β-Amino-n-butyric Acid Regulates Seedling Growth and Disease Resistance of Kimchi Cabbage

  • Non-protein amino acid, ${\beta}$-amino-n-butyric acid (BABA), has been involved in diverse physiological processes including seedling growth, stress tolerance and disease resistance of many plant species. In the current study, treatment of kimchi cabbage seedlings with BABA significantly reduced primary root elongation and cotyledon development in a dose-dependent manner, which adverse effects were similar to the plant response to exogenous abscisic acid (ABA) application. BABA was synergistically contributing ABA-induced growth arrest during the early seedling development. Kimchi cabbage leaves were highly damaged and seedling growth was delayed by foliar spraying with high concentrations of BABA (10 to 20 mM). BABA played roles differentially in in vitro fungal conidial germination, mycelial growth and conidation of necrotroph Alternaria brassicicola causing black spot disease and hemibiotroph Colletotrichum higginsianum causing anthracnose. Pretreatment with BABA conferred induced resistance of the kimchi cabbage against challenges by the two different classes of fungal pathogens in a dose-dependent manner. These results suggest that BABA is involved in plant development, fungal development as well as induced fungal disease resistance of kimchi cabbage plant.

  • Somatic embryogendesis is one of good examples of the basic research for plant embryo development as well as an important technique for plant biotechnology. This paper describes the direct somatic embryogenesis from zygotic embryos of Panax ginseng is reversely related to normal axis growth of zygotic embryos by the experiment of various chemical treatments. Under the normal growth condition, the apical tips of embryo axis produced an agar-diffusible substance, which suppressed somatic embryo development from cotyledons. Although the cells of zygotic embryos were released from the restraint of embryo axis, various factors were still involved for somatic embryo development. Electron microscopic observation revealed that the ultrastructure of cells of cotyledon epidermis markedly changed before initiation of embryonic cell division, probably indicating reprogramming events into the cells embryogenically determined state. Polar accumulation of endogenous auxin or cell-cell isolation by plasmolysis pre-treatment is the strong inducer for the somatic embryo development. The cells for the process of somatic embryogenesis might be determined by the physiological conditions fo explants and medium compositions. Direct somatic embryos from cotyledons fo ginseng were originated eithrer from single or multiple cells. The different cellular origin of somatic embryos was originated either from single or multiple cell. The different cellular origin of somatic embryos was depended on various developmental stages of cotyledons. Immature meristematic cotyledons produced multiple cell-derived somatic embryos, which developed into multiple embryos. While fully mature cotyledons produced single cell-derived single embryos with independent state. Plasmolysis pretreatment of cotyledons strongly enhanced single cell-derived somatic embryogenesis. Single embryos were converted into normal plantlets with shoot and roots, while multiple embryos were converted into only multiple shoots. GA3 or a chilling treatment was prerequisite for germination and plant conversion. Low concentration of ammonium ion in medium was necessary for balanced growth of root and shoot of plantlets. Therefore, using above procedures, successful plant regeneration of ginseng was accomplished through direct single embryogenesis, which makes it possible to produce genetically transformed ginseng efficently.

  • Structural changes of the endosperm of Panax ginseng C.A. Meyer from fertilization to germination were investigated by light microscope. The endosperm of the ginseng seed is cellular type. Since endosperm cells adjacent embryo continuously breakdown and disappear with the elongation of embryo, the real of endosperm is gradually decreased. As the anatropous ovules of immature seed with green seed coat developes more and more, ovary cells adjacent ovary cavity become abundant by the periclinal division, their size is decreased, hypotrophy of cell wall discern, and they are gradually differentiated in seed coat. Though embryo responds strongly to basic dye at the stage of completion of endosperm formation, tissue of endosperm responds to acidic dye positively Cell wall of embryo and endosperm are composed of primary cell wall not lignified. Endosperm cells adjacent embryo begin to breakdown in the endosperm tissue of indehiscent seed before the beginning of the after-ripening. Dehiscent seed of which seed coat is opened through after-ripening represent the form as a seedling in the result of embryo developments with the formation of organs; radicle, cotyledon, plumule. Umbilifom layer represents strong positive response to the toluidine blue and the basic function. Umbiliform layer that endosperm cells breakdown and disappear is observed clearly at the periphery of the embryo cotylemon, while slightly at the periphery of the radicle.

