Variation of Anthocyanin and Protein Contents in Glycine max L. (Merr) (Soybean) Germplasms from Korea

  • Soybean (Glycine max L. (Merr) is commonly consumed and found in major foods including soymilk, soy sauce, tofu, and soy sprout in Korea and east Asia. In addition, it is common to cook the whole seeds with rice. Soybean is known to have ranges of health benefits including antiaging, anticancer, neuroprotective and antidiabetic taken either as supplement or dietary food. Anthocyanins and flavonoids in G. max are found to be the main contributors to such wide arrays of health benefits. Due to increasing economic values of soybean, development of specialty soybean cultivars is becoming an area of interest worldwide. In this study, 746 black soybean accessions from National Agrobiodiversity Center were characterized as part of an attempt to identify important germplasms of G. max. Seed coats of each accession were analyzed for their total anthocyanin, cyanidin 3-O-Glucoside (C-3-O-G), delphinidin 3-O-glucoside (D-3-O-G), petunidin-3-O-glucoside (Pt-3-O-G), and their whole seeds for crude protein contents. HPLC was used to determine and quantify the anthocyanin compositions while crude protein was determined using Kjeldahl method by Kjeltec auto-analyzer (Kjeltec 8400, Foss, Sweden). Accessions were grouped according to their anthocyanins and protein contents; the mean content of which were correlated to agronomic traits including maturity date, one hundred seed weight, cotyledon color and seed lust color. The results indicated that the total anthocyanin content (TAC) ranged from 273.77 to 6250.52 mg/100 g, with mean value of 1853.03 mg/100 g while the crude protein content (CPC) being between 33.43 and 47.51%, with mean value of 40.81%. The highest number of accessions (45.97%) showed TAC between 1000~1900 mg/100 g while 30.96% of accessions showed CPC between 41~43%. Among the 746 accessions considered, 11 (IT142935, 175818, 175855, 177191, 177209, 177211, 177214, 177216, 177218, 177220, 177274) of them showed TAC above 4000 mg/100 g. C-3-O-G was found to be the major contributor to TAC showing strong correlation. Accessions with green cotyledon color showed high mean TAC compared to those having yellow cotyledon color, and accessions with dull seed lust color showed high mean TAC than those having shiny seed lust color. One hundred seeds weight and maturity date showed positive correlation with all anthocyanin contents, except for Pt-3-O-G in the latter case. The overall result of the present study could be used as background for developing new black soybean cultivars and breeds with high anthocyanin and protein contents. The result depicted that many of the accessions could be used as potential parental lines.

  • 유소직 세포들은 aldose인 D-glucose ketose인 D-fructose에 대하여 다른 능동 수송계를 소유하고 있었다. D-glucose와 D-fructose 수송계의 $K_{m}$ 값은 각각 0.28 mM과 15.02mM이었다. D-mannose는 $K_{m}$ 값이 0.44 mM로 D-glucose와 유사하였지만, 그러나 D-glucose와는 다른 수송계를 소유하고 있었다. L-glucose도 고유한 수송계를 통하여 세포내로 수송되었으며, 그러나 그 기능을 전혀 알지 못하고 있다. 유조직 원형질체는 단당류 능동 수송계만을 소유하고, 이당류인 sucrose능동 수송계는 소유하지 않고 있었다. 발달 초기단계에 있는 사부 원형질체는 glucose와 sucrose 수송계를 동시에 소유하고 있었다. 완전히 발달된 사부세포에서는 이미 존재하였던 glucose 능동 수송계는 사라지고, 새로 유도된 sucrose 능동 수송계만 존재하는 것으로 측정되었다.

Comparison of Agrobacterium-mediated of Five Alfalfa (Medicago sativa L.) Cultivars Using the GUS Reporter Gene

  • Alfalfa (Medicago sativa L.) is one of the most important forage legumes in the world. It has been demanded to establish the efficient transformation system in commercial varieties of alfalfa for forage molecular breeding and production of varieties possessing new characteristics. To approach this, genetic transformation techniques have been developed and modified. This work was performed to establish conditions for effective transformation of commercial alfalfa cultivars, Xinjiang Daye, ABT405, Vernal, Wintergreen and Alfagraze. GUS gene was used as a transgene and cotyledon and hypocotyl as a source of explants. Transformation efficiencies differed from 0 to 7.9% among alfalfa cultivars. Highest transformation efficiencies were observed in the cultivar Xinjiang Daye. The integration and expression of the transgenes in the transformed alfalfa plants was confirmed by polymerase chain reaction (PCR) and histochemical GUS assay. These data demonstrate highly efficient Agrobacterium transformation of diverse alfalfa cultivars Xinjiang Daye, which enables routine production of transgenic alfalfa plants.

