed. Compared to 0 IU vitamin E, 40 IU in corn oil and 1,000 IU in fish oil groups were effective in decreasing G57-P positive foci area. Fish oil groups tended to have smaller area of GST-P positive foci. fish oil groups showed lower body weight, lower activities of Cu/Zn-SOD and TGPx, higher TBARS contents, higher activities of GST, catalase, G6Pase, GR and higher liver/body ratio than corn oil groups. As the level of vitamin I increased, GST-P positive foci count, catalase activities, and TBARS tended to decrease. G6Pase activities tended to increase in both groups. At higher vitamin E levels, GST activities tended to decrease in fish oil groups. These results suggest that vitamin I has suppressive offects on hepatocellular chemical carcinogenesis probably through antioxidant eH:cts decreasing TBARS contents, $H_2O$$_2$, and organic peroxides. fish oil tended to have greated suppressive offects than corn oil on hepatocellular carcinogenesis. Performance of Male Crossbred Calves as Influenced by Substitution of Grain by Wheat Bran and the Addition of Lactic Acid Bacteria to Diet

  • To study the effect of wheat bran and lactic acid producing bacteria (LAB) on the performance of calves, 20 crossbred male cattle calves (day old), distributed into two groups were fed on calf starters containing 50 or 0% maize grain, along with green berseem ad libitum and milk as per body weight. Each group was further divided into two sub groups and one subgroup of each group was supplemented with mixed culture of LAB (Lactobacillus acidophilus L. casei, L. Jugarti). Milk feeding was discontinued after 8 weeks of age. The addition of culture increased (p<0.05) DM intake in calves receiving grainless diet from eighth week to the thirteenth one. There was about 21% higher body weight gain and 14% lower feed : gain ratio in culture supplemented calves. DM digestibility was significantly lower (p<0.05) in calves getting grain without culture. The crude protein NDF and ADF digestibility was higher (p<0.05) in grainless than the grain fed group. No major change on rumen fermentation pattern among different treatments was found. The concentration of total volatile fatty acids (TVFA) and protozoa count was higher (p<0.05) in grain fed group. However, lactic acid concentration was higher and rumen pH was lower due to culture feeding. The incidence as well as severity of diarrhoea was reduced in culture supplemented group. The results indicate that crossbred calves can be reared successfully on grainless diet and berseen fodder. The performance of calves was also improved by LAB supplementation.

Antineoplastic Activity of Crude Saponin Mixture from the Roots of Luffa tuberosa (Roxb.) in Ehrlich Ascites Carcinoma Bearing Mice

  • The antitumor activity of crude saponin mixture obtained from Luffa tuberosa (Roxb.) (Fam; Cucurbitaceae) hairy roots (CSLT) in mice transplanted with Ehrlich ascites carcinoma (EAC) was investigated. The EAC-bearing mice receiving 150 and $300{\mu}g/kg$ body weight, (i.p) of CSLT have shown a dose dependent elevation in tumor-tree survival and a highest number of survivors were observed after administration of CSLT $(300{\mu}g/kg)$, which was considered as an optimum dose for its antineoplastic action. The mean survival time (MST) for this dose was approximately $47.1{\pm}0.74d$, when compared with $19.0{\pm}0.36d$ of untreated control. Administration of $300{\mu}g/kg$ CSLT resulted in 130% long-term increased survival time. The measurement of body weight, tumor volume, packed cell volume, viable and non-viable count indicated the efficacy of CSLT in tumor-bearing mice, there was a significant recovery in hematological profiles, and there was depletion in lipid peroxidation levels, and the antioxidant enzyme activities such as GSH, SOD and CAT were restored to near the normal levels. The CSLT was found to be devoid of conspicuous short-term toxicity in the mice when animals were intraperitoneally injected with 250, 500, 750 and $1000{\mu}g/kg$ bodyweight. The treated mice showed conspicuous toxic symptoms only at a dose of $1500{\mu}g/kg$. Mortality of the animals was monitored up to 14 d post drug treatment, $1/7^{th}$ of the $LD_{50}$ dose has been considered for the optimal antineoplastic activity.