Agrobacterium-mediated Transformation of PAT into Platycodon grandiflorum A. De. candolle (Agrobacterium을 이용한 Phosphinothricin Acetyl Transferase의 도라지로의 형질전환)

  • This study was conducted to introduce phosphinothricin acetyl transferase (PAT) gene, resistant to basta which was non-selective herbicide, into balloon flower (Platycodon grandiflorum A. De. candolle). Seeds were germinated on MS medium, and 10-day-old immature cotyledon explants and 30-day-old leaf explants were cocultured with Agrobacterium tumefaciens strain MP 90 (pBinSyn) on 1/10 MS medium for 48 hours in the dark at $25^{\circ}C$. The cultures were transferred for selection of kanamycin-resistant shoots to the MS medium supplemented with 0.2 $mg/{\ell}$ NAA, 1.0 $mg/{\ell}$ BA, 3% sucrose, 100 $mg/{\ell}$ kanamycin, 500 $mg/{\ell}$ carbenicillin. Shoots were obtained from 10-day-old immature cotyledon explants after 4 weeks of culture. The shoots were subcultured twice every 4 weeks on the same medium for growth of transgenic shoots. Successful transformation was confirmed by histochemical GUS assay, PCR analysis, RT-PCR analysis, 10 $mg/{\ell}$ phosphinothricin treatment and 0.3% basta spray. The basta-resistant transgenic plants flowered normally.

REDIFFERENTIATION FROM TISSUE CULTURE AND ISOLATION OF VIABLE PROTOPLASTS IN PANAX GINSENG C.A. MEYER (고려인삼의 조직배양에 의한 기관형성과 원형질체배양에 관한 연구)

  • Ginseng cotyledon calli were cultured on 1/2MS media supplemented with combination of various growth regulators to induce more embryoids and plantlets in a short period. And tissues of ginseng root and calli were also incubated under various factors or conditions to establish methods for the isolation of viable protoplasts in Panax ginseng C.A. Meyer. The calli derived from cotyledon produced numerous embryoids in 1/2MS media containing 0.5mg/$\ell$ 2,4-D and 0.5mg/$\ell$ kinetin after 2 months' culture. But only shoot formation was less frequent. Further development of these embryoids occurred on 1/2MS medium supplemented with the same concentration of BA and GA. Viable protoplasts were isolated from the root tissue and callus of ginseng. The specific conditions for the isolation of viable protoplasts were required of ginseng materials, root tissue and callus, being processed. For the production of viable protoplasts from 1-year old ginseng root tissue, an enzyme mixture of $2\%$ cellulase 'Ono-zuka' and $0.5\%$ macerozyme, an enzyme solution pH of 5.2 to 5.8, a 7- to 8- hour incubation period at $28{\pm}1^{\circ}C$, and 0.9M mannitol as osmoticum in the cell enzyme mixture were optimum, while the treatments with an enzyme mixture of $2\%$ cellulase 'Onozuka', $2\%$ macerozyme and $1\%$ driselase, and 25-hour incubation period at $28{\pm}1^{\circ}C$, were more efficient for the production of viable protoplasts from ginseng callus.

Adventitious Shoot Formation and Plant Regeneration from Explants of Solanum nigrum L. (까마중(Solanum nigrum L.)의 유식물 절편체에서 부정아 형성 및 식물체 재분화)

  • In the present study, the effects of plant growth regulators on adventitious shoot and root formation of various explants of $in$ $vitro$ seedlings of $Solanum$ $nigrum$ L. were investigated to determine the optimum conditions for the high-efficiency plant regeneration of this species. The formation of adventitious shoots was higher in leaf explants than in cotyledon, hypocotyl, or epicotyl explants at low concentrations (0.5~2.0 mg $L^{-1}$ ) of 6-benzylaminopurine (BAP). The number of adventitious shoots and the shoot length were also higher in both leaf and cotyledon explants. In particular, 2.0 mg $L^{-1}$ BAP was most effective for stimulating the induction and multiplication of adventitious shoots. In terms of root formation and root development from shoots that were separated from multiple shoots, indole butyric acid (IBA) and indole acetic acid (IAA) were more effective than ${\alpha}$-naphthalene acetic acid (NAA). The percentage of rooting as well as the number of roots per shoot (4.0), root length (7.82 cm), and shoot length (8.76 cm) was highest on MS media supplemented with 0.05 mg $L^{-1}$ IAA. Furthermore, 100% of the regenerated plantlets survived when transplanted to compost soil. These results suggest that leaf explants are the best source for the high-efficiency regeneration of $S.$ $nigrum$ L., and that 2.0 mg $L^{-1}$ BAP and 0.05 mg $L^{-1}$ IAA are the best conditions for shoot and root induction, respectively.