배추 원형질체 배양에 미치는 다양한 처리의 효과와 원형질체 유래 캘러스로부터 신초 재분화 (Effects of a variety of treatments affecting Chinese cabbage protoplast culture, and plant regeneration from protoplast-derived callus)

  • 본 연구에서는 배추 원형질체 배양에 있어 다양한 처리의 효과를 구명함으로써 배추 원형질체 배양을 통한 재분화 체계를 확립하고자 하였다. 배추 원형질체 분리 수율은 효소액, 사용된 조직 및 효소액 처리 시간에 따라 달리 나타났는데, 효소액 처리 약 7시간 이후부터는 분리되는 원형질체의 수가 크게 증가하지 않았다. 따라서 효소 자체에 의한 부정적 영향을 고려하였을 때 최장 7시간 동안 효소처리를 하는 것이 적정한 것으로 판단하였다. 떡잎유래 원형질체를 2,4-D 1.0 mg/L, NAA 0.5 mg/L 및 BA 1.0 mg/L 첨가 액체배지에서 배양하였을 때 초기 세포분열과 이어지는 micro-callus 생장이 가장 양호하였다. 다양한 배추 인자형에 대한 검토 결과 본 연구의 배양조건 하에서는 초기 세포분열과micro-callus 생장이 배추 인자형에 따라 뚜렷하게 영향을 받는 것으로 나타났다. 노랑봄 품종의 떡잎에서 분리한 원형질체로부터 생장한 micro-callus 현탁액을 zeatin 0.2 mg/L외 spermidine 0.1 mM이 첨가된 반고체 MS 배지에 plating하였을 때 가장 왕성하게 callus colony가 형성 및 생장하였다. 이들 callus colony는 BA 5.0 mg 및 NAA 1.0 mg/L가 첨가된 배지에서 4.3%의 빈도로 재분화 신초가 유도되었다 얻어진 신초는 PGR 무첨가 MS배지에서 손쉽게 발근하였고 발근된 식군체는 기외에서 정상 식물체로 생장하였다. 본 연구의 결과는 배추 단독 원형질체 배양을 통한 고빈도 식물체 재분화 연구에 중요한 정보를 제공함으로써 유용하게 활용될 것으로 사료된다.

세포내 소기관과 물질대사의 관점에서 오이 떡잎의 발달 (Development of Cucumber Cotyledon in View of Metabolic Pathways and Organelle)

  • 오이 씨앗의 발아는 세포의 지방체내 저장지방의 분해 결과인 acyl-CoA의 글라이옥시좀으로 이동 후 베타 산화의 결과물인 acetyl-CoA의 글라이옥실산 회로로의 유입과 지방의 유동으로 촉발된다. Acetyl-CoA는 글라이옥실산 회로의 가동을 위한 탄소원을 제공하며 시트르산과 말산을 생성하며 글라이옥실산 회로의 작동을 유도한다. 지방 저장 종자의 발아에 있어서 글라이옥실산 회로는 필수적 요소이며, 그 결과물인 말산 및 숙신산의 미토콘드리아로의 이동은 TCA 회로의 가동과 옥살초산의 생성 및 세포질로의 유동으로 PEPCK에 의한 당신생을 가능하게 한다. 즉, 저장 지방을 원료로 여러 대사물질의 생산 및 이동과 다중의 대사경로를 통하여 발아 시 사용 가능한 에너지원인 포도당의 형태로 전환이 이루어진다. 이에 동반하여 많은 유전자의 발현 조절이 이루어지고, 세포내 소기관 특히 미소체로 대표되는 글라이옥시좀은 말산 합성효소(malate synthase)와 이소 시트르산 분해효소(isocitrate lyase)로 특화된다. 또 다른 acetyl-CoA의 유동은 carnitine을 매개로 하는 BOU (A BOUT DE SOUFFLE)의 작동이다. 이것은 카니틴의 대사와 관련하여 고등식물의 발달과 대사과정에서의 중요성이 확인된 것으로 사료된다.

  • Lipase specific activity in cocoa beans varied from 70 to 40 ${\mu}mol/min/mg$ protein during six days of fermentation. At the end of this period most parts of the cotyledon has turned to brown color which would be more distinguishable after drying. The beans were slightly swollen thus causing its testa to disintegrate. During fermentation there was a decrease in pH from 6.4 to 5.8. Whereas the percentage of acetic acid was increased by 0.04% of wet weight beans on the third day but decreased progressively with time.

발아중 빛에 의한 무 유식물의 자엽 Microbody의 활성 변화 (Effect of Light on Developmental Changes and Activities of Microbody in the Cotyledons of Radish Seedlings)

  • The enzyme patterns and the food storage changes in radish (Raphanus sativus L. cv. Taewang) cotyledons during seedling development were studied. The radish seeds were germinated for 8 days at $25^{\circ}C$ under light (7, 000 lux) or dark condition. The lipid and protein contents per seed were 4.3 mg and 2.85 mg respectively. In 8-day-old light-grown seedling, the lipid and protein contents per cotyledon pair were 1.5 mg and 2.08 mg; in 8-day-old dark-grown seedling, they were 0.8 mg and 1.24 mg respectively. The heterotrophic phase of seedlings continued for 3 days after sowing and followed by autotrophic phase (3~6 day) and senescence phase (6~8 day). The food storage function decreased in response to time course. During heterotrophic phase, the activities of glyoxysomal enzymes (malate synthetase, isocitrate lyase, and catalase) were high at 2~3 day. Those patterns were somewhat more prominent in darkness. During the autotrophic phase, the activities of peroxysomal enzymes (glycolate oxidase and catalase) increased at 4~5 day.