Assessment of Daily Steps, Physical Activity and Activity Coefficient of the Elementary School Children in the Rural Area (농촌지역 초등학생의 1일 평균 보행수, 활동량 및 활동계수 평가)

  • The purpose of this study was to assess the daily steps, physical activities and activity coefficient of the elementary school children in the rural area. Body weight, height and daily steps were measured and one-day activity diaries were collected by interviewing children. The average age of the subjects was $9.96{\pm}1.02$ years. Average height, weight, obesity index, body fat and muscle of subjects were $136.2{\pm}8.9cm,\;35.2{\pm}8.5kg,\;99.6{\pm}18.8%,\;22.9{\pm}8.5%\;and\;35.8{\pm}6.0%$, respectively. The average daily steps of the subjects was 17,584 and daily steps (19,314) of 3rd grade students was significantly higher than that (15,712) of 5th grade children. But there was no significant difference in daily steps and activity coefficients between boys and girls. Daily steps (23,347) of exercise group showed the significantly higher than that (16,144) of non-exercise group. Gender and grade of subjects did not have significant influence on activity coefficients, but there was a significant difference in activity coefficient on weekdays between the exercise group ($1.82{\pm}0.30$) and non-exercise group ($1.50{\pm}0.21$). Analysis of variance revealed stronger associations between daily steps and body fat (%) than between daily steps and BMI. Daily steps showed significant negative correlation with body fat (%) measured using two methods r=-0.321 and r=-0.365, respectively. Activity coefficient was significantly correlated (r=0.436) with daily steps, thus increasing daily steps cm prevent and treat childhood obesity by increasing the energy expenditure. The higher activity coefficients (weekday 1.56, weekend 1.53) of the subjects was caused by the fact that rural students spent more time outside and enjoyed stronger activities than urban students. The results of this study can be used to estimate energy requirements for Korean children based on exercise levels and living areas.

Nutrition Intakes and Relations to the Obesity and the Prevalence of Anemia in Preschool Children Living in Metropolitan Area of Korea (대도시 지역 학령 전 아동의 영양소 섭취량과 비만 및 빈혈과의 관계 연구)

  • This study was designed to investigate nutrition intakes and its relation to the obesity and the prevalence of anemia in 252 children(136 boys, 116 girls) aged $2{\sim}6$ years. The hematological parameters, daily nutrient intakes and height and weight were measured. Calorie intakes of 2 and 3year-old-children were over their RDA while those of 4-6 years were below the RDA. Intakes of protein, P, and vit B complex were far over the RDA in all ages of children. Fe and vit A intakes were insufficient in all ages except 3 years while Ca intakes were insufficient in all ages except 2 and 3 years. About 18.2% of the children were evaluated as obese. However, very few children were anemic by hematologic parameters. The mean Hb concentrations were 12.2mg/dl in boys and also in girls. The mean Hct was 36.2% in boys, 35.8% in girls. Serum Fe concentration was 100.1mg/dl in boys, 101.1mg/dl in girls. RBC count was significantly higher in boys while MCH was significantly higher in girls. Intakes of protein, P, Fe, vit $B_1$, niacin, vit C were significantly higher in obese group compared to under weight group. There were significant positive correlations between protein intake and Hb, Hct, MCH, and MCHC. The correlation between Fe intake and Hb or Hct was not significant. In conclusion, it seems that the nutritional status of the most children was in very good condition. However, they needed to take more foods supplying Fe, Ca, and vit A.

Bycatch of sharks in Korean tuna longline fishery (한국다랑어연승어업에 있어서 상어류의 부수어획)

  • Data collected by on-board observers and from experimental surveys during 2004-2006 were analyzed to figure out the status of shark bycatch in Korean tuna longline fishery. Results obtained from 10 surveys indicated that 14, 13 and 1 species of shark were incidentally caught in Korean tuna longline fishery operated in the Pacific, Indian and Atlantic (Mediterranean) Ocean, respectively, and that shark bycatch accounted for about 29-31% of the total catch. Sharks brought aboard were processed in 3 ways; out of 1,127 sharks observed, 575 sharks (51.0%) were discarded after finning, 299 sharks (26.5%) were stored frozen after finning for future use and 253 sharks (22.4%) were released into the sea immediately after caught. The fin to body weight ratio of sharks was estimated to be about 4.7% which is similar to the guideline of 5% established by the international fisheries organizations. The underestimate of shark bycatch in Korean tuna longline fishery was significant because it was general practice that fishermen on-board did not count the discarded shark as a catch.

Effect of Presoaking Treatments on Growth and Rot of Soybean Sprouts (침지처리가 콩나물의 생육 및 부패에 미치는 영향)

  • In order to inhibit rot of soybean sprouts, presoaking treatments of soybeans with various solutions were studied. Optimal soaking time was 20min and citric acid, chitosan, GFSE(Grapefruit seed extract) were more effective. Most of presoaking treatments decreased the rot ratio of soybeans considerably, and didn't decrease germination ratio of them compared to control. Particularly GFSE, chitosan and phosphate buffer were effective. Presoaking treatments besides ethanol showed increased weight and yield of soybean sprouts compared to control, and particularly chitosan treatment increased yield of 67%, weight of 6.9% and length of 9.5% of soybean sprouts compared to control. Presoaking treatments decreased total microbial count of soybeans and rot of soybean sprouts during cultivation.