  • Effects of polyamines on chlorophyll and protein content, and $\delta$-aminolevulinate dehydratase(ALAD) activity were investigated during the greening of mung bean cotyledons. Polyamines stimulated chlorophyll formation in greening cotyledons, and this effect was enhanced by KCl. The changes in protein content were similar to the changes for chlorophyll content. The excision entailed an increase in ALAD activity. Then a decrease appeared after 48 h incubation on water in the dark. It was more precocious in the light, but was accelerated when the cotyledons were illuminated after a dark preincubation. Putrescine had little effect on ALAD activity in the dark. In the light, putrescine prevented the decrease in ALAD activity and enhanced this activity when a dark preincubation preceded illumination. KCl had a slight stimulating effect in the dark, but was uneffective in the light. The combination putrescine+KCl was devoid of stimulating effect. The results obtained suggest that plastid development of mung bean cotyledons during greening was affected by polyamines and light and that polyamines may play a role in the regulation of plastid development.

In vitro seed germination and callus formation on flower bud of Korean mistletoe ( Viscum album L. var. cololatum [Kom.] Ohwi) (겨우살이 종자 발아 및 화아 배양에 의한 캘러스 형성)

  • Effects of growth regulators and culture conditions on seed germination, haustorium development, and callus formation of Korean mistletoe (Viscum album var. coloratum (Kom.) Ohwi) were described. Histological examination showed that seed of V. album contained one or two zygotic embryos with rod shape, and actively dividing cells were mainly distributed in radicle region rather than cotyledon of zygotic embryo. The most significant factor for seed germination and haustorium development of V. album was the requirement of the light. Various growth regulators examined in this study failed to substitute the effect of the light on seed germination. The frequency of callus formation was highest at 27.3% when flower buds were cultured onto B5 medium containing $0.1\;mgl^{-1}$ IAA. Explants from other organs were recalcitrant in forming calluses. Culture conditions described in this study could be applied for production of useful metabolites and multiplication of V. album in future.

Quality Characteristics and Antioxidant Activities of Lotus (Nelumbo nucifera Gaertn.) Sprouts Grown Under Different Conditions

  • Lotus (Nelumbo nucifera Gaertn.) is an economically important aquatic ornamental herb with multiple uses, including food, tea, natural pigments, and/or healthcare product. The objective of this study was to evaluate the physicochemical properties and antioxidant potential of lotus sprouts grown in three media: sprouting machine (LSSG), soil (LSSC), and mud (LSMC). The longest sprout was obtained in LSMC (4.79 and 26.79 cm) followed by LSSC (1.95 and 5.4 cm), and LSSG (0.60 and 2.85 cm) at 5 and 10 days, respectively. Higher amounts of total free amino acids were found in cotyledons (33.96, 21.45, and 38.90 mg/g) than in hypocotyls (15.77, 7.90, and 15.29 mg/g ) for LSSG, LSSC, and LSMC, respectively. The ratios of total essential to total non-essential amino acids were higher in hypocotyls (0.36, 0.31, and 0.46) than in cotyledons (0.34, 0.25, and 0.40), respectively. Similarly, the total polyphenol content of the hypocotyl of LSMC (50.33 ㎍ GAE/g) was the highest and that of the husk of LSSG (24.08 ㎍ GAE/g) was the lowest. Overall, the antioxidant potential of hypocotyl was higher than that of husk and cotyledon. The results indicated that the lotus sprouts grown in mud could be a good source of nutritional and natural antioxidants.

Study on the Optimum Time of Seed Production and Development of Embryo in Panax ginseng C.A.Meyer (인삼종자의 채종적기구명과 배발달에 관한 연구)

  • The characteristics such as characters of seed, fruit and endosperm, development of embryo, dehisced and germination rate of seeds which harvested at 10 days intervals from 20 days to 60 days after pollination were determined to clarify the optimum time for seed production of ginseng. The sizes of length, width and thickness of fruits, seeds and endosperms investigated on 20 days after pollination were not largely different from those that harvested after 60 days. But the weights of fruits and seeds were increased along with the delay of harvesting time. The seeds which harvested from 30 to 60 days showed comparatively high dehisced and germinated rates, especially 40 days in dehisced rates and 50 days in germinated rates. The embryos were rapidly grown in early stage. Lengths of the embryo on 20 days after pollination were approximately grown to 30 ${\mu}{\textrm}{m}$. And after 30 days pollinated, embryos were not only more rapidly grown to 213 ${\mu}{\textrm}{m}$ but also cotyledon premodia were developed in this time and completely formed after 40 days.