Changes of Chemical Composition during Seedling Development in Soybean

  • Seedling establishment of soybean [Glycine mar (L.) Mer-rill] is an important factor for soybean production in the field. The objective of this study was to determine the distribution of chemical composition in the emerging organs during seedling development in soybeans. Three soybean cultivars (Hill, Paldalkong, and Jangyeobkong) were planted at the Research Farm of College of Natural Resources, Korea University, on May 26, June 5, and June 14. Protein, oil, sugar, and starch contents were measured in each organ at each developing stage. Mean dry weight of three soybean cultivars decreased until VE stage and increased after this stage. Protein content of whole seedling did not change significantly during the seedling growth stage, but the amount in cotyledons markedly decreased with each growth stage increment. About 88% of the cotyledon protein was translocated to the other parts of the seedling at the V2 stage. Oil content of cotyledons sharply decreased until the V1 stage. Sugar content of the seedling was not detected at VE stage and starch content of seedlings increased slightly at VE and VC stages. For the changes of each metabolic component, the amount for whole plants decreased until the V1 stage and started to increase after this stage. The results of this study provide evidence for the breakdown of carbohydrates and oil at the initial stage of seedling growth.

Germination of Hemerocallis Seeds as Influenced by Seed Development and Temperature Treatments

  • Germination of Hemerocallis seeds as influenced by cold stratification at $5^{\circ}C$ (CS) and maturity of seeds evaluated using X-ray imaging has not been well investigated. Seeds of H. lilioasphodelus, H. citrina, H. citrina 'April Flower', and H. minor collected from China and H. thunbergii collected from Korea were germinated at $20^{\circ}C$ without pre-temperature treatment, while H. hongdoensis, H. dumortieri, H. minor, and H. vespertina seeds were treated with CS. Harvesting 'Stella de Oro' capsules at 35-40 days after anthesis yielded mature seeds with well-developed embryo and cotyledons analyzed by X-ray images with a 92% germination in 17 days after sowing. Seeds of H. thunbergii and H. citrina germinated in less than 13 days without CS; two weeks of CS did not accelerate seed germination. Seeds of H. hongdoensis germinated in 24 days when seeds were stored at $25^{\circ}C$ without CS and in less than 27 days when cold stratified. Therefore, 'Stella de Oro' capsules should be harvested at 35-40 days after anthesis to harvest mature seeds. Cold stratification is not required to accelerate seed germination in the Hemerocallis taxa evaluated in this study.

수도기계이앙 육묘에 관한 연구 -제5보 상자육묘시 배유양분의 소모가 묘생육 및 활착에 미치는 영향- (Rice Seedling Establishment for Machine Transplanting V. Effect on Endosperm Weight Change on the Seedling Growth and Regrowth After Transplanting)

  • 수도 기계이앙 육묘에서 종자내 배유양분의 소모가 묘생육에 미치는 영향을 검토코자 초형이 다른 통일형 품종을 공시, 육묘온도를 2수준으로 실시하여 다음과 같은 결과를 얻었다. 1. 묘령의 증가추이는 육묘초기는 주간 3$0^{\circ}C$/야간 1$0^{\circ}C$(고온구)에서 배유양분소모가 빨랐으며 엽수도 증가되었으나 후기에는 반대로 주간 $25^{\circ}C$/야간 1$0^{\circ}C$(저온구)에서 배유양분소모가 완만하였고 배유양분이 늦게까지 시속되고 엽수도 증가하였다. 2. 배유양분의 소진기는 엽기로 보면 고온구 2.5영, 저온구 3.0영이며. 육묘일수로는 22일, 27일에 완전히 소진되었다. 3. 대, 소입종간에는 대립종 품종이 육묘초부터 늦게까지 배유양분이 완만히 소모되었으나, 소립종은 배유양분의 소진기가 대입종보다 다소 빨리 왔다. 4. 이앙후 초기활착과 배유양분과의 관계는 배유양분이 9~10% 남아있는 치묘는 활간이 중. 성묘보다 2~3일 촉진되었으나 배유양분이 6% 남아있는 치묘에서는 촉진효과가 없었다. 5. 따라서 배유양분소모와 묘령 진전에 알맞은 경화기 온도는 $25^{\circ}C$/1$0^{\circ}C$이며. 출아장은 5mm, 치묘의 이앙적기는 배유양분이 9~10% 이상 남아있는 2.0~2.1 엽기이다.
